Abstract
本发明涉及Spexin活性多肽在制备抑制心房颤动药物中的应用,属于防治心房颤动药物技术领域。本发明提供了Spexin活性多肽的药物新用途,即Spexin活性多肽在制备防治心房颤动药物中的应用。Spexin活性多肽的氨基酸序列为NWTPQAMLYLKGAQ。动物实验证实Spexin活性多肽能够降低房颤诱发率、降低房颤发作次数、缩短房颤持续时间、延长心房有效不应期,降低静息膜电位负值,延长动作电位复极时间,进而降低房颤易感性。本发明将Spexin活性多肽应用于防治心房颤动的药物制备,开发其新的临床用途,具有生物学多效性、安全性高、毒副作用小等特点,有望成为房颤治疗的新型有效手段,具有重要转化意义。

技术领域
本发明属于防治心房颤动药物技术领域,尤其涉及Spexin活性多肽在制备抑制心房颤动药物中的应用。
背景技术
心房颤动(atrial fibrillation,AF),简称房颤,是临床最常见心律失常之一,发病率随着年龄增长而增加,80岁以上老年人中发生率为10~12%。房颤能够导致动脉栓塞和心力衰竭等并发症,具有较高的致残率和致死率,是严重威胁人类健康的重大疾病。房颤发生的病理生理学机制复杂,主要包括离子通道改变、纤维化、氧化应激、炎症及能量代谢异常等。目前房颤治疗仍存在诸多问题,抗心律失常药物疗效欠佳且容易诱发其他心律失常,射频消融术后复发率仍较高且花费大。房颤是一种自身进展性疾病,主要是由于房颤使心房发生重构,而心房重构又促进房颤持续,形成恶性循环。因此深入探析房颤心房重构机制,寻找新的有效防治靶点是目前该领域的主要研究方向。
Spexin活性多肽是一种新型内源性生物活性肽类激素,已有研究证实Spexin活性多肽在机体痛觉、食欲、排便、糖脂代谢、水盐平衡及抗炎等生理功能调节中发挥关键作用。但Spexin活性多肽是否能够用于防治心房颤动尚未见报道。

本发明提供了Spexin活性多肽的药物新用途,即Spexin活性多肽在制备防治心房颤动药物中的应用。

本发明的有益效果:
本发明提供了一种Spexin活性多肽防治心房颤动的药物新用途,动物实验结果证实Spexin活性多肽能够抑制房颤的发生,降低房颤诱发率、降低房颤发作次数、缩短房颤持续时间、延长AERP,降低静息膜电位(Resting membrane potential,RMP)负值,延长动作电位复极90%时间(90%repolarization time ofaction potential,APD90),进而降低房颤易感性。
本发明将Spexin活性多肽应用于防治心房颤动的药物制备,开发其新的临床用途。由于外源性给予Spexin活性多肽治疗可降低房颤的发生。而Spexin活性多肽作为内源性存在的多肽,具有生物学多效性、安全性高、毒副作用小等特点,有望成为房颤治疗的新型有效手段,具有重要的转化意义。

下面结合实施例对本发明的技术方案做进一步的说明,但并不局限于此,凡是对本发明技术方案进行修改或者等同替换,而不脱离本发明技术方案的精神和范围,均应涵盖在本发明的保护范围中。下列实施例中未具体注明的工艺设备或装置均采用本领域内的常规设备或装置,若未特别指明,本发明实施例中所用的原料等均可市售获得;若未具体指明,本发明实施例中所用的技术手段均为本领域技术人员所熟知的常规手段。
实施例1
本实施例考察了Spexin活性多肽对心房颤动的抑制效果。
本发明使用的实验动物均为C57BL/6J小鼠(雄性,年龄为8-10周,体重为20±5g),购自辽宁长生动物技术有限公司。。
建模、分组、给药方法:
建立血管紧张素Ⅱ(Angiotensin II,AngⅡ)诱导的房颤小鼠模型,提前两天将Ang-Ⅱ的缓释泵放在培养皿内,加入无菌生理盐水,然后放进无菌恒温37℃孵箱,浸泡48小时备用。现配现用Ang-Ⅱ(25mg/ml),用1ml注射器向每一个缓释泵里面装100μl溶解好的Ang-Ⅱ。将小鼠麻醉后,从小鼠头部纵行或者横行剪开其上皮,然后用止血钳进行钝性分离,即用止血钳伸入,分离上皮和皮下组织,然后把缓释泵放进止血钳撑开而形成的缝隙,然后用手术针线进行缝合,术后饲养3周即为小鼠房颤模型。
将小鼠分为阴性对照组、房颤阳性对照组和Spexin活性多肽治疗组,分别标记为CTL组、Ang-Ⅱ组和Ang-Ⅱ+SPX组。
如图1所示,在Ang-Ⅱ的缓释泵植入皮下一周后,对Spexin活性多肽治疗组小鼠连续腹腔注射Spexin活性多肽,给药量为50μg/kg/d,连续给药两周。阴性对照组和房颤阳性对照组给药均为等量生理盐水;各组给药时间均为14天。建模3周后进行心脏电生理检测。
房颤的诱发方法:
用1.5%的异弗烷麻醉各组实验小鼠。一根八极电生理导管经颈静脉插入小鼠的右心房。使用河南华南医学科技有限公司的刺激仪与数据采集系统GY6000收集数据。将麻醉后小鼠仰卧位固定于鼠板上,剪开右侧颈部皮肤,使用显微镊分离右侧颈静脉,将小鼠电生理导管沿颈静脉逆行插入至右心房。将电极连接至电刺激仪器上,应用S1S1(40毫秒开始2毫秒递减至20毫秒)连续刺激,电压分别是3.5V,5V和8V刺激诱发房颤,记录诱发率及持续时间。
建模结束后对各组小鼠进行心脏电生理检测,图2为各组小鼠房颤诱发的代表图,图中AF表示心房颤动,SR表示正常窦性心律;图3为各组小鼠房颤诱发率对比图。如图2和图3所示,与CTL组相比,Ang-Ⅱ组房颤诱发率显著增加;与Ang-Ⅱ组相比,Ang-Ⅱ+SPX组房颤诱发率显著降低。
图4为各组小鼠房颤事件对比图;如图4所示,与CTL组相比,Ang-Ⅱ组房颤事件增加;与Ang-Ⅱ组相比,Ang-Ⅱ+SPX组房颤事件明显降低(n=11-13,*P<0.05vs.CTL,#P<0.05vs.Ang-II)。
图5为各组小鼠房颤持续时间对比图;如图5所示,与CTL组相比,Ang-Ⅱ组房颤持续时间明显延长;与Ang-Ⅱ组相比,Ang-Ⅱ+SPX组房颤持续时间明显缩短(*P<0.05vs.CTL,#P<0.05vs.Ang-II)。
图6为各组小鼠AERP对比图;如图6所示,与CTL组相比,Ang-Ⅱ组AERP明显缩短;与Ang-Ⅱ组相比,Ang-Ⅱ+SPX组AERP明显延长(n=11-13,*P<0.05vs.CTL,#P<0.05vs.Ang-Ⅱ)。
试验结果证实,Spexin活性多肽治疗能够明显抑制Ang-Ⅱ引起的房颤易感性增加。
实施例2
本实施例采用离体实验考察了Spexin活性多肽对心房颤动的抑制效果。
在实施例1建模及治疗的基础上,进一步用Langendorff灌流小鼠心脏,用高频电刺激诱导灌流心脏房颤的发生,并用Optical mapping记录小鼠心房动作电位。
Langendorff离体心脏灌注模型制备方法及Optical mapping流程:
先配制心脏灌流液并充氧30分钟。清洗和润洗灌流系统。开启循环水浴锅,水浴温度为38℃。小鼠腹腔注射肝素钠注射液0.1ml(浓度为50mg/ml)。10分钟后,颈部脱臼处死小鼠,打开小鼠胸腔立即取出心脏,置于盛有灌流液的玻璃皿中,去除多余血液和腺体组织,利用体视显微镜将心脏主动脉挂于灌流针上,将灌流针与Langendorff灌流装置连接,注意此过程不得有气泡进入导管和接头处。将心脏置于恒温水浴槽中,待灌流5分钟稳定后向灌流管内加入解偶联剂-[-]Blebbistatin(浓度为20μmol/L)。10分钟后,待心脏停止搏动,向灌流柱中加入电压敏感染料RH-23715μl(母液浓度1.25mg/mL)再灌流10分钟后进行检测,观察心脏染色均匀后,调整心脏位置后采集心脏图片信息。利用电刺激仪给予心脏5Hz大小的刺激。应用MiCAM05 CMOS系统采集心脏图片(分辨率100×100像素,每秒1000帧)。应用BV-ana软件分析小鼠心脏传导速度和动作电位时程。
对于离体灌流心脏房颤的诱发,待检测完心脏传导速度和动作电位后,利用电刺激仪以S1S1连续起搏给予小鼠心脏60毫秒,50毫秒,40毫秒,30毫秒,20毫秒的刺激5秒,并用MiCAM05 CMOS系统采集图片看是否有房颤发生。
图7为Optical mapping检测各组小鼠心脏房颤诱发情况的代表图、图8为各组小鼠离体灌流心脏检测房颤诱发率对比图;如图7和图8所示,与CTL组相比,Ang-Ⅱ组房颤诱发率明显增加;与Ang-Ⅱ组相比,Ang-Ⅱ+SPX组房颤诱发率明显降低。
此外,进一步检测各组小鼠离体心脏的动作电位,图9为Optical mapping检测各组小鼠离体灌流心脏动作电位代表照片;图10为各组小鼠离体灌流心脏动作电位代表图;图11和图12分别为各组小鼠离体灌流心脏APD50和APD80的结果对比图。如图11和图12结果显示,Spexin活性多肽治疗可以抑制Ang-Ⅱ引起的小鼠心房APD80缩短,但对APD50无延长作用(n=9,*P<0.05vs.CTL,#P<0.05vs.Ang-II)。以上电刺激结果显示,Ang-Ⅱ增加离体灌流心脏的房颤易感性,而Spexin活性多肽治疗能够抑制Ang-II引起的房颤易感性增加。
实施例3
本实施例采用膜片钳检测考察了Spexin活性多肽对小鼠心房肌细胞动作电位时程的影响。
在实施例1各组小鼠建模及治疗的基础上,我们进一步分离小鼠心房肌细胞利用全细胞膜片钳记录心房肌细胞RMP和动作电位时程(Action potential duration,APD)。

利用灌流法急性分离实验小鼠心房肌细胞和新生大鼠心房肌细胞记录动作电位和IK1电流。实验开始前打开膜片钳系统和电极拉制仪预热仪器。使用P-97拉制仪拉制玻璃电极,调节拉制仪温度使拉制的电极入液阻值在2-4MΩ之间。全细胞膜片钳实验在37℃恒温下进行。实验开始前将灌注有电极内液的电极安装在夹持器上,并在浴槽中加入动作电位外液,外液中滴加适量细胞。利用放大器进行细胞电流和动作电位记录,信号经1kHz过滤并且通过A/D转换得到数据(Digi-data 1440,Axon Instrument)。在电压钳模式下记录电流和电流钳模式下记录电压。实验结果为组间比较,电流以膜电容归一化处理,电流密度以pA/pF表示。
图13为各组小鼠心房肌细胞的动作电位对比图;图14为各组小鼠心房肌细胞的RMP对比图;如图14所示,与CTL组相比,Ang-Ⅱ增加小鼠心房肌细胞静息膜电位负值,而Spexin活性多肽治疗能够使小鼠心房肌细胞静息膜电位负值减小(n=11-18,*P<0.05vs.CTL,#P<0.05vs.Ang-Ⅱ)。
图15和图16分别为各组小鼠心房肌细胞的APD50和APD90的对比图;如图15和16所示,Ang-Ⅱ组小鼠心房肌细胞APD50和APD90明显缩短,Spexin活性多肽治疗对APD50(图15,n=11-18,*P<0.05vs.CTL)无影响,但延长了APD90(图16,n=11-18,*P<0.05vs.CTL,#P<0.05vs.Ang-Ⅱ)。由此证实Spexin活性多肽能够抑制Ang-Ⅱ诱导的小鼠心房肌细胞APD缩短。
以上结果证实,Spexin活性多肽治疗能够降低小鼠的房颤易感性。

1.Spexin活性多肽在制备防治心房颤动药物中的应用。
2.根据权利要求1所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述Spexin活性多肽的氨基酸序列为NWTPQAMLYLKGAQ。
3.根据权利要求1或2所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动药物以Spexin活性多肽为唯一活性成分或活性成分之一。
4.根据权利要求3所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动的药物中Spexin活性多肽的含量为0.1wt%~99wt%。
5.根据权利要求4所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动的药物还包括药学上可接受的辅料和/或载体。
6.根据权利要求5所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动的药物用于防治哺乳动物的心房颤动。
7.根据权利要求6所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述哺乳动物为人或小鼠。
8.根据权利要求7所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动的药物为非经胃肠道给药剂型,具体为注射给药剂型。
9.根据权利要求8所述Spexin活性多肽在制备防治心房颤动药物中的应用,其特征在于,所述防治心房颤动的药物能够降低房颤诱发率、降低房颤发作次数、缩短房颤持续时间、延长心房有效不应期、降低心房肌细胞静息膜电位、延长动作电位复极时间,进而降低房颤易感性。
