Abstract
本发明公开了一种聚乙二醇修饰的IL‑21衍生物及其应用。通过对IL‑21的结构进行分析,在不影响IL‑21整体结构的情况下,将IL‑21中特定的位点突变成半胱氨酸,再通过巯基偶联将合适的PEG分子连接到IL‑21上突变后的半胱氨酸上。PEG修饰后的IL‑21血浆半衰期延长,具有优异的剂量依赖的抗肿瘤药效。此外,PEG修饰的IL‑21具有更好的水溶性,有利于药物的制剂配方。相较于其他长效化的IL‑21融合蛋白需要在哺乳动物细胞中表达,本发明中PEG修饰运用的IL‑21骨架蛋白是在大肠杆菌中表达并通过变复性纯化得到的,具有低成本,高产率的产业化优势。

背景技术
白细胞介素-21(Interleukin-21,IL-21)属于IL-2家族的成员,是一个多功能性的细胞因子,对淋巴细胞和骨髓细胞具有多种生物学效应,在自然免疫和获得性免疫中都发挥重要的作用。IL-21主要由活化的CD4+T细胞、NK细胞、Tfh细胞、Th17细胞分泌,能提高免疫细胞的抗原特异性反应。IL-21对T细胞的作用是多效应的,包括促进细胞增殖和增强细胞毒性。IL-21可以诱导CD8+T细胞和NK细胞增殖和成熟并增强其细胞毒性,从而增强机体免疫反应。IL-21通过调节B细胞的增殖,分化成浆细胞来进行免疫应答1。IL-21还可以促进记忆CD8+T细胞分化和抑制Treg细胞2。
人源的IL-21含有134个氨基酸,结构上形成了四个α螺旋束。IL-21中有四个半胱氨酸残基(C42,C49,C93和C96),其中第42位和93位的半胱氨酸形成二硫键,第49位和96位的半胱氨酸形成二硫键3。
IL-21受体是一个由IL-21独有的亚单位IL-21R和γ链组成的异二聚体,其中IL-21R为配体识别结合部位,γ链为信号转导单位。IL-21R在T细胞、B细胞、NK细胞、树突状细胞和单核/巨噬细胞的表面广泛表达。IL-21与IL-21R具有很高的亲和力,高达70pM。IL-21与其异二聚体受体结合后,主要通过蛋白酪氨酸激酶JAK/信号转导和转录激动子SATA通路进行信号传导,活化JAK(JAK1和JAK3),随后磷酸化STAT1、STAT3、STAT4和STAT5,最后进入细胞核内调节相应基因的表达4。γ受体为IL-2,IL-4,IL-7,IL-9,IL-15和IL-21的通用受体。虽然IL-21与γ受体的亲和力较低(160μM),但是IL-21与γ受体的相互作用却是信号转导所必须的5。
自上世纪八十年代以来,利用白细胞介素类蛋白,特别是IL-2治疗癌症被广泛应用于临床,利用高剂量的IL-2治疗黑色素瘤和肾细胞癌的效果良好6,7。但是高剂量的IL-2会引起较强的副作用,比如血管渗漏综合征(VLS),导致人体器官的内积水,如肺水肿和肝细胞损伤。近年来的研究发现低剂量的IL-2能诱导调节性T细胞Treg,抑制免疫反应,促进肿瘤逃逸8。因此低剂量的IL-2不能被用于治疗肿瘤,IL-2在临床上的应用前景被剂量和毒副问题大大限制。IL-21作为一个新兴的抗肿瘤效应的细胞因子因其明显低于IL-2的毒性越来越引起人们的关注。同时低剂量的IL-21并不会诱导调节性T细胞Treg,仍然保持对免疫细胞的激活作用。重组人IL-21在黑色素瘤和肾细胞癌的临床实验中同样展现出了良好的抗肿瘤效果9。在结肠癌和乳腺癌小鼠模型中,低剂量的IL-21便能诱导出良好的抗肿瘤效应10,11。虽然IL-21对机体的毒副作用较低,但临床实验表明人IL-21的最大耐受剂量是30μg/kg。随着IL-21用药量的增加,毒副作用便开始出现。IL-21最常见的不良反应包括头痛、疲劳、发热、恶心、肌痛/关节痛、皮疹、腹泻、血小板减少等12,13。
IL-21较短的半衰期和较差的稳定性也大大限制了IL-21的作为抗肿瘤药物的开发8。野生型IL-21的血浆半衰期较短,在小鼠中仅0.2小时,在食蟹猴中是0.4-0.8小时,在人体内约为2小时10。因此,设计一种可靠的长效且稳定的IL-21对抗肿瘤药物的开发具有重要意义。

发明内容
本发明针对现有技术的现状及存在的不足,通过对IL-21的结构进行分析,在不影响IL-21整体结构的情况下,将IL-21中特定的位点突变成半胱氨酸,再通过巯基偶联将合适的PEG分子连接到IL-21上突变后的半胱氨酸上。PEG修饰后的IL-21血浆半衰期延长,具有优异的剂量依赖的抗肿瘤药效。此外,PEG修饰的IL-21具有更好的水溶性,有利于药物的制剂配方。相较于其他长效化的IL-21融合蛋白需要在哺乳动物细胞中表达,本发明中PEG修饰运用的IL-21骨架蛋白是在大肠杆菌中表达并通过变复性纯化得到的,具有低成本,高产率的产业化优势。
为实现上述目的,本发明采用以下技术方案施行:
在第一方面,本发明提供一种IL-21突变体,所述IL-21突变体选自如下任一种:
(a)其氨基酸序列由SEQ ID NO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N25,K52,K56,N59,G61,N82,S98;
或,
(b)所述IL-21突变体与(a)所述的氨基酸序列具有95%,优选98%,更优选99%的序列相同性,并且具有(a)所述蛋白的功能,其中对应于SEQ ID NO:1所示氨基酸序列的N25,K52,K56,N59,G61,N82,S98位氨基残基与(a)所述的氨基酸序列中的相同;
或,
(c)所述IL-21突变体由在(a)所述的氨基酸序列的C末端和/或N末端添加或缺失1-30个,更优选1-10个,还要优选1-6个,最优选1-3个氨基酸残基而形成,并且具有(a)所述IL-21突变体的功能,其中对应于SEQ ID NO:1所示氨基酸序列的N25,K52,K56,N59,G61,N82,S98位与(a)所述的氨基酸序列中的相同。
在优选的实施方式中,所述IL-21突变体(a)其氨基酸序列由SEQ ID NO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N25,K56,N59,G61,N82,S98。

在第四方面,本发明提供一种宿主细胞,所述宿主细胞包含本发明第三方面所述的表达载体或者其基因组中整合有本发明第二方面所述的核苷酸序列。
在优选的实施方式中,所述宿主细胞是细菌;更优选地,所述宿主细胞是大肠杆菌(E.coli)、谷氨酸棒杆菌(Corynebacterium glutamicum);最优选地,所述宿主细胞是大肠杆菌(E.coli)。
在第五方面,本发明提供一种第一方面所述的IL-21突变体的聚乙二醇衍生物,所述聚乙二醇修饰剂连接于IL-21突变体突变位点的半胱氨酸上。
在优选的实施方式中,所述的聚乙二醇修饰剂为巯基聚乙二醇修饰剂,是具有选自乙烯砜、马来酰亚胺或碘乙酰胺的官能团的多元醇衍生物。
在优选的实施方式中,所述的聚乙二醇修饰剂的分子量为20kDa。
在第六方面,本发明提供一种制备第五方面所述IL-21突变体的聚乙二醇衍生物的方法,包括如下步骤:用硫醇反应性多元醇剂和第一方面所述的IL-21突变体反应,所述IL-21突变体具有单一游离的半胱氨酸残基,所述多元醇试剂和所述IL-21突变体的单一游离的半胱氨酸残基特异性共价结合,形成共价硫醚键,得到多元醇-IL-21突变体偶联物,回收纯化产生的目的多元醇-IL-21突变体偶联物,即得。
在优选的实施方式中,其中所述IL-21突变体的聚乙二醇衍生物的聚乙二醇部分具有10~40kDa之间的分子量。
进一步优选地,所述IL-21突变体的聚乙二醇衍生物的聚乙二醇部分具有20kDa的分子量。在本发明中,计算IL-21突变体聚乙二醇衍生物的浓度和质量时并未将聚乙二醇的分子量包含在内,仅计算了IL-21突变体蛋白的分子量。
在第七方面,本发明提供一种药物组合物,所述药物组合物包含第一方面所述的IL-21突变体,或第五方面所述IL-21突变体的聚乙二醇衍生物,以及药学上可接受的载体或赋形剂。
在第八方面,本发明提供第一方面所述的IL-21突变体、或第五方面所述IL-21突变体的聚乙二醇衍生物,或第七方面所述的药物组合物在制备药物中的应用,所述的药物是调节或激活免疫的药物或抗肿瘤药物。
在第九方面,本发明提供第一方面所述的IL-21突变体、或第五方面所述IL-21突变体的聚乙二醇衍生物,或第七方面所述的药物组合物在制备促进B细胞分化增殖,促进T细胞分化增殖,促进NK细胞分化增殖的制剂中的应用。
在第十方面,本发明提供第一方面所述的IL-21突变体、或第五方面所述IL-21突变体的聚乙二醇衍生物,或第七方面所述的药物组合物在制备预防和/或治疗肿瘤药物中的用途。
本文中所有的氨基酸序列编号均以天然IL-21(SEQ ID NO:1)序列为准,第一个氨基酸编号为1,依次类推。由于本发明中所有蛋白均使用大肠杆菌表达,所以本发明中所有蛋白均在-1位含有起始氨基酸甲硫氨酸,其不影响IL-21活性,其中仅在天然IL-21的-1位添加起始氨基酸甲硫氨酸的IL-21在本文中定义为野生型IL-21(wt-IL-21)。
本发明的有益效果:
1、本发明通过对IL-21的结构进行分析,成功找到了七个位点并突变成半胱氨酸。这七个位点远离IL-21与IL-21R的相互作用界面,突变后并不会显著改变IL-21的结构,也不会影响IL-21与IL-21R的相互作用。进一步通过化学偶联在点突变引入的半胱氨酸上进行PEG修饰。PEG修饰后的IL-21突变体随着分子量的增大降低了肾小球透过率,从而延长了血浆半衰期。另一方面,PEG分子会阻碍蛋白酶靠近IL-21突变体,防止IL-21突变体被蛋白酶水解从而延长IL-21的血浆半衰期。
2、PEG修饰的IL-21突变体具有更好的水溶性,有利于药物的制剂配方。
3、相较于其他长效化的IL-21融合蛋白需要在哺乳动物细胞中表达,本发明中PEG修饰运用的IL-21突变体骨架蛋白是在大肠杆菌中表达并通过变复性纯化得到的,具有低成本,高产率的产业化优势。
4、PEG修饰后的IL-21突变体分子药效温和,保持了低毒性的特征,在高达18mg/kg的给药剂量下仍然没有表现出明显的毒副作用,具有很高的用药安全性。
5、小鼠肿瘤模型的药效实验表明PEG修饰后的长效化IL-21突变体具有优异的剂量依赖的抗肿瘤药效,相比之下野生型IL-21并未明显抑制小鼠体内肿瘤生长。

除非另有说明,以下实施例中使用的原料和试剂均为市售商品,或者可以通过已知方法制备。下列实施例中未注明具体条件的实验方法,通常按照常规条件如Sambrook等人,分子克隆:实验室手册(New York:Cold Spring Harbor Laboratory Press,1989)中所述的条件,或按照制造厂商所建议的条件。
定义与说明:
除非另外定义或由背景清楚指示,否则在本公开中的全部技术与科学术语具有如本公开所属领域的普通技术人员通常理解的相同含义。
“白介素-21”或“IL-21”指来自任何脊椎动物来源,包括哺乳动物如灵长类(例如人)和啮齿动物(例如小鼠和大鼠)的任何天然的IL-21。该术语涵盖未加工的IL-21以及源自细胞中的加工的任何形式的IL-21。该术语还涵盖天然存在的IL-21变体,例如剪接变体或等位变体。示例性天然人IL-21的氨基酸序列如SEQ ID NO:1所示。
如本文所用,术语“IL-21突变体”、“IL-21衍生物”、“IL-21衍生物蛋白”、“改造后的IL-21分子”可互换使用,指在本发明第一方面中所述的一种IL-21突变体,与天然IL-21SEQ ID NO:1相比,所述IL-21突变体在SEQ ID NO:1所示的氨基酸序列的第25位、52位、56位、59位、61位、82位、98位的其中之一的氨基酸突变为半胱氨酸。
“衍生物”旨在被广义地解释,包括任意IL-21相关的产品。包括但不限于人和非人的IL-21同系物、片段或截短体、融合蛋白(如与信号肽融合或其他活性、非活性成份融合,活性成份例如抗体或其抗原结合片段)、修饰形式(如PEG化、糖基化、白蛋白缀合/融合、Fc缀和/融合、羟乙基化等)和保守修饰的蛋白等。
如本文中使用的,术语“氨基酸突变”意为涵盖氨基酸取代、缺失、插入和修饰。可以进行取代、缺失、插入和修饰的任意组合来实现最终构建体,只要最终构建体拥有期望的特性。优选的氨基酸突变是氨基酸取代,即将一个氨基酸用具有不同结构和/或化学特性的另一种氨基酸替换。优选的氨基酸取代包括一些亲水的带电或者不带有电荷的氨基酸,如Ser,Thr,Ala,Gly,Glu,Arg,His和Lys。可以使用本领域中公知的遗传或化学方法生成氨基酸突变。遗传方法可以包括定点诱变、PCR、基因合成等。
当提及来自野生型蛋白的变体时,提及氨基酸取代诸如“N82C”是指原始残基天冬酰胺(N)的位置数目(82),随后是取代的残基半胱氨酸(C)。
一般方法
通常,本发明的IL-21突变体的制备可由本文公开的程序和由公认的重组DNA技术实现,所述公认的重组DNA技术包括例如,聚合酶链式反应(PCR)、质粒DNA的制备、用限制性酶裂解DNA、寡核苷酸的制备、DNA的连接、mRNA的分离、将DNA引入适当的细胞、转化或转染宿主、宿主的培养。另外,可以使用离液剂和熟知的电泳、离心和色谱方法分离和纯化融合分子。关于这些方法的公开内容一般参见,Sam brook等,Molecular Cloning:ALaboratory Manual(第二版(1989);和Ausubel等,Current Protocols in MolecularBiology,John Wiley&Sons,New York(1989))。
编码本发明变体蛋白的基因包括限制性酶消化和连接作为用于产生编码所需融合物的DNA的基本步骤。DNA片段的末端可需要在连接之前修饰,且这可通过用填充突出端、核酸酶(例如,ExoIII)缺失片段的末端部分、定点诱变或通过PCR添加新的碱基对来实现。
可使用多连接子和衔接子来促进所选片段的连接。表达构建体通常以使用数轮限制性酶切、连接和大肠杆菌转化的阶段组装。适合于构建表达构建体的许多克隆载体是本领域已知的(lambda.ZAP,Agilent;pET,EMD Millipore)并且特定的选择对于本发明不是关键的。
克隆载体的选择将受被选择用于将表达构建体引入宿主细胞中的基因转移系统的影响。在每个阶段结束时,可通过限制性酶切、DNA测序、杂交和PCR分析来分析所得的构建体。
定点诱变通常用于通过本领域已知的方法将特定突变引入编码本发明的IL-21突变体的基因中。参见例如,美国专利申请公布2004/0171154;Storici等人,2001,NatureBiotechnology 19:773-776;Kren等人,1998,Nat.Med.4:285-290;和Calissano和Macino,1996,Fungal Genet.Newslett.43:15-16。可在本发明中使用任何定点诱变程序。存在许多可用于制备本发明的变体的商业试剂盒。
可以根据本发明使用各种启动子(转录起始调节区)。合适的启动子的选择取决于所提议的表达宿主。可以使用来自异源来源的启动子,只要它们在所选宿主中有功能。
IL-21突变体可在无信号肽序列的大肠杆菌(E.coli)中表达,且蛋白从包涵体中回收并重折叠成活性形式。
术语“载体”或“表达载体”与“表达构建体”同义并指用于导入与其可操作联合的特定基因并指导其在靶细胞中表达的DNA分子。该术语包括作为自主复制核酸结构的载体以及掺入到其所导入的宿主细胞的基因组中的载体。本发明的表达载体包含表达盒。表达盒允许转录大量稳定的mRNA。一旦表达载体在靶细胞内,就通过细胞转录和/或翻译体系生成基因编码的核糖核酸分子或蛋白质。在一个实施方案中,本发明的表达载体包含表达盒,其包含编码本发明的IL-21衍生物的多核苷酸序列。
本文中的术语“转化”具有本领域技术人员普遍理解的意思,即将外源性的DNA导入宿主的过程。所述转化的方法包括任何将核酸导入细胞的方法,这些方法包括但不限于电穿孔法、磷酸钙(CaPO4)沉淀法、氯化钙(CaCl2)沉淀法、微注射法、聚乙二醇(PEG)法、DEAE-葡聚糖法、阳离子脂质体法以及乙酸锂-DMSO法。
术语“宿主细胞”、“宿主细胞系”和“宿主细胞培养物”可交换使用并指已引入外源核酸的细胞,包括这类细胞的后代。宿主细胞包括“转化体”和“经转化的细胞”,其包括初始转化的细胞和自其衍生的后代(不考虑传代数)。后代在核酸内含物上可能与亲本细胞不完全相同,但可以含有突变。本文中包括具有与原始转化细胞中所筛选或选择的功能或生物学活性相同的功能或生物学活性的突变后代。
本文的宿主细胞的培养可以根据本领域的常规方法进行,包括但不限于孔板培养、摇瓶培养、批次培养、连续培养和分批补料培养等,并可以根据实际情况适当地调整各种培养条件如温度、时间和培养基的pH值等。
如本文中使用的,术语“IL-21受体”指IL-21受体是一个由IL-21独有的亚单位IL-21R和γ链组成的异二聚体,其中IL-21R为配体识别结合部位,γ链为信号转导单位。
“亲和力”指分子(例如受体)的单一结合位点与其结合配偶体(例如配体)之间全部非共价相互作用总和的强度。除非另外指示,如本文中使用的,“结合亲和力”指反映结合对的成员(例如受体和配体)之间1:1相互作用的内在结合亲和力。分子X对其配偶体Y的亲和力通常可以用解离常数(KD)来表述,其为解离与结合速率常数(分别为K解离和K结合)的比率。如此,相等的亲和力可能包含不同的速率常数,只要速率常数的比率保持相同。亲和力可以通过本领域知道的确立方法来测量,包括本文中描述的那些方法。
可以依照实施例中提出的方法通过表面等离子共振(SPR),使用标准仪器如Biacore仪(GE Healthcare)和受体亚基(如可以通过重组表达获得的)来测定突变的或野生型IL-21对IL-21受体的各种形式的亲和力。或者,可以评估IL-21突变体对IL-21受体的不同形式的亲和力,其使用已知表达一种或另一种这类受体形式的细胞系。用于测量结合亲和力的特定的说明性和例示性实施方案在下文中描述。
药剂的“有效量”指引起接受其施用的细胞或组织的生理学变化所必需的量。药剂例如药物组合物的“治疗有效量”指在必需的剂量和时间段上有效实现期望的治疗或预防结果的量。治疗有效量的药剂例如消除、降低、延迟、最小化或预防疾病的不良效果。
术语“药物组合物”指其形式使得其中含有的活性成分的生物学活性有效,且不含对会接受该组合物施用的受试者有不可接受的毒性的别的成分的制剂。
“药学可接受载体”指药物组合物中活性成分以外对受试者无毒的成分。药学可接受载体包括但不限于缓冲剂、赋形剂、稳定剂或防腐剂。
本文所用的术语“包含”、“具有”、“包括”或“含有”是指包括在内的或开放式的,并不排除额外的、未引述的元件或方法步骤。
本文所用的“约”表示:一个值包括测定该值所使用的装置或方法的误差的标准偏差。
本文所用的“或”的定义仅为替代物以及“和/或”,但除非明确表示仅为替代物或替代物之间相互排斥外,权利要求中的术语“或”是指“和/或”。本公开中,“IL-21”可以与“IL21”互换使用。
实施例1:IL-21突变位点的选择
通过巯基偶联将PEG偶联到蛋白质上是一种常用的蛋白质长效化手段。天然IL-21中有4个半胱氨酸,且恰好形成了两对二硫键,所以要想通过巯基偶联的方法对IL-21进行PEG修饰,便需要将IL-21中其他的氨基酸突变成单独的半胱氨酸。同时,半胱氨酸巯基偶连相对于其它偶连方式,如氨基偶连等,具备方便易操作和特异性强等优点。
本发明对天然IL-21的结构进行了分析和计算,寻找不影响IL-21功能的可突变成半光氨酸的氨基酸。首先为了不影响IL-21的整体结构,突变位点排除了IL-21中参与形成二级结构(如α螺旋)的氨基酸。同时为了不影响IL-21与IL-21R的结合,突变位点需要远离IL-21与IL-21R相互作用界面中的关键氨基酸(如Q12)。此外,本发明还充分考虑到突变位点可能会影响IL-21中原有的二硫键的形成,所以也尽量远离原有的二硫键(原则上大于)。
综上所述,最终该专利设计了七个(N25,K52,K56,N59,G61,N82,S98)可突变氨基酸,并分别突变成半胱氨酸,这些突变位点与Q12和相近二硫键的距离信息如下表1所示。

实施例2.IL-21及其突变体质粒构建、蛋白表达与纯化
1.质粒构建
在本发明中,对天然IL-21进行突变,将第25位的N,第52位的K,第56位的K,第59位的N,第61位的G,第82位的N和第98位的S分别突变为C,得到七个不同的突变体,分别命名为1327(N25C),1328(K52C),1329(K56C),1330(N59C),1331(G61C),1332(N82C)和1333(S98C)。
委托北京擎科生物科技有限公司合成wt-IL-21以及IL-21突变体(1327,1328,1329,1330,1331,1332,1333)的基因。按照《分子克隆》中的操作方法,PCR得到目的片段。然后将目的片段与通用载体pET41a重组连接、转化、测序和保菌,从而得到能够表达相应蛋白质粒。按照《Qiagen Mini-prep Kit》中的操作方法,对wt-IL-21和IL-21的突变体进行质粒制备。
天然IL-21和IL-21突变体的氨基酸序列如表2。

2.蛋白的表达
将上述构建成功的质粒转化至大肠杆菌表达菌株BL21(DE3)中,从平板上挑取单克隆菌落按1:50的体积比转接到含Kana抗性的500mL TB培养基的三角瓶中,初始OD600大约为0.1,37℃,220rpm,培养至OD600为2.0,加入终浓度为0.5mM的IPTG,37℃、220rpm培养4h后收菌。使用SDS-PAGE检测蛋白表达情况,如图1所示,所有蛋白均以包涵体(InclusionBody)的形式成功表达,且表达量较高。
3.包涵体破碎洗涤
本发明中所有的野生型和突变体的质粒转化至大肠杆菌宿主工程菌中进行表达,表达产生不可溶性包涵体。菌体经高压破碎,洗涤后,回收包涵体,为后期蛋白复性做准备。
4.蛋白纯化
4.1蛋白复性
上述wt-IL-21和IL-21突变体的包涵体,使用20倍(v/w)体积的变性液(6M盐酸胍(Guanidine Hydrochloride),50mM Tris,pH 9.0)在室温下溶解30分钟后加入DTT,继续还原30分钟。将变性液滴入30倍体积的复性液中(20mM Tris,0.5M精氨酸(Rrginine),2.5mM半胱氨酸(Cysteine),pH 8.0),在室温下搅拌过夜,第二天检测复性效果。
4.2蛋白纯化
以上复性好的蛋白用醋酸钠(Sodium Acetate)调整pH至5.3,电导用去离子水稀释至20ms/cm以下,离心后上清直接流经GE公司的SP BB柱进行捕获。
操作如下:纯化前用5倍柱体积的平衡液(20mM醋酸钠,pH 5.3)平衡SP BB柱;将复性液流经柱子,再用5倍柱体积的平衡液清洗柱子,去除非特异性结合蛋白;用15倍柱体积的洗脱缓冲液(20mM醋酸钠,1M NaCl,pH 5.3)梯度洗脱,收集含目标蛋白的洗脱液。
结果如图2所示,wt-IL-21和IL-21突变体只经一步SP BB捕获便可以得到较高纯度的目的蛋白。在投入等量的包涵体的情况下,1327,1329,1330和1332的复性产率要明显高于1328,1331和1333,具体复性和纯化效率排序为:1330>1329>1332>1333=1331>1327=1328。
实施例3.wt-IL-21和IL-21突变体的PEG修饰和纯化
1.PEG修饰
聚乙二醇(PEG)是一种惰性、非致癌性聚合物。聚乙二醇修饰又被称作分子的PEG化(PEGylation),可以增强疏水性药物、蛋白质、脂质体、核酸的溶解性、提高其稳定性和延长半衰期,同时还可以降低修饰物免疫原性,提高修饰物临床应用范围等。
为了延长IL-21的血浆半衰期,本发明采用了PEG修饰的方法。通过巯基偶联对含有游离半胱氨酸的IL-21分子(1327,1328,1329,1330,1331,1332和1333)进行了PEG修饰,具体方法如下:
将含有游离半胱氨酸的IL-21突变体的浓度控制在0.5-2.0mg/mL之间。将甲氧基PEG马来酰亚胺(mPEG-MAL,委托厦门赛诺邦格生物科技股份有限公司合成)溶解于50mM磷酸氢二钠溶液,pH值7.0,终浓度为100mg/mL。用超纯水配制10mM的(对-磺苯基)苯瞵二水合二甲盐(BSPP)溶液。
用氢氧化钠溶液调节IL-21突变体溶液的pH值至7.0-7.5之间。加入终浓度为0.015-0.05mM的BSPP,将IL-21突变体中的被保护半胱氨酸的进行还原,还原反应室温静置进行16-20h。添加mPEG-MAL前进行质谱分析,确定骨架蛋白的内部二硫键没有被打开,只有突变位点处游离的半胱氨酸被还原。向还原后的IL-21突变体溶液中添加100mg/mL的mPEG-MAL溶液,使IL-21与PEG的摩尔浓度比为1:10。搅拌均匀,室温静置反应30-60min后再次纯化,以去除杂质和过量的PEG。
2.PEG修饰后纯化
反应完成后,离心去除沉淀。上清用醋酸钠调整pH至5.3,电导用去离子水稀释至20ms/cm以下,流经GE公司的30S进行精纯。
操作如下:先用5倍柱体积的平衡液(20mM醋酸钠,pH 5.3)平衡30S柱;将上清流经柱子,再用5倍柱体积的平衡液清洗柱子,去除非特异性结合蛋白;用15倍柱体积的洗脱缓冲液(20mM醋酸钠,1M NaCl,pH 5.3)梯度洗脱,收集含目标蛋白的洗脱液。用脱盐柱将蛋白换到1×PBS的溶液环境中。纯化后的最终产物如图3所示。进一步计算PEG修饰后蛋白的产率,如表3所示,排序为:1330>1332>1333=1331>1329>1327>1328。

实施例4.wt-IL-21和IL-21突变体与IL-21受体(IL-21R)体外亲和力检测
本发明改造后的IL-21突变体在不同位置上引入了半胱氨酸的突变,因此需要检测突变位点是否会影响IL-21与其受体IL-21R的结合。此外对突变体进行PEG修饰后,又有进一步引入了20kDa的PEG,同样需要检测PEG修饰是否会影响IL-21与IL-21R的结合。体外亲和力检测是通过BiacoreSPR 8K仪器测定IL-21与IL-21R的解离常数(KD),具体方法如下:
1)使用Protein-A芯片(Cytiva)将带有Fc标签的IL-21R固定在芯片表面,流动相为含有0.05%Tween20的PBS缓冲液。
2)使用不同浓度(50nM,25nM,12.5nM,6.25nM,3.125nM,1.56nM,0.78nM,0nM)的IL-21及其突变体或衍生物作为分析物,检测其与IL-21R的响应值。在本发明中,计算PEG修饰的IL-21衍生物的浓度和质量时均未将聚乙二醇的分子量包含在内,仅计算了IL-21突变体蛋白的分子量。
3)实验结束后,依次计算出各个分子与IL-21R的解离常数,评估各个分子与IL-21R的亲和力差异。
1.SPR检测点突变后的IL-21与IL-21R的亲和力
结果如表4所示,相较于wt-IL-21,大多数的IL-21突变体(1327,1329,1330,1332,1333)与IL-21R的亲和力无明显的变化,说明N25C,K56C,N59C,N82C和S98C的突变并不影响IL-21与IL-21R的相互作用。而1328和1331分子与IL-21R的亲和力有一定程度的下降,说明K52C和G61C的突变会减弱IL-21与IL-21R之间的相互作用。
2.SPR检测PEG修饰后的IL-21突变体与IL-21R的亲和力
结果如表4所示,相较于wt-IL-21,PEG修饰后的IL-21突变体(1327-PEG,1329-PEG,1330-PEG,1331-PEG,1332-PEG,1333-PEG)与IL-21R的亲和力小幅度的提高。而1328-PEG与IL-21R的亲和力仍然明显低于wt-IL-21,说明K52C突变对IL-21与IL-21R的相互作用有较大的影响,后续实验可以排除1328分子。
此外,IL-21各种突变体蛋白的复性和纯化效率有较大差异的,复性和纯化效率排序为1330>1329>1332>1333=1331>1327=1328。PEG修饰过程中蛋白也会产生一定出损失,PEG修饰蛋白的产率排序为1330>1332>1333=1331>1329=1327>1328。综合蛋白复性和纯化效率以及PEG修饰后蛋白的产率,本发明初步筛选出1330,1331,1332和1333四个分子进行后续细胞实验,检验PEG修饰后蛋白促进免疫细胞增殖和激活免疫细胞的能力。

实施例5.PEG修饰的IL-21突变体对小鼠脾脏中免疫细胞的激活实验
为了进一步验证和筛选本发明中所设计的IL-21突变体的生物学活性,我们选用小鼠脾脏细胞,分别用1330-PEG,1331-PEG,1332-PEG,1333-PEG和wt-IL-21作用于小鼠脾脏细胞,检测这些分子对免疫细胞的促进增殖和激活能力。
具体操作如下:取适量mouse anti-CD3e用PBS稀释到0.25μg/mL,1mL/孔加入到24孔板中,将板子置于37℃孵育2小时。孵育好的24孔板,弃掉包被溶液,每孔加入1mL PBS洗两次,备用。取1只C57小鼠脾脏,研磨后通过40μm细胞筛分离单细胞。单细胞中加入40mLPBS,300g离心5min。去上清后使用15mL培养基重悬,转移到T75培养瓶,37℃,5%CO2培养箱静置1h。取未贴壁细胞进行细胞计数,将细胞数调整为2×106细胞/mL,0.5mL/孔铺于24孔板中。待测蛋白样品使用培养基稀释到起始浓度2000nM,然后进行10倍梯度稀释共计4个点。0.5mL/孔加入铺好的细胞中。将小鼠脾脏细胞与待测样品在37℃,5%CO2培养箱中共孵育4天。
每个样品取1×106的细胞,300g离心5min弃上清。加入1mL洗液,300g离心5min弃上清。使用100μL staining buffer重悬细胞,每管加入(CD3、CD4、CD8、CD335、CD127、CD25)混合抗体。混匀,4℃避光孵育30min。每管加入1mL staining buffer,300g离心5min弃上清。重复洗一次。每管加入400μL staining buffer重悬细胞,使用流式细胞仪进行检测。
当wt-IL-21和PEG修饰的IL-21突变体单独用药时,对小鼠脾脏中的免疫细胞无刺激作用。连用CD3抗体后,wt-IL-21和PEG修饰的IL-21突变体均可以促进CD4-CD8+T细胞和NKT细胞增殖,并可以激活CD4-CD8+T细胞,CD4+CD8-T细胞和NKT细胞。不同的是wt-IL-21介导的刺激效应是呈剂量依赖的,而PEG修饰的IL-21突变体介导的刺激效应在低浓度时上升,高浓度时下降。如图4所示,与对照组wt-IL-21相比,发现PEG修饰后的IL-21突变体促进免疫细胞增殖的能力和激活免疫细胞的能力均有不同程度下降,表明PEG修饰后的IL-21分子药效更温和,临床上具有更高的用药安全性。
实施例6.PEG修饰的IL-21突变体的小鼠体内毒性实验
IL-21分子相较于广泛研究的IL-2分子,具有明显低于后者的细胞毒性。目前文献报道的小鼠肿瘤模型中,wt-IL-21最高的用药剂量达到200μg/mouse,并且未表现出异常的毒副作用。为了检测PEG修饰后IL-21的毒性,本发明选取1330-PEG和1332-PEG作为研究对象,进一步将给药剂量提高至18mg/kg,在小鼠体内进行实验。
选取6-8周龄雌性C57BL/6小鼠随机分成3组(Vehicle,1330-PEG,1332-PEG),每组2只,给药周期为每周两次,给药途径为经尾静脉重复注射受试药物,共计3周。动物进行单独监测。每周测量两次动物体重。日常健康观察包括动物死亡率、外观、自发活动、身体姿势、食物和水的摄入量。任何其他病变和不良反应都会被记录。在实验终点,进行解剖,检查脏器有无肉眼可见病变。
注射三周以后,实验组和对照组的小鼠状态均良好。如图5所示,六只小鼠的体重变化均在合理的范围内,无明显地减少。将小鼠安乐死以后,小鼠的主要脏器未见明显的病变。由此可见,PEG修饰后的IL-21分子的毒副作用很低,在高达18mg/kg的给药剂量下仍然没有表现出明显的毒副作用,具有很高的用药安全性。
实施例7.PEG修饰的IL-21突变体在小鼠体内的药代动力学特征
人源IL-21的血浆半衰期较短,在小鼠中仅0.2小时,在食蟹猴中是0.4-0.8小时,在人体内约为2小时。过短的血浆半衰期极大地限制了IL-21的抗肿瘤药效。PEG修饰是一种常用且高效的蛋白质长效化方式,本实施例中检测了PEG修饰后的IL-21突变体的血浆半衰期。
本实施例中选取6-8周龄雌性C57BL/6小鼠随机分成4组(wt-IL-21,1330-PEG,1332-PEG,1333-PEG),每组6只,以0.15mg/kg的剂量为经尾静脉注射受试药物,分别在0,0.5,2,6,24,48,72,96h时检测小鼠外周血中受试药物的浓度。
由于检测方法的限制,1332-PEG分子对检测方法不敏感,无法检测其浓度,而wt-IL-21,1330-PEG和1333-PEG则不受影响。结果如图6所示,wt-IL-21进入小鼠血浆中以后被快速代谢消除,半衰期与文献报道的0.2h基本吻合。而PEG修饰后,1330-PEG的半衰期约为22h,1333-PEG的半衰期约为18h。由此可见PEG修饰后,IL-21的血浆半衰期得到了显著的延长。
实施例8. 1330-PEG,1332-PEG和1333-PEG体内药效实验
IL-21作为一个多效的免疫激活因子,IL-21可以促进免疫细胞增殖并抑制肿瘤的生长,但是由于野生型IL-21的半衰期较短导致IL-21抑制肿瘤的能力受到限制。PEG修饰后的IL-21具有明显长于野生型的半衰期,所以在本实施例中进一步检测了半衰期的延长对IL-21抗肿瘤药效的影响。本实施例中分别选用MC38结肠癌模型小鼠和EMT-6乳腺癌模型小鼠进行实验,在小鼠体内检测长效化的IL-21(1330-PEG,1332-PEG,1333-PEG)的抗肿瘤药效。
实验方法具体如下:复苏MC38或EMT-6细胞,体外培养,获得1.7×108细胞。选取7-8周龄雌性C57BL/6小鼠经1周适应性饲养,取1×106个细胞,接种于实验动物皮下。肿瘤体积50-100mm3后分组给药,共5组,每组6只,腹腔给药,每周给药两次,共计三周,定期测量小鼠体重和肿瘤的体积。给药信息和分组信息如表5。

MC38结肠癌肿瘤模型结果如图7所示,与对照组相比,wt-IL-21对肿瘤生长的无抑制作用。而长效化的IL-21(1330-PEG,1332-PEG,1333-PEG)相较于对照组均展现出了显著的抑制肿瘤生长的药效。特别的1330-PEG的抗肿瘤药效最优,1333-PEG次之,1332-PEG的抗肿瘤药效相比之下最弱,但1332-PEG相对对照组仍然将肿瘤体积缩小了近40%。
EMT-6乳腺癌肿瘤模型结果如图8所示,wt-IL-21组相对于对照组有较微弱的肿瘤生长抑制作用。而在注射长效化的IL-21(1330-PEG,1332-PEG,1333-PEG)后,小鼠体内的肿瘤增长速度受到明显抑制。比较三个分子在EMT-6肿瘤模型中的药效,1332-PEG的药效最优,1333-PEG次之,1330-PEG最弱,但差异较小,基本属于相似的水平。
由此可见,三个不同的IL-21突变体(1330,1332,1333)在进行PEG修饰后,随着半衰期的延长其抗肿瘤药效也得到了显著地增强,并且在MC38和EMT-6两种肿瘤模型中都展现出了稳定的肿瘤生长抑制作用,为未来临床上运用到多种肿瘤的治疗提供可能。
实施例9. 1332-PEG的抗肿瘤药效呈剂量依赖
实施例8表明低剂量的PEG修饰的IL-21便能诱导明显的抗肿瘤药效。为了最大化IL-21的抗肿瘤药效,选取1332-PEG作为研究对象,在MC38结肠癌小鼠模型中设置不同给药剂量进行抗肿瘤药效的实验。
实验方法与实施例8基本一致。给药信息和分组信息如表6。

结果如图9所示,与对照组相比,wt-IL-21(0.625mg/kg)对肿瘤生长的无抑制作用。而1332-PEG相较于对照组展现出了显著的抑制肿瘤生长的药效,并且抗肿瘤药效呈现强烈的剂量依赖。特别地,当给药剂量达到2.5mg/kg时,随着不断的给药,小鼠体内肿瘤的体积在逐步缩小。
Claims (10)
1.一种IL-21突变体,其特征在于,所述IL-21突变体选自如下任一种:
(a)其氨基酸序列由SEQ ID NO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N25,K52,K56,N59,G61,N82,S98;
或,
(b)所述IL-21突变体与(a)所述的氨基酸序列具有95%,优选98%,更优选99%的序列相同性,并且具有(a)所述蛋白的功能,其中对应于SEQ ID NO:1所示氨基酸序列的N25,K52,K56,N59,G61,N82,S98位氨基酸残基与(a)所述的氨基酸序列中的相同;
或,
(c)所述IL-21突变体由在(a)所述的氨基酸序列的C末端和/或N末端添加或缺失1-30个,更优选1-10个,还要优选1-6个,最优选1-3个氨基酸残基而形成,并且具有(a)所述IL-21突变体的功能,其中对应于SEQ ID NO:1所示氨基酸序列的N25,K52,K56,N59,G61,N82,S98位与(a)所述的氨基酸序列中的相同。
2.根据权利要求1所述的IL-21突变体,其特征在于,所述(a)的氨基酸序列由SEQ IDNO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N25,K56,N59,G61,N82,S98;所述IL-21突变体的氨基酸序列如SEQ ID NO:3、5、6、7、8、9所示;
优选地,所述(a)的氨基酸序列由SEQ ID NO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N59,G61,N82,S98;所述IL-21突变体的氨基酸序列如SEQ ID NO:6、7、8、9所示;
进一步优选地,所述(a)的氨基酸序列由SEQ ID NO:1所示序列突变而来,在选自下组的一个氨基酸残基位点突变为半胱氨酸C:N59,N82,S98,所述IL-21突变体的氨基酸序列如SEQ ID NO:6、8、9所示。
3.一种表达载体,其特征在于,所述载体包含权利要求1或2所述的IL-21突变体的核苷酸序列。
4.一种宿主细胞,其特征在于,所述宿主细胞包含权利要求1或2所述的IL-21突变体的核苷酸序列的表达载体;
优选地,所述宿主细胞是细菌;更优选地,所述宿主细胞是大肠杆菌、谷氨酸棒杆菌;最优选地,所述宿主细胞是大肠杆菌。
5.一种权利要求1或2所述的IL-21突变体的聚乙二醇衍生物,所述聚乙二醇修饰剂连接于IL-21突变体突变位点的半胱氨酸上;
优选地,所述的聚乙二醇修饰剂为巯基聚乙二醇修饰剂,是具有选自二硫对吡啶、乙烯砜、马来酰亚胺或碘乙酰胺的官能团的多元醇衍生物;
进一步优选地,所述的聚乙二醇修饰剂的分子量为20kDa。
6.一种制备权利要求5所述IL-21突变体的聚乙二醇衍生物的方法,其特征在于,包括如下步骤:用硫醇反应性多元醇剂和权利要求1或2所述的IL-21突变体反应,所述IL-21突变体具有单一游离的半胱氨酸残基,所述多元醇试剂和所述IL-21突变体的单一游离的半胱氨酸残基特异性共价结合,形成共价硫醚键,得到多元醇-IL-21突变体偶联物,回收纯化产生的目的多元醇-IL-21突变体偶联物,即得;
优选地,其中所述IL-21突变体的聚乙二醇衍生物的聚乙二醇部分具有10~40kDa之间的分子量;
进一步优选地,所述IL-21突变体的聚乙二醇衍生物的聚乙二醇部分具有20kDa的分子量。
7.一种药物组合物,其特征在于,所述药物组合物包含权利要求1或2所述的IL-21突变体,或权利要求5所述IL-21突变体的聚乙二醇衍生物,以及药学上可接受的载体或赋形剂。
8.权利要求1或2所述的IL-21突变体、或权利要求5所述IL-21突变体的聚乙二醇衍生物,或权利要求7所述的药物组合物在制备药物中的应用,所述的药物是调节或激活免疫的药物或抗肿瘤药物。
9.权利要求1或2所述的IL-21突变体、或权利要求5所述IL-21突变体的聚乙二醇衍生物,或权利要求7所述的药物组合物在制备促进B细胞分化增殖,促进T细胞分化增殖,促进NK细胞分化增殖的制剂中的应用。
10.权利要求1或2所述的IL-21突变体、或权利要求5所述IL-21突变体的聚乙二醇衍生物,或权利要求7所述的药物组合物在制备预防和/或治疗肿瘤药物中的用途。
