CVN的一级结构包含101个残基,分为两个结构域A(残基1-50)和B(残基51-101),两个结构域具有高度的内部序列和结构相似性。我们发现,在晶体学实验条件下(低pH和26%异丙醇),两个对称相关的单体通过结构域交换形成二聚体,这样一个单体的结构域A与其晶体学对称伴侣的结构域B0相互作用,反之亦然。由于两个交换的域彼此距离较远,域交换不会导致额外的分子内相互作用。尽管其中一种用于结晶的蛋白质样品溶液明显含有100%的CVN分子,但根据各种方法判断,我们只能获得含有结构域交换二聚体的晶体。

CVN对各种理化降解(physicochemical degradation)均表现出高度的抗性,用变性剂、洗涤剂、有机溶剂、多次冻融循环和热处理对其抗病毒活性没有影响。因此,CVN及其功能类似物可能被证明是一种很好的候选药物,用于各种治疗和预防用途。
在赖氨酸残基上的无规PEG化(PEGylation)导致了抗病毒活性的不良保留。因此,需要确定定点突变体被用来测试位点特异性的PEG化。CVN其中一个突变体中,谷氨酰胺62被半胱氨酸取代(CVN(Q62C))取代,并被马来酰亚胺激活的PEG化,在体外保留了显著的抗HIV活性(H.Zappe,et al.,Advanced Drug Delivery Reviews,60(2008):79–87)。因此,CVN的位点特异性、选择性修饰和衍生化能提供更多样化的分子,为治疗目的和需要提供不同的化学/物理/生物/药理特性。
发明内容
本发明提供了一种位点特异性修饰和标记的抗病毒肽氰病毒素N及其类似物,以及含有它的组合物,以及使用和获得他们的方法,特别是在临床和诊断中的应用,如在抗病毒治疗和预防中的应用。
本申请第一个方面是提供一种抗病毒肽氰病毒素N类似物I,所述类似物I为在氰病毒素N的Gln14、Asn26、Ser38、Ser52、Gln62、Ser66、Asp89作为突变位点进行修饰,其中,修饰基团来自含有氨基酸的叠氮化物和炔烃之间点击化学反应产生的共价产物基团,
本申请第二个方面是提供一种抗病毒肽氰病毒素N类似物II,所述类似物II为在氰病毒素N的Gln14、Asn26、Ser38、Ser52、Gln62、Ser66、Asp89作为突变位点进行修饰,其中,修饰基团来自含有氨基酸的叠氮化物。

本申请还提供了一种合成所述抗病毒肽氰病毒素N类似物I的方法,包括将含有氨基酸的叠氮化合物链接到抗病毒肽氰病毒素N的肽链Gln14、Asn26、Ser38、Ser52、Gln62、Ser66、Asp89中的至少一个位点上。
其中,含有氨基酸的叠氮化合物可以是通过酰基叠氮法等现有技术链接抗病毒肽氰病毒素N的肽链的Gln14、Asn26、Ser38、Ser52、Gln62、Ser66、Asp89位点上。
本申请还提供了一种合成所述抗病毒肽氰病毒素N类似物II的方法,通过点击化学反应,将炔烃化合物链接到抗病毒肽氰病毒素N类似物I上(即叠氮化合物的基团上)。
本申请选择了Gln14X、Asn26X、Ser38X、Ser52X、Gln62X、Ser66X、Asp89X等作为突变位点,可用于测试位点特异性的PEG化等。

本发明提供一种抗病毒肽氰病毒素N的位点特异性修饰和标记方法,为使本发明的目的、技术方案及效果更加清楚、明确,以下参照附图并举实例对本发明进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。

C端酰胺,如肽酰胺,通常使用Rink Amide树脂制备;C端酸,如肽酸,使用三丁基氯树脂或Wang树脂制备。尤其是,组装的肽链仍然附着在树脂载体上时,可以在固相上形成二硫键;也可以通过本领域已知的各种方法在溶液中形成二硫键。具有完全或部分保护侧链的肽片段的溶解缩合/连接,在适当的偶联剂和催化剂存在下,在与完全或部分保护侧链的肽片段的溶液中进行。那些用于天然化学连接的肽硫酯片段是根据已知的技术制备(如:Dey B,Lerner DL,Lusso P,Boyd MR,Elder JH,Berger EA.2000.J.Virol.74:4562–4569。Lagenaur LA,Sanders-Beer BE,Brichacek B,Pal R,Liu X,Liu Y,Yu R,Venzon D,LeePP,Hamer DH.2011.Mucosal Immunol.4:648–657)。

实施例4:三片段缩合制备方法
第一个片段含有27个氨基酸残基:Fmoc-LGKFSQTCYN SAIQGSVLTS TCERTNG-acid;
第二个片段含有38个氨基酸残基:Fmoc-GYN TSSIDLNSVI ENVDGSLKWQ PSNFIETCRN TQLAG;
第三个片段含有36个氨基酸残基:SSELA AECKTRAQQF VSTKINLDDH IANIDGTLKYE-amide;
合成的肽为:Ac-LGKFSQTCYN SAIQGSVLTS TCERTNGGYN TSSIDLNSVI ENVDGSLKWQGSNFIETCRN TQLAGSSELA AECKTRAQQF VSTKINLDDH IANIDGTLKYE-amide
通过两步缩合获得完整长度的线性肽,所述线形肽然后在氧化还原反应条件下折叠为自然构型。
在合成上面这7个片段来组装整个CVN蛋白分子的过程中,分别进行了以下位点特异性突变体(如氨基酸残基置换):Gln14X、Asn26X、Ser38X、Ser52X、Gln62X、Ser66X、Asp89X(数字代表位点)。

相应的氨基酸构成单元的例子包括:
等。而且Fmoc-和Boc-保护的氨基酸构成单元是可以市场上买到的。
最终产物通过反相HPLC纯化,并通过分析和质谱进一步表征。从反相HPLC中纯化的多肽通常是三氟乙酸(TFA)的形式。这种盐通常被转化为更具有药物友好性的盐形式,如醋酸或盐酸盐形式。将TFA盐中的肽转化为盐酸盐可以通过在稀盐酸溶液中在TFA盐中反复冻干来实现。对于将TFA盐中的肽转化为醋酸盐,最简单的方法是使用如下方法制备的阴离子交换树脂:
强阴离子交换树脂(氯离子形式,取代量3mmole/g,含水量50%,每克肽使用2克树脂)首先用milli Q水洗涤三次,然后用1N氢氧化钠溶液洗涤3次,5分钟/次,然后用milli Q水洗涤5次,5分钟/次。用75%乙醇水清洗树脂,直到pH达到7.4。这种树脂用10%的醋酸溶液处理3次,每次5分钟。然后用1%的醋酸溶液清洗树脂3次,每次5分钟。该树脂可以用于纯化肽的盐转化。
将纯化的冻干肽溶解在1%醋酸溶液中,并添加到上述制备的树脂中。将混合物在室温下搅拌或磁性搅拌1小时。上清液分离。该树脂用1%的醋酸溶液洗涤三次。将上清液和洗涤液结合,通过0.22μm的膜过滤并冻干,在醋酸盐中生成一种肽。
那些具有位点特异性叠氮基基团的合成CVN类似物可以在温和的点击化学条件下进一步选择性地与达因、PEGs(PEG:聚乙二醇)、聚氨基酸、蛋白质、肽或带有炔官能团的放射性试剂衍生,如下商业上可获得的含炔的试剂,包括荧光染料,可以在温和的水介质条件下通过点击化学偶联到含有叠氮化物基团的肽化合物上。
以上对本发明的具体实施例进行了详细描述,但其只是作为范例,本发明并不限制于以上描述的具体实施例。对于本领域技术人员而言,任何对本发明进行的等同修改和替代也都在本发明的范畴之中。因此,在不脱离本发明的精神和范围下所作的均等变换和修改,都应涵盖在本发明的范围内。

1.一种抗病毒肽氰病毒素N类似物,其特征在于,所述类似物为在氰病毒素N的Gln14、Asn26、Ser38、Ser52、Gln62、Ser66、Asp89作为突变位点进行修饰,其中,修饰基团来自含有氨基酸的叠氮化物。
