Abstract
本发明涉及一种动脉重塑标志物及其应用;所述标志物包括PPARa和YAP中的至少一种,所述标志物可用于制备或筛选具有如下用途的药物,A1)治疗动脉重塑;A2)抑制肌成纤维细胞生成;A3)抑制动脉巨噬细胞浸润;A4)抑制动脉炎症反应;A5)抑制动脉巨噬细胞向M1极化。所述的标志物也可以用于制备动脉重塑辅助诊断和/或疗效评价产品中。
Description
技术领域
本发明属于生物医药技术领域,特别是涉及一种动脉重塑标志物及其应用。
背景技术
动脉重塑通过细胞重组和基质过度生成,损害动脉正常的机械功能,增加心肌梗塞、脑卒中、肾功能衰竭及失明等并发症的风险,是心血管病死亡率的独立危险因素之一。动脉在收缩期储存弹性能量,在舒张期维持血流。动脉重塑导致动脉顺应性降低,增加左心室后负荷影响心脏收缩,且降低舒张压,导致冠状动脉灌注减少,同时升高微血管床内的搏动负荷影响大脑和肾脏的功能[1,2]。血压和年龄是动脉重塑的主要决定因素,目前临床上以降脂和降压来控制血管重塑[3,4]。然而,由于血压血脂控制的达标率低,且有些顽固性高血压患者即使联合使用三种或三种以上的降压药物也无法达到正常血压[5,6]。因此寻找新的有效延缓动脉重塑的生物学靶点具有迫切的临床需求与价值。
动脉重塑包括内膜内皮细胞功能异常,中膜平滑肌表型转换,外膜胶原沉积和炎症反应[7-9],涉及细胞肥大、增殖、凋亡、炎症反应、氧化应激等,其发生发展与巨噬细胞介导的炎症反应紧密相关。
过氧化物酶体增殖物激活受体α(PPARα)是一种配体诱导的转录因子,属于核受体(NR)超家族PPAR,其家族由PPARα(NR1C1)、PPARβ/δ(NR1C2)和PPARγ(NR1C3)组成[10]。已有报道PPARα在单核细胞/巨噬细胞、心脏、血管平滑肌细胞、内皮细胞中表达。PPAR与类视黄醇X受体(RXR)形成异二聚体,以识别特定的DNA序列并诱导靶基因的表达。PPARα参与一系列生理过程,包括线粒体脂肪酸氧化、分解代谢、炎症反应和应激反应[11]。尽管PPARα是一种相对已知的分子,但其在动脉重塑中的作用尚不明确。
Yes相关蛋白(YAP)是Hippo信号通路的主要调控因子[12]。YAP磷酸化后在细胞质中降解和失活,反之,YAP入核后结合转录激活因子TAZ启动基因转录调控细胞增殖、分化、炎症激活等生物学过程[13]。近期研究表明,在动脉粥样硬化、心梗后修复中,YAP入核后促进巨噬细胞向M1型极化,炎症反应加重[14,15]。既往研究显示,PPARδ通过与YAP的相互作用促进胃肿瘤发生。PPARγ通过与TAZ相互作用调节脂肪细胞炎症反应。在巨噬细胞中PPARα与YAP之间是否存在相互作用,尚未见报道。

发明内容
基于此,为解决现有技术中存在的至少一种技术问题,本发明提出一种动脉重塑标志物及其应用。
为实现上述目的,本发明采用如下技术方案:
本发明的第一方面,提供了一种动脉重塑标志物,所述标志物包括PPARα和YAP中的至少一种。
优选地,所述动脉重塑为压力超负荷诱导的升主动脉重塑。
本发明的第二方面还提供了上述的标志物在制备/或筛选药物中的应用,所述药物用途为如下A1)-A5)中的至少一种:
A1)治疗动脉重塑;
A2)抑制肌成纤维细胞生成;
A3)抑制动脉巨噬细胞浸润;
A4)抑制动脉炎症反应;
A5)抑制动脉巨噬细胞向M1极化。
优选地,所述药物包括有提升PPARα表达量的试剂和/或抑制YAP表达量的试剂。
进一步优选地,所述提升PPARα表达量的试剂有效成分为PPARα激动剂Wy14643,所述抑制YAP表达量的试剂为维替泊芬。
优选地,所述药物可通过口服、舌下含服、静脉注射、皮下注射、肌肉注射、吸入方式给药。
本发明的第三方面还提供了上述的标志物在制备动脉重塑辅助诊断和/或疗效评价产品中的应用,所述产品通过检测样本中的PPARα和/或YAP的表达水平来辅助诊断动脉重塑和/或疗效评价。
所述的产品可以是各种检测试剂,也可以是试剂盒或芯片,如基因芯片、蛋白质芯片、基因检测试剂盒、蛋白免疫检测试剂盒等。
优选地,所述产品包括,通过RT-qPCR、WB、免疫荧光标记或转录组测序检测PPARα和/或YAP表达水平的产品。
进一步优选地,所述用RT-qPCR检测PPARα基因表达水平的产品至少包括一对特异性扩增PPARα基因的引物。
进一步优选地,所述引物由引物F和引物R组成,
所述引物F为如下B1)或B2):
B1)序列表的序列1所示的单链DNA分子;
B2)将序列1经过一个或几个核苷酸的取代和/或缺失和/或添加且与序列1具有相同功能的DNA分子;
所述引物R为如下B3)或B4):
B3)序列表的序列2所示的单链DNA分子;
B4)将序列2经过一个或几个核苷酸的取代和/或缺失和/或添加且与序列2具有相同功能的DNA分子。
与现有技术相比,本发明具有以下有益效果:
本研究通过构建主动脉弓缩窄(Transverse Aortic Constriction,TAC)模型,运用转录组测序筛选压力超负荷诱导的重塑动脉中的差异表达转录本,筛选出参与动脉重塑发生的重要分子靶标过氧化物酶体增殖物激活受体alpha(Peroxisome Proliferator-Activated Receptor alpha,PPARα)。通过体内外实验,明确了PPARα在动脉重塑后表达降低,激活PPARα可抑制YAP入核,减少巨噬细胞M1极化,降低血管外膜炎症反应与胶原沉积,改善动脉重塑。另外,进一步实验发现,PPARα和YAP存在相互作用,抑制YAP可通过减少巨噬细胞向M1极化,改善动脉重塑;本研究的发现为PPARα靶向治疗动脉重塑提供了分子基础,填补了巨噬细胞PPARα/YAP与压力诱导动脉重塑之间的理论空白,对早期干预和预防动脉重塑具有重要意义。

具体实施方式
下面将结合附图,对本发明的优选实施例进行详细的描述。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实验动物
小鼠:8周龄的C57BL/6J雄性小鼠,平均为体重20-25g,购于浙江维通利华实验动物技术有限公司。所有动物操作遵循南方医科大学实验动物伦理规范。小鼠被饲养在南方医科大学南方医院实验动物中心,饲养环境为清洁级,给予小鼠每日自由饮水、进食及在笼内自由活动,不受限,环境清洁,温度和湿度适宜,12小时照明,12小时黑暗。
主要试剂的配制
(1)脂多糖(LPS)
10mg粉末中加入无菌水2mL,稀释为5.0mg/mL。取40μL加入无菌水960μL,浓度为200μg/mL。再加入无菌水9mL,浓度为20μg/mL,分装后冻存于-80℃冰箱。
(2)干扰素γ(IFNγ)
20μg粉末中加入无菌水20μL,稀释为1.0mg/mL,加入980μLPBS缓冲液,浓度为20μg/mL。取100μL加入400μLPBS缓冲液,浓度为4μg/mL,分装后冻存于-80℃冰箱。
(3)PPARα激动剂(Wy14643):
50mg粉末中加入DMSO1250μL,浓度为40mg/mL。
(4)YAP抑制剂(维替泊芬)
100mg维替泊芬粉末中加入DMSO1000μL,剧烈震荡混匀,原液浓度为100mg/mL,避光放置于-80℃冰箱保存。工作液由10%母液+40%PEG300+5%吐温80+55%双蒸水配制,现配现用。
主要实验方法:
1、TAC小鼠制备(图1)
(1)采用C57BL/6雄性小鼠(7-8周龄,体重10-24g)制备TAC模型。
(2)氯胺酮和盐酸二甲苯胺噻嗪联合麻醉,通过观察夹尾反射强弱来判断麻醉深度。
(3)将小鼠以仰卧位胶带固定在温控手术台上,用脱毛膏小心去除胸部的毛发,酒精消毒。
(4)用冷光源照射辅助压舌板做气管插管后,连接小动物呼吸机。通过观察胸廓起伏情况判断气管插管是否成功。
(5)剪开小鼠左前胸第二肋骨处皮肤。用镊子小心钝性分离,并依次分离胸大肌、胸小肌和肋间肌,探入第二肋间间隙暴露胸腺。将胸腺拨向左侧,充分暴露主动脉弓。钝性分离主动脉弓周围组织,分离部位局限于头臂干和左颈总动脉之间。
(6)用6-0丝线从主动脉弓下缘穿出,而后在其上放置一枚27号垫扎针,对主动脉及垫扎针结扎。
(7)抽出垫扎针,缝合,等待小鼠恢复自主呼吸时再脱离呼吸机。术后保温护理。
手术后1周,用异氟烷麻醉后,用多普勒超声检测TAC组和Sham组主动脉弓结扎部位(图1E)、右颈总动脉和左颈总动脉处的血流速度(图1F),当压力阶差大于40mmHg及RC/LC大于5时,视为TAC模型构建成功。
2、组织RNA提取
(1)升主动脉组织总RNA的提取
1)采集新鲜升主动脉血管组织,剪碎、离心、液氮中保存;
2)在每50-100mg组织中加入1000μL Trizol试剂;然后匀浆、振荡、离心、异丙醇沉淀、洗涤、室温晾干;
3)待沉淀由白色转为无色透明时,视样品量加入10-20μL无RNase的水溶解RNA,吹打混匀,可置于4℃下重复溶解10min。
(2)总RNA质量检测
用NanoDrop ND-1000微量分光光度计测定总RNA的浓度。并以OD260/OD230和OD260/OD280的吸光度比值来判断提取的总RNA的浓度和纯度。
(3)逆转录,cDNA合成
逆转录体系20μL为5×PrimeScript RT Master 4μL,RNA5μL,DEPC11μL。RNA取总质量1000μg。按上述体系加入ep管后,微量离心。PCR仪器设置37℃15min,85℃5s,4℃保存。
(4)实时荧光定量PCR(qRT-PCR)
反应体系为10μL,其中SYBR TBGreen IImix 5μL、上、下游引物各为0.4μL、cDNA模板加RNase-free水共4.2μL,反应条件:预变性:95℃反应30s;95℃、60℃分别反应5s、30s,40个循环;95℃反应5s,60℃保温l min后制作溶解曲线,检测引物的特异性,50℃降温30s结束反应。每一样本设置3个复孔,以Gapdh作为内参,采用2-△△Ct法分析基因相对表达量。
(5)引物设计
根据Genebank数据库(网址为:http://www.ncbi.nlm.nih.gov/genbank)查找待检测基因的mRNA序列并用Primer-BLAST在线软件设计特异性的引物(网址为:https:// www.ncbi.nlm.nih.gov/tools/primer-blast),
相关引物设计如表1所示。

3、组织蛋白提取
(1)采集新鲜升主动脉血管组织,剪碎,放入离心管中,迅速放置在液氮中保存;
(2)在每10-20mg组织中加入150-200μL蛋白裂解试剂RIPA(含1%蛋白酶抑制剂+蛋白磷酸酶抑制剂+PMSF);用匀浆机进行匀浆,30秒每次间隔5秒,共进行匀浆3次;
(3)将组织匀浆置于冰上静置30分钟;
(4)4℃,12000g离心15分钟;
(5)将蛋白上清液转移至新EP管中,吸取2μL用于蛋白浓度测定;
(6)总蛋白的浓度测定:用BCA法检测蛋白浓度,将标准蛋白用RIPA稀释为0.5mg/mL,8个标准孔中分别加入0μL、1μL、2μL、4μL、8μL、12μL、16μL、20μL,不到20μL的孔用RIPA裂解液补充至20μL。每孔2μL样品,各孔加入200μL BCA工作液。37℃孵育30min。用酶标仪检测每个样品在562nm波长处的吸光值,从而计算相应的蛋白质浓度;
(7)加入1/4体积量的上样缓冲液loading buffer(5×);100℃煮沸10分钟,分装后-80℃保存。
4骨髓巨噬细胞提取
(1)采用C57BL/6雄性小鼠(7-8周龄,体重10-24g)提取小鼠骨髓巨噬细胞。
(2)小鼠断颈处死,用70%乙醇浸泡小鼠消毒。
(3)分离胫骨股骨,将离体胫骨股骨放入装有PBS(含1%双抗)的离心管中。
(4)转移离心管至细胞台中,无菌PBS(含1%双抗)反复清洗后,用剃净残余肌肉,剪断骨骺端。
(5)用注射器吸取PBS(含1%双抗),反复冲洗骨髓腔,直到骨髓从红到白。
(6)用70μm滤网过滤出单细胞骨髓悬液,加入3倍体积的0.8%NH4Cl溶液冰上裂解红细胞10min。
(7)4℃,500g离心5min收集细胞。
(8)DMEM培养基(含20%L929上清+10%血清+1%双抗)培养细胞7天诱导为骨髓巨噬细胞,第三天换液一次。其中L929上清为小鼠上皮成纤维细胞(L929)种在T75培养瓶中连续培养7天不换液收集而来。
(9)收集细胞,流式染色判断巨噬细胞分化纯度。
5细胞常规培养
RAW264.7细胞培养基为DMEM高糖加10%的胎牛血清,小鼠骨髓巨噬细胞(BMDM)培养基为DMEM高糖加10%的胎牛血清、1%青霉素、链霉素及20%的L929上清。
细胞传代
待RAW264.7细胞生长至80%左右的融合度时将原先的细胞培养液弃去,加入PBS,洗涤细胞1次,然后加入冰的PBS,轻柔吹打皿底,使贴壁细胞脱落。收集细胞至离心管中,1000rpm,5min离心,弃上清,用新的细胞培养基重悬细胞,按1:2~1:3的比例将细胞接种于新的培养皿中,置于细胞孵箱中继续培养。
6细胞处理
细胞传代后,调整细胞至合适密度后接种于六孔板中。待细胞贴壁,弃去培养基,加入PBS,洗涤细胞1次,用无血清的DMEM高糖培养液中进行培养,培养基中加入PBS、10μM和100μM的Wy14643处理24小时,或者,培养基中加入PBS、2μM的维替泊芬处理24小时。更换培养基并用PBS洗涤一次,加入无血清的DMEM高糖培养液,培养基中加入100ng/mL的LPS和20ng/mL的IFN-γ处理24小时。
7细胞转染
本实验用到的si-PPARα及空白对照si-RNA均由上海和元生物技术有限公司合成。
(1)将细胞种植到六孔板中,待细胞融合密度为50%~70%,弃去旧培养基,PBS缓冲液洗涤一次。用不含血清的DMEM培养基饥饿细胞6小时。
(2)取两只无菌ep管,各加入250μL减血清培养基opti,一管中加5μLLipofectamin RNAiMAX,一管中加入si-RNA稀释浓度为100nM;
(3)将两只ep管的液体轻柔混匀,室温静置15min;
(4)将混合液均匀滴加到细胞培养液中,轻轻混匀;
(5)转染8小时后,将培养基更换为含10%FBS的DMEM;
(6)培养48~72小时后提取RNA或蛋白进行转染效果验证。
8胞核胞浆蛋白抽提
(1)PBS洗涤细胞,用细胞刮刀将贴壁细胞从培养皿刮下,将细胞悬液转移到预冷的离心管中。
(2)吸干PBS,加入胞浆提取试剂A(含1%蛋白酶抑制剂+蛋白磷酸酶抑制剂+PMSF);
(3)剧烈震荡5s,冰浴10-15min;
(4)加入试剂B10μL,剧烈震荡5s,冰浴1min;
(5)剧烈震荡5s,4℃,12000g离心5分钟(6)吸取上清即为胞浆蛋白,留取少量上清,以免触及沉淀
(6)吸尽残余上清,往沉淀中加入50μL细胞核蛋白抽提试剂(含1%蛋白酶抑制剂+蛋白磷酸酶抑制剂+PMSF),不吸尽上清会带来细胞浆蛋白的污染
(7)用超声机进行破碎,65hz,30s每次间隔5s;将超声后的细胞置于冰上静置30分钟;
(8)4℃,12000g离心5分钟,取上清即为细胞核蛋白。
实施例采用SPSS16.0统计软件对数据进行处理,实验结果以平均值±标准偏差表示,差异性检验水准均设为P=0.05(双侧)。多组间比较采用单向方差分析:若方差齐性,组间多重比较采用Bonferroni’s法。*P<0.05表示具有显著性差异,**P<0.01表示具有极显著性差异,***P<0.001表示具有极显著性差异。
实施例1、PPARα可以作为压力超负荷诱导的动脉重塑的生物标志物
一、采用主动脉弓缩窄(TAC)的动物模型评价组织中PPARα的表达量对于动脉重塑的早期预测作用
1、TAC模型的构建与验证
TAC模型组:参照上述试验方法构建。
Sham(假手术)组:与模型组操作基本相同,但只穿线不进行结扎。
2、压力超负荷诱导升主动脉发生明显重塑(图2)
手术后两周处死小鼠,分离主动脉组织,多聚甲醛固定后行常规石蜡包埋及切片,切片制作完成后常规脱蜡水化,行HE染色和天狼星红染色分析形态学变化(图2A)。结果显示,在压力负荷升高的情况下,主动脉血管壁发生明显病变。与Sham组相比,TAC组升主动脉中膜面积(图2B左)和外膜面积(图2B右)明显增加,以外膜最为显著。天狼星红染色结果显示TAC组升主动脉外膜胶原面积(图2C左)和胶原面积占外膜面积的百分比(图2C右)均显著增加。
以上结果提示在压力负荷升高诱导主动脉发生明显重塑,伴随主动脉壁增厚及胶原沉积增加。
3、动脉重塑后升主动脉外膜巨噬细胞浸润显著增加及炎症因子上调(图3)
TAC手术后分别在第3天、7天及14天时处死小鼠,分离主动脉组织,多聚甲醛固定后行常规石蜡包埋及切片,切片制作完成后常规脱蜡水化,对分离的升主动脉切片做免疫荧光检测,用F4/80标记巨噬细胞。结果可见与Sham组相比,TAC手术3天、7天及14天后,巨噬细胞在升主动脉血管壁内浸润逐渐增加,且巨噬细胞主要浸润在动脉外膜(图3A、B)。
流式分选出升主动脉的巨噬细胞,可见,巨噬细胞占比在TAC术后3天达到高峰(图3C)。考虑因主动脉壁在术后14天显著增厚,成纤维细胞数目增加较巨噬细胞快,导致巨噬细胞占比相对第3天时降低。
qRT-PCR检测升主动脉炎症因子表达水平,TAC组升主动脉炎症因子Mcp1、Tnfα、Il1β、Il6、Mmp2的mRNA水平显著高于Sham组(图3D)。以上结果提示,在压力负荷升高诱导主动脉发生明显重塑,伴随动脉外膜巨噬细胞浸润增加,且炎症因子表达上调。
4、TAC小鼠升主动脉中免疫应答升高伴PPAR信号通路表达下调(图4)
对TAC和Sham组小鼠的升主动脉组织进行转录组测序(Illumina Novaseq 6000测序平台),测序结果已上传至NCBI(PRJNA659049)。对转录组测序数据进行生物信息学分析,共筛选出1019个差异表达的mRNAs,其中包含722个上调mRNAs和297个下调mRNAs(图4A)。GO生物过程富集分析显示,差异基因主要富集在免疫反应(图4B)。进一步对免疫相关的基因进行聚类分析,使用热图来呈现聚类结果,结果显示TAC术后升主动脉的免疫应答显著上调(图4C)。对差异基因进行基因富集分析(GSEA),筛选出明显差异表达的PPAR信号通路(表1)。ES反映PPAR信号通路相关基因集成员在样品的基因表达排序列表的富集程度。折线图中的峰值就是这个基因集的ES值,ES值越高,说明样品在该通路中富集越显著。结果显示PPAR信号通路主要富集在Sham组,即在TAC组中表达下调。
以上结果提示,TAC小鼠升主动脉中免疫应答升高伴PPAR信号通路表达下调。
5、巨噬细胞中PPARα基因表达显著下调(图5)
进一步对PPAR通路的基因进行分析,测序数据显示,与Sham组相比,TAC组升主动脉Pparα转录水平明显降低,而同家族的Pparδ和Pparγ基因表达并无显著差异(图5A)。PPARα的qRT-PCR和WB检测结果与测序结果一致(图5B、C)。
流式分选出升主动脉的巨噬细胞,TAC组的巨噬细胞中PPARα表达水平较Sham组显著降低(图5D)。
以上结果提示,PPAR信号通路中PPARα的表达在动脉重塑中被抑制。验证了在动脉重塑后巨噬细胞中的PPARα表达显著下调。提示巨噬细胞中PPARα的表达量变化可能与动脉重塑的进展相关,PPARα或可成为调控动脉重塑的新靶点。
实施例2、激活PPARα改善动脉重塑
1、TAC模型的构建与验证
与实施例1相同。
2、靶向激活PPARα改善TAC诱导的升主动脉重塑(图6)
术前3天至术后14天,于小鼠胃内给药Wy14643 10mg/kg/d,具体地:50mg Wy14643粉末中加入DMSO1250μL,浓度为40mg/mL。若每只小鼠体重为20g,则每日需要给药0.2mg,换算为原液为5μL。用玉米油稀释至100μL胃内给药,根据小鼠体重具体换算原液剂量。对照组等体积玉米油给药。给药过程如图所示(图6A)。超声评估主动脉弓结扎缩窄处的血流速度,Wy14643未改变TAC小鼠的血流动力学(图6B)。手术后两周处死小鼠,分离升主动脉组织,对分离的升主动脉切片做HE染色和形态学分析,Wy14643灌胃治疗后,与TAC组相比,其升主动脉外膜面积显著降低(图6C)。天狼星红和马松染色将胶原纤维染为红色及蓝色,结果显示与TAC组相比,TAC+Wy14643组升主动脉中外膜的胶原蛋白沉积比例显著降低(图6D、图6E)。
以上结果提示激活PPARα改善TAC诱导的升主动脉重塑,且不依赖于血流动力学的改变。
3、靶向激活PPARα显著抑制肌成纤维细胞生成(图7)
在受到伤害和应激时,成纤维细胞活化为肌成纤维细胞,显着增殖,分泌I型和III型原纤维胶原,I型胶原纤维的扩展性较差,有助于承受压力负荷,是血管僵硬的重要因素。成纤维细胞的活化被认为是血管纤维化的标志。肌成纤维细胞表达平滑肌细胞的标记物SM22α。对升主动脉切片(制作方法同上)进行Col I和SM 22α免疫组化染色结果显示(图7),激活PPARα后,肌成纤维细胞数目减少,分泌的I型胶原蛋白减少,动脉纤维化降低。以上结果提示激活PPARα显著抑制肌成纤维细胞生成。
4、靶向激活PPARα显著抑制TAC小鼠升主动脉巨噬细胞浸润(图8)
由实施例1结果得出,动脉重塑与巨噬细胞浸润及其分泌的炎症因子相关,且重塑后巨噬细胞PPARα表达下调。因此对血管壁浸润的巨噬细胞数量进行进一步评估。手术后两周处死小鼠,分离主动脉组织,对分离的升主动脉切片(制作方法同上)做免疫荧光检测,结果如图8所示。与TAC组相比,TAC+Wy14643组升主动脉外膜中巨噬细胞数目显著降低(图8A)。进一步对分离的主动脉组织做流式细胞学检测,与Sham组相比,TAC组升主动脉巨噬细胞浸润明显增加,而Wy14643灌胃治疗后,升主动脉巨噬细胞浸润显著减少(图8B)。以上结果提示,激活PPARα显著抑制TAC小鼠升主动脉巨噬细胞浸润。
5、靶向激活PPARα抑制TAC小鼠升主动脉炎症反应(图9)
手术后两周处死小鼠,分离升主动脉组织,对分离的升主动脉组织切片(制作方法同上)行免疫组化检测,结果如图9所示,与Sham组相比,TAC组升主动脉外膜MCP-1、IL-6和p-NF-κB蛋白表达明显增加,而Wy14643灌胃治疗后,与TAC组相比,TAC+Wy14643组升主动脉外膜MCP-1、IL-6和p-NF-κB蛋白表达显著减少。以上结果提示,靶向激活PPARα抑制TAC小鼠升主动脉炎症反应。
6、靶向激活PPARα抑制TAC小鼠升主动脉巨噬细胞向M1极化(图10)
通过Western-blot检测升主动脉组织PPARα、iNOS、IL6和TNFα蛋白水平情况,结果显示TAC组升主动脉PPARα表达较Sham组下调,但激活PPARα后蛋白水平无明显变化。提示激活PPARα不通过改变其蛋白水平发挥作用。TAC组升主动脉iNOS、IL6和TNFα蛋白水平水平显著高于Sham组,而Wy14643灌胃治疗后,与TAC组相比,TAC+Wy14643组升主动脉iNOS、IL6和TNFα蛋白水平显著降低(图10A)。
qRT-PCR检测升主动脉中炎症因子Mcp1、Tnfα、Il1β和Il6的mRNA表达水平,可见激活PPARα显著抑制升重塑主动脉炎症因子的表达(图10B)。
分离主动脉组织,对分离的升主动脉组织做流式细胞学检测,与Sham组相比,TAC组升主动脉M1巨噬细胞占比升高。而Wy14643灌胃治疗后,与TAC组相比,TAC+Wy14643组升主动脉M1巨噬细胞占比显著降低(图10C)。
以上结果提示靶向激活PPARα抑制TAC小鼠升主动脉巨噬细胞向M1极化。
7、靶向激活PPARα抑制巨噬细胞向M1极化(图11)
用PBS、10μM和100μM的Wy14643处理RAW264.7细胞24小时后,培养基中加入100ng/mL的LPS和20ng/mL的IFN-γ处理24小时诱导巨噬细胞分化为M1型。
Western-blot检测结果显示,巨噬细胞向M1极化后,PPARα表达下调(图11A、B),与动脉重塑后M1型巨噬细胞比例增高伴随PPARα表达下调相对应。iNOS蛋白表达水平和流式分选M1型巨噬细胞结果显示,Wy14643处理显著抑制巨噬细胞向M1极化,并且具有浓度依赖性,100μM的Wy14643较10μM对巨噬细胞向M1极化的抑制作用更强(图11A-C)。
以上结果提示,靶向激活PPARα抑制巨噬细胞向M1极化。
8、敲低PPARα减少Wy14643抑制巨噬细胞M1极化的作用(图12)
培养RAW264.7细胞,构建三条小干扰RNA敲低PPARα。通过转录组水平筛选出siRNA-2序列的敲低效率最高(图12A)。
通过Western Blot验证siRNA-2敲低PPARα后,PPARα蛋白表达水平降低50%(图12B)。在RAW264.7细胞培养基中加入100ng/mL的LPS和20ng/mL的IFN-γ处理24小时诱导巨噬细胞分化为M1型。相对于对照组,加入100μM的Wy14643处理同时,可以显著降低炎症因子Inos、Tnfα、Il6、Il1β的转录水平,敲低PPARα显著减弱了Wy14643的抗炎作用(图12C)。以上结果提示,敲低PPARα减少Wy14643抑制巨噬细胞M1极化的作用。
人和鼠的PPARα在核苷酸水平上显示出85%的同源性,在氨基酸水平上显示出91%的同源性。基于实施例1筛选出PPARα是调控动脉重塑的新靶点。
本实施例分别从体内外实验探索,首先通过PPARα的特异激动剂Wy14643胃内给药,观察到激活PPARα显著改善了动脉重塑,中膜外膜面积降低,但以外膜改变最为显著,且不依赖于血流动力学的改变。激活PPARα后,外膜中SM22α及COLI表达量减少,即抑制了成纤维细胞向肌成纤维细胞的转换。升主动脉巨噬细胞浸润减少,又分别从转录学水平、蛋白水平及组织水平验证了激活PPARα后炎症反应降低,流式结果显示升主动脉中巨噬细胞向M1极化减少。同时通过体外实验探索PPARα对巨噬细胞极化的调控作用。
说明了靶向激活PPARα可抑制巨噬细胞向M1极化,而敲低PPARα减少Wy14643同样具有抑制巨噬细胞M1极化的作用。
实施例3、激活PPARα抑制YAP入核减少巨噬细胞M1极化
1、提取小鼠骨髓原代巨噬细胞(图13)
提取小鼠骨髓原代巨噬细胞,在集落因子的刺激下,巨噬细胞分化成熟。用流式进行检测F4/80染色阳性的细胞比例,测试巨噬细胞纯度高达98.7%,如图13所示。
2、测序分析靶向激活PPARα后骨髓巨噬细胞的转录组变化(图14)
将提取的小鼠骨髓巨噬细胞,用集落刺激因子刺激分化培养7天后,分为PBS组、LPS+IFN-γ组和LPS+IFN-γ+Wy14643组。培养24小时后收集细胞进行转录组测序(图14A)。
测序结果验证激活PPARα抑制炎症相关基因,并且激活PPARα的下游基因。同时激活PPARα后可见YAP靶基因显著下调(图14B)。
3、激活PPARα抑制YAP入核(图15)
将小鼠骨髓巨噬细胞分为PBS组、LPS+IFN-γ组,LPS+IFN-γ+Wy14643组,通过免疫荧光标记YAP。可见,当巨噬细胞向M1极化时,YAP入核增加,当激活PPARα后,可以抑制YAP入核(图15A)。
进一步分离细胞的胞浆胞核蛋白进行Western Blot分析,可见当巨噬细胞向M1极化时,胞浆胞核的蛋白都表达上调,而激活PPARα后,可以降低YAP表达总量,且抑制YAP入核(图15B)。
4、PPARα与YAP存在相互作用(图16)
将小鼠骨髓巨噬细胞用LPS+IFN-γ刺激极化为M1,免疫荧光标记PPARα与YAP,发现PPARα与YAP存在共同定位(图16A)。
进一步通过免疫共沉淀,发现PPARα与YAP存在相互作用(图16B),且在激活PPARα后,PPARα与YAP之间的相互作用增强(图16C)。
以上结果提示,Wy14643增强PPARα与YAP之间的相互作用,从而抑制YAP入核。
本实施例通过免疫共沉淀及免疫荧光共定位发现,PPARα和YAP存在相互作用,且激活PPARα后相互作用增强。提示激活PPARα通过结合YAP,使YAP滞留在胞浆中,无法入核,从而抑制巨噬细胞向M1极化。
实施例4、抑制YAP降低巨噬细胞M1极化改善动脉外膜重塑
1、抑制YAP减少巨噬细胞向M1极化(图17)
培养基中加入LPS和IFN-γ处理24小时诱导巨噬细胞分化为M1型,在加入YAP抑制剂维替泊芬(Verteporfin)的同时,巨噬细胞向M1极化减弱(图17A)。提取RNA进一步分析,在YAP被抑制的情况下,炎症因子Inos、Tnfα、Il6、Il1β表达下调(图17B)。提取RNA进一步分析,在YAP被抑制的情况下,iNOS、Tnfα的蛋白表达水平下调(图17C)。提示抑制YAP减少巨噬细胞向M1极化。
2、抑制YAP改善TAC诱导的升主动脉重塑(图18)
TAC模型建立同实施例1,术前3天至术后14天,每两日维替泊芬腹腔注射处理(100mg/kg/d),具体地,100mg维替泊芬粉末中加入DMSO1000μL,浓度为100mg/mL。工作液由10%母液+40%PEG300+5%吐温80+55%双蒸水配制,浓度为10mg/mL。若每只小鼠体重为20g,则每日需要给药2mg,换算为工作液为200μL。腹腔注射给药,根据小鼠体重具体换算工作液剂量。对照组等体积溶剂给药。
手术后两周处死小鼠,分离升主动脉组织,对分离的升主动脉切片做HE染色和形态学分析,结果如图18所示,维替泊芬腹腔注射治疗后,与TAC组相比,其升主动脉外膜面积显著降低。天狼星红和马松染色将胶原纤维染为红色及蓝色,结果显示与TAC组相比,TAC+Verteporfin组升主动脉中外膜的胶原蛋白沉积比例显著降低。以上结果提示,抑制YAP改善TAC诱导的升主动脉重塑。
3、抑制YAP减轻TAC小鼠升主动脉炎症反应(图19)
手术后两周处死小鼠,分离升主动脉组织,对分离的升主动脉组织行免疫组化检测,如图19所示,与Sham组相比,TAC组升主动脉外膜MCP-1和IL-6蛋白表达明显增加,维替泊芬腹腔注射治疗后,与TAC组相比,TAC+Verteporfin组升主动脉外膜MCP-1和IL-6蛋白表达显著减少。以上结果提示,靶向抑制YAP减轻TAC小鼠升主动脉炎症反应。
本实施例说明,YAP可能是NF-κB激活核转位的共同效应物,依次发挥促炎反应。在体外实验中,给予能够抑制YAP-TEAD相互作用的小分子化合物如维替泊芬后可抑制巨噬细胞向M1极化,降低炎症因子的表达。在体内实验中,腹腔注射能够抑制YAP-TEAD相互作用的小分子化合物如维替泊芬能够显著改善压力超负荷诱导的动脉重塑,炎症因子及胶原沉积减少。以上结果提示,抑制YAP可通过减少巨噬细胞向M1极化改善动脉重塑。为早期干预和预防动脉重塑提供一个新的靶点。
综上,本研究通过构建主动脉弓缩窄(TAC)模型,运用转录组测序筛选压力超负荷诱导的重塑动脉中的差异表达转录本,筛选出参与动脉重塑发生的重要分子靶标PPARα。通过体内外实验,明确了PPARα在动脉重塑后表达降低,激活PPARα可抑制YAP入核,减少巨噬细胞M1极化,降低血管外膜炎症反应与胶原沉积,改善动脉重塑。另外,在本研究中确定了PPARα/YAP相互作用在调节巨噬细胞M1极化中的作用。通过实验发现,PPARα和YAP存在相互作用,抑制YAP可通过减少巨噬细胞向M1极化,改善动脉重塑;本研究为研究为PPARα和YAP靶向治疗动脉重塑提供了分子基础,填补了巨噬细胞PPARα/YAP与压力诱导动脉重塑之间的理论空白,丰富了动脉重塑发病机制的相关理论。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (10)
1.动脉重塑标志物,其特征在于,所述标志物包括PPARα和YAP中的至少一种。
2.根据权利要求1所述的标志物,其特征在于,所述动脉重塑为压力超负荷诱导的升主动脉重塑。
3.如权利要求1所述的标志物在制备或筛选药物中的应用,所述药物用途为如下A1)-A5)中的至少一种:
A1)治疗动脉重塑;
A2)抑制肌成纤维细胞生成;
A3)抑制动脉巨噬细胞浸润;
A4)抑制动脉炎症反应;
A5)抑制动脉巨噬细胞向M1极化。
4.根据权利要求3所述的应用,其特征在于,所述药物包括有提升PPARα表达量的试剂和/或抑制YAP表达量的试剂。
5.根据权利要求4所述的应用,其特征在于,所述提升PPARα表达量的试剂有效成分为PPARα激动剂Wy14643,所述抑制YAP表达量的试剂为维替泊芬。
6.根据权利要求3所述的应用,其特征在于,所述药物可通过口服、舌下含服、静脉注射、皮下注射、肌肉注射、吸入给药。
7.如权利要求1所述标志物在制备动脉重塑辅助诊断和/或疗效评价产品中的应用,所述产品通过检测样本中的PPARα和/或YAP的表达水平来辅助诊断动脉重塑和/或疗效评价。
8.根据权利要求7所述的应用,其特征在于,所述产品包括,通过RT-qPCR、WB、免疫荧光标记、或转录组测序检测PPARα和/或YAP表达水平的产品。
9.根据权利要求8所述的应用,其特征在于,所述用RT-qPCR检测PPARα基因表达水平的产品至少包括一对特异性扩增PPARα基因的引物。
10.根据权利要求9所述的应用,其特征在于,所述引物由引物F和引物R组成,所述引物F为如下B1)或B2):
B1)序列表的序列1所示的单链DNA分子;
B2)将序列1经过一个或几个核苷酸的取代和/或缺失和/或添加且与序列1具有相同功能的DNA分子;
所述引物R为如下B3)或B4):
B3)序列表的序列2所示的单链DNA分子;
B4)将序列2经过一个或几个核苷酸的取代和/或缺失和/或添加且与序列2具有相同功能的DNA分子。
