Abstract
本发明公开了一种用于手术过程中腔镜镜头冲洗的含碘清洗消毒液及制备方法,属于碘消毒液技术领域,其特征在于所述含碘清洗消毒液中包含按重量份数计的以下组分:单质碘0.1~5份,碘复合络合剂5~45份,稳定剂1~5份,表面活性剂1~10份,分散剂1~2.5份,pH调节剂1~5份,渗透压调节剂9份,保湿剂0.5~5份,所述含碘清洗消毒液的pH值范围为2.0~4.0;本发明的有益效果是:采用了羧甲基纤维素钠作为分散剂,羧甲基纤维素钠能够与碘络合剂聚乙烯吡咯烷酮协同起效,共同起到防雾效果,对于镜头防雾具有较为积极的效果;羧甲基壳聚糖对于碘络合剂在碘络合效果上具有增益作用。

技术领域
本发明属于碘消毒液技术领域,更具体地涉及用于手术过程中腔镜镜头冲洗的含碘清洗消毒液及制备方法。
背景技术
随着经济的发展,医疗水平的提升,人们对手术室安全追求越来越高,含碘清洗消毒液成为一款手术室中必备的消毒产品。
黄染即在碘伏使用过程中,与物体接触一定时间后,用清水难以除去的现象。目前,公众所知的是碘伏在使用的过程中会对皮肤、地面造成黄染,在手术室中,由于碘伏的黄染特性,此时再使用清水很难除去,辅以除碘剂,虽然可以解决黄染,但是成本会有所增加,操作亦较繁琐。
专利号为201510834421.9的国家发明专利公开了一种无痕型聚维酮碘消毒液,将碘与聚乙烯吡咯烷酮络合后辅以一定的螯合与包裹技术,使其在地板上残留24个小时内,用清水洗去,肉眼观测几乎无黄色痕迹。相对于普通聚维酮碘(碘伏)消毒液,在对地板黄染方面,有较大的优势。
该产品能对手术室的工作面进行清洗,但对于手术过程中腔镜镜头冲洗存在一定的技术瓶颈:该聚维酮碘消毒液将碘与聚乙烯吡咯烷酮络合后辅以一定螯合与包裹技术,当用在腔镜镜头上时,用清水洗去,肉眼观测可以呈现几乎无黄色痕迹,但是会产生较为严重的起雾现象。
发明内容
为解决上述问题,克服现有技术的不足,本发明提供了一种手术过程中腔镜镜头冲洗的含碘清洗消毒液及制备方法,能够有效解决手术过程中腔镜镜头冲洗时产生的黄染和起雾问题。
本发明解决上述技术问题的具体技术方案为:用于手术过程中腔镜镜头冲洗的含碘清洗消毒液,其特征在于所述含碘清洗消毒液中包含按重量份数计的以下组分:单质碘0.1~5份,碘复合络合剂5~45份,稳定剂1~5份,表面活性剂1~10份,分散剂1~2.5份,pH调节剂1~5份,渗透压调节剂9份,保湿剂0.5~5份。其中,所述含碘清洗消毒液的pH值范围为2.0~4.0。
进一步地,所述的分散剂为羧甲基纤维素钠。
进一步地,所述碘复合络合剂为聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物。
进一步地,所述聚乙烯吡咯烷酮和羧甲基壳聚糖的质量比为20:1~10:1,所述碘复合络合剂的制备方法包括:将聚乙烯吡咯烷酮和羧甲基壳聚糖按一定的质量比加入到纯化水中,在80℃条件下搅拌至溶解,随后在搅拌状态下自然冷却至室温即可得到聚乙烯吡咯烷酮和羧甲基壳聚糖的混合溶液,即碘复合络合剂溶液。
进一步地,所述的稳定剂为碘化钾或碘酸钾的其中一种。
进一步地,所述的表面活性剂为脂肪醇聚氧乙烯醚、脂肪醇聚氧乙烯醚硫酸钠、烷基糖苷、吐温-80、聚氧代乙基壬基苯基醚以及烷基芳香基聚氧乙烯醚中的一种或多种。
进一步地,所述的渗透压调节剂为氯化钠。
进一步地,所述的保湿剂为甘油、丁二醇、丙二醇以及D-泛醇中的一种或多种。
一种用于手术过程中腔镜镜头冲洗的含碘清洗消毒液的制备方法,其特征在于,根据含碘清洗消毒液的组分配置而成,包括以下步骤:
S1、在常温、常压条件下,将单质碘和稳定剂加入碘络合剂中进行络合反应,搅拌反应2.5~4小时,得到溶液B;
S2、将渗透压调节剂、表面活性剂、分散剂以及保湿剂加入上述溶液B中,搅拌溶解1~2小时,制成溶液C备用;
S3、将S2中所得溶液C加入pH调节剂,调节溶液pH为2.0~4.0,制得含碘清洗消毒液。
本发明的有益效果是:
(1)本发明采用了羧甲基纤维素钠作为分散剂,在执行分散剂效果的同时,意外地发现羧甲基纤维素钠能够与碘络合剂聚乙烯吡咯烷酮协同起效,共同起到防雾效果,对于镜头防雾具有较为积极的效果;
(2)本发明采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘复合络合剂;对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭效果上具有更为积极的效果,说明了羧甲基壳聚糖对于碘络合剂在碘络合效果上具有增益作用,
与此同时,从防雾效果上看,采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘络合剂相对于聚乙烯吡咯烷酮作为碘络合剂来说,具有更为清晰的视野,羧甲基壳聚糖与聚乙烯吡咯烷酮在防雾效果上的协同起效,共同起到防雾效果。

制备方法:
S1、将聚乙烯吡咯烷酮加入纯化水中,在80℃条件下搅拌溶解2小时,随后在搅拌状态下自然冷却至室温,即可得到络合剂溶液;
S2、在常温、常压条件下,将单质碘和碘化钾加入上述络合剂溶液中进行络合反应,搅拌反应2.5~4小时,得到溶液B;

本发明的含碘清洗消毒液用于冲洗腔镜镜头,不仅能够做到防雾、快速去污以及保证镜头清洁及视野清晰,还具有消毒杀菌效果,避免重复冲洗在穿刺过程中造成的感染,
其中,消毒杀菌效果具体表现为能够杀灭金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌以及白色念珠菌,具体杀灭试验可按照如下方法进行;
1、金黄色葡萄球菌杀灭试验
本发明含碘清洗消毒液原液用含5g/LNa2S2O3、30g/L吐温-80的PBS作中和剂,对金黄色葡萄球菌作用1min,可有效中和悬液中残留的消毒剂,且中和剂及中和产物对试验菌和培养基无不良影响;
本发明含碘清洗消毒液原液(有效碘含量:100mg/L)对金黄色葡萄球菌作用1min,杀灭对数值>5.00;
2、大肠杆菌杀灭试验
本发明含碘清洗消毒液原液(有效碘含量:100mg/L)对大肠杆菌作用1min,杀灭对数值>5.00;
3、铜绿假单胞菌杀灭试验
本发明含碘清洗消毒液原液(有效碘含量:100mg/L)对铜绿假单胞菌作用1min,杀灭对数值>5.00;
4、白色念珠菌杀灭试验
本发明含碘清洗消毒液原液用含5g/LNa2S2O3、30g/L吐温-80的PBS作中和剂,对白色念珠菌作用1min,可有效中和悬液中残留的消毒剂,且中和剂及中和产物对试验菌和培养基无不良影响;

由表1数据分析可知:
(1)实施例1和对比例1相比:
在消毒杀菌效果方面,对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌杀灭差异性不大,但是从防雾效果上看,虽然经过清洗以后,不具有黄染,但是腔镜镜头起雾,视野存在严重的模糊问题,详见附图1和附图3;
这可能是由于:聚乙烯吡咯烷酮在防雾效果上并没有实质性的贡献,而羧甲基纤维素钠能够对于镜头防雾具有较为积极的效果;
(2)实施例1和对比例2相比:
由于聚乙烯吡咯烷酮在体系中起到碘络合剂的作用,因此,不加聚乙烯吡咯烷酮将会使得该体系无法建立,杀菌效果及防雾效果无从验证;
(3)实施例1和对比例3相比:
在消毒杀菌效果方面,对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌杀灭差异性不大,但是从防雾效果上看,虽然经过清洗以后,对比例3不具有黄染,但是腔镜镜头起雾,视野存在严重的模糊问题,且与对比例1的模糊程度无显著性差异,详见附图1和附图4;
这可能是:对比例3中的十二烷基苯磺酸钠虽然与羧甲基纤维素钠都属于分散剂,但是,十二烷基苯磺酸钠并不能与碘络合剂聚乙烯吡咯烷酮协同起效,共同起到防雾效果,而单独的聚乙烯吡咯烷酮在防雾效果上并没有实质性的贡献。
为了更加直观的展现本发明的碘复合络合剂对于防雾效果和杀菌效果工艺优势,特以本发明含碘清洗消毒液和相同工艺采用等效替换的方法进行对比,

其中,消毒杀菌效果具体表现为能够杀灭金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌以及白色念珠菌,具体杀灭试验按照如下方法进行;
1、金黄色葡萄球菌杀灭试验
本发明含碘清洗消毒液原液用含5g/LNa2S2O3、30g/L吐温-80的PBS作中和剂,对金黄色葡萄球菌作用1min,可有效中和悬液中残留的消毒剂,且中和剂及中和产物对试验菌和培养基无不良影响;
本发明含碘清洗消毒液原液(有效碘含量:100mg/L)对金黄色葡萄球菌作用1min,杀灭对数值>5.00;
2、大肠杆菌杀灭试验
本发明含碘清洗消毒液原液(有效碘含量:100mg/L)对大肠杆菌作用1min,杀灭对数值>5.00;

(1)实施例1和实施例4对比可知:
作为实施例1的优选实施方案,将碘络合剂创造性采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘复合络合剂;
其中,从对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭效果上看,实施例4优于实施例1,说明了羧甲基壳聚糖对于碘络合剂在碘络合效果上具有增益作用,
与此同时,从防雾效果上看,采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘络合剂相对于聚乙烯吡咯烷酮作为碘络合剂来说,具有更为清晰的视野,详见附图1和附图2;
这可能是,羧甲基壳聚糖与聚乙烯吡咯烷酮在防雾效果上的协同起效,共同起到防雾效果;而单独加入聚乙烯吡咯烷酮并不存在该协同起效的防雾效果,
(2)实施例4和对比例4对比可知:
由于单纯的羧甲基壳聚糖对碘的络合效果较差,因此,等量的羧甲基壳聚糖并不能络合所述量的碘,因此该体系无法构建,杀菌效果和防雾效果也无从验证;
(3)实施例4和对比例5、对比例1对比可知:
从对比例5对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭效果上看,均具有一定的杀菌效果,环糊精和羧甲基壳聚糖均属于碘络合剂,均能够对碘产生络合效果,从而产生杀菌效果,但是效果不如实施例4,详见附图2、附图3和附图5;
由此来看,羧甲基壳聚糖相对于同为络合剂的环糊精来说,羧甲基壳聚糖与聚乙烯吡咯烷酮在对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭效果上具有协同增益效果;
尤为重要的是:环糊精和羧甲基壳聚糖均属于络合剂,但是环糊精在防雾方面并没有显著性效果,对比例5在防雾效果上明显低于本发明的实施例4,两者的区别仅在于碘复合络合剂中的分散剂羧甲基壳聚糖替换成其他常用分散剂环糊精,因此,羧甲基壳聚糖不仅仅在体系中作为分散剂,羧甲基壳聚糖与聚乙烯吡咯烷酮在防雾效果上协同起效,共同起到防雾作用;
(4)实施例4与对比例6对比可知:
对比例6在消毒杀菌效果方面,对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭率有一定程度的降低,这说明了碘化钾作为稳定剂,能够在聚乙烯吡咯烷酮与碘进行络合时,起到很好的稳定效果,从而促进消毒杀菌效果;这个与对比例3的结果是一致的,详见附图2和附图6;
所不同的是:从防雾效果上看,对比例6与实施例4相比,视野存在模糊的问题,模糊程度远低于对比例5,说明了碘化钾对于防雾效果上存在一定的积极效果,但是效果远低于羧甲基壳聚糖与聚乙烯吡咯烷酮在防雾效果。
综上所述:
(1)本发明采用了羧甲基纤维素钠作为分散剂,在执行分散剂效果的同时,意外地发现羧甲基纤维素钠能够与碘络合剂聚乙烯吡咯烷酮协同起效,共同起到防雾效果,对于镜头防雾具有较为积极的效果;
(2)本发明采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘复合络合剂;对金黄色葡萄球菌、大肠杆菌、铜绿假单胞菌、白色念珠菌的杀灭效果上具有更为积极的效果,说明了羧甲基壳聚糖对于碘络合剂在碘络合效果上具有增益作用,
与此同时,从防雾效果上看,采用了聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物作为碘络合剂相对于聚乙烯吡咯烷酮作为碘络合剂来说,具有更为清晰的视野,羧甲基壳聚糖与聚乙烯吡咯烷酮在防雾效果上的协同起效,共同起到防雾效果。
Claims (10)
1.一种用于手术过程中腔镜镜头冲洗的含碘清洗消毒液,其特征在于所述含碘清洗消毒液中包含按重量份数计的以下组分:单质碘0.1~5份,碘络合剂5~45份,稳定剂1~5份,表面活性剂1~10份,分散剂1~2.5份,pH调节剂1~5份,渗透压调节剂9份,保湿剂0.5~5份,所述含碘清洗消毒液的pH值范围为2.0~4.0。
2.根据权利要求1所述的含碘清洗消毒液,其特征在于所述的分散剂为羧甲基纤维素钠。
3.根据权利要求1所述的含碘清洗消毒液,其特征在于所述碘络合剂为为聚乙烯吡咯烷酮。
4.根据权利要求1所述的含碘清洗消毒液,其特征在于所述碘络合剂为复合络合剂,具体为聚乙烯吡咯烷酮和羧甲基壳聚糖的混合物。
5.根据权利要求4所述的含碘清洗消毒液,其特征在于所述聚乙烯吡咯烷酮和羧甲基壳聚糖的质量比为20:1~10:1,所述碘复合络合剂的制备方法包括:将聚乙烯吡咯烷酮和羧甲基壳聚糖按所述的质量比加入到纯化水中,在80℃条件下搅拌至溶解,随后在搅拌状态下自然冷却至室温即可得到聚乙烯吡咯烷酮和羧甲基壳聚糖的混合溶液,即碘复合络合剂溶液。
6.根据权利要求1所述的含碘清洗消毒液,其特征在于所述的稳定剂为碘化钾或碘酸钾的其中一种。
7.根据权利要求1所述的含碘清洗消毒液,其特征在于所述的表面活性剂为脂肪醇聚氧乙烯醚、脂肪醇聚氧乙烯醚硫酸钠、烷基糖苷、吐温-80、聚氧代乙基壬基苯基醚以及烷基芳香基聚氧乙烯醚中的一种或多种。
8.根据权利要求1所述的含碘清洗消毒液,其特征在于所述的渗透压调节剂为氯化钠。
9.根据权利要求1所述的含碘清洗消毒液,其特征在于所述的保湿剂为甘油、丁二醇、丙二醇以及D-泛醇中的一种或多种。
10.一种用于手术过程中腔镜镜头冲洗的含碘清洗消毒液的制备方法,其特征在于,根据权利要求1-9任意一项所述的含碘清洗消毒液的组分配置而成,包括以下步骤:
S1、在常温、常压条件下,将单质碘和稳定剂加入碘络合剂溶液中进行络合反应,搅拌反应2.5~4小时,得到溶液B;
S2、将渗透压调节剂、表面活性剂、分散剂以及保湿剂加入上述溶液B中,搅拌溶解1~2小时,制成溶液C备用;
S3、将S2中所得溶液C加入pH调节剂,调节溶液pH为2.0~4.0,制得含碘清洗消毒液。
