Abstract
本发明涉及纳米材料技术领域,具体公开了一种金纳米颗粒及其制备方法与应用。本发明提供了一种5‑巯基‑1,3,4‑噻二唑‑2‑基硫代乙酸包裹的金纳米颗粒。制备该金纳米颗粒的方法,包括:(1)在黑暗条件下,将5‑巯基‑1,3,4‑噻二唑‑2‑基硫代乙酸和氯金酸在碱性水溶液中混合,获得反应体系;所述反应体系中的碱为氢氧化钠或氢氧化钾,所述反应体系中5‑巯基‑1,3,4‑噻二唑‑2‑基硫代乙酸、氯金酸和碱的摩尔质量比为(1.8~2.2):1:(8~12);(2)以波长360~370 nm的紫外光照射所述反应体系,进行光化学还原反应至荧光强度不再升高。本发明金纳米颗粒对多种细胞的细胞膜进行特异性标记。

技术领域
本发明涉及纳米材料技术领域,具体地说,涉及一种金纳米颗粒及其制备方法与应用。
背景技术
细胞膜是一项至关重要的细胞结构,它位于细胞外部,与环境直接接触。细胞膜在多个方面发挥着关键作用,包括营养物质的吸收、代谢废物的排泄以及信息的交换。事实上,细胞膜在维持细胞的生存中扮演着关键的角色。
细胞膜充当了一个屏障,防止细胞内外的物质自由进出。这种屏障功能确保了细胞内环境的相对稳定性,从而使各种生物化学反应能够有序地进行。细胞膜的存在是原始生命向细胞进化的一个重要标志,它由脂质和蛋白质构成,其厚度约为7-8纳米。
此外,细胞膜还是免疫系统中识别受损细胞和肿瘤细胞的最关键通道之一。在人体内,T细胞具有识别异常细胞的功能,一旦它们的识别机制被肿瘤细胞阻碍,癌症可能会得以发展。设计能够定向作用于细胞膜的纳米材料,并改变肿瘤细胞表面抗原的特性,对于免疫系统主动攻击肿瘤组织具有巨大的潜力。因此,基于细胞膜的抗肿瘤药物研发预计将成为肿瘤治疗领域的新兴热点。
荧光标记技术在细胞生物学研究中扮演着关键的角色,它有助于科研人员准确识别细胞的边界。目前已有报道的细胞膜标记荧光物质主要是小分子化合物,但它们存在一些问题,如毒性高、水溶性差、稳定性差、有效期短、价格昂贵,而且它们的表面往往缺乏可修饰位点,这限制了它们在应用中的发展。
至今,还没有关于使用未经靶向分子修饰的金纳米颗粒来实现针对多种细胞膜的研究的报道。这种方法可能会为细胞膜标记领域带来重大突破。
发明内容
本发明的目的之一在于提供一种可特异性靶向细胞膜的金纳米颗粒。
为了实现该目的,本发明的技术方案如下:
一种制备金纳米颗粒的方法,其包括:
(1)在黑暗条件下,将5-巯基-1,3,4-噻二唑-2-基硫代乙酸和氯金酸在碱性水溶液中混合,获得反应体系;所述反应体系中的碱为氢氧化钠或氢氧化钾,所述反应体系中5-巯基-1,3,4-噻二唑-2-基硫代乙酸、氯金酸和碱的摩尔质量比为(1.8~2.2):1:(8~12),优选为2:1:10;
(2)以波长360~370nm(优选为365nm)的紫外光照射所述反应体系,进行光化学还原反应至荧光强度不再升高。
本发明以氯金酸为原料,以5-巯基-1,3,4-噻二唑-2-基硫代乙酸((5-mercapto-1,3,4-thiadiazole-2-ylthio) acetic acid,TMT)为还原剂,以碱性的水溶液为反应体系,按特定反应条件和物料比例进行金纳米颗粒的制备。特别使用固定的光照模式进行照射反应,赋予了本发明金纳米颗粒靶向多种细胞膜的特性。
本发明所述金纳米颗粒为一种荧光纳米材料,其是具有发光性能的金纳米颗粒(Luminescent gold nanoparticles,L-AuNPs),其特征是其表面配机为5-巯基-1,3,4-噻二唑-2-基硫代乙酸,核心为发光金纳米颗粒,即本发明纳米颗粒为TMT包被的发光金纳米颗粒(L-AuNP@TMT)。L-AuNP@TMT是通过光化学还原的方法制备获得的,其具有较好的生物相容性,在≤70 μg/mL的浓度下对细胞的生长和增殖无显著影响,具备激发发光的功能,可以特异性地靶向多种细胞膜,作为细胞膜的一种特征标记物。且其表面暴露的氨基或羧基,可增加其水溶性和生物相容性。
本发明的反应体系中,氯金酸的浓度为0.9-1.1 M,优选为1 M(mol/L)。
本发明步骤(2)中,所述光化学还原反应的反应温度为23~25℃。
本发明步骤(2)中,使用光谱检测仪检测反应过程中的荧光强度。
本发明步骤(2)中,使用功率为4.5~5.5W(优选为5W)的紫外灯进行紫外光照射,所述光化学还原反应的反应时间为15-20分钟。
本发明还提供一种金纳米颗粒,其由上述方法制备得到。
本发明的金纳米颗粒,所述金纳米颗粒的电镜尺寸为2.42±0.53 nm;所述金纳米颗粒的水动力学直径为4.75±1.63 nm。
本发明的金纳米颗粒的激发波长为350-550 nm,发射波长为500-700 nm;所述金纳米颗粒发出绿色荧光。
本发明再提供一种上述金纳米颗粒在作为细胞膜标记物中的应用。
本发明的金纳米颗粒的应用为非疾病诊断或治疗领域的应用,可用于评价、制备以细胞膜为靶点的药物。
本发明的有益效果至少在于:
本发明的金纳米颗粒L-AuNP@TMT不需要其它任何靶向标记分子及荧光基团辅助,即可以对多种细胞的细胞膜进行特异性标记。且其原材料易得、价格低廉,工艺简单、反应温和,具有很好的水溶性(浓度为2mg/mL的L-AuNP@TMT水溶液于4℃放置3年无沉淀)、光稳定性和生物安全性。

下述实施例中所使用的实验方法如无特殊说明,均为常规方法。下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到或按本领域常规方法制备。
实施例1
本实施例通过光化学还原的方法制备5-巯基-1,3,4-噻二唑-2-基硫代乙酸包裹的金纳米颗粒L-AuNP@TMT。
具体包括以下步骤:
1) 称取5-巯基-1,3,4-噻二唑-2-基硫代乙酸(TMT),均匀溶解在超纯水中,配置浓度为20 mM的TMT母液。
2) 称取氯金酸,均匀溶解在超纯水中,配置浓度为10 mM的氯金酸母液。
3) 称取氢氧化钠,均匀溶解在超纯水中,配置浓度为500 mM的氢氧化钠母液。
4) 在容量为100 ml的圆底烧瓶中加入39 mL的超纯水,加入转子,在磁力搅拌器的作用下进行搅拌。
5) 在磁力搅拌器的作用下边搅拌边向反应体系中逐滴加入配置好的TMT母液5ml,环境温度为25℃。
6) 向反应体系中逐滴加入配置好的氢氧化钠母液1 ml。
7) 向反应体系中逐滴加入配置好的氯金酸母液5 ml。
8) 在避光条件下,使得反应物充分混合。
9) 打开功率为5 W,波长为365 nm的紫外灯,照射反应瓶,反应温度为25℃,使用荧光光谱仪检测反应体系的反应过程,待荧光稳定不再升高时,关闭紫外灯,停止反应,反应时间为10分钟。
10) 使用超滤法去除未反应完的残留:使用5000 KD 的超滤管,将反应物用超纯水洗3次。
11) 将制备好的金纳米颗粒置于4℃冰箱保存。
使用投射电镜观察及统计所合成的纳米颗粒L-AuNP@TMT的大小,并进行统计。电镜观察结果见图1中的(A)。通过统计拍摄的250个纳米颗粒的直径,得出其直径为2.42±0.53 nm,参见图1中的(B)。
所合成的金纳米颗粒L-AuNP@TMT不仅含有金核,还有表面配基,可通过动态光散射的方法(仪器:纳米粒度及Zeta电位分析仪)测量其包括配基在内的水动力学直径大小,根据动态光散射结果,其直径为4.75±1.63 nm。通过测其zeta电势,可得出其表面的带点性,其表面为强负电性,-53.3 mV,参见图1中的(C)。
测量金纳米颗粒L-AuNP@TMT的光谱特性,结果参见图1中的(D)。根据金纳米颗粒的吸收谱可知,其在500 nm无显著光吸收,表明其不存在表面等离子体吸收现象;根据其激发光谱可知,其最适激发波长为467 nm;根据其发射光谱可知,其最强发射波长为548 nm。
实施例2
本实施例对实施例1制备的金纳米颗粒L-AuNP@TMT的细胞靶向性进行研究,具体方法如下:
1)在共聚焦培养皿铺Hepa 1-6细胞,密度为3万/mm2。将金纳米颗粒L-AuNP@TMT分散在PBS缓冲液中,浓度为70μg/mL,孵育Hepa 1-6细胞3分钟,将上清液吸走,PBS洗三次,加入细胞的完全培养基。
2)激光共聚焦显微镜成像,用488 nm的激光作为激发光源,接受500-700 nm的荧光信号,结果如图2中的(A)的左图。
3)与传统的细胞膜染料进行对比:使用Cellmask Deep Red Plasma MembrMarker染细胞膜,按照产品说明中染料推荐方法对细胞膜染色,结果如图2中的(A)的中间图。
4)将两张图重叠,研究其是否共定位,结果如图2中的(A)的右图。
5)对图2中的(A)的右图白线处的红色荧光(Cellmask Deep Red Plasma MembrMarker)和绿色荧光(L-AuNP@TMT)进行分析,结果如图2中的(B),红色荧光(在图中为强度较高的曲线)和绿色荧光(在图中为强度较低的曲线)的强度起伏一致,为共定位。
6)计算L-AuNP@TMT的细胞膜共定位系数:通过Image J 中的ColocalizationFinder的共定位评估,其共定位系数Pearson=0.938,具有很好的共定位关系(一般认为大于0.85即共定位)。参见图2中的(C)。
实施例3
本实施例研究上述金纳米颗粒L-AuNP@TMT对其它肿瘤细胞膜的靶向性。具体如下:
1)金纳米颗粒L-AuNP@TMT对人肝癌细胞HepG2细胞膜的靶向性:
在共聚焦培养皿铺HepG2细胞,密度为3万/mm2。将金纳米颗粒L-AuNP@TMT分散PBS缓冲液中,浓度为70μg/mL,孵育HepG2细胞3分钟,将上清液吸走,PBS洗三次,加入细胞的完全培养基。激光共聚焦显微镜成像,用488 nm的激光作为激发光源,接受500-700 nm的荧光信号,结果如图3所示。从左到右依次是荧光场图片、明场图片和合并图片,上面一行是加了金纳米颗粒L-AuNP@TMT的实验结果,下面一行是没有加金纳米颗粒L-AuNP@TMT(对照)的实验结果。结果显示金纳米颗粒L-AuNP@TMT具有很好的HepG2细胞膜的靶向性。
2)金纳米颗粒L-AuNP@TMT对小鼠结肠癌细胞CT26细胞膜的靶向性:
在共聚焦培养皿铺CT26细胞,密度为3万/mm2。将金纳米颗粒L-AuNP@TMT分散PBS缓冲液中,浓度为70μg/mL,孵育CT26细胞3分钟,将上清液吸走,PBS洗三次,加入细胞的完全培养基。激光共聚焦显微镜成像,用488 nm的激光作为激发光源,接受500-700 nm的荧光信号,结果如图4所示。从左到右依次是荧光场图片、明场图片和合并图片。上面一行是加了金纳米颗粒L-AuNP@TMT的实验结果,下面一行是没有加金纳米颗粒L-AuNP@TMT(对照)的实验结果。结果显示金纳米颗粒L-AuNP@TMT具有很好的CT26细胞膜的靶向性。
实施例4
本实施例研究金纳米颗粒L-AuNP@TMT在正常细胞(非肿瘤细胞)人胚肾细胞293T细胞上的定位。实验方法同实施例3,仅所使用的细胞为293T。结果如图5所示,金纳米颗粒L-AuNP@TMT具有很好的293T细胞膜靶向性。
实施例5
本实施例研究实施例1制备的金纳米颗粒L-AuNP@TMT对细胞生长的影响。具体如下:
使用仪器:活细胞分析仪incucyte S3;
方法:使用含不同浓度L-AuNP@TMT的完全培养基孵育HepG2细胞,通过incucyteS3检测72小时内细胞数目的变化,判断L-AuNP@TMT的细胞安全性。结果见图6。
应当理解的是,对上述实施例所用试剂或原料的用量进行等比例扩大或者缩小后的技术方案,与上述实施例实质相同。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (2)
1.金纳米颗粒在作为细胞膜标记物中的应用,其特征在于,所述金纳米颗粒的制备方法包括:
(1)在黑暗条件下,将5-巯基-1,3,4-噻二唑-2-基硫代乙酸和氯金酸在碱性水溶液中混合,获得反应体系;所述反应体系中的碱为氢氧化钠或氢氧化钾,所述反应体系中5-巯基-1,3,4-噻二唑-2-基硫代乙酸、氯金酸和碱的摩尔质量比为2:1:10,所述反应体系中,氯金酸的浓度为1mM;
(2)以波长365 nm的紫外光照射所述反应体系,进行光化学还原反应至荧光强度不再升高;使用功率为4.5~5.5 W的紫外灯进行紫外光照射,所述光化学还原反应的反应时间为15-20分钟;所述光化学还原反应的反应温度为23~25℃;
所述金纳米颗粒的电镜尺寸为2.42±0.53 nm;所述金纳米颗粒的水动力学直径为4.75±1.63 nm;所述金纳米颗粒的最适激发波长为467 nm,最强发射波长为548 nm;所述金纳米颗粒发出绿色荧光;
作为细胞膜标记物中的应用方法为:
将所述金纳米颗粒先分散在PBS缓冲液中,浓度为70μg/mL,孵育细胞3分钟,将上清液吸走,PBS洗涤后,再加入细胞的完全培养基;采用激光共聚焦显微镜成像,用488 nm的激光作为激发光源,接受500-700 nm的荧光信号。
2.根据权利要求1所述的应用,其特征在于,所述金纳米颗粒的制备方法的步骤(2)中,使用光谱检测仪检测反应过程中的荧光强度。
