Abstract
The invention provides a hair-blacking essence composition, and a preparation method and application thereof, and belongs to the technical field of cosmetics. The hair-blacking essence composition comprises the following components in parts by weight: 6-14 parts of plant extract, 2-5 parts of humectant, 0.1-0.5 part of penetrating agent, 0.4-0.6 part of preservative and 70-90 parts of water. The hair blackening essence composition provided by the invention is also added with the synergistic components of beta-carotene and psoralen, the beta-carotene and the psoralen which are accidentally sent out and the plant extract can effectively penetrate into scalp and hair follicles through mutual renting, tyrosinase is activated, damaged melanocytes are repaired, the growth of melanocytes and the generation of melanin are promoted, regulation and repair are carried out on a melanin formation channel, the purpose of blackening hair is achieved, and the addition of the beta-carotene and the psoralen can improve the durability of blackening hair.

Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a hair-blacking essence composition, and a preparation method and application thereof.
Background
Hair is a part of human tissue and is an organism composed of cells. Hair cells divide from the follicle and, in multiples, new hair is pushed from the follicle to the scalp surface. One end of the hair is black, so that people are young, healthy and mental, and therefore, one end of the hair is expected to have a natural black hair.
Along with the development of society, the life rhythm is accelerated, the working and life pressures of people are also increased, the immunity is also reduced, and the hair is white and the hair is lost more and more. Excessive hair loss or whitening is closely related to hypofunction, heredity, mental stress, excessive brain use, malnutrition, etc.
The color of hair depends on the number, size, distribution of pigment particles in the hair shaft and the nature of the pigment contained. White hair is the hair that is fully or partially whitened, and white hair is produced due to reduced melanin particles in the medulla and cortex of the hair or air-filled, both congenital and acquired. The main physiological or pathological factors responsible for acquired grey hair include 4 general categories: (1) The accumulation of hydrogen peroxide causes methionine to oxidize, and as the age increases, the content of oxygen radicals in the hair increases, and when its concentration in the hair roots increases to a certain level, it causes a decrease in the activity of melanocyte tyrosinase in the hair, and the synthesis of melanin decreases, resulting in the formation of white hair. The prior hair-blackening composition has more researches, for example, china patent CN112220719B discloses a hair-blackening composition, a preparation method and application thereof, and the hair-blackening composition comprises the following components in parts by weight: 0.05-10 parts of sakura extract, 0.01-7 parts of glycyrrhetinic acid, 0.1-8 parts of ginseng extract and 0.01-5 parts of glossy privet fruit extract. The hair blackening composition can effectively penetrate into scalp and hair follicles through synergistic combination of four active ingredients, activate tyrosinase, repair damaged melanocytes, promote growth of melanocytes, mature transformation of functional melanocytes and generation of melanin, regulate and repair from a melanin formation path, and achieve the purposes of blackening hair, improving grey hair and white hair; meanwhile, the hair blackening composition can regulate and control the microecological system for scalp and hair growth and maintain the stable and healthy growth of scalp and hair, and the oxidation effect of scalp hair follicles of the composition is not ideal.
And as Chinese patent CN115998630A discloses a hair blackening essence and a preparation method thereof, the hair blackening essence can improve the white hair phenomenon of some adult men and has higher white hair to black hair efficiency. The hair blackening essence comprises a solvent and a main agent, wherein the main agent comprises, by mass, 10-20 parts of 2-hydroxy chalcone, 5-15 parts of acetyl tetrapeptide-3, 8-18 parts of imidazo [1,2-A ] pyridine-3-carboxylic acid, 6-16 parts of copper pyrrolidone carboxylate, 3-9 parts of hydrogen oxalate, 6-12 parts of hydrolyzed collagen, 1-7 parts of 2-deoxy-2-hexylglycerol, 2-8 parts of phenylacrylol, 3-13 parts of N-acetyl-D-glucosamine amino alcohol and 2-8 parts of 5-bromo-2' -deoxycytidine. The hair blackening essence can penetrate into hair follicles to clean, and each component can penetrate into hair follicles efficiently to nourish hair follicles and hair papilla, promote repair and development of melanocyte, strengthen connection of hair papilla and blood vessel, repair nutrition supply channel of hair papilla, promote synthesis and transfer efficiency of melanin, effectively repair adult male white hair, promote white hair to turn into black hair, and the composition can not activate the aminopeptidase better.
Therefore, there is a need to develop a hair-blacking essence composition which can effectively nourish hair follicles, and can effectively activate tyrosinase and promote the growth of melanocytes, and a preparation method and application thereof.
Disclosure of Invention
Based on the defects existing in the prior art, the invention aims to provide the hair blacking essence composition which has multiple plant components, high activity and small irritation, can effectively nourish hair follicles, activate tyrosinase and promote the growth of melanocytes, and a preparation method thereof.

The hair-blacking essence composition comprises the following components in parts by weight: 6-14 parts of plant extract, 2-5 parts of humectant, 0.1-0.5 part of penetrating agent, 0.4-0.6 part of preservative and 70-90 parts of water.

The humectant is one or more selected from glycerol, propylene glycol, 1, 3-butanediol, sodium hyaluronate, sodium lactate, hydrolyzed collagen, beta-glucan and panthenol;

As a preferred embodiment, the humectant comprises 3 parts by weight, specifically 2 parts of sodium hyaluronate, 0.8 part of sodium lactate and 0.2 part of beta-glucan.
In the implementation process, the invention discovers that the mixture of the sodium hyaluronate, the sodium lactate and the beta-glucan is used as the humectant, so that not only can the moisturizing effect of the hair-blacking essence composition be improved, but also the integrity of a fiber structure can be maintained, the living environment of skin cells can be improved, the metabolism of skin tissues can be promoted, and the effect of moisturizing the skin can be achieved.
The penetrating agent is a mixture of glycerol, tetrahydropiperine and inositol, and the mass ratio of the glycerol to the tetrahydropiperine to the inositol is 10:7:1.
As a preferred embodiment, the penetrant comprises 0.45 parts by weight, specifically 0.25 parts of glycerin, 0.175 parts of tetrahydropiperine and 0.025 parts of inositol.
In the implementation process, the invention discovers that the mixture of glycerol, tetrahydropiperine and inositol is adopted as the penetrating agent, so that each component in the hair blackening essence composition can efficiently penetrate into hair follicles, nourish the hair follicles and hair papilla, promote the repair and development of melanocytes, strengthen the connection between the hair papilla and blood vessels, repair the nutrition supply channel of the hair papilla, and promote the synthesis and transfer efficiency of melanin.

The hair-blacking essence composition also comprises 0.05-0.1 part of synergist, wherein the synergist is a mixture of beta-carotene and psoralen, and the mass ratio of the beta-carotene to the psoralen is 1:1.
As a preferred embodiment, the synergistic agent comprises 0.08 parts by weight, specifically 0.04 parts of beta-carotene and 0.04 parts of psoralen.
The invention discovers that beta-carotene, psoralen and plant extracts can effectively penetrate into scalp and hair follicles through mutual renting, activate tyrosinase, repair damaged melanocytes, promote melanocyte growth and melanin generation, regulate and repair from a melanin formation path, achieve the purpose of blackening hair, and the addition of the beta-carotene and the psoralen can improve the durability of blackening hair.
As a preferred embodiment, the hair-blacking essence composition comprises the following components in parts by weight: 5 parts of snakehead intestinal leaf extract, 0.5 part of mint leaf extract, 3 parts of polygonum multiflorum extract, 1.5 parts of Chinese gall extract, 2 parts of sodium hyaluronate, 0.8 part of sodium lactate, 0.2 part of beta-glucan, 0.25 part of glycerin, 0.175 part of tetrahydropiperine, 0.025 part of inositol, 0.5 part of p-hydroxyacetophenone, 0.04 part of beta-carotene, 0.04 part of psoralen and 86.15 parts of water.
The invention also provides a preparation method of the hair-blacking essence composition, which comprises the following steps:
Adding penetrant, humectant and water into a stirring pot, heating to 50-60 ℃ while stirring, cooling to below 40 ℃ after complete dissolution, adding the rest components, stirring uniformly, and standing to room temperature to obtain the hair blackening essence composition.
The invention also provides application of the hair-blacking essence composition in preparation of hair-blacking products.
The hair blackening product is hair blackening spray, hair nourishing lotion, hair blackening essence, shampoo, hair conditioner or hair dye.
Compared with the prior art, the invention has the beneficial effects that:
(1) The hair-blacking essence composition provided by the invention comprises snakehead intestine leaf extract, peppermint leaf extract, polygonum multiflorum extract and gallnut extract, wherein the plant extracts have the effect of blacking hair; in the implementation process, synergistic components of beta-carotene and psoralen are added into the hair blackening essence composition, the beta-carotene and the psoralen which are accidentally sent out and the plant extract can effectively penetrate into scalp and hair follicles through mutual renting, tyrosinase is activated, damaged melanocytes are repaired, the growth of melanocytes and the generation of melanin are promoted, regulation and repair are carried out from a melanin formation channel, the purpose of blackening hair is achieved, and the addition of the beta-carotene and the psoralen can improve the durability of blackening hair;
(2) The invention adopts the mixture of sodium hyaluronate, sodium lactate and beta-glucan as the humectant, which not only can improve the moisturizing effect of the hair-blacking essence composition, but also can maintain the integrity of fiber structure, improve the living environment of skin cells and promote the metabolism of skin tissues, thereby achieving the effect of moisturizing the skin.
(3) It is known in the art that hair blackening essence compositions accumulate on the surface of the scalp if the active ingredients thereof cannot be absorbed, which is liable to cause clogging of pores of the scalp, which brings a heavy burden to the scalp and even causes bacteria to grow; however, in the implementation process, the invention discovers that the mixture of glycerol, tetrahydropiperine and inositol is adopted as the penetrating agent, so that each component in the hair blackening essence composition can efficiently penetrate into hair follicles, nourish the hair follicles and hair papilla, promote the repair and development of melanocytes, strengthen the connection between the hair papilla and blood vessels, repair the nutrition supply channel of the hair papilla, and improve the synthesis and transfer efficiency of melanin.
(4) The hair-blacking essence composition provided by the invention has simple components, contains various natural plant components, has small irritation, can deeply nourish hair follicles, and does not generate dependence.

Example 5 Hair blacking essence composition and preparation method thereof
The coating comprises the following components in parts by weight: 5 parts of snakehead intestinal leaf extract, 0.5 part of mint leaf extract, 3 parts of polygonum multiflorum extract, 1.5 parts of Chinese gall extract, 2 parts of sodium hyaluronate, 0.8 part of sodium lactate, 0.2 part of beta-glucan, 0.25 part of glycerin, 0.175 part of tetrahydropiperine, 0.025 part of inositol, 0.5 part of p-hydroxyacetophenone, 0.08 part of beta-carotene and 86.15 parts of water.
The preparation method comprises the following steps:

Example 6 Hair blacking essence composition and preparation method thereof

Comparative example 1
The difference from example 1 is that: the snakehead intestinal leaf extract and the mint leaf extract are replaced with ginseng extract, namely 5.5 parts of ginseng extract, and other components and preparation methods are the same as in example 1.
Comparative example 2
The difference from example 1 is that: the mass ratio of the beta-carotene to the psoralen was 7:1, namely, 0.07 part of the beta-carotene, 0.01 part of the psoralen, and other components and preparation methods were the same as in example 1.
Comparative example 3
The difference from example 1 is that: the mass ratio of the beta-carotene to the psoralen was 1:7, namely, 0.01 part of the beta-carotene and 0.07 part of the psoralen, and other components and preparation methods are the same as in example 1.
Effect experiment
1. Irritation detection
The hair-blacking essence compositions provided in examples 1-6 were used for the skin irritation test of human body spot patches.
Dripping hair blackening essence composition (25 μl) onto filter paper sheets attached to the spot tester, and placing into the spot tester; applying the patch test with the hair blackening essence composition dropwise onto the curved side of forearm of a subject with a non-irritating adhesive tape, and applying to skin with palm light pressure for 24 hr; after removing the spot tester for 30min, the skin reaction is observed after the indentation disappears. If the result is negative, observing again after 24h and 48h respectively after the patch test; the skin reaction evaluation criteria for the closed patch test are shown in table 1.

60 Volunteers 20-50 years old with healthy skin were randomly selected, and were randomly divided into 6 groups, and scores of the closed patch test were recorded using the hair blackening essence compositions provided in examples 1-6, respectively, to obtain skin irritation test results of the human patch, and specific results are shown in Table 2 below.

According to the detection results of table 1, it can be seen that the hair blackening essence compositions provided by examples 1-6 of the invention show good safety in skin irritation experiments of human body spots, and the hair blackening essence composition provided by the invention is proved to be mild and non-irritating to human skin and can be safely used.
2. Tyrosinase activation rate
0.5Mg/mL of L-tyrosine solution (the solvent is phosphate buffer) is prepared, 35mg/mL of mushroom tyrosinase solution (the solvent is phosphate buffer) is prepared, and 1mg/mL of sample solution to be tested (the hair blackening essence compositions provided in examples 1-6 and comparative examples 1-3 are respectively tested) is prepared by using propylene glycol as the solvent to serve as a comparative test group.
The 4 groups of solutions were prepared according to the following proportions:
A:0.7mL of L-tyrosine solution+0.7 mL of pure propylene glycol+0.6 mL of phosphate buffer;
b:0.7mL of pure propylene glycol+1.3 mL of phosphate buffer;
c:0.7mL of L-tyrosine solution+0.7 mL of propylene glycol solution of the sample to be tested+0.6 mL of phosphate buffer;
D:0.7mL of propylene glycol solution of the sample to be tested plus 0.6mL of phosphate buffer;
After the above 4 groups of solutions were kept at a constant temperature in a water bath of 37℃for 10 minutes, 0.3mL of tyrosinase solution was added to each solution, and after 5 minutes of reaction, the solution was quickly transferred into a cuvette, absorbance (OD) was measured at 475nm, OD was recorded, and tyrosinase activation rate=100% × ((OD 3-OD4)-(OD1-OD2))/(OD1-OD2); where (OD 1-OD2) represents the change in OD value of the reaction solution in the absence of an activator and (OD 3-OD4) represents the change in OD value of the reaction solution in the presence of an activator (hair-blacking concentrate compositions provided in examples 1 to 6, comparative examples 1 to 3), and the detection data were shown in Table 3 below.

As can be seen from the detection results of the above Table 3, the activation rate of tyrosinase of the hair blackening essence compositions provided by the embodiments 1-3 of the present invention can reach 25% at the highest; in example 4, the activating rate of tyrosinase by the obtained hair-blacking essence composition is obviously reduced by 18.7 percent; the variety of the synergists in examples 5-6, i.e., the use of beta-carotene alone or psoralen alone, affects the activation rate of the hair blackening essence composition to tyrosinase to some extent; in comparative example 1, the plant extracts of snakehead intestinal leaf extract and peppermint leaf extract are replaced by ginseng extract, so that the activation rate of tyrosinase by the hair-blacking essence composition is obviously reduced; changing the mass ratio of the synergistic agent ingredients in comparative examples 2-3 also affects the activation rate of tyrosinase by the hair blackening essence composition to some extent, but the activation rate is significantly higher than in examples 4-6.
3. Melanin content test
The density of the B16 melanoma cell suspension was adjusted to 1X 10 5 cells/mL, and 1mL of the cell suspension per well was inoculated into a 12-well plate to give a cell density of 5X 10 4 cells/well, and the cell suspension was cultured in an incubator (37 ℃ C., 5% carbon dioxide) for 24 hours. The 12-well plate was taken out, the culture medium was removed, 1mL of the culture medium containing the sample to be tested (the hair-blacking essence composition provided in examples 1 to 6 and comparative examples 1 to 3, the concentration was 1 mg/mL), 1mL of the culture medium containing no sample to be tested was added to the blank control group and the zeroing group, and after the culture was continued in the incubator for 12 hours, the culture medium was removed. 1mL of alpha-MSH solution prepared by RPMI 1640 medium is added into each hole of the experimental group and the zeroing group, 1mL of medium without alpha-MSH is added into the blank group, and the blank group is placed into an incubator for culturing for 36 hours. The wells were washed twice with 1mL of PBS, 100. Mu.L of 1N NaOH solution (prepared with PBS) containing 10% DMSO was added to each well, after 1 hour of water bath in a water bath at 80℃the cell lysates of each group in 12 well plates were transferred to 96 well plates, and the absorbance OD of each well was measured at 475nm by an ELISA reader. The formula for calculating the intracellular melanin content is as follows: melanin content = 100% × (OD 1-OD3)/(OD2-OD3), where OD 1 is the absorbance of the experimental group, OD 2 is the absorbance of the blank group, OD 3 is the absorbance of the zeroed group, and the test data are shown in table 4 below.

From the test data of table 4 above, it can be seen that the hair blacking essence compositions provided in examples 1-3 of the present invention increased the melanin content in B16 melanoma cells to 8.15% at maximum; however, in example 4, the hair-blacking essence composition obtained does not contain a synergistic agent, so that the melanin content in B16 melanoma cells is not improved as in examples 1-3, but is only 7.61%; examples 5-6, in which the variety of the potentiating agent is varied, i.e., the use of β -carotene alone or psoralen alone, affects melanin production to some extent; in comparative example 1, the plant extracts of snakehead intestinal leaf extract and peppermint leaf extract were replaced with ginseng extract, and changing the mass ratio of the synergistic agent components in comparative examples 2-3 all affected the production of melanin, so that the content of melanin in B16 melanoma cells was not improved as in examples 1-3, but was superior to examples 4-6.
Finally, it should be noted that the above description is only for illustrating the technical solution of the present invention, and not for limiting the scope of the present invention, and that the simple modification and equivalent substitution of the technical solution of the present invention can be made by those skilled in the art without departing from the spirit and scope of the technical solution of the present invention.
Claims (14)
1. A hair blacking essence composition, characterized in that: the coating comprises the following components in parts by weight: 6-14 parts of plant extract, 2-5 parts of humectant, 0.1-0.5 part of penetrating agent, 0.4-0.6 part of preservative and 70-90 parts of water;
The plant extract is selected from one or more of snakehead intestinal leaf extract, peppermint leaf extract, black sesame extract, polygonum multiflorum extract and gallnut extract.
2. The hair blacking concentrate composition of claim 1 wherein: the plant extract is a mixture of snakehead intestinal leaf extract, peppermint leaf extract, polygonum multiflorum extract and gallnut extract; the mass ratio is 4-6:0.4-0.6:1-3:1-3.
3. The hair blacking concentrate composition of claim 1 wherein: the plant extract is a mixture of snakehead intestinal leaf extract, peppermint leaf extract, polygonum multiflorum extract and gallnut extract; the mass ratio is 5:0.5:3:1.5.
4. The hair blacking concentrate composition of claim 1 wherein: the humectant is one or more selected from glycerol, propylene glycol, 1, 3-butanediol, sodium hyaluronate, sodium lactate, hydrolyzed collagen, beta-dextran and panthenol.
5. The hair blacking concentrate composition of claim 4 wherein: the humectant is a mixture of sodium hyaluronate, sodium lactate and beta-glucan; the mass ratio is 8-12:3-5:1.
6. The hair blacking concentrate composition of claim 5 wherein: the mass ratio of the sodium hyaluronate to the sodium lactate to the beta-glucan is 10:4:1.
7. The hair blacking concentrate composition of claim 1 wherein: the penetrating agent is a mixture of glycerol, tetrahydropiperine and inositol, and the mass ratio of the glycerol to the tetrahydropiperine to the inositol is 10:7:1.
8. The hair blacking concentrate composition of claim 1 wherein: the hair-blacking essence composition also comprises 0.05-0.1 part of synergist.
9. The hair blacking concentrate composition of claim 8 wherein: the synergist is selected from the group consisting of beta-carotene and psoralen.
10. The hair blacking concentrate composition of claim 9 wherein: the mass ratio of the beta-carotene to the psoralen is 1:1.
11. The hair blacking concentrate composition of any one of claims 1-10, wherein: the coating comprises the following components in parts by weight: 5 parts of snakehead intestinal leaf extract, 0.5 part of mint leaf extract, 3 parts of polygonum multiflorum extract, 1.5 parts of Chinese gall extract, 2 parts of sodium hyaluronate, 0.8 part of sodium lactate, 0.2 part of beta-glucan, 0.25 part of glycerin, 0.175 part of tetrahydropiperine, 0.025 part of inositol, 0.5 part of p-hydroxyacetophenone, 0.04 part of beta-carotene, 0.04 part of psoralen and 86.15 parts of water.
12. A process for preparing a hair blacking concentrate composition according to any one of claims 1 to 10, characterized in that: the method comprises the following steps:
Adding penetrant, humectant and water into a stirring pot, heating to 50-60 ℃ while stirring, cooling to below 40 ℃ after complete dissolution, adding the rest components, stirring uniformly, and standing to room temperature to obtain the hair blackening essence composition.
13. Use of the hair blacking concentrate composition according to any one of claims 1 to 10 for the preparation of hair blacking products.
14. The use according to claim 13, characterized in that: the hair blackening product is hair blackening spray, hair nourishing lotion, hair blackening essence, shampoo, hair conditioner or hair dye.
