Abstract
The invention discloses a cosmetic composition mainly comprising a mushroom extract for promoting skin repair, which is characterized in that a fermentation method of the extract is enriched, the mushroom extract is obtained by fermenting mushroom with mixed bacteria, and the mixed bacteria and the natural extract are matched, so that the composition is purely natural, free of addition, fresh, easy to absorb, moisture-keeping, release, improve closing, refreshing, transparent, tough, repair skin, maintain water-oil balance, resist oxidation, resist aging and remove free radicals.

Moreover, the urban pollution is serious nowadays, and the oil is too large for a long time, so pores are easily blocked, dust and dirt are easily adhered, and skin infection, acne and the like are caused.
The skin health is maintained, firstly, the water-oil balance of the skin is maintained, namely the skin is in the healthiest and ideal state without oil and dryness, the pH value is between 5 and 5.6, the secretion of sweat glands and sebaceous glands is moderate, and the skin is in a non-greasy and non-dry neutralization state, so that the skin is ruddy and fine, and is not sensitive to external stimulus.

the invention obtains the bacillus methylotrophicus (the preservation number is CCTCCNO: M2011201) for fermenting and producing the gamma-polyglutamic acid by searching in a targeting way and knowing the bacillus methylotrophicus in contact with a preservative; bacillus mucilaginosus SM-01 (CN 0141004782720140212) with high yield of polysaccharide and moisturizing effect; burkholderia cepacia G1-7-4 for fermenting high-yield lipase can be used for producing surfactants and aromatic compounds by establishing cooperation to obtain the strain, and the lipase can be used for well promoting the lipidation, fat transfer, alcoholysis, aminolysis and the like of fermentation products. The inventor further improves the rapid absorption, water supplementing, water locking, antioxidation, anti-aging and free radical removal effects of the flat ground mushrooms, the Raschia and the agaricus bisporus, further ferments the extract in bacteria, inactivates the bacterial strain after fermentation is finished to prevent the interference effect of the bacteria on the skin, and the functions of the obtained sterilized fermented product are correspondingly improved.

The preparation method of the fermentation product comprises pulverizing 1kg of Agaricus blazei Murill, 1kg of Agaricus blazei Murill and 1kg of Agaricus bisporus, adding 10 times of ultrapure water, extracting with ultrasonic water for 60min, extracting at 55-65deg.C for 120min, filtering, adding 5-6% of activated carbon, stirring at room temperature for 40-50min, and filtering to obtain filtrate; in a culture medium added with glucose, naCl and urea, 10% of bacillus methylotrophicus (preservation number is CCTCCNO: M2011201), 10% of bacillus mucilaginosus SM-01 and 10% of Burkholderia cepacia G1-7-4 are taken, seed liquid (the common strain culture medium comprises 15-20 of glucose in G/L, 20-15 of yeast extract, 0.4-0.6 of MgSO4.7H2O and K2HPO40.5-1.5 of shaking bottle with the rotating speed of 200rpm and the culture temperature of 35-37 ℃ for 8-12 hours) is taken, fermentation culture medium (25G/L of glucose, 10G/L of peptone, 2G/L of NH4NO3, 2G/L of KNO3, 0.1G/L of KH2PO4, 0.4G/L of magnesium sulfate, 0.5mg/L of nicotinic acid and 0.1mg/L of vitamin B1) is added for fermentation culture, the fermentation temperature is set to be 30-32 ℃ and the ventilation amount to be 0.5dm3/min, and the fermentation liquid is adjusted to be 280.8-7.2 r-300 min according to the fermentation condition; obtaining fermentation liquor, sterilizing at high temperature, centrifuging, taking supernatant, filtering and purifying. Through adjusting common parameters in the extraction fermentation process, the method finds that the method has little influence on the functions of the final composite fermentation products of the three fungi of the flat ground mushrooms, the poking mushrooms and the agaricus bisporus, namely M2011201+SM-01+G1-7-4, so that the subsequent research can be in the direction, but the current repeated test finds that the composite fermentation products of the three mushrooms are stable in functions and do not change along with the fine adjustment of test conditions, and the fermentation products are stable and have far market prospects.

Detailed Description
Embodiments of the present invention will be further described with reference to examples.
The invention divides experiments into a first group of blank basic substrates, a second group of basic substrates and a flat ground mushroom ferment (the initial raw materials of the ferment in the invention content are replaced by flat ground mushrooms, agaricus blazei and agaricus bisporus, the rest is unchanged, the rest is the same), a third group of basic substrates and a Agaricus blazei ferment, a fourth group of basic substrates and an agaricus bisporus ferment, a fifth group of basic substrates and a flat ground mushroom, agaricus blazei and agaricus bisporus M2011201+SM-01 ferment, a sixth group of basic substrates and a flat ground mushroom, agaricus blazei and agaricus bisporus G1-7-4 ferment, a seventh group of basic substrates and a flat ground mushroom, agaricus blazei and agaricus bisporus M2011201+SM-01+G1-7-4 ferment, and the basic substrates are 10G of 1, 3-propanediol, 10G of butanediol, 5G of alcohol, 1G of sodium hyaluronate, 1G ergothione, 43G of water (73G of first group of water) and the like. 70g of basic matrix, 30g of ferment, and 5% of ferment is used in the culture medium in experiments for verifying the function of ferment in examples without special description.
EXAMPLE 1 cell irritation
In order to confirm the cell irritation of each group, a common cell irritation test method for ginseng was invented, and a cytotoxicity test was performed on keratinocytes (HaCat).
Keratinocytes (HaCat) were seeded in 96-well plates at a density of 5 x 104 cells/well. After 48 hours, 10% ez-Cytox was mixed in DMEM medium to measure cell viability and proliferation, and each group of experiments was added to the medium. After 2 hours of incubation, absorbance was measured with a microplate reader at a wavelength of 450 nm.
Cell viability (%) = (absorbance of experimental group 2-7/absorbance of experimental group 1) ×100 results: experimental groups 2-7 showed cytotoxicity of 2.75%, 2.45%, 2.55%, 3.05%, 2.85%, 3.75%, respectively. The above groups each have low cytotoxicity of 5.0% or less, and the above groups can be used as materials for cosmetic safety.
Example 2 enhancement of DNA repair barrier protection system.
Referring to the common quantitative RT-PCR gene expression detection method, we select genes related to barrier functions OCLN, CLDN1, KLK7 and cell damage SMAD7, examine whether the gene expression changes are given to the ferment, thereby achieving the purpose of accurately targeting skin balance and repairing skin by biological means and exploring whether the ferment can reach the degree of deep penetration gene layer repair.
Human normal erythrocytes were purchased and inoculated in 96-well plates at an inoculation density of 5×104 cells per well in culture medium. Culturing for 24h until the cell confluence reaches 80%, adding experimental group culture for 5d, adding lysis buffer according to the kit to extract total RNA of the cells, measuring gene expression by quantitative RT-PCR, using a reverse transcription kit (High-Capacity), using a primer and a TaqMan probe (purchased from Jin Kairui biological company, designed by the company), using GAPDH gene as an internal reference gene, and obtaining the relative expression amount of the target gene (the expression amount of each gene of experimental group 1 is set as 100). The PCR reaction conditions were 95℃for 15 minutes, 95℃for 10-15 seconds, 60℃for 10-15 seconds, and 72℃for 10-15 seconds, and 30 cycles and 72℃for 7 minutes were performed.

Results: the ferment can promote the expression of OCLN and CLDN1 at the gene level, promote the tight connection between epithelial cells, promote the expression of SMAD7, relieve the stimulation of the outside on cells, inhibit the skin peeling process of KLK7, and promote the balance repair of skin at the gene level.
Example 3 alleviation of cell stimulation
Referring to the usual cell stimulation remission test method, fibroblasts were cultured in medium until cell confluence reached 80%. Cells were transferred to 96-well plates at a density of 5×104 cells/well and cultured for 48 hours. After culturing, the cell attachment and growth state were observed under a microscope. Lactic acid (0.2%) was used as skin irritant. MTT solution 3mg/ml was added and then the cell viability of the experimental group 1 was 100% using ELISA reader (absorbance at 570nm was measured to compare the cell viability of each group).
Results: experimental groups 2-7 showed cell viability of 105%, 108%, 121%, 155%, 185%, 275%, respectively. Microscopic examination of cell results: each group of cells survived well and was in a proliferative state, unaffected by cell stimulation.
EXAMPLE 4 antioxidant Effect
The cell culture method of example 4 was used to examine the clearance of hydroxyl and superoxide anion radicals in cells according to the kit instructions, and the results are shown in Table 2. Calculated as hydroxyl and superoxide anion radical scavenging rate of 0% per cell initial state.

Results: experiments 2-7 all had hydroxyl and superoxide anion radical scavenging effects.
Example 5 performance test:
280 volunteers (not dividing men and women) aged 20-45 were selected as trial subjects. The test object is detected by a skin detector of a company, and is identified as skin health, the history of no skin allergy is 140, and the damaged skin and water lack need to be repaired to 140; each group was then randomized into 7 subgroups of 20 persons. The using method comprises the following steps: after the test object cleans the face, the compound toner is uniformly smeared on the face, is gently beaten to be fully absorbed, is used once in the morning and evening every day, and is continuously used for 50 days. The compositions were then scored for freshness, resistance to sensitivity, wrinkle resistance, and water smoothness to the skin. The total usage effect is divided into 5 minutes. The average scores are as follows. The results are shown in Table 4.

A cosmetic composition mainly containing mushroom extract for promoting skin repair is characterized in that the composition is added with fermentation products of flat ground mushrooms, raschia mushrooms and agaricus bisporus, wherein the fermentation products are preserved by bacillus methylotrophicus (CCTCCNO: m2011201 Bacillus mucilaginosus (Bacillus mucilaginosus) SM-01 with preservation number of CGMCC No.5766, burkholderia cepacia G1-7-4 (Burkholderia cepacia) G1-7-4, and preservation number of CCTCCNO: M2014656, fermenting Agaricus blazei Murill, agaricus praecox and Agaricus bisporus, sterilizing, separating, and purifying.
2. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein said composition further comprises aloe vera leaf extract.
3. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein said composition further comprises white willow bark extract.
4. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein the composition further comprises centella asiatica extract.
5. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein the composition further comprises a cortex phellodendri extract.
6. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein said composition further comprises geranium oil.
7. A cosmetic composition for promoting skin repair comprising a mushroom extract as claimed in claim 1, wherein said composition further comprises lavender oil.
8. A cosmetic composition for promoting skin repair comprising mushroom extract as claimed in claim 1, wherein the composition further comprises 1, 3-propanediol and butanediol.
9. A cosmetic composition for promoting skin repair comprising a fungus mushroom extract as claimed in claim 1, wherein said composition further comprises panthenol and sodium hyaluronate.
10. A cosmetic composition for promoting skin repair comprising a mushroom extract according to claim 1, wherein ergothioneine is further included in the composition.
