技术领域
本发明涉及人血清白蛋白-硫氧还蛋白融合体的修饰体,进一步详细而言,涉及通过修饰而提高了硫氧还蛋白活性以及其药理作用的稳定性及安全性的融合体。
背景技术
硫氧还蛋白(Trx)于1964年作为大肠杆菌的核糖核苷酸还原酶的辅酶被发现(非专利文献1)。另外,已知在20世纪80年代后期,淀井等人通过基因克隆发现人Trx与成人T细胞白血病源性因子(ATL-derived factor:ADF)为同源体,其中,ADF被报告为白细胞介素2受体α链的诱导因子(非专利文献2)。
Trx在人中由第9号染色体的TXN基因编码为分子量约为14,000的蛋白质,在所有生物物种的各种细胞中表达。对于人而言,Trx基因的功能丧失型突变体在胚胎发生早期的四细胞期的时间点已经致死。Trx具有-Cys-Gly-Pro-Cys-这一活性部位,由紫外线、放射线、氧化剂、病毒感染、缺血再灌注损伤、施予抗癌药等各种因素引起的氧化应激而被诱导表达,去除细胞内的活性氧类,发挥抗氧化作用(非专利文献1及3)。另外,具有细胞因子、核因子-κB(nuclear factor-κB:NF-κB)、凋亡信号调节激酶1(apoptotic signal-regulating kinase-1:ASK-1)等转录因子的活性控制,以及抑制抗凋亡作用、抑制炎症性细胞因子、人巨噬细胞移动抑制因子(Human Macrophage Migration Inhibitory Factor:MIF)的抗炎作用等药理活性(非专利文献4~7)。
另外,人血清白蛋白(HSA)是在肝脏合成的分子量为66,000的蛋白质,占血清蛋白的约50~65%。HSA除了血液的浸透压的保持、pH缓冲作用、向各组织供给氨基酸、抗氧化作用以外,还具有结合脂肪酸、胆红素、无机离子、药物等外来物质的功能。
该HSA的血液中的半衰期为14~21天,与HSA结合的低分子化合物不会被器官摄取,因此能够不被代谢·排泄而在血液中循环。如上所述,Trx虽然表现出各种药理作用,但是在作为药物施予时,迅速在肾小球滤过,因此血液中的半衰期短至约1小时,因此,为了维持血药浓度而得到治疗效果,需要定速静脉内施予、频繁施予。
此前,发明人中的一部分报告了使用白蛋白融合技术(专利文献1)使HSA融合于Trx(HSA-Trx),由此,能够使血液中的半衰期达到1小时至10小时,从而能够延长血液中的滞留时间(非专利文献8)。另外,报告了该HSA-Trx在各种病况模型中的效果。例如,报告了在顺铂肾病(非专利文献9)、对乙酰氨基酚诱发的肝损伤(非专利文献10)、博来霉素诱发的肺纤维化(非专利文献11)及流感病毒诱发的肺损伤(非专利文献12)等病况模型中,经单次或低频率的施予显示出有效的治疗效果。
Trx是由105个氨基酸残基形成的蛋白质,具有被称为硫氧还蛋白折叠(thioredoxin fold)的结构,具有催化二硫(S-S)键的形成、异构化的酶中共通包含的α-螺旋和β-折叠这二者。并且,Trx为4个β-折叠夹在2个α-螺旋中的结构。
Trx具有5个半胱氨酸残基,且均为游离巯(SH)基。自N端起第32位和第35位的Cys32和Cys35是Trx的氧化还原活性的中心,并且是ASK-1结合的调整部位。其余的自N端起第62位、第69位及第73位的Cys62、Cys69及Cys73是担负别构效应的部位,Cys69为亚硝基化部位,Cys62和Cys69附近是硫氧还蛋白还原酶(TrxR)的结合部位,当Cys62与Cys69进行二硫键合时,TrxR不能结合。Cys73是谷胱甘肽加成部位,并且是与Trx的二聚体化相关的部位(非专利文献13)。据报告,由丝氨酸取代了Csy32及Cys35的修饰体表现出迅速向细胞内移动的性质,具有高的细胞增殖抑制活性(专利文献2)。另外,还有报告称,若由丝氨酸取代Cys62、Cys69及Cys73,则与野生型相比,Trx活性(还原活性)下降(非专利文献14)。有报告称,Trx具有的活性中心的半胱氨酸残基不进行修饰、将除此以外的半胱氨酸基中的至少1个或全部取代为其他的氨基酸残基,从而使其在非还原条件下稳定(专利文献3)。
另一方面,HSA是由585个氨基酸残基形成的单纯蛋白质,由结构域I(1~195残基)、结构域II(196~383残基)及结构域III这3者折叠的相同区域构成,并且,它们分别具有亚结构域A及B,IIA(SiteI)及IIIA(SiteII)是多数药物进行结合的部位,在N端侧有金属进行结合的部位。在分子内,具有17对二硫(S-S)键,仅自N端起第34位的半胱氨酸残基(Cys34)具有游离巯(SH)基。该游离巯基残基为游离状态的HSA称为还原型HSA、形成二硫键的称为氧化型HSA,氧化型HSA是各种疾病的标志物。另外,Zhao等人发现,干扰素-α2b(IFN-alpha2b)与HSA的融合蛋白(IFN-alpha2b-HSA)通过由丝氨酸取代HSA的Cys34能够抑制凝集体的形成,并且热稳定性也增加(非专利文献15)。

发明所要解决的课题
根据本申请发明人的见解,观察到血清白蛋白-硫氧还蛋白融合体(非修饰型)得不到稳定的药效,这作为医药品而言,具有品质、安全性上的偏差,很难说是优选的。
本申请发明人发现,本次通过在血清白蛋白-硫氧还蛋白融合体的硫氧还蛋白的序列中,由其它氨基酸取代至少自N端起第73位的半胱氨酸,从而显著提高硫氧还蛋白的活性,并且得到稳定的药效。并且,还得到了其免疫原性减少、安全性优异的见解。本发明是基于上述见解的发明。
因此,本发明的目的在于提供活性得到提高、且活性稳定的血清白蛋白-硫氧还蛋白融合体。另外,本发明的目的在于提供安全性优异的血清白蛋白-硫氧还蛋白融合体。

本说明书中,人血清白蛋白-硫氧还蛋白融合体是指人血清白蛋白与硫氧还蛋白伴随使硫氧还蛋白的血液中的半衰期延长的作用,而使人血清白蛋白融合(fusion)或接合(conjugate)。根据本发明的一个方式,人血清白蛋白与硫氧还蛋白通过接头结合,例如通过多肽连结而成。
进一步,本发明中的硫氧还蛋白是至少其氨基酸序列中的自N端起第73位的半胱氨酸被其它氨基酸取代而成的修饰体。硫氧还蛋白在其序列中具有自N端起第32位、第35位、第62位、第69位及第73位共计5个半胱氨酸残基。本发明中,其中至少第73位的半胱氨酸被其它氨基酸取代。本发明中,取代该半胱氨酸的其它氨基酸没有特别限定,根据本发明的优选方式,可举出中性氨基酸,更优选由丙氨酸、丝氨酸、或甘氨酸取代。本发明中,最优选由丙氨酸取代。另外,进行取代的氨基酸也可以是其结构的一部分被修饰等的氨基酸。

根据本发明的一个优选方式,经修饰的人血清白蛋白-硫氧还蛋白融合体具有以下氨基酸序列。

另外,根据本发明的其它方式,本发明的经修饰的人血清白蛋白-硫氧还蛋白融合体具备的“人血清白蛋白”和“硫氧还蛋白”的数量不限定于各一个,也可以是多个。另外,只要它们伴随使硫氧还蛋白的血液中的半衰期延长的作用而结合,则其结合位置也没有限定。例如可举出将Trx连接于HSA的C端而成的HSA-Trx,连接于N端而成的Trx-HSA,连接于N端、C端这二者而成的Trx-HSA-Trx,将2个Trx连接于C端而成的HSA-Trx-Trx,同样地将Trx连接于所连接的2个HSA而成的HSA-HSA-Trx、Trx-HSA-HSA、Trx-HSA-HSA-Trx、HSA-HSA-Trx-Trx等修饰体。
本发明的结合“人血清白蛋白”与“硫氧还蛋白融合体”的接头的种类及长度没有特别限定,优选通过如上所述的多肽连结的接头,其优选的长度为0~40氨基酸残基左右。
经修饰的人血清白蛋白-硫氧还蛋白融合体的制备
本发明的经修饰的人血清白蛋白-硫氧还蛋白融合体能够按照已知的方法制备。例如,能够首先通过专利文献1中记载的白蛋白融合技术使人血清白蛋白融合于硫氧还蛋白。在此,通过将所使用的各序列的半胱氨酸的密码子改变为其它氨基酸的密码子,能够制成修饰体。
经修饰的人血清白蛋白-硫氧还蛋白融合体的用途
对于本发明的经修饰的血清白蛋白-硫氧还蛋白融合体而言,通过其修饰,首先能够提高硫氧还蛋白自身的活性,并且具有能够稳定发挥其活性的优点。另外,本发明的经修饰的血清白蛋白-硫氧还蛋白融合体的免疫原性减少,在安全性上也是优选的。特别是本发明的经修饰的血清白蛋白-硫氧还蛋白融合体与非修饰血清白蛋白-硫氧还蛋白融合体相比,不形成被认为是其二聚体的凝集体,从而被认为是由其导致的副反应的抗体诱导减少,并且,在几乎观察不到抗体这一点上也是有利的。抗体诱导的减少与不发生过敏反应有关。
伴随以上的有利效果,本发明的经修饰的血清白蛋白-硫氧还蛋白融合体还被期待目前已知的硫氧还蛋白活性带来的用途、特别是目前已知的预防或治疗效果,此外,这些治疗效果能够有效地用于已阐明的疾病的预防或治疗。作为这样的用途,可举出能够通过抗氧化作用、细胞因子转录因子的活性控制、抗凋亡及抗炎作用等药理活性治疗的疾病(例如,酒精性肝病、非酒精性肝病、对乙酰氨基酚肝炎、急性重型肝炎(fulminanthepatitis)、肝硬化、急性肾损伤、慢性肾损伤、顺铂肾病、横纹肌溶解症、造影剂肾病、慢性肾脏病、糖尿病性肾脏病、器官纤维化、自身免疫性疾病、阿尔茨海默病、帕金森病、周围神经病(peripheral neuropathy)、脑梗死、动脉硬化、心肌梗死、心力衰竭、心肌梗死、急性呼吸窘迫综合征、慢性阻塞性肺疾病、过敏性肺炎、流感病毒肺炎、支气管哮喘等)。
因此,根据本发明的一个方式,提供包含本发明的经修饰的血清白蛋白-硫氧还蛋白融合体而成的医药组合物、及本发明的经修饰的血清白蛋白-硫氧还蛋白融合体用于制造医药组合物的用途。另外,根据本发明的其它方式,提供包含本发明的经修饰的血清白蛋白-硫氧还蛋白融合体的医药组合物及本发明的经修饰的血清白蛋白-硫氧还蛋白融合体用于能够通过抗氧化作用、细胞因子转录因子的活性控制、抗凋亡及抗炎作用的药理活性治疗的疾病的预防或治疗的用途。
实施例
以下,通过实施例详细地说明本发明,但本发明不受实施例的任何限定。
实施例1:对半胱氨酸进行取代而得的修饰体的活性
分别制作HSA-Trx的HSA序列的第34位的半胱氨酸,以及Trx的第62位、第69位、第73位的半胱氨酸中的1个、2个、3个及全部(4个)被丙氨酸取代的经修饰的HSA-Trx。构建在HSA-Trx的CHO细胞表达质粒中、将编码上述半胱氨酸的序列改变为编码丙氨酸的密码子的质粒。通过将质粒转染至CHO细胞从而得到产生经修饰的HSA-Trx的培养上清液。转染、培养使用ExpiCHO表达系统(ThermoFisher)并按照产品说明书实施。使用捕获选择人白蛋白亲和基质(Capture Select Human Albumin Affinity Matrix)(Thermo Scientific)、CaptoMMC(GE Healthcare)由所得到的培养上清液纯化修饰HSA-Trx,并使用Amicon Ultra-1530K(Millipore)浓缩,将缓冲液更换为PBS。使用硫氧还蛋白检测试剂盒(IMCO)按照产品说明书测定它们的Trx的活性,算出相对于试剂盒附属的重组人Trx(rTrx)的比活性(以rTrx为100%)。结果如图1所示。
相对于rTrx而言,对C62进行取代而得的修饰体为100%、对C69进行取代而得的修饰体为101%、对C73进行取代而得的修饰体为189%、对C62和C69进行取代而得的修饰体为102%、对C62和C73进行取代而得的修饰体为204%、对C69和C73进行取代而得的修饰体为186%、对C62和C69和C73进行取代而得的修饰体为189%、对H34和C62进行取代而得的修饰体为103%、对H34和C69进行取代而得的修饰体为97%、对H34和C73进行取代而得的修饰体为189%、对H34和C62和C69进行取代而得的修饰体为87%、对H34和C62和C73进行取代而得的修饰体为164%、对H34和C69和C73进行取代而得的修饰体为184%、对H34和C62和C69和C73进行取代而得的修饰体为207%,显示出几乎同等或更高的活性。特别是对C73的进行取代而得的修饰体全部显示出164%~207%的高值。由以上结果可知,H34、C62、C69、C73成为丙氨酸的取代与rTrx相比,Trx活性为同等或同等以上,并且,当进行包含C73的取代时,活性提高。
实施例2:C73修饰体相对于非修饰体(HT)的比活性
测定C73修饰体和HT的Trx活性,算出C73修饰体相对于HT的比活性(以HT为100%)。测定与实施例1同样地实施。结果如图2所示。
相对于HT而言,C73修饰体为175%、C62和C73的修饰体为177%、C69和C73的修饰体为141%、C62和C69和C73的修饰体为189%、H34和C73修饰体为159%、H34和C62和C73的修饰体为134%、H34和C69和C73的修饰体为142%、H34和C62和C69和C73的修饰体为186%,C73修饰体相对于HT均表现出134~189%的高值。由以上可知,包含C73的半胱氨酸的取代与未进行取代的HT相比,Trx活性提高。
实施例3:修饰体的电泳
为了阐明对半胱氨酸进行取代而得的修饰体的分子形状而进行电泳。将100ng样品混合于进样液(由十二烷基硫酸钠、甘油、溴酚蓝、蒸馏水构成,pH6.8),于100℃煮沸3分钟后,注入至10%电泳用凝胶(ATTO)中,使用Tricine电泳缓冲液(由Tris、Tricine、十二烷基硫酸钠、蒸馏水构成,约pH8.6),以25mA电泳约55分钟。用固定液(由甲醇、乙酸、蒸馏水构成)固定凝胶约20分钟后,使用银染KANTOIII(Silver Stain KANTOIII)(东京化学)按照产品说明书进行银染。结果如图3所示。
图中,双箭头为HSA-Trx的分子大小,单一箭头为凝集体的分子大小。在作为非取代体的HT中,除了HSA-Trx的分子大小以外,还观察到大量凝集体。并且,在H34、C62、C69、C62和C69的取代体中,与HT同样地观察到凝集体。与之相对,在作为包含C73的取代体的C73、C62和C73、C69和C73、C62和C69和C73、H34和C73、H34和C62和C73、H34和C69和C73、H34和C62和C69和C73中,确认到凝集体的减少,在H34和C62和C69和C73中,几乎未确认到凝集体的条带。由以上结果可知,包含C73的半胱氨酸的取代使得凝集体减少,并且,当对所有半胱氨酸进行取代时,几乎不能确认到凝集体。
实施例4:取代成丝氨酸、甘氨酸而得的修饰体
通过与实施例1同样的方法制作将H34、C62、C69及C73这4个半胱氨酸取代成丝氨酸或甘氨酸而得的修饰HSA-Trx(以下,将取代成丝氨酸而得的修饰体简称为H34T3Ser、将取代成甘氨酸而得的修饰体简称为H34T3Gly、将取代成丙氨酸而得的修饰体简称为H34T3)。按照按照实施例1及2的方法对它们进行电泳及Trx活性的测定。结果如图4及图5所示。
图中,图4中的a表示取代成丝氨酸的修饰体、b表示取代成甘氨酸的修饰体的电泳图像。均与H34T3同样地未观察到凝集体的条带,因此氨基酸取代使凝集体减少。图5示出Trx活性的测定结果,相对于HT而言,H34T3的比活性为145%、H34T3Ser的比活性为115%、H34T3Gly的比活性为94%,几乎为同等或同等以上。相对于HSA-Trx的Trx活性而言,将半胱氨酸取代成丙氨酸、丝氨酸、或甘氨酸而得的修饰体的Trx活性的研究结果表明,丙氨酸取代的活性上升最大。
实施例5:修饰体的安全性
为了确认修饰体的安全性,研究了对小鼠进行2次施予时的过敏反应及抗体诱导的有无。
动物使用ICR小鼠(雄性、6周龄、Slc),动物数为各组4只,进行HT与经修饰的H34T3的比较。以1周的间隔,以0.1μmol/kg通过尾静脉对HT、H34T3进行2次施予。从第2次施予后即刻至第120分钟为止,根据下表1的评分评价过敏反应。

对于抗体而言,在第1次施予后的第6天通过尾静脉进行局部采血,于37℃进行30分钟血液凝固反应后,以3500rpm离心10分钟,得到血清试样。在进行ELISA测定前将血清试样保存于-80℃。通过以下方法实施ELISA。用固相化溶液(0.1mol/L碳酸盐缓冲液pH9.6)将H34T3配制成1μg/mL浓度,以100μL/孔添加于ELISA板,利用自然吸附于4℃静置过夜进行固相化。用0.05%吐温20PBS溶液(以下为PBST)清洗3次后,以250μL/孔添加封闭液(用PBST将Block Ace(KAC)原液稀释成1/4浓度),于室温反应2小时。用PBST清洗3次后,用PBST/BlockAce溶液(用PBST将Block Ace(KAC)原液稀释成1/10浓度)将试样血清稀释200倍,以100μL/孔进行添加,于37℃反应2小时。用PBST清洗3次后,用PBST/Block Ace溶液将抗小鼠IgG-HRP(ZyMax、81-6720)稀释2000倍,以100μL/孔进行添加,于37℃反应1.5小时。用PBST清洗2次、用蒸馏水清洗2次后,以100μL/孔稀释TMB+(Dako),于室温反应30分钟。以100μL/孔添加1N硫酸使反应停止,测定450-650nm的吸光度(光学浓度:OD值)。
结果如图6所示。在HT中,确认到4例中的3例引起了中度以上(比评分3严重)的过敏反应,2例中有明显的抗体诱导。在H34T3中未确认到过敏反应,并且也几乎未确认到抗体诱导。由以上结果可以认为,进行了半胱氨酸的氨基酸取代的修饰体对免疫应答的安全性提高。
实施例6:修饰体对肝疾病的有效性
使用伴刀豆球蛋白A(Concanavalin A)的肝炎诱导模型确认修饰体的有效性,然后进行HT与经修饰的H34T3的比较。动物使用C57BL/6小鼠,以1组2~4只进行研究。在修饰体的有效性的研究中,将检体以0.05μmol/kg对尾静脉进行施予。在HT与H34T3的比较研究中,以0.4μmol/kg对尾静脉进行施予。在检体施予后5~15分钟之间,将伴刀豆球蛋白A(WAKO)以12mg/kg对尾静脉进行施予。在施予伴刀豆球蛋白A后的第12小时进行采血,得到血清,使用转氨酶CII Test Wako(FUJIFILM),按照产品说明书测定丙氨酸氨基转移酶(ALT)。其结果确认到通过伴刀豆球蛋白A施予使ALT上升。在修饰体有效性的研究中,模型的ALT值为565IU/mL(正常小鼠(n=1)为14IU/mL),相对于此,对C73进行取代而得的修饰体为79IU/mL,H34、C73修饰体为37IU/mL,C62、C69、C73修饰体为141IU/mL,H34、C62、C69、C73修饰体为102IU/mL,C62、C73修饰体为54IU/mL,H34、C62、C73修饰体为57IU/mL,C69、C73修饰体为47IU/mL,H34、C69、C73修饰体为28IU/mL,从而抑制了肝炎的诱导(图7)。在HT与H34T3的比较研究中,模型的ALT值为4241IU/mL,相对于此,HT为2681IU/mL,H34T3为1858IU/mL,从而HT、H34T3均抑制了肝炎诱导,而H34T3的抑制更强(图8)。由以上结果可知,进行了包含C73的半胱氨酸的取代而得的修饰体抑制伴刀豆球蛋白A诱导的急性肝炎,并且,修饰体比非修饰体有更强的抑制。
实施例7:修饰体对肾疾病的有效性(其1)
使用顺铂诱导急性肾损伤(AKI)模型进行评价。动物使用ICR小鼠(雄性、6周龄、Slc),动物数为各组3只,进行非修饰体的HT与经修饰的H34T3的比较。将HT、H34T3以0.25μmol/kg向尾静脉施予,然后,通过将顺铂(顺铂点滴静注10mg“MARUKO”、Yakult)以15mg/kg向尾静脉内施予来诱导AKI。在诱导AKI后第96小时,使用Urea Nitrogen,Detection Kit,Colorimetric,DetectX(Arbor Assays),按照产品说明书测定血清尿素氮(BUN)。
结果如图9所示。确认到顺铂施予导致BUN上升(图中模型、BUN的平均值为161mg/dL,正常小鼠(n=1)为26mg/dL)。HT施予组为186mg/dL,未确认到诱导AKI的抑制效果。H34T3施予组为90mg/dL,确认到抑制效果。通过以上结果能够得到下述结论:将半胱氨酸取代成其它氨基酸而得的经修饰的HSA-Trx对肾疾病显示出药效。
实施例8:修饰体对肾疾病的有效性(其2)
使用缺血再灌注诱导的AKI模型进行评价。动物使用C57BL/6小鼠(雄性、7周龄以上、Slc),动物数为各组2~3只,进行作为非修饰体的THT(在HT的N端连接Trx)与经修饰的H34T3的比较。在诱导AKI、施予受验物质1周前,在麻醉下取出右肾。将THT、H34T3以0.1μmol/kg向尾静脉施予。在麻醉下进行开腹,使左肾缺血,于33分钟后进行再灌注,从而诱导AKI。在诱导AKI后第24小时,与实施例7同样地测定BUN。研究实施2次,其总结果如图10所示。
通过进行缺血再灌注,确认到相对于正常小鼠的BUN值为29mg/dL而言,平均值为141mg/dL(模型),BUN上升。作为非修饰体的THT的BUN值为147mg/dL,未确认到AKI的抑制效果,而H34T3施予组为58mg/dL,确认到抑制效果。通过以上结果能够得到下述结论:与实施例7同样,将半胱氨酸取代成其它氨基酸而得的经修饰的HSA-Trx对急性肾损伤显示出药效。
实施例9:修饰体对肺疾病的有效性
使用博来霉素诱发的小鼠肺纤维化模型研究修饰体对急性肺炎的效果。动物使用ICR小鼠(雄、6周龄、Slc),动物数为各组4只,进行非修饰体的HT与经修饰的H34T3的比较。肺纤维化通过在麻醉下向气管内施予1.2mg/kg博来霉素(日本化药)来诱导。在施予博来霉素后第1、3、5及8天,将HT、H34T3以0.4μmol/kg向尾静脉施予。评价通过在施予博来霉素后第18天测定左肺重量及羟脯氨酸的量进行。羟脯氨酸的量的测定按照以下步骤实施。向试样中添加蒸馏水1mL和50w/v%TCA 125μL,进行匀浆化(homogenize),于4℃静置20分钟。以10,000rpm、4℃离心10分钟,除去上清液。添加500μL盐酸(Wako),轻拍使试样沉渣漂浮后,于110℃保温一个昼夜(约18小时)。将干燥的试样沉渣用蒸馏水1mL悬浮。使用L-羟脯氨酸(L-Hydroxyproline)(Wako)作为标准物质,配制0、0.0156、0.03125、0.0625、0.125、0.25mg/mL用于制作标准曲线。将200μL的试样、标准物质分装于试管,添加500μL的氯胺-T溶液(由氯胺-T、乙酸钠、2-丙醇、蒸馏水构成)。于室温静置20分钟后,添加500μL的Ehnoh’s试剂(由4-(二甲基氨基)苯甲醛(4-(Dimethylamino)benzaldehyde)、2-丙醇、高氯酸构成)。于65℃保温15分钟,将100μL分装于96孔板,测定波长540nm的吸光度。
结果如图11及12所示。在通过施予博来霉素诱导的肺纤维化中,相对于正常小鼠平均肺重量为53mg而言,肺重量为76mg(模型组),肺重量上升。HT施予组为89mg,不仅没有抑制效果,反而增强。与之相对,H34T3施予组为56.8mg,几乎未确认到上升。相对于正常小鼠的羟脯氨酸的量为6.1mg/mL而言,模型组为8.7mg/mL,确认到伴随纤维化的上升。HT施予组为10.5mg/mL,不仅没有抑制效果,反而增强。与之相对,H34T3施予组为7.1mg/mL,几乎未确认到上升。通过以上结果能够得到下述结论:将半胱氨酸取代成其它氨基酸而得的经修饰的HSA-Trx对急性肺炎显示出药效。

所述硫氧还蛋白是至少其氨基酸序列中的自N端起第73位或与其等同的位置的半胱氨酸被其它氨基酸取代而成的修饰体。
2.如权利要求1所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,所述硫氧还蛋白是其氨基酸序列中的自N端起第62位或与其等同的位置的半胱氨酸及/或第69位或与其等同的位置的半胱氨酸进一步被其它氨基酸取代而成的。
3.如权利要求1或2所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,所述血清白蛋白是其氨基酸序列中的自N端起第34位或与其等同的位置的半胱氨酸被其它氨基酸取代而成的。
4.如权利要求1~3中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,取代所述半胱氨酸的氨基酸是中性氨基酸。
5.如权利要求1~4中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,取代所述半胱氨酸的氨基酸选自丙氨酸、丝氨酸、及甘氨酸。
6.如权利要求1~5中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,取代所述半胱氨酸的氨基酸为丙氨酸。
7.如权利要求1~6中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,所述硫氧还蛋白的氨基酸序列中的自N端起第62位或与其等同的位置的半胱氨酸及/或第69位或与其等同的位置的半胱氨酸进一步被其它氨基酸取代,并且,
所述血清白蛋白是其氨基酸序列中的自N端起第34位或与其等同的位置的半胱氨酸被其它氨基酸取代而成的。
8.如权利要求7所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其中,取代所述半胱氨酸的氨基酸为丙氨酸。
9.如权利要求1~8中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体,其是血清白蛋白与硫氧还蛋白通过接头结合而成的。
10.医药组合物,其包含权利要求1~9中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体。
11.如权利要求10所述的医药组合物,其用于能够通过抗氧化作用、细胞因子转录因子的活性控制、抗凋亡及抗炎作用的药理活性治疗的疾病的预防或治疗。
12.如权利要求11所述的医药组合物,其中,所述疾病为酒精性肝病、非酒精性肝病、对乙酰氨基酚肝炎、急性重型肝炎、肝硬化、急性肾损伤、慢性肾损伤、顺铂肾病、横纹肌溶解症、造影剂肾病、慢性肾脏病、糖尿病性肾脏病、器官纤维化、自身免疫性疾病、阿尔茨海默病、帕金森病、周围神经病、脑梗死、动脉硬化、心肌梗死、心力衰竭、心肌梗死、急性呼吸窘迫综合征、慢性阻塞性肺疾病、过敏性肺炎、流感病毒肺炎、支气管哮喘。
13.权利要求1~9中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体用于制造医药组合物的用途。
14.权利要求1~9中任一项所述的经修饰的血清白蛋白-硫氧还蛋白融合体用于预防或治疗能够通过抗氧化作用、细胞因子转录因子的活性控制、抗凋亡及抗炎作用的药理活性治疗的疾病的用途。
15.如权利要求14所述的用途,其中,所述疾病为酒精性肝病、非酒精性肝病、对乙酰氨基酚肝炎、急性重型肝炎、肝硬化、急性肾损伤、慢性肾损伤、顺铂肾病、横纹肌溶解症、造影剂肾病、慢性肾脏病、糖尿病性肾脏病、器官纤维化、自身免疫性疾病、阿尔茨海默病、帕金森病、周围神经病、脑梗死、动脉硬化、心肌梗死、心力衰竭、心肌梗死、急性呼吸窘迫综合征、慢性阻塞性肺疾病、过敏性肺炎、流感病毒肺炎、支气管哮喘。
