背景技术

氰病毒素N(CVN)是一种培养蓝藻(Nostoc ellipsosporumwith)的组成部分,具有强大的抗HIV活性,是一种独特的环肽,有101个氨基酸,长11kDa。一级氨基酸结构与任何已知的蛋白质具有<20%的同源性,因此提供了一个独特的三维结构和序列。

CVN对所有免疫缺陷病毒(HIV-1、HIV-1的M-和T型毒株、HIV-2、SIV(猿类)、FIV(猫))以及其他包膜病毒:包括流感病毒、埃博拉病毒、马尔堡病毒、严重急性呼吸综合征冠状病毒、丙型肝炎病毒、疱疹病毒6和麻疹病毒,具有强活性。

CVN对各种理化降解(physicochemical degradation)均表现出高度的抗性,用变性剂、洗涤剂、有机溶剂、多次冻融循环和热处理对其抗病毒活性没有影响。因此,CVN及其功能类似物可能被证明是一种很好的候选药物,用于各种治疗和预防用途。
CVN及其一些突变体已经通过重组产生,这需要先进的分子生物学和昂贵的发酵设施。目前,即使经过全面的文献研究,也还没有实用和直接的化学合成CVN的报道。
发明内容
本发明提供一种抗病毒肽、尤其是氰病毒素N的化学合成、其类似物以及应用。
本申请第一个方面提供一种肽,其含有、或组成为SEQ ID No.2-SEQ ID No.21的任意一个序列或更多个序列(即大于一个序列)。
本申请第二个方面是提供一种所述肽的制药学上可接受的盐。优选地,所述盐可以是羧酸盐或无机酸盐,如醋酸盐、盐酸盐、硝酸盐、硫酸盐、柠檬酸盐、琥珀酸盐等。

一种合成所述肽的方法,包括:C端为羧酸的第一个肽片段与第二个肽片段通过树脂上形成二硫键连接构成肽,第一片段和第二片段均固相形成二硫键。

实施例1:两片段合成方法(two-fragment process),通过树脂上形成二硫键(on-resin disulfide bond formation)
第一个片段,含有45个氨基酸残基(固相形成二硫键):LGKFSQTCYNSAIQGSVLTSTCERTNGGYN TSSIDLNSVI ENVDG-acid
第二个片段,含有56个氨基酸残基(固相形成二硫键):SLKWQPSNFIETCRNTQLAGSSELA AECKTRAQQF VSTKINLDDHIANIDGTLKYE

实施例4:三片段缩合制备方法
第一个片段含有27个氨基酸残基:Fmoc-LGKFSQTCYN SAIQGSVLTS TCERTNG-acid;
第二个片段含有38个氨基酸残基:Fmoc-GYN TSSIDLNSVIENVDGSLKWQ PSNFIETCRN TQLAG;
第三个片段含有36个氨基酸残基:SSELA AECKTRAQQF VSTKINLDDHIANIDGTLKYE-amide;
合成的肽为:Ac-LGKFSQTCYN SAIQGSVLTS TCERTNGGYNTSSIDLNSVI ENVDGSLKWQGSNFIETCRN TQLAGSSELA AECKTRAQQFVSTKINLDDH IANIDGTLKYE-amide
通过两步缩合获得完整长度的线性肽,所述线形肽然后在氧化还原反应条件下折叠为自然构型。
本申请其他肽SEQ ID No.6-21可以通过上述三种方式中的一种来获得。
上述实施例的最终产物通过反相HPLC纯化,并通过分析和质谱进一步表征。从反相HPLC中纯化的多肽通常是三氟乙酸(TFA)的盐的形式。
这种盐通常被转化为更具有药物友好性的盐形式,如醋酸或盐酸盐形式。将TFA盐中的肽转化为盐酸盐可以通过在稀盐酸溶液中在TFA盐中反复冻干来实现。对于将TFA盐中的肽转化为醋酸盐,最简单的方法是使用如下方法制备的阴离子交换树脂:
强阴离子交换树脂(氯离子形式,取代量3mmole/g,含水量50%,每克肽使用2克树脂)首先用milli Q水洗涤三次,然后用1N氢氧化钠溶液洗涤3次,5分钟/次;然后用milli Q水洗涤5次,5分钟/次。
用75%乙醇水溶液清洗强阴离子交换树脂,直到pH值达到7.4。这种树脂用10%的醋酸溶液处理3次,每次5分钟。然后用1%的醋酸溶液清洗树脂3次,每次5分钟。该树脂可以进行纯化的肽的盐转化。
将纯化的冻干肽溶解在1%醋酸溶液中并添加到上述制备的强阴离子交换树脂中。将混合物在室温下搅拌或磁性搅拌1小时。上清液分离。该强阴离子交换树脂用1%的醋酸溶液洗涤三次。将上清液和洗涤液结合,通过0.22μm的膜过滤并冻干,在醋酸盐中生成一种肽。
本申请通过上述方式可以获得具有突变的肽,从而获得具有抗病毒活性的肽,可用于替代CVN。
以上对发明的具体实施例进行了详细描述,但其只是作为范例,本发明并不限制于以上描述的具体实施例。对于本领域技术人员而言,任何对本发明进行的等同修改和替代也都在本发明的范畴之中。因此,在不脱离本发明的精神和范围下所作的均等变换和修改,都应涵盖在本发明内。

1.一种肽,其特征在于,所述肽含有、或组成为SEQ ID No.2-SEQ ID No.21的任意一个序列或更多个序列。
2.一种权利要求1所述肽的制药学上可接受的盐。
3.根据权利要求3所述的盐,其特征在于,所述盐选自羧酸盐或无机酸盐。
4.根据权利要求4所述的盐,其特征在于,所述盐选自醋酸盐、盐酸盐、硝酸盐、硫酸盐、柠檬酸盐、琥珀酸盐。
5.一种药物组合物,其特征在于,所述药物组合物含有权利要求1所述的肽、或权利要求2所述肽的制药学上可接受的盐。
6.根据权利要求5所述的药物组合物,其特征在于,所述药物组合物,还含有一种或更多种制药学上可接受的辅料或稀释剂。
7.根据权利要求6所述的药物组合物,其特征在于,所述药物组合物的剂型是:口服、经鼻、含片、舌下、肺、体表、阴道、直肠、眼部、经皮或肠外给药中的任意一种或更多种。
8.一种权利要求5所述药物组合物在准备治疗病毒感染药物中的应用。
9.一种合成权利要求1所述的肽的方法,其特征在于,选自:
C端为羧酸的第一个肽片段与第二个肽片段通过树脂上形成二硫键连接构成肽,第一片段和第二片段均固相形成二硫键;
C端为羧酸的第一个肽片段与第二个肽片段通过树脂上形成二硫键连接构成带有P51G突变的肽,第一片段和第二片段均固相形成二硫键;
C端为硫酯的第一个肽片段与C端为半胱氨酸的第二个肽片段通过自然化学连接形成肽,然后肽进行折叠,优选为自然氧化还原条件下进行折叠;
N端为Fmoc的三个肽片段通过两步缩合获得肽,第一个肽片段C端为羧酸,第三个片段C端为氨基,缩合后,所述肽进行折叠,,优选为自然氧化还原条件下进行折叠。
