Abstract
本发明涉及iPS诱导定向分化成类肾器官的方法及应用。本发明针对GSK‑3α和GSK‑3β双靶点,设计了保守的二者共有的免疫多肽片段,以该多肽片段来制备获得了相应的单克隆抗体,所述抗体能够较好的抑制GSK‑3α/β的活性,克服了现有技术的CHIR99021化学药还对CDC2、ERK2等蛋白激酶也有抑制作用导致的对细胞产生额外的其他副作用,导致诱导的类肾器官功能会收到的影响。本发明的单抗可以用于制备诱导IPS诱导定向分化成类肾器官的方法,应用价值较高。

技术领域
本申请涉及生物领域,具体的涉及一种iPS诱导定向分化成类肾器官的方法及应用。
背景技术
多能干细胞(pluripotent stem cell,PSC)包括胚胎干细胞(ESC)和诱导多能干细胞(iPSC),由于PSC具有分化为体内所有类型细胞的潜能,PSC体外培养技术已被广泛应用于生物医学及生物科学领域中。近十年,随着体外3D培养技术的发展,类器官的构建已被认为是体内发育程序的体外培养系统。类器官可由PSC或祖细胞分化而成,拥有与体内器官相似的细胞类型和组织结构,并能还原该器官在体内的部分功能。类器官构建是一种不可替代的在体外模拟人体组织的方法,对疾病模型、器官再生、细胞发育和药效测试等研究都有重要意义。近年来,PSC体外诱导分化产生功能性的肾单位方法越来越成熟。其中肾脏类器官的出现是肾脏再生领域中一个突破性的进展。
肾脏类器官的形成分为3个主要阶段。干细胞首先分化成PSM,进一步分化成IM,IM再分化成MM和UB。第一阶段:干细胞分化成PSM。PSM是中胚层和内胚层的祖细胞群,可以由小鼠ESC经过激活素A(activinA)与BMP4诱导而成,其中activinA决定了内胚层向前体节中胚层分化而中胚层向后PSM分化。在小鼠和人的ESC分化过程中,经典的Wnt信号也可以作为PSM的诱导剂。第二阶段:PSM分化成IM。原肠胚形成后,中胚层进一步产生IM、傍轴中胚层(PM)和侧板中胚层(LPM)。过去的研究认为转录因子OSR1是IM甚至MM形成的标志最近研究发现OSR1在躯体中胚层延伸至LPM。PSM经BMP4/activinA,诱导后3d会有OSR1的表达,而IM其他标志物,比如PAX2和LHX1并无表达,这一结果提示第二阶段分化需要新的生长因子的参与,FGF信号也可能参与该过程,FGF8从PSM至后躯干中胚层阶段均有表达,而FGF9在IM和PM中有表达。在体外MM的生存需要培养基中加入FGF2/BMP7或FGF9。在FGF2或FGF9因子的刺激下,OSR1、PAX2和LHX1共同表达,其中PAX2的表达率超过80﹪,这些都是IM的分化特征[16]。FGF9诱导IM产生是浓度依赖性的(最佳浓度为200ng/ml)抑制LPM的分子标志FOXF1和OSR1的表达。因此,FGF信号可以有效并特异性地诱导PSM分化成IM。第三阶段:IM分化成MM和UB。在哺乳动物发育过程中,IM分化成肾脏、性腺和肾上腺,肾管是首先形成的结构,沿着肾管从头至尾依次排列着三对排泄器官(前肾、中肾、后肾),他们均发源于同一肾源性脊髓,只有后肾一直保留,并且成为动物永久性器官,后肾形成的关键是重要细胞组分之间相关诱导作用。MM通过表达GDNF促进UB生成,UB通过表达FGF9为MM的生存提供了必要条件,通过Wnt信号通路诱导肾单位的形成,每个肾单位延长或分段形成许多不同功能的细胞类型。研究证实维甲酸(RA)具有促进UB生长的功能,RA和BMP7能够诱导小鼠ESC向肾系分化,而FGF9可以维持小鼠肾单位祖细胞的状态。利用BMP4/activinA诱导人体胚胎干细胞(hESC)分化后,从第6天至第17天加入200ng/mlFGF9、50ng/mlBMP7和0.1μmol/LRA。整个诱导过程利用RT-PCR检测,结果显示:hESC逐渐从PSM中胚层(MIXL1,LHX1)经IM(OSR1、PAX2、LHX1)分化成MM(SIX2、WT1、GDNF、HOXD11)。HOXD11的表达提示了后肾的出现,重要的是,肾管(输尿管)上皮基因(C-RET、HOXB7)也同时被诱导表达,在诱导程序的第14天,ECAD+PAX2+上皮结构就已经出现,RA以浓度依赖的作用方式对这些早期上皮结构的形成起了促进作用,这些上皮结构和周围间质被证明有增殖的作用。在肾脏发育过程中,阳性表达SIX2和WT1的初始间质区被ECAD+输尿管上皮所包围,WT1+细胞的百分比在输尿管上皮出现后继续增加,提示WT1在肾单位祖细胞和分化更好的肾结构中均有表达,在诱导分化的第22天,足细胞标志基因(SYNPO、NPHS1和WT1)、肾小管标志基因(AQP1和SLC3A1)和集合管标志基因(AQP2和SCNNB1)都有明显表达。
申请人在研究发现,PSC在Wnt激动剂CHIR99021刺激下分化成PSM,这是类肾器官研究的关键,由于CHIR99021不仅是一种有效的、选择性、特异性较高的糖原合酶激酶-3(GSK-3α/β)抑制剂,其还对CDC2、ERK2等蛋白激酶也有抑制作用,导致CHIR99021的使用时还对细胞产生额外的其他副作用,导致类肾器官功能会导致一定的影响。因此,开发一种特异性的GSK-3抑制剂而不影响其他通路的活性是目前研究的关键。
发明内容
本发明开发了一种特异性的GSK-3抑制剂,即针对GSK-3α/β的单克隆抗体。
具体的,所述的GSK-3α/β-2A9单克隆抗体单克隆抗体其轻链可变区序列如SEQ IDNO:1所示,重链可变区序列如SEQ ID NO:2所示。
具体的,所述的GSK-3α/β-2A9单克隆抗体针对GSK-3α的IC50通过检测达到3nM,针对GSK-3β为4nM,效果较好。
进一步的,本发明的抗体还可以采用本领域常规的抗体可变区修饰,包括以下步骤:1)将抗体可变区进行氨基酸序列改造,以改变其免疫学特性;2)将改造后的抗体可变区进行糖基化修饰,以改变其血清半衰期、免疫原性及药代动力学特性;3)将修饰后的抗体可变区进行二硫键还原,以增加其稳定性及生物活性。
进一步的,本发明还提供了所述单克隆抗体的同源物。
“同源物”是在单抗的编码的核苷酸序列、多肽序列、功能或结构水平上相似的生物活性分子。同源物可以包括与参考序列具有一定百分比同一性的序列衍生物。因此,在一个实施方案中,同源或衍生序列共有至少70%的序列同一性。在一个具体的实施方案中,同源或衍生序列共有至少80%或85%的序列同一性。在一个具体的实施方案中,同源或衍生序列共有至少90%的序列同一性。在一个具体的实施方案中,同源或衍生序列共有至少95%的序列同一性。在一个更具体的实施方案中,同源或衍生序列共有至少50、55、60、65、70、75、85、86、87、88、89、90、91、92、93、94、95、96,97、98或99%的序列同一性。同源或衍生核酸序列也可以通过它们在高严格性杂交条件下保持与参考核酸序列结合的能力来定义。与参考分子具有结构或功能相似性的同源物可以是参考分子的化学衍生物。检测、产生和筛选结构和功能同源物以及衍生物的方法是本领域已知的。
具体的,本发明的抗体可以是小鼠腹水制备也可以是通过基因工程和细胞工程手段,实现抗体可变区的改造和糖基化修饰,操作简单,生产效率较高,可实现工业化生产。
进一步的,本发明提供了一种将人皮肤成纤维细胞诱导生成类肾器官的方法,所述方法包括使用本发明的GSK-3α/β-2A9单克隆抗体用于加速诱导成纤维细胞生成高活性的类肾器官的步骤。
进一步的,所述的成纤维细胞是从皮肤组织分离的,或者采用商业化手段购买获得的。
进一步的,本发明提供了一种促进成纤维细胞诱导分化成为类肾器官的培养基,其中所述培养基含有本发明的GSK-3α/β-2A9单克隆抗体单克隆抗体。
具体的,本发明还提供了将成纤维细胞重编程为iPS细胞的步骤。其中分化步骤中使用的是本领域常规的类肾器官诱导分化培养基。
一般而言,iPSC是通过宿主细胞中一种或多种“重编程因子”的瞬时表达(通常使用附加体载体引入的)而产生的。在这些条件下,少量的细胞被诱导成为iPSC(通常,此步骤的效率很低,因为没有使用选择标志物)。一旦细胞被“重新编程”并变得多能,它们就会失去附加体载体并利用内源基因产生因子。附加体载体的这样的损失导致被称为“零足迹”细胞的细胞。这是期望的,因为遗传修饰越少(尤其是在宿主细胞的基因组中)越好。因此,优选所得的hiPSC不具有永久的遗传修饰。
在一些实施方案中,使用单个重编程因子OCT4。在其他实施方案中,使用两个重编程因子OCT4和KLF4。在其他实施方案中,使用三个重编程因子OCT4、KLF4和SOX2。在其他实施方案中,使用四个重编程因子OCT4、KLF4、SOX2和c-Myc。在其他实施方案中,可以使用选自SOKMNLT:SOX2、OCT4(POU5F1)、KLF4、MYC、NANOG、LIN28和SV40L T抗原的5、6或7个重编程因子。
更进一步的,所述的重编程可以采用本领域较为成熟的重编程试剂盒转染获得。
进一步的,本发明提供一种将ips细胞分化为类肾器官的方法,具体的,所述方法包括将基质胶铺在细胞培养皿的表面当作细胞外基质,在37℃细胞培养箱中培养所述的iPSCs至80%~90%汇合度时进行传代,2~3d后汇合度增长至40%~50%,即可进入分化阶段。分化阶段一(第0天)细胞达到40%~50%汇合度,利用含1-50μmol GSK-3α/β-2A9单克隆抗体的APEL培养基培养至第4天,每2天更换上述培养基。第4天,利用含有200μg/LFGF9因子+1mg/L肝素的APEL培养基培养至第7天,每2天更换上述培养基。第7天,消化、离心并重悬细胞后进行细胞计数,按照每个类肾体大约5×105个细胞的量再次离心,弃去培养基后将细胞球放在Transwell过滤器上,利用含1-50μmol GSK-3α/β-2A9单克隆抗体的APEL培养基在37℃孵育lh。然后更换为含200μg/L FGF9+lmg/L肝素的APEL培养基培养至第12天,每2天更换培养基。去除FGF9(第12天)在第12天换为纯APEL培养基并培养至第25天,每2天更换培养基,收集获得类肾体。
有益效果
本发明针对GSK-3α和GSK-3β双靶点,设计了保守的二者共有的免疫多肽片段,以该多肽片段来制备获得了相应的单克隆抗体,所述抗体能够较好的抑制GSK-3α/β的活性,克服了现有技术的CHIR99021化学药还对CDC2、ERK2等蛋白激酶也有抑制作用导致的对细胞产生额外的其他副作用,导致诱导的类肾器官功能会收到的影响。本发明的单抗可以用于制备诱导IPS诱导定向分化成类肾器官的方法,应用价值较高。

筛选鉴定获得了其中在2个蛋白中均保守序列,通过抗原肽段分析,获得了6个较好的抗原表位,通过计算机分析和活性位点分析,最终筛选得到如下的抗原表位:分值=1.209长度=22,SIDVWSAGCVLAELLLGQPIFP,将所述多肽肽段进行人工合成,利用Sulfo-SMCC将多肽偶联于BSA载体蛋白,制备免疫原。
取8周龄BALB/c小鼠,腹腔注射免疫抗原(重组多肽加等体积弗氏完全佐剂乳化)进行免疫80μg/只,两周后改用弗氏不完全佐剂,抗原剂量为100μg/只。三免、四免同前。摘取脾脏3d前腹腔注射不加佐剂抗原100μg/只加强免疫1次。细胞融合、筛选与克隆参照本领域常规的方法进行,SP2/0和免疫鼠脾细胞融合后,共接种192孔,9d后有175孔有杂交瘤细胞生长。用多肽抗原包被酶标板,常规间接ELISA方法检测杂交瘤细胞生长孔上清液,阳性孔为38孔。用有限稀释法对分泌抗体阳性的细胞生长孔进行亚克隆,经过4次亚克隆至所有单克隆孔上清抗体阳性率为100%,获得1株阳性反应最强稳定性最好的GSK-3α/β单克隆抗体的杂交瘤细胞株,命名为GSK-3α/β-2A9。
将GSK-3α/β-2A9杂交瘤细胞扩大培养,腹腔注射BABL/c小鼠每只约1×106个/0.2mL。无菌收集腹水,用饱和硫酸铵盐析法纯化腹水单抗,分装后置-80℃保存备用。
Westernblot分析:将多肽肽段和BSA采用常规处理上样。经SDS-PAGE电泳分离。半干转膜后加入封闭液4℃轻摇封闭过夜。在杂交袋中加入纯化单抗,以HRP-羊抗鼠IgG为二抗,用DAB显色试剂盒对膜进行显色,进行Western blot鉴定。结果如图1所示。
从图1的结果可以看出,本发明的单克隆抗体具有较好的特异性,只能与多肽肽段反应,而不与BSA蛋白反应。
实施例2 GSK-3α/β-2A9单克隆抗体特性鉴定
亲和力测定公式:K=([Ag′]/[Ag]t-1)/2(n[Ab′]-[Ab]t),其中:[Ab′]表示抗原浓度为[Ag′]时OD=1/2ODmax对应的抗体摩尔浓度,[Ab]t表示抗原浓度为[Ag]t时OD=1/2ODmax对应的抗体摩尔浓度,n为抗原[Ag′]与[Ag]t间的稀释倍数。结果如表1所示。

从表1的结果可以看出,本发明的单克隆抗体具有较强的亲和力,活性较好,适宜用于生物学实验。
委托快序生物进行抗体可变区序列分析,结果显示,所述GSK-3α/β-2A9单克隆抗体其轻链可变区序列如SEQ ID NO:1所示,重链可变区序列如SEQ ID NO:2所示。
实施例3皮肤成纤维细胞制备iPS细胞
本实施例的方法是申请人前期构建的成熟的方法。具体的,取手术后分离的儿童包皮皮肤约0.3cm×0.3cm,将其置于含有青霉素(100kU/L)和链霉素(100mg/L)的HBSS中反复漂洗后,去除皮下脂肪。在60mm培养皿中用眼科剪将皮肤剪成1mm2大小,置于含有青霉素(100kU/L)和链霉素(100mg/L)的DMEM/F12(1:1)液3ml37℃下置于体积分数为5%的CO2、95%空气、饱和湿度孵箱中过夜。次日挑除汗腺腺体,待成纤维细胞生长明显时,吸弃旧培养液,用无Na+、Mg2+的HBSS洗2次,加入含有质量分数为0.25%胰酶和0.02%乙二胺四乙酸(EDTA)的D-Hank消化液约1ml37℃下置于5%CO2、95%空气、饱和湿度条件下孵育2min,加入含10%胎牛血清的DMEM约2ml终止消化,1000r/min离心6min并收集细胞,用含胎牛血清(10%)、青霉素(100kU/L)和链霉素(100mg/L)的DMEM分散细胞,细胞以1x104/ml接种4ml至25cm²的培养瓶中,在37℃、5%CO2、95%空气、饱和湿度条件下培养,经过连续5次传代培养后,可得到纯化的成纤维细胞。去少量细胞进行HE染色发现,细胞为长梭形,细胞核为淡蓝色,细胞浆为粉红色,通过染色体核型分析显示为46条染色体,鉴定制备获得了较为纯净的成纤维细胞。
将分离培养的皮肤成纤维细胞调整细胞数为1×106个,100μl电转缓冲液中加入3μlEpi5TMEpisomaliPSC重编程试剂盒中的重编程因子,采用常规电转染细胞按2ml/孔接种于基质胶包被的6孔板中,置于培养箱中培养。并使用ReproTeSRTM重编程培养基隔天换液,转染后第8天,将细胞消化后重新接种在5代小鼠成纤维细胞制作的饲养层上,第18天,观察iPSCs可见典型的克隆,采用试剂盒检测干细胞多能性基因发现OCT4、SOX2、NANOG、KLF4和LIN28均阳性表达,表明制备获得了iPS细胞。将iPS克隆重新接种于基质胶包被的6孔板中,置于培养箱中培养,使用mTeSRTM培养基继续培养,筛选纯化后备用。
进一步的,为了检验iPS细胞的活性,将制备的iPS细胞体外悬浮培养形成EB,裂解细胞提取RNA,用于RT-PCR检测EB细胞基因表达量,结果显示,EB细胞的胚层分化基因外胚层Nestin、中胚层Eomes、内胚层AFP的表达水平均显著高于对照组iPS细胞5倍以上(P<0.01),实验结果表明所建立的人iPS细胞具有体外分化能力。
实施例4 iPS细胞诱导分化为类肾器官
实验组:将基质胶铺在细胞培养皿的表面当作细胞外基质,在37℃细胞培养箱中培养实施例3所述的iPSCs至80%~90%汇合度时进行传代,2~3d后汇合度增长至40%~50%,即可进入分化阶段。分化阶段一(第0天)细胞达到40%~50%汇合度,利用含8μmolGSK-3α/β-2A9单克隆抗体的APEL培养基培养至第4天,每2天更换上述培养基。第4天,利用含有200μg/L FGF9因子+1mg/L肝素的APEL培养基培养至第7天,每2天更换上述培养基。第7天,消化、离心并重悬细胞后进行细胞计数,按照每个类肾体大约5×105个细胞的量再次离心,弃去培养基后将细胞球放在Transwell过滤器上,利用含5μmol GSK-3α/β-2A9单克隆抗体的APEL培养基在37℃孵育lh。然后更换为含200μg/L FGF9+lmg/L肝素的APEL培养基培养至第12天,每2天更换培养基。去除FGF9(第12天)在第12天换为纯APEL培养基并培养至第25天,每2天更换培养基,收集获得类肾体。

取第7天的类肾体使用4%多聚甲醛固定、0.1%TritonX-100透化、5%牛血清白蛋白封闭、一抗4℃孵育过夜、二抗室温孵育lh,然后加入4,6-二氨基一2-苯基吲哚,利用激光共聚焦显微镜观察。结果如图2所示,从图2可以看出,实验组采用单克隆抗体诱导制备的形状较为规则,饱满,对照组的形状稍微不规则,不饱满,这也说明采用靶标特异性的单克隆抗体能够减少对细胞的副作用影响,效果较好。
诱导25天的类肾体,实验组已经看到明显的管状结构分化,其边缘部位可以明显的比对照组更多的高折光的管状结构轮廓,这说明,采用单抗诱导的方法比采用多靶点的化合物具有更好的诱导效果。
同时,采用Western bolt进行了E-cad和SIX2二种种肾脏组织标志物的鉴定,以GAPDH作为相对表达量。鉴定的分别是第1天,第7天和第25天的类肾体,结果如表2所示。

在第1天,基本检测不到E-cad和SIX2的表达。在第7天和第25天E-cad和SIX2相对表达量均显著的得到表达,表明ips细胞成功的向类肾器官分化。并且与第7天相比25天的结果也是显著提高差异显著,P<0.05。与对照组相比,采用单克隆抗体特异性诱导的结果值也比采用化合物诱导的效果要好,说明单抗能够克服化合物的其它副作用,效果显著。
尽管以上结合附图对本发明的实施方案进行了描述,但本发明并不局限 于上述的具体实施方案和应用领域,上述的具体实施方案仅仅是示意性的、 指导性的,而不是限制性的。本领域的普通技术人员在本说明书的启示下和 在不脱离本发明权利要求所保护的范围的情况下,还可以做出很多种的形 式,这些均属于本发明保护之列。
Claims (3)
1.一种特异性抑制GSK-3α和GSK-3β的单克隆抗体,所述单克隆抗体其轻链可变区序列如SEQ ID NO:1所示,重链可变区序列如SEQ ID NO:2所示。
2.如权利要求1所述的单克隆抗体在制备用于促进ips细胞分化为类肾器官的培养基中的用途;其中所述的ips细胞是从人皮肤成纤维细胞重编程后制备获得的;所述的重编程是采用Epi5TM Episomal iPSC 重编程试剂盒实现。
3.一种将ips细胞分化为类肾器官的方法,所述方法包括将基质胶铺在细胞培养皿的表面当作细胞外基质,在37℃细胞培养箱中培养iPS至80%~90%汇合度时进行传代,2~3d后汇合度增长至40%~50%,即可进入分化阶段,分化阶段一第0天细胞达到40%~50%汇合度,利用含1-50μmol GSK-3α/β-2A9单克隆抗体的APEL培养基培养至第4天,每2天更换上述培养基;第4天,利用含有200μg/L FGF9因子+1mg/L肝素的APEL培养基培养至第7天,每2天更换上述培养基,第7天,消化、离心并重悬细胞后进行细胞计数,按照每个类肾体大约5×105个细胞的量再次离心,弃去培养基后将细胞球放在Transwell过滤器上,利用含1-50μmol GSK-3α/β-2A9单克隆抗体的APEL培养基在37℃孵育lh;然后更换为含200μg/L FGF9+lmg/L肝素的APEL培养基培养至第12天,每2天更换培养基;去除FGF9在第12天换为纯APEL培养基并培养至第25天,每2天更换培养基,收集获得类肾体;其中所述的单克隆抗体是权利要求1中所述的抗体,所述的iPS细胞是是从人皮肤成纤维细胞重编程后制备获得的;所述的重编程是采用Epi5TM Episomal iPSC 重编程试剂盒实现。
