Abstract
本发明公开了一种复合型类胡萝卜素及其制备方法与应用,属于类胡萝卜素技术领域,制备方法包括如下步骤:将植物油、类胡萝卜素和多酚稳定剂混合,超声分散后得到芯材,将明胶溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,用高速分散机乳化,调节pH值为弱酸性,搅拌,降温至15‑20℃,加入交联剂,调节pH值为弱碱性,搅拌,搅拌结束后,经过过滤、水洗、干燥,即可。该复合型类胡萝卜素以明胶和阿拉伯胶为壁材,在溶液中形成凝聚物,然后经过交联剂交联固化,形成微囊,对内部芯材可以实现缓慢释放,不仅可以对芯材中的成分进行有效地保护,而且可以实现有效成分的缓慢释放,延长作用时间。
Description
技术领域
本发明属于类胡萝卜素技术领域,具体涉及一种复合型类胡萝卜素及其制备方法与应用。
背景技术
类胡萝卜素是一类具有多种生物活性的天然色素,主要存在于动物、高等植物、真菌、藻类的黄色、橙红色或红色的色素之中。从微生物,动物,植物等中已经鉴定出超过750种类胡萝卜素。例如,西红柿,胡萝卜,火烈鸟和龙虾的颜色是由类胡萝卜素颜料着色的。类胡萝卜素在自然界中的生理作用是多种多样的,并且在光合作用,光保护和抗氧化作用中的辅助采光作用中起着重要作用。此外,类胡萝卜素是人类和其他动物的人体所需的营养素是维生素A的前体,近年来在癌症和心脏疾病的也有报道预防作用。
类胡萝卜素通常具有式C40H56的基本骨架,该骨架由连接在一起的八个异戊二烯单元组成。构成类胡萝卜素化学结构的高度不饱和长烃链降低了它的水溶性和生物利用度,还增加了它们对光、pH、氧和高温的敏感性,限制了它们的工业适用性。
发明内容
本发明的目的在于提供一种复合型类胡萝卜素及其制备方法与应用,以解决类胡萝卜对温度、光照敏感性较强,限制其工业适应性的问题。
本发明的目的可以通过以下技术方案实现:
一种复合型类胡萝卜素的制备方法,包括如下步骤:
将植物油、类胡萝卜素和多酚稳定剂混合,超声分散后得到芯材,将明胶溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,用高速分散机乳化,调节pH值为弱酸性,搅拌10min-20min,降温至15-20℃,加入交联剂,调节pH值为弱碱性,搅拌30min-40min,搅拌结束后,经过过滤、水洗、干燥,得到一种复合型类胡萝卜素。
进一步地,芯材中植物油、类胡萝卜素和多酚稳定剂的用量比为70-75:20:0.5-1;明胶和阿拉伯胶的质量比为1:1;明胶溶液和阿拉伯胶溶液的质量分数为1%;明胶和阿拉伯胶的总用量和芯材的质量比为1:1.5-1.7;交联剂的添加量为明胶和阿拉伯胶的总用量的1%。耐热性更好,间接影响抗氧化效果。
进一步地,所述类胡萝卜素包括为β-胡萝卜素、叶黄素、玉米黄质、岩藻黄素和虾青素中两种或两种以上按照任意比例混合。
进一步地,所述植物油包括奇亚籽油、核桃油、杏仁油和玉米油。
进一步地,所述交联剂通过如下步骤制备:
将氧化海藻酸钠和水混合,调节pH值为9,在35℃下分批加入经过无水乙醇稀释的辛烯基琥珀酸酐,加完后,维持反应体系的温度为9,搅拌反应3h,反应结束后,调节pH值为7,用无水乙醇水溶液洗涤,干燥后得到交联剂。
进一步地,所述氧化海藻酸钠通过如下步骤制备:
将海藻酸钠分散在无水乙醇中,搅拌分散得到分散液,加入高碘酸钠水溶液,避光搅拌反应4h,反应结束后加入乙二醇中止反应,将得到的反应液与无水乙醇混合,析出沉淀后,经过过滤干燥,得到氧化海藻酸钠。氧化海藻酸钠的氧化度为80%。
进一步地,所述多酚稳定剂为儿茶素、山柰酚和槲皮素中的一种。
一种复合型类胡萝卜素,由上述制备方法制备而成。
一种复合型类胡萝卜素应用于食品、化妆品中,作为营养增补剂和着色剂使用。
本发明的有益效果:
本发明提供了一种复合型类胡萝卜素,以明胶和阿拉伯胶为壁材,在溶液中形成凝聚物,然后经过交联剂交联固化,形成微囊,对内部芯材可以实现缓慢释放,不仅可以对芯材中的成分进行有效地保护,而且可以实现有效成分的缓慢释放,延长作用时间。
本发明提供的复合型类胡萝卜素中含有植物油、类胡萝卜素和多酚稳定剂,类胡萝卜素长期处于高温条件下,其清除自由基的能力会下降,在高温下容易加速芯材的氧化变质,本发明中的加工过程温度低,能减少在加工过程中有效成分的损失,配合多酚稳定剂能更好的发挥清除自由基的作用,此外,植物油可以根据需要选择,不能可以提高芯材中各物质的混合效果,而且在成分上选择性多,可以满足不同领域的需求。
本发明中在选择交联剂的过程中选择自制的交联剂,相对于常规的甲醛作为交联剂的能形成复杂的交联网络,具有更好的耐热性,并且,相对于甲醛更加安全,不用考虑残留、安全性等问题;更为突出的是,交联剂在使用前经过了辛烯基琥珀酸酐的修饰,提高了交联剂的两亲性,对于芯材的载量也具有一定的提升,能满足高载量的需求。

下面将结合本发明实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其它实施例,都属于本发明保护的范围。
实施例1
本实施例提供一种交联剂,通过如下步骤制备:
将5g海藻酸钠分散在25mL无水乙醇中,搅拌分散得到分散液,加入高碘酸钠水溶液,避光搅拌反应4h,反应结束后加入乙二醇中止反应,将得到的反应液与无水乙醇混合,析出沉淀后,经过过滤干燥,得到氧化海藻酸钠。控制氧化海藻酸钠的氧化度为80%;海藻酸钠:Mn=357475,Mn/Mw=1.392,食品级。
将2g氧化海藻酸钠和10mL水混合,调节pH值为9,在35℃下分批加入经过1mL无水乙醇稀释的0.2g辛烯基琥珀酸酐,加完后,维持反应体系的温度为9,搅拌反应3h,反应结束后,调节pH值为7,用无水乙醇水溶液洗涤,干燥后得到交联剂。
实施例2
本实施例提供一种复合型类胡萝卜素的制备方法,包括如下步骤:
将奇亚籽油、类胡萝卜素和儿茶素混合,超声分散后得到芯材,将明胶(A型明胶)溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,用高速分散机乳化,调节pH值为弱酸性(pH值为5),搅拌10min,降温至15℃,加入按照实施例1中的方法制备的交联剂,调节pH值为弱碱性(pH值为9),搅拌30min,搅拌结束后,经过过滤、水洗、干燥,得到一种复合型类胡萝卜素。类胡萝卜素为β-胡萝卜素和虾青素等质量混合而成;
芯材中奇亚籽油、类胡萝卜素和儿茶素的用量比为70:20:0.5;明胶和阿拉伯胶的质量比为1:1;明胶溶液和阿拉伯胶溶液的质量分数为1%;明胶和阿拉伯胶的总用量和芯材的质量比为1:1.5;交联剂的添加量为明胶和阿拉伯胶的总用量的1%。
实施例3
本实施例提供一种复合型类胡萝卜素的制备方法,包括如下步骤:
将奇亚籽油、类胡萝卜素和儿茶素混合,超声分散后得到芯材,将明胶(A型明胶)溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,用高速分散机乳化,调节pH值为弱酸性(pH值为5),搅拌10min,降温至15℃,加入实施例1中的方法制备的交联剂,调节pH值为弱碱性(pH值为9),搅拌40min,搅拌结束后,经过过滤、水洗、干燥,得到一种复合型类胡萝卜素。类胡萝卜素为β-胡萝卜素和虾青素等质量混合而成;
芯材中奇亚籽油、类胡萝卜素和儿茶素的用量比为75:20:1;明胶和阿拉伯胶的质量比为1:1;明胶溶液和阿拉伯胶溶液的质量分数为1%;明胶和阿拉伯胶的总用量和芯材的质量比为1:1.6;交联剂的添加量为明胶和阿拉伯胶的总用量的1%。
实施例4
本实施例提供一种复合型类胡萝卜素的制备方法,包括如下步骤:
将奇亚籽油、类胡萝卜素和儿茶素混合,超声分散后得到芯材,将明胶(A型明胶)溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,用高速分散机乳化,调节pH值为弱酸性(pH值为5),搅拌20min,降温至20℃,加入实施例1中的方法制备的交联剂,调节pH值为弱碱性(pH值为9),搅拌40min,搅拌结束后,经过过滤、水洗、干燥,得到一种复合型类胡萝卜素。类胡萝卜素为β-胡萝卜素和虾青素等质量混合而成;
芯材中奇亚籽油、类胡萝卜素和儿茶素的用量比为75:20:1;明胶和阿拉伯胶的质量比为1:1;明胶溶液和阿拉伯胶溶液的质量分数为1%;明胶和阿拉伯胶的总用量和芯材的质量比为1:1.7;交联剂的添加量为明胶和阿拉伯胶的总用量的1%。
对比例1
本对比例与实施例4相比,交联剂换成2%的甲醛其余原料及制备过程保持不变。
对比例2
本对比例与实施例4相比,交联剂换成实施例1中的氧化海藻酸钠其余原料及制备过程保持不变。
对比例3
对比例与对比例2相比,不添加儿茶素余原料及制备过程保持不变。
将装有300mL、2%的微胶囊悬浮液的烧杯在20℃条件下放置20min,然后放置在80℃水浴中,在200rpm下搅拌,每隔20min取出一个过滤,收集过滤后的溶液,测定溶液吸光度值变化。

从测试结果可知,本发明中的微囊结构能更好地储存以及缓释类胡萝卜素,在高温条件下,能对其内部包埋的类胡萝卜复合物更好地保护。
以质量分数100%计,饮料中含有白砂糖20%、柠檬酸0.5%、甜橙香料0.15%,在饮料中添加0.5%的实施例4的复合型类胡萝卜素,余量为水。将饮料置于50℃,光照条件下放置1年,观察沉淀和浊度。采用浊度计测定饮料的浊度,浊度单位NTU,测定角度90°±2.5°,测定波长860nm±10nm,测试结果如下表2;

由表2可以看出,本发明制备的复合型类胡萝卜素具有良好的缓释效果,可以延长饮料的货架期,提高饮料的稳定性。

Claims (9)
1.一种复合型类胡萝卜素的制备方法,其特征在于,包括如下步骤:
将植物油、类胡萝卜素和多酚稳定剂混合,超声分散后得到芯材,将明胶溶液和阿拉伯胶溶液混合,然后加入混合好的芯材,在温度为40℃条件下,搅拌分散,调节pH值为弱酸性,搅拌10min-20min,降温至15-20℃,加入交联剂,调节pH值为弱碱性,搅拌30min-40min,搅拌结束后,经过过滤、水洗、干燥,得到一种复合型类胡萝卜素。
2.根据权利要求1所述的一种复合型类胡萝卜素的制备方法,其特征在于,芯材中植物油、类胡萝卜素和多酚稳定剂的用量比为70-75:20:0.5-1;明胶和阿拉伯胶的质量比为1:1;明胶溶液和阿拉伯胶溶液的质量分数为1%;明胶和阿拉伯胶的总用量和芯材的质量比为1:1.5-1.7;交联剂的添加量为明胶和阿拉伯胶的总用量的1%。
3.根据权利要求1所述的一种复合型类胡萝卜素的制备方法,其特征在于,所述类胡萝卜素包括为β-胡萝卜素、叶黄素、玉米黄质、岩藻黄素和虾青素中两种或两种以上按照任意比例混合。
4.根据权利要求1所述的一种复合型类胡萝卜素的制备方法,其特征在于,所述植物油包括奇亚籽油、核桃油、杏仁油和玉米油。
5.根据权利要求1所述的一种复合型类胡萝卜素的制备方法,其特征在于,所述交联剂通过如下步骤制备:
将氧化海藻酸钠和水混合,调节pH值为9,在35℃下分批加入经过无水乙醇稀释的辛烯基琥珀酸酐,加完后,维持反应体系的温度为9,搅拌反应3h,反应结束后,调节pH值为7,用无水乙醇水溶液洗涤,干燥后得到交联剂。
6.根据权利要求5所述的一种复合型类胡萝卜素的制备方法,其特征在于,所述氧化海藻酸钠通过如下步骤制备:
将海藻酸钠分散在无水乙醇中,搅拌分散得到分散液,加入高碘酸钠水溶液,避光搅拌反应4h,反应结束后加入乙二醇中止反应,将得到的反应液与无水乙醇混合,析出沉淀后,经过过滤干燥,得到氧化海藻酸钠;氧化海藻酸钠的氧化度为80%。
7.根据权利要求1所述的一种复合型类胡萝卜素的制备方法,其特征在于,所述多酚稳定剂为儿茶素、山柰酚和槲皮素中的一种。
8.一种复合型类胡萝卜素,由权利要求1-7任意一项所述的制备方法制备而成。
9.一种根据权利要求8所述的复合型类胡萝卜素应用于食品、化妆品中,作为营养增补剂和着色剂使用。
