技术领域
本发明涉及辣椒色素提取技术领域,具体涉及一种辣椒色素的提取方法。
背景技术
椒椒红色素是一种天然食用色素,不溶于水,具有较好的热稳定性、耐酸碱性、耐光性和乳化分散性,同时浓度不同时可以呈现深红、橘红、黄色、淡黄色等颜色,广泛应用于食品、医药以及化妆品等工业产品的着色。
辣椒色素的提取方法主要包括油溶法和有机溶液获取法,其中,油溶法,即用动、植物油类(如鱼油、棉籽油、豆油、菜油、椰子油、玉米油等)浸泡辣椒皮或辣椒粉,然后分离出溶解在油中的色素,这种方法的缺点是油与色素的分离比较困难,难以得到浓稠的色素。有机溶液获取法是现阶段工业生产上运用最大多数的获取方式,其基本原理是相似相溶,具备一次性大批获取、加工工艺简易、低成本等优势,但存在提取率较低的难题,同时,不管在油溶法还是有机溶液获取法中,都存在辣椒脱籽流程费时费力的情况。
发明内容
为解决上述问题,本发明提供了一种辣椒色素的提取方法,可显著提高辣椒色素的提取率。
为实现上述目的,本发明采取的技术方案为:
一种辣椒色素的提取方法,包括如下步骤:
S1、将辣椒清理除杂、烘干、脱籽后,在0~22℃粉碎至100目,得辣椒粉;
S2、将所述的辣椒粉置于酶解罐内,调节pH至5.4,加入适量纤维素酶,控制温度在30~50℃,压力在0.06~0.1Mpa,真空渗透处理5~7s后,酶解2h,离心分离,取上清液干燥,并造粒,得辣椒素颗粒;
S3、将所述辣椒素颗粒置于萃取器内,以丁烷为萃取剂,进行3次亚临界逆流萃取,萃取温度30~35℃,得到深红色的膏状萃取物;
S4、将所述膏状萃取物与适量75%(体积分数)的乙醇混合后,利用辣椒红色素相和乙醇相的密度差进行离心分离,得富含辣椒碱的乙醇相和油膏。
进一步地,所述步骤S1中,将辣椒去梗后,切段,置于干燥网上,采用负压脉冲喷动干燥,喷动温度为45~50℃、喷动频率为间隔10秒喷动3秒。
进一步地,所述步骤S3中,辣椒素颗粒与丁烷的质量体积比为1:1.1。
进一步地,所述步骤S3中,辣椒素颗粒为Φ4mmx8mm的棒状颗粒。

本发明具有以下有益效果:
1)采用喷动干燥技术,协同干燥工艺参数的设计,可以在干燥的同时实现辣椒的脱籽操作;
2)采用酶解结合亚临界逆流萃取的方式,可以显著提高辣椒红素的提取率;
3)通过酶解工艺的优化,可以显著缩短酶解所需的时间。

实施例1
一种辣椒色素的提取方法,包括如下步骤:
S1、将辣椒去梗后,切段,置于干燥网上,采用负压脉冲喷动干燥,喷动温度为45℃、喷动频率为间隔10秒喷动3秒,干燥至含水率低于10%后,置于粉碎机内,在10℃粉碎至100目,得辣椒粉;干燥完成后,辣椒脱籽率基本可以达到100%;
S2、将所述的辣椒粉置于酶解罐内,调节pH至5.4,按8.1mg/g辣椒粉的比例加入适量纤维素酶,控制温度在30℃,压力在0.06Mpa,真空渗透处理7s后,恢复常压,酶解的温度为45℃,酶解2h,离心分离,取上清液干燥,并造粒,得Φ4mmx8mm的棒状辣椒素颗粒;
S3、将所述辣椒素颗粒置于萃取器内,以丁烷为萃取剂,进行3次亚临界逆流萃取,辣椒素颗粒与丁烷的质量体积比为1:1.1,萃取温度30℃,得到深红色的膏状萃取物;
S4、将所述膏状萃取物与适量75%(体积分数)的乙醇混合后,利用辣椒红色素相和乙醇相的密度差进行离心分离,得富含辣椒碱的乙醇相和油膏,富含辣椒碱的乙醇相浓缩回收乙醇,得到辣椒碱。获得的辣椒红色素的色阶为186.1,提取率为99.5%;获得的辣椒碱的纯度为 79.8%,提取率为96.7%。
实施例2
一种辣椒色素的提取方法,包括如下步骤:
S1、将辣椒去梗后,切段,置于干燥网上,采用负压脉冲喷动干燥,喷动温度为45℃、喷动频率为间隔10秒喷动3秒,干燥至含水率低于10%后,置于粉碎机内,在15℃粉碎至100目,得辣椒粉;干燥完成后,辣椒脱籽率基本可以达到100%;
S2、将所述的辣椒粉置于酶解罐内,调节pH至5.4,按8.1mg/g辣椒粉的比例加入适量纤维素酶,控制温度在40℃,压力在0.08Mpa,真空渗透处理6s后,恢复常压,酶解的温度为45℃,酶解2h,离心分离,取上清液干燥,并造粒,得Φ4mmx8mm的棒状辣椒素颗粒;
S3、将所述辣椒素颗粒置于萃取器内,以丁烷为萃取剂,进行3次亚临界逆流萃取,辣椒素颗粒与丁烷的质量体积比为1:1.1,萃取温度32℃,得到深红色的膏状萃取物;
S4、将所述膏状萃取物与适量75%(体积分数)的乙醇混合后,利用辣椒红色素相和乙醇相的密度差进行离心分离,得富含辣椒碱的乙醇相和油膏,富含辣椒碱的乙醇相浓缩回收乙醇,得到辣椒碱。获得的辣椒红色素的色阶为193.6,提取率为99.8%;获得的辣椒碱的纯度为 80.6%,提取率为97.2%。
实施例3
一种辣椒色素的提取方法,包括如下步骤:
S1、将辣椒去梗后,切段,置于干燥网上,采用负压脉冲喷动干燥,喷动温度为45℃、喷动频率为间隔10秒喷动3秒,干燥至含水率低于10%后,置于粉碎机内,在22℃粉碎至100目,得辣椒粉;干燥完成后,辣椒脱籽率基本可以达到100%;
S2、将所述的辣椒粉置于酶解罐内,调节pH至5.4,按8.1mg/g辣椒粉的比例加入适量纤维素酶,控制温度在50℃,压力在0.1Mpa,真空渗透处理7s后,恢复常压,酶解的温度为45℃,酶解2h,离心分离,取上清液干燥,并造粒,得Φ4mmx8mm的棒状辣椒素颗粒;
S3、将所述辣椒素颗粒置于萃取器内,以丁烷为萃取剂,进行3次亚临界逆流萃取,辣椒素颗粒与丁烷的质量体积比为1:1.1,萃取温度35℃,得到深红色的膏状萃取物;
S4、将所述膏状萃取物与适量75%(体积分数)的乙醇混合后,利用辣椒红色素相和乙醇相的密度差进行离心分离,得富含辣椒碱的乙醇相和油膏,富含辣椒碱的乙醇相浓缩回收乙醇,得到辣椒碱。获得的辣椒红色素的色阶为187.7,提取率为99.6%;获得的辣椒碱的纯度为 80.3%,提取率为97.1%。
对比例1
S2、将所述的辣椒粉造粒,得Φ4mmx8mm的棒状辣椒素颗粒;其余设计同实施例2。获得的辣椒红色素的色阶为180.3,提取率为92.7%;获得的辣椒碱的纯度为 73.6%,提取率为93.2%。
对比例2
S2、将所述的辣椒粉置于酶解罐内,调节pH至5.4,按8.1mg/g辣椒粉的比例加入适量纤维素酶,酶解的温度为45℃,酶解2h,离心分离,取上清液干燥,并造粒,得Φ4mmx8mm的棒状辣椒素颗粒;其余设计同实施例2。获得的辣椒红色素的色阶为181.7,提取率为93.6%;获得的辣椒碱的纯度为 75.6%,提取率为94.3%。
由实施例1和对比例1的对比,可以看出,酶解工艺和丁烷亚临界逆流萃取的结合可以显著提高辣椒红色素的提取率。由实施例2和对比例2的对比可以发现,对比例2的辣椒红色素的提取率显著低于实施例2,究其原因,发现其酶解反应没有充分完成,可见,负压渗透处理可以使得酶解工艺在2h内即可充分完成。

S3、将所述辣椒素颗粒置于萃取器内,以丁烷为萃取剂,进行3次亚临界逆流萃取,萃取温度30~35℃,得到深红色的膏状萃取物;
S4、将所述膏状萃取物与适量75%的乙醇混合后,利用辣椒红色素相和乙醇相的密度差进行离心分离,得富含辣椒碱的乙醇相和油膏。
