本发明提供了一种耐热κ‑卡拉胶酶及其应用,耐热κ‑卡拉胶酶是由如SEQ ID NO.1所示序列的野生型κ‑卡拉胶酶经过其第154位氨基酸突变而成。与野生型κ‑卡拉胶酶相比,突变酶在50℃的残余活力比野生型酶高出50%左右,在55℃的残余活力比野生型高出10%左右,从而使该耐热κ‑卡拉胶酶在卡拉胶寡糖工业生产中具有良好的应用前景。

技术领域
本发明属于生物工程技术领域,具体涉及一种耐热κ-卡拉胶酶及其应用。
背景技术
卡拉胶是一种由α-1,3-D-半乳糖和β-1,4-D-半乳糖组成的线性硫酸化多糖,具有成胶能力,可作为食品增稠剂、凝胶剂、澄清剂等。根据结构中是否含有3,6-脱水-D-半乳糖,以及半乳糖上硫酸基团的位置和数量,卡拉胶主要分为κ-、ι-和λ-卡拉胶三种类型,其中κ-型应用最为广泛。κ-卡拉胶分子量大、粘度高、溶解性差,生物利用度低,其应用受到了限制。κ-卡拉胶的降解产物为κ-卡拉胶寡糖,具有抗炎、抗凝血、抗肿瘤和抗血栓形成等作用,广泛应用在食品、医药等领域。卡拉胶寡糖的制备方法有物理法、化学法和酶解法。其中,酶解法由于条件温和、反应特异性好等特点,应用最为广泛。κ-卡拉胶酶可用于寡糖的制备,该酶的来源广泛,主要来源于海洋细菌,如假单胞菌属(Pseudomonas sp.)、弧菌属(Vibrio sp.)、希瓦氏菌属(Shewanella sp.)等。高温可以降低κ-卡拉胶的粘度,促进κ-卡拉胶酶处理κ-卡拉胶,因此提高κ-卡拉胶酶的耐热性对于κ-卡拉胶的高值资源化利用具有重要意义。
发明内容
本发明旨在至少在一定程度上解决上述技术中的技术问题之一。为此,本发明提出了一种耐热κ-卡拉胶酶,其具有较高的热稳定性和水解卡拉胶能力。
为此,在本发明的第一个方面中,提供了一种耐热κ-卡拉胶酶,其由如SEQ ID NO:1所示序列的野生型κ-卡拉胶酶(WT)经过其第154位氨基酸突变而成。
根据本发明的实施例,所述耐热κ-卡拉胶酶是将野生型κ-卡拉胶酶的第154位氨基酸进行突变。与野生型κ-卡拉胶酶相比,突变酶在50℃的残余活力比野生型酶高出50%左右,在55℃的残余活力比野生型高出10%左右,从而使κ-卡拉胶酶在卡拉胶寡糖工业生产中具有良好的应用前景。
可选地,所述野生型κ-卡拉胶酶的第154位氨基酸由Glu变为Ala。
可选地,所述突变方式为定点突变。
在本发明的第二方面中,提供了一种编码上述耐热κ-卡拉胶酶的基因,所述基因的核苷酸序列如SEQ ID NO:2所示。

实施例1构建κ-卡拉胶酶突变体
突变酶重组表达载体的构建:
以携带野生型κ-卡拉胶酶编码基因(SEQ ID NO:1)的载体为模板进行定点突变,构建突变酶重组表达载体,引物按照突变试剂盒Mut ExpressⅡFast Mutagenesis Kit V2引物设计要求设计定点突变引物,突变引物如表1所示。其中,定点突变参照突变试剂盒MutExpressⅡFast Mutagenesis Kit V2的说明书进行操作。

包括目标质粒扩增、扩增产物Dpn I消化、重组反应三个步骤。
目标质粒扩增:
依次加入上述试剂后,混合均匀,然后进行PCR。PCR反应参数:95℃ 30s,95℃15s,60℃ 15s,72℃ 2min,30次循环,72℃ 5min,4℃保存。
扩增产物DpnⅠ消化:
DpnⅠ 1μL
扩增产物 40~50μL
在PCR反应液中加入1μL Dpn I并混匀,37℃反应1h 30min。
重组反应:
置于37℃恒温反应30min后,降至4℃或立即置于冰上冷却。
重组产物转化表达宿主:
(1)在冰上解冻E.coli BL21感受态细胞。
(2)取10μL重组产物加入到100μL感受态细胞中,轻弹管壁混匀,冰上静置30min。
(3)42℃水浴热激45s后,立即置于冰上冷却2~3min。
(4)加入900μL LB液体培养基(不添加抗生素),37℃摇菌1h(转速200rpm)。
(5)将卡那霉素抗性的LB固体培养基平板在37℃培养箱中预热。
(6)4,000rpm离心5min,弃掉900μL上清。用剩余培养基将菌体重悬,在37℃恒温箱中倒置培养12~16h。利用菌落PCR进行重组质粒鉴定,并经测序验证突变酶的基因序列。

从LB平板(含卡那霉素100μg/mL)挑取阳性克隆的单菌落移至5mL LB培养基(含卡那霉素100μg/mL),37℃摇动(180rpm)培养过夜。将过夜培养物按照1%的接种量接种于300mL LB液体培养基(含100μg/mL卡那霉素),转接完后在37℃继续振摇培养至细菌OD600值达1.0~1.2。加入异丙基-β-D-硫代半乳糖苷(IPTG)至终浓度0.5mmol/L,16℃低温诱导表达,用于后续的亲和层析纯化。诱导表达的菌液于5000rpm离心5min,细胞重悬于10~20mL预冷的溶解缓冲液(50mmol/L NaH2PO4,300mmol/L NaCl,15mmol/L咪唑;pH8.0)。冰浴条件下进行超声波破菌(处理总时间约15min;每次可以处理5s,间隔5s后再处理,间隔的时间内摇动菌液使其混匀),4℃下15000rpm离心20min。适量Ni-NTA agarose用预冷的溶解缓冲液平衡后,与上清(上清用0.22微米滤膜过滤)混匀,4℃下摇动反应0.5~1h。用预冷的洗涤缓冲液(50mmol/L NaH2PO4,300mmol/L NaCl,30mmol/L咪唑;pH8.0)洗去杂蛋白,至流出液的OD280趋近于零。用预冷的洗脱缓冲液(50mmol/L NaH2PO4,300mmol/L NaCl,250mmol/L咪唑;pH8.0)洗脱目的蛋白。利用亲和层析对重组蛋白进行分离纯化。采用SDS-PAGE分析纯化后目标蛋白的分子量及其纯度。
SDS-PAGE分析如图1所示,图中,泳道M为蛋白质分子量标准,1为WT,2为E154A。突变酶的分子质量与野生型酶一致,大小约为35.0kDa。
实施例2耐热κ-卡拉胶酶的酶学性质测定
κ-卡拉胶酶活力的测定:将WT和突变体E154A的蛋白质浓度稀释至相同浓度水平。将10酶液与490μL含有0.5%(w/v)浓度的卡拉胶底物溶液(以50mmol/L NaH2PO4-Na2HPO4缓冲液配制,pH 8.0)在40℃下反应15min,加入500μL DNS试剂终止反应,沸水浴10min冷却,测定520nm处吸光值。利用半乳糖标准曲线,确定产生的还原糖含量。每分钟释放1μmoL还原糖(以半乳糖计)所需的酶量为一个酶活力单位(U)。酶活性分析结果如图2所示,突变酶的比活力比野生型高出29%左右。
κ-卡拉胶酶的最适反应温度和最适反应pH测定:将WT和突变体E154A的蛋白质浓度稀释至相同浓度水平。在30–60℃的温度范围内,以5℃的间隔,分别测定野生型和突变体在不同温度下的活力,分析酶的最适反应温度。在6.0–10.0的pH范围内,以1.0的间隔,分布测定野生型和突变体在不同pH下的活力,分析酶的最适反应pH。
κ-卡拉胶酶的耐热性测定:将纯化的酶在45-60℃处理30min后,测定酶的残留活性,分析酶的耐热性。蛋白质在X分钟(T50 X)后保持一半剩余活性的温度是蛋白质失活的常用指标,计算酶的T50 30值。将酶在50℃和60℃下分别处理不同时间后,测定酶的残余活力,计算酶在50℃和60℃下的酶活性的半衰期(t1/2)。
耐热性分析结果如图3所示,突变酶在50℃的残余活力比野生型酶高出50%左右,在55℃的残余活力比野生型高出10%左右。其他酶学性质的分析结果表2所示,突变酶和野生型酶的最适反应温度和pH不变,分别为40℃和pH8.0。在50℃,野生型酶的半衰期是原始型酶的15.4倍。突变酶的T50比原始酶高出4.0℃。

实施例3κ-卡拉胶酶降解κ-卡拉胶
用50mmol/L的NaH2PO4-Na2HPO4缓冲液(pH 8.0)制备15mL含有0.5%(w/v)的κ-卡拉胶底物溶液,加入2U酶,40℃恒温反应2h,分别于15、30、60、90、120min取样500μL,沸水浴灭活10min,利用DNS法测定野生型酶及突变酶的还原糖生成量。
结果如图4所示,在40℃下,15–90min时,突变酶对κ-卡拉胶的降解率高于野生型,表明突变酶对κ-卡拉胶的降解能力高于野生型酶,能够使κ-卡拉胶酶更好的应用于卡拉胶寡糖工业生产。
综上,根据本发明的实施例,采用定点突变技术将野生型酶进行定点突变,通过将其第154位氨基酸由Glu变为Ala,突变酶的热稳定性得到了一定的提高,在一定程度上提高了对卡拉胶的降解,在卡拉胶寡糖的工业生产中有良好的应用潜力。

1.一种耐热κ-卡拉胶酶,其特征在于,由如SEQ ID NO 1:所示序列的野生型κ-卡拉胶酶经过其第154位氨基酸突变而成。
2.如权利要求1所述的耐热κ-卡拉胶酶,其特征在于,所述野生型κ-卡拉胶酶的第154位氨基酸由Glu变为Ala。
3.如权利要求1所述的耐热κ-卡拉胶酶,其特征在于,所述突变方式为定点突变。
4.一种编码权利要求1所述的耐热κ-卡拉胶酶的基因,其特征在于,所述基因的核苷酸序列如SEQ ID NO 2:所示。
5.一种构建体,其特征在于,包含如权利要求4所述的基因。
