Abstract
本发明涉及抗体技术领域,具体公开了一种靶向TROP2和CD3的双特异性抗体及其制备方法与应用。本发明的靶向TROP2和CD3的双特异性抗体具有对称形式的类IgG结构,包括:结合TROP2的一对Fab分子、结合人T细胞表面抗原CD3的2条序列相同的单链抗体、铰链区以及Fc;单链抗体的N端与Fab分子的CH1的C端连接,单链抗体的C端与铰链区的N端连接。本发明提供了具备良好的结合人TROP2抗原和结合人CD3的细胞桥双特异性抗体,其在交联T细胞和表达TROP2的癌细胞后具有激活T细胞的能力,同时能有效介导交联的T细胞对表达TROP2的靶细胞进行高效杀伤。
Description
技术领域
本发明涉及抗体技术领域,具体地说,涉及一种靶向TROP2(人滋养层细胞表面抗原2)和人T细胞表面抗原CD3的双特异性抗体及其制备方法与应用。
背景技术
抗体药物是以细胞工程技术和基因工程技术为主体的抗体工程技术制备的生物大分子药物,具有特异性高、性质均一、可针对特定靶点定向制备等优点。单克隆抗体在临床上主要应用于以下三个方面:肿瘤治疗、免疫性疾病治疗以及抗感染治疗。其中,肿瘤治疗是目前单克隆抗体应用最为广泛的领域,目前已经进入临床试验和上市的单克隆抗体产品中,用于肿瘤治疗的产品数量占比大概为50%。单克隆抗体治疗肿瘤是一种针对病变细胞特异靶点刺激免疫系统杀伤靶细胞的免疫疗法,为了增强抗体的效应功能,特别是提高杀伤肿瘤细胞的效果,人们尝试多种方法改造抗体分子。
获取特异性抗体的方法有多种。传统的杂交瘤技术通过免疫小鼠或大鼠,将其脾脏细胞与骨髓瘤细胞融合,然后进行稀释(至一个细胞每孔)培养,用酶联免疫法(ELISA)检测杂交瘤细胞上清与免疫抗原的结合,进而筛选可分泌与抗原特异性结合抗体的单克隆细胞株。这种鼠源抗体如果应用于临床药物研发,则需要通过杂合抗体技术(chimerization)、人源化技术(humanization)等基因工程手段进行去免疫原性改造。为了从起始阶段就消除未来临床必须解决的免疫原性问题,转基因鼠技术(transgenic mouse)通过替换鼠源抗体可变区为人源抗体可变区,使得获得的单克隆抗体的可变区序列主体上就是人源的,然后再进一步替换恒定区就可以获得全人源单克隆抗体。另外一个技术就是噬菌体表面呈现(phage display)技术。噬菌体表面呈现技术是在试管中构建工程抗体,一般是单链抗体或Fab形式。但是组成工程抗体的基因片段来源于人免疫细胞的基因库,所以获得的抗体是全人源的。已有多个噬菌体技术获得的抗体进入临床研究或获得批准,用于各个方面的疾病治疗。
在单克隆抗体的基础上,为了进一步提高药效和减少毒副作用,以单克隆抗体为基础的新抗体药物,包括药物偶联抗体(Antibody drug conjugate, ADC),双特异性抗体(bispecific antibody,bsAb)以及CAR-T (chimeric antigen receptor T cells)等新型蛋白和细胞药物的发展极为迅速。其中,双特异性抗体能够同时识别两个不同的靶标,因此产生了更多的生物学作用机制,带来了新的应用途径。例如,针对两个不同癌肿瘤抗原的双抗(如抗EGFR和cMET双抗,Amivantamab®),用于同时抑制EGFR和cMet信号通路,已被FDA批准用于治疗铂类化疗后进展的EGFR外显子20插入突变的转移性非小细胞肺癌(NSCLC)患者。Hemlibra是一种同时结合凝血IXa和X的双特异性抗体,能够将参与天然凝血级联反应的两种关键蛋白IXa和X聚集在一起,触发凝血反应,可用于A型血友病患者的凝血治疗,已被批复用于临床。
基于双特异性抗体分子构建的细胞桥(cell-bridging)双抗,又称engager。如果双抗分子其中一个靶标是免疫细胞的信号受体,如人T细胞的CD3,另一个靶标是肿瘤细胞表面的抗原,则engager可同时连接免疫细胞和肿瘤靶细胞,形成肿瘤细胞-双抗分子-T细胞复合体,该复合体交联并活化免疫细胞受体(如T细胞的CD3)。T细胞因此进入活化、增殖状态,除了数量的增加,还释放多种细胞因子(如IL2,TNF-α,IFN-γ,IL-6等等)及毒性分子(颗粒酶B和穿孔素等)。细胞因子系统性动员了免疫系统,毒性分子成为了T细胞特异性杀伤靶细胞的武器。这些T engager表现出了突破性的临床效果,因而在血癌等疾病治疗上受到了极大的关注。Blincyto®就是交联表达抗原CD1 9的B细胞和TCR亚基(CD3)的T细胞的双特异性抗体,已批复用于临床急性淋性白血病(ALL)的治疗。
双特异性抗体可通过多种途径获得,其制备方法主要有:化学偶联法、杂交-杂交瘤法和基因工程抗体制备法。化学偶联法是将2个不同的单克隆抗体用化学偶联的方式连接在一起,制备的双特异性单克隆抗体,这是最早的双特异性单克隆抗体。杂交-杂交瘤法是通过细胞杂交法或者三元杂交瘤的方式产生双特异性单克隆抗体,这些细胞杂交瘤或者三元杂交瘤是通过建成的杂交瘤融合,或者建立的杂交瘤和从小鼠的淋巴细胞融合而得到的,只能用于生产鼠源的双特异性抗体,因此,其应用受到了极大的限制。而随着分子生物学技术的迅速发展,出现了基因工程人源化或全人源的双特异性抗体的多种构建模式,主要包括双特异性微抗体、双链抗体、单链双价抗体、多价双特异性抗体四类。目前,国际上已有数种基因工程双特异性抗体药物进入临床试验阶段或被批复用于不同的临床适应症,并显示有较好的应用前景。
滋养层细胞表面抗原2(TROP2)是由TACSTD2基因编码表达的细胞表面糖蛋白,又名肿瘤相关钙离子信号转导子2(TACSTD2)、表皮糖蛋白1(EGP-1)、胃肠肿瘤相关抗原(GA733-1)、表面标志物1(M1S1),最早于人胎盘滋养层鉴定到的细胞表面糖蛋白,后来被发现高度表达于大部分人的实体脏器肿瘤癌细胞中,且仅受限或有限地表达于正常人组织。TROP2属于GA733 蛋白家族,与上皮细胞黏附分子(EpCAM,又称 TROP1、TACSTD1)有较高结构序列相似性,同源性达49%。TROP2由323个氨基酸构成,其中信号肽含26个氨基酸,胞外区248个氨基酸,跨膜区23个氨基酸,胞质区26个氨基酸,其结构示意图如图1中的A所示。TROP2主要通过调节钙离子信号通路、细胞周期蛋白表达及降低纤黏蛋白黏附作用促进肿瘤细胞生长、增殖和转移。TROP2也可以与Wnt信号级联中的β-连环蛋白相互作用,因而对细胞核癌基因的转录及细胞的增殖起作用。大量临床研究和文献报道表明,TROP2在乳腺癌、胰腺癌、胆囊癌、结肠癌、胃癌、非小细胞肺癌、前列腺癌、子宫癌和口腔鳞癌等上皮癌中过表达,而在成年人正常组织中很少表达或不表达;TROP2在肿瘤组织中的过表达与患者的预后不良和癌细胞的转移密切相关,同时影响患者的总生存率。因此,TROP2已成为肿瘤分子靶向治疗中引人注目的靶标。
TROP2是一种跨膜蛋白,其细胞外结构域在多种肿瘤细胞上分布广泛,因此成为了靶向治疗的天然候选。TROP2的组织表达局限性使得靶向治疗的毒性降低,这也是靶向TROP2治疗的优势所在。以TROP2为靶点的抗体、抗体偶联物以及联合用药等多种形式的药物正处于研发中。抗TROP2抗体与其他化疗药物偶联的效用已在各种临床前研究中得到证实。用于治疗TROP2过表达的上皮恶性肿瘤的抗体偶联药物 IMMU-132已获FDA批准上市(2020年4月)。新型抗体偶联药物Sacituzumab govitecan (IMMU-132)是以TROP2为靶点,利用人源化抗体hRS7作为靶向载体与伊立替康活性代谢产物SN38偶联而成,可用于治疗多种上皮恶性肿瘤如乳腺癌(三阴乳腺癌)、卵巢癌、小细胞肺癌等。此外,其它人源化的抗TROP2的 IgG-SN-38偶联物,如抗TROP2 hRS7-CL2A-SN-38抗体偶联药物,已被证明在多种肿瘤细胞系(Calu-3、Capan-1、BxPC-3和COLO-205)的异种移植模型中具有显著的特异性抗癌作用。综上所述,开发一种靶向TROP2的双特异抗体,尤其是具备潜在药效良好的TROP/CD3抗原结合性的engager免疫双抗具有重要意义。
T细胞表面的CD3分子是由5个亚基α、β、γ 、δ、ε组成的6聚体(一个聚体中有两个ε亚基),其分子质量分别为14.9kDa,15.3kDa,20.0kDa,18.7kDa和22.8 kDa,其长度分别为135,139、182、171,207个氨基酸残基,共同组成胞外区的6条肽链,加上胞内的两个ζ亚基,共同形成T细胞受体(T cell receptor,TCR)复合物(TCR complex),其结构示意图见图1中的B。
TCR复合体具有信号转导、T细胞活化以及稳定TCR结构的功能。CD3胞质段含有免疫受体酪氨酸活化基序(immunoreceptor tyrosine-based activation motif,ITAM),TCR识别并结合由MHC(major histo-compatibility complex)分子提呈的抗原肽,造成TCR复合体寡聚,导致ζ亚基和CD3的ITAM的保守序列中的酪氨酸残基被T细胞内的酪氨酸蛋白激酶p56lck磷酸化,然后募集其它含有SH2 (Scr homology 2)结构域的酪氨酸蛋白激酶(如ZAP-70)。ITAM的磷酸化以及与ZAP-70的结合是T细胞活化信号传导过程早期阶段的重要生化反应之一。同样,结合CD3亚基(ε/γ/σ)的抗体如OKT3等也会造成TCR复合体的寡聚并活化T细胞,因此,可有望开发一种效果好的同时结合TROP2和CD3的、可用于免疫治疗的双特异性抗体。
发明内容
双特异性抗体(bispecific antibody,BsAb,简称双抗)是指能同时特异性结合两个不同抗原或不同抗原表位的人工抗体。形象比喻,它就像一座连接2个抗原(表位)的桥梁。交联免疫T细胞或NK细胞和肿瘤细胞的双抗又称T细胞/NK细胞交联剂/分子胶(T /NKengager),在免疫治疗肿瘤、自身免疫病等疾病上价值巨大,亦称免疫双抗。
免疫双抗在抗肿瘤治疗中的主要作用机制是:双抗结合肿瘤表达抗原,形成分子集束,然后结合招募(结合)T细胞或NK细胞,形成靶细胞-双抗集束-T或NK细胞复合体。此时的T或NK细胞被激活,释放细胞因子及毒性分子,就近杀伤其附近的肿瘤细胞。广泛意义的双抗还有其它的作用机制,例如(1)同时激活免疫细胞活化进程中两个不同的信号传导通路而发挥独特或重叠的功能,增强免疫细胞的生长增殖及存活;(2)同时抑制免疫细胞衰竭进程中两个不同的信号传导通路而发挥独特或重叠的功能,延迟免疫细胞功能衰竭和存活;(3)同时阻断癌变进程中两个不同的生长信号传导通路而发挥独特或重叠的功能,干扰/抑制肿瘤细胞的生长增殖及存活;(4)同时靶向细胞表面不同的抗原或表位,增强/扩大其与肿瘤细胞的特异性结合并直接利用ADCC杀伤肿瘤细胞;(5)同时靶向两个蛋白,通过物理相互作用拉近这两个蛋白,使之形成活化状态而发挥生物学功能,如结合凝血因子Ⅸ和Ⅹ的双抗可替代凝血因子Ⅷ而实现促凝血作用,以及其它新型机制。
双特异性抗体在自然条件下并不存在,而是通过细胞融合或重组DNA技术制备实现的。由于其特异性和双功能性,现已成为抗体工程领域的研究热点,尤其是免疫双抗在肿瘤治疗及自身免疫病等领域中具有广阔的应用前景。除此之外,双特异性抗体还被应用于治疗骨质疏松、血友病等其他领域。
对研发双抗药物而言,在设计融合两个抗体结合单元时,尽管首先需要考虑的是保持原有两个抗体结合单元的各自生物学活性,但还有一系列其它因素需要慎重考虑,才能保证双抗产品的临床前研究推进,临床效果发挥和工业化生产。首先,这两个单元的各自亲和力问题;其次,双抗的组成形式;再次,表达水平、稳定性、半衰期等等问题。例如Blincyto是由两个单链抗体形成的T细胞免疫双抗,靶向人B细胞的CD19和人T细胞的CD3亚基,已经批复用于成人及儿童急性淋性白血病(ALL)。此外还有基于Knob-into-Hole(KIH)形式的双抗,已经研发了多款临床双抗项目。不同形式的双抗结构既影响抗体功效的发挥,也影响工业化制备,怎样设计才能兼顾各方效果是需要反复摸索的。进一步的,双抗两个功能单元之间的距离问题,有数据表明这个距离会影响到包括双抗分子的生物活性在内的多种性质。不管是靶向两个(可溶性)靶标还是细胞膜靶标,保持双抗同时结合到各自抗原的能力往往是其发挥生物学功能的基础,双抗单元间的空间距离会影响同时结合的效率和速度。对于免疫双抗,两个单元间的距离还显著影响整个双抗分子的药效,包括对免疫细胞的刺激强度和双抗介导杀伤的能力。因此,针对特定抗体的组合,也需要对其之间的距离进行设计摸索。最后,成药性是双抗融合中最重要的因素。当采取非对称形式的双抗时,用于不同的重链/轻链基因共同在一个细胞内表达,往往出现错配问题。这些错配分子给纯化工艺及CMC(化学、制造和质量控制)带来极大的复杂性和不确定性,同时也会对双抗药物的稳定性带来挑战。因此,如何选取合适的双抗形式才能在兼顾各方效果的同时,实现产业化制造也是需要特别考量的。
本发明设计时所用的技术平台称之为FIST(Fusion of IgG and scFvTechnology),是设计2+2对称性双特异性抗体的构建平台。其特点具体来讲有以下方面:对称类抗体,生产纯化易:类经典IgG分子的“Y”字型结构和性质,左右对称;多个融合点,活性窗可调:可以根据不同靶标,改变scFv在IgG不同结构域之间的位置,以便筛选最佳活性分子;双价双靶标,强效细胞桥:完整的FIST分子分别有两对结合臂,每对都是双价的;结构超稳定,长效半衰期:得益于其类IgG的对称结构和Fc部分的存在,FIST分子具有类似于经典IgG的理化性质和超长的体内半衰期,因而带来长效效应;哺乳细胞生产,成熟工艺路线:FIST表达、生产和纯化系统高度类似于经典IgG工艺。总之,本发明的FIST平台产生的双抗/多抗具有多个优点:无错配、高产、长效、活性可调、分子形式因靶而变等等,其中几个优点都是其它非对称分子所不具备的。
本发明的FIST双抗,具有显著不同于其它双抗类别的特点,相互比较如下表1所示。

可以看出,相比于其它双抗平台,FIST首先具有生产方面的显著优势。
本发明的目的在于以FIST平台提供一种新的靶向TROP2和CD3的双特异性抗体(及其活性片段)及其制备方法与应用。
本发明研究发现,具有本发明对称结构的双特异性抗体与其它结构的双特异性抗体相比,能够更好地保留第一功能单位和第二功能单位的各自的特异性抗原结合能力,同时具有优异的成药性和同时结合TROP2和CD3表达细胞的细胞桥功能,在生产工艺和药用性能等方面具有明显优势。本发明开发的具有特定抗体分子结构的结合TROP2和CD3的双特异性抗体,该双特异性抗体具有特异性的靶向作用,并能够高效激发、导向T细胞免疫反应,杀伤高表达TROP2的肿瘤细胞。
具体地,本发明提供以下技术方案:
第一方面,本发明提供靶向TROP2和CD3的双特异性抗体,所述双特异性抗体具有(2+2)对称形式的类IgG结构,所述双特异性抗体包括:一对结合TROP2(人滋养层细胞表面抗原2)的Fab分子、结合人T细胞表面抗原CD3的2条序列相同的单链抗体(scFv)、铰链区(Hinge)以及Fc;2条所述单链抗体的N端分别通过连接肽与Fab分子的两个CH1(第一恒定区)的C端连接,2条所述单链抗体的C端分别通过连接肽与铰链区中2条铰链的N端连接。
本发明的双特异性抗体,所述Fab分子包括重链可变区和轻链可变区,所述重链可变区具有如SEQ ID NO.3所示的氨基酸序列,所述轻链可变区具有如SEQ ID NO.4所示的氨基酸序列;所述单链抗体具有如SEQ ID NO. 7-10任一条所示的氨基酸序列;
和/或,所述双特异性抗体的轻链具有如SEQ ID NO.6所示的氨基酸序列。
本发明的双特异性抗体,所述Fc具有如SEQ ID NO.11或SEQ ID NO.12所示的氨基酸序列。
本发明的所述双特异性抗体具有SEQ ID NO. 13-20任一条所示的重链氨基酸序列和SEQ ID NO. 6所示的轻链氨基酸序列。
本发明提供的抗体可变区氨基酸序列模式为FR1 - CDR1 - FR2 - CDR2 - FR3 -CDR3 - FR4。在本发明中,FR和CDR的区域划分基于Kabat命名系统。在此,FR1~4代表4个框架区域,CDR1~3代表3个高变区。FR1~4可以是从恒定区序列分离而来(比如人免疫球蛋白轻重链类、亚类或亚家族最常用的氨基酸),也可以是从单独的人抗体框架区分离而来或从不同的框架区基因组合而来。
在上述重链和轻链的基础上,本发明具体提供这些抗TROP2/CD3的双抗抗体的重链和轻链的序列的克隆:
重链具有SEQ ID NO.13所示的氨基酸序列,轻链具有SEQ ID NO.6的氨基酸序列;

本发明提供的抗体结合于表达TROP2的细胞,所述细胞可以为人上皮恶性肿瘤(胃癌、宫颈癌、乳腺癌、肺癌、前列腺癌、结肠癌、食道癌、胰癌、头颈癌、卵巢癌、子宫内粘膜浆液性乳头癌等肿瘤)细胞。
本发明的双特异性抗体中,所述连接肽的氨基酸序列为(GGGGX)n,其中,X为Gly或Ser,n为1-5的自然数(SEQ ID NO.21:GGGGX)。
优选,单链抗体中重链可变区和轻链可变区中的连接肽为(GGGGX)n,其中,X为Ser,n为3。
以上所公开和要求保护的SEQ ID NO.1-21所示序列包括“保守序列修饰”,即不明显影响和改变所述抗体或含有所述氨基酸序列抗体的结合特征的核苷酸和氨基酸序列修饰。所述保守序列修饰包括核苷酸或氨基酸替换、添加或缺失。可以通过本领域的标准技术,如定点诱变和PCR介导的诱变等将修饰导入其中,保守氨基酸替换包括氨基酸残基被具有相似侧链的氨基酸残基或其他氨基酸残基代替。本领域中,已经定义了具有相似侧链的氨基酸残基的家族。这些家族包括具有碱性侧链的氨基酸(如赖氨酸、精氨酸、组氨酸),具有酸性侧链的氨基酸(如天冬氨酸、谷氨酸),具有不带电的极性侧链的氨基酸(如甘氨酸、天冬酰胺、谷氨酰胺、丝氨酸、苏氨酸、酪氨酸、半胱氨酸、色氨酸),具有非极性侧链的氨基酸(如丙氨酸、缬氨酸、亮氨酸、异亮氨酸、脯氨酸、苯丙氨酸、甲硫氨酸),具有β分支侧链的氨基酸(如苏氨酸、缬氨酸、异亮氨酸)和具有芳香侧链的氨基酸(如酪氨酸、苯丙氨酸、色氨酸、组氨酸)。因此,优选用来自于同一侧链家族的另一种氨基酸残基代替人抗TROP2抗体中的非必须氨基酸残基。
因此,以上公开的具有所述氨基酸序列的抗体和/或含有以上公开的氨基酸序列的抗体包括基本上由经保守序列修饰的相似序列编码的,或含有经保守序列修饰的相似序列的抗体,均应视为本发明的范畴。
第二方面,本发明提供一种核酸分子,其编码上述双特异性抗体。
考虑到密码子的简并性,编码本发明所述抗体的基因可在其编码区,在不改变氨基酸序列的条件下,对编码上述抗体的基因序列进行修改,获得编码相同抗体的基因。本领域技术人员可以根据表达抗体宿主的密码子偏爱性,人工合成改造基因,以提高抗体的表达效率。
第三方面,本发明提供含有上述核酸分子的生物材料,所述生物材料包括重组DNA、表达盒、载体、宿主细胞、工程菌或细胞系。
其中,所述载体包括但不限于克隆载体、表达载体,可为质粒载体、病毒载体、转座子等载体。
所述宿主细胞或细胞系可以为来源于微生物或动物的细胞或细胞系。
第四方面,本发明提供一种制备上述双特异性抗体的方法,其包括:将编码所述双特异性抗体的核酸导入宿主细胞中,获得稳定表达所述双特异性抗体的宿主细胞;培养所述宿主细胞,经分离纯化获得所述双特异性抗体。
在制备上述双特异性抗体时,本领域技术人员可根据需要选择本领域常规的宿主细胞、表达载体、将表达载体导入宿主细胞的方法以及抗体的分离纯化方法。
第五方面,本发明提供一种产品,其包含上述双特异性抗体,所述产品为多特异性抗体、融合蛋白、免疫毒素、药物、检测试剂或检测试剂盒。
以上所述的免疫毒素包含以各种形式连接于细胞毒性剂的双特异性抗体。
所述的各种连接形式为抗体被标记、体外交联或分子偶联。所述细胞毒性剂包括化学分子、放射性同位素、多肽、毒素及其他对细胞具有杀伤或诱导细胞死亡特性的物质。
以上所述的融合蛋白,包含本发明提供的上述双特异性抗体以及具有某种功能的其他蛋白或多肽分子的复合物。
具体地,所述的融合蛋白可为将抗体基因与免疫毒素或细胞因子基因连接构建重组表达载体,通过哺乳动物细胞或其他表达系统获得重组融合蛋白分子。
以上所述的所述药物、检测试剂还可包含药学领域和检测试剂领域允许的其它有效成分或辅料。例如:抗体的组分和药理学上可接受的递送分子或溶液。其中,治疗用组分为无菌,可低温冻干。
所述试剂盒用于检测CD3和/或TROP2。上述双特异性抗体可与CD3和/或TROP2蛋白进行结合,可用于定性或定量检测生物样本中的CD3和/或TROP2蛋白。
第六方面,本发明提供上述双特异性抗体或核酸分子或生物材料的如下任一种应用:
(1)在制备用于诊断、预防或治疗TROP2表达的相关疾病的药物中的应用;
(2)在制备用于诊断、预防或治疗以TROP2为靶标的疾病的药物中的应用;
(3)在制备用于杀伤TROP2表达的细胞的药物中的应用;
(4)在制备TROP2和/或CD3的检测试剂或试剂盒中的应用;

以上所述的TROP2表达的相关疾病优选为高表达/过度表达TROP2的肿瘤,特别是表达TROP2的上皮恶性肿瘤,包括但不限于胃癌、宫颈癌、乳腺癌、肺癌、前列腺癌、结肠癌、食道癌、胰癌、头颈癌、卵巢癌、子宫内粘膜浆液性乳头癌等肿瘤。
优选地,所述药物为抗肿瘤药物。
以上所述的标记抗体可为放射性同位素标记抗体。
本发明提供的双特异性抗体本身可用于治疗和诊断。抗体可以被标记、交联或偶联及与其他蛋白或多肽分子融合表达形成复合物(如细胞毒性物质、放射性毒素和/或化学分子等)用于诊断和治疗。
本发明提供的双特异性抗体,能够抑制TROP2所诱导的一种或多种生物学活性。这些抗体通过与TROP2结合后,复合物被内化从而消耗细胞表面的TROP2而发挥作用。TROP2拮抗剂所具有的所有干扰功能均应平等地视为本发明的目的。

本发明利用基因工程和抗体工程方法构建包含单链抗体和完整单克隆抗体结构的结合TROP2和CD3的双特异性抗体,该双特异性抗体融合蛋白保留了完整的单克隆抗体结构,而且具有高度稳定的对称结构,更好地保留了抗CD3单链抗体和抗TROP2单克隆抗体的生物学功能,实现了一个双特异性抗体分子同时具有优异的抗TROP2和抗CD3两个单克隆抗体的生物学功能,能够在肿瘤细胞和免疫效应细胞之间搭建桥梁,有效激活免疫效应细胞和导向性免疫反应,显著增强了免疫细胞杀伤肿瘤细胞的功效,同时基本消除了ADCC效应和CDC效应,具有较高的安全性。
此外,本发明提供的双特异性抗体由于具有结构完全对称的特点,在进行宿主表达时,不会产生其它结构的蛋白异构体,从而大大降低了提取和纯化工艺的难度,具有制备简单、产量高的优势,在肿瘤的免疫治疗中具有广阔的应用前景,为以TROP2为靶标的临床肿瘤免疫治疗抗体药物开发提供了基础。
本发明的双特异性抗体能够同时结合免疫细胞和肿瘤细胞,导向T免疫反应,特异性有效杀伤肿瘤细胞,可研发为上皮恶性肿瘤的抗体药物。

具体实施方式
下面将结合实施例对本发明的优选实施方式进行详细说明。需要理解的是以下实施例的给出仅是为了起到说明的目的,并不是用于对本发明的范围进行限制。本领域的技术人员在不背离本发明的宗旨和精神的情况下,可以对本发明进行各种修改和替换。
实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规产品。
实施例1 FACS分析验证TROP2特异性抗体hRS7与TROP2细胞的结合
hRS7抗体的氨基酸序列来自文献(中国专利授权公告号 CN 101264325 B)。为了验证hRS7是否结合纯化的TROP2抗原以及细胞膜表面表达的TROP2,hRS7抗体的IgG重链基因及轻链基因(其编码氨基酸序列如SEQ ID NO.5和6所示)被克隆至真核表达载体pCEP4(赛默飞,ThermoFisher Scientific),共转染表达,得到hRS7蛋白。用Protein-A进行亲和纯化,也可以用(HRP或荧光素)标记抗人Fc抗体进行检测。
取表达TROP2的乳腺癌细胞系MDA-MB-468细胞,按1 × 106细胞/反应管重悬于1%BSA的PBS中,以1 ml 结合缓冲液(含有0.5% w/v BSA+2 mM EDTA的PBS)漂洗细胞一次,离心(350 ×g,4℃,5 min),后用200 µl 结合缓冲液重悬细胞。二抗对照样品管加入200 µl 结合缓冲液,待测样品管加入特异抗体上述制备获得的hRS7蛋白至终浓度为5 µg/ml,冰上孵育30 min。如上漂洗细胞3次后用100 µl结合缓冲液重悬细胞。所有样品管加入5 µl荧光标记二抗抗体(Goat anti-human IgG Fc PE Antibody, Ebioscience)。如上漂洗细胞一次。用200 µl PBS重悬细胞后,上机(Cytoflex,Beckman)检测。对MDA-MB-468细胞系的流式分析图如图2所示。图2的左图为对照组检测结果,右图为实验组检测结果。
实施例2 抗CD3单链抗体人源化克隆的结合验证
抗人CD3特异性抗体来自专利WO 2016/116626 Al中描述的克隆18F5。其抗体轻链可变区和重链可变区序列分别如SEQ ID NO. 2 和SEQ ID NO. 1所示。18F5的可变区经本发明特定人源化改造后获得了四株亲和力不同的单链抗体克隆,分别命名为18F5-A,18F5-B,18F5-C,18F5-D,这些单链抗体序列依次如SEQ ID NO. 7~10所示。
对获得的CD3抗体克隆18F5-A,18F5-B,18F5-C,18F5-D对人T细胞表面CD3抗原的结合进行验证。将18F5-A,18F5-B,18F5-C,18F5-D的单链抗体形式(VH-linker-VK)基因克隆至包含有人Fc的pcDNA3.1中,转染、表达纯化后获得scFv-Fc形式的蛋白。以流式细胞仪检测了这些scFv-Fc对表达CD3的人T细胞(Jurkat)的结合。具体方法如下:取培养的Jurkat细胞,按1 × 106细胞/管重悬于1%BSA的PBS中,以1 ml结合缓冲液(含有0.5% w/v BSA+2mM EDTA的PBS)漂洗细胞一次,离心(350 ×g,4℃,5 min),后用200 µl结合缓冲液重悬细胞。二抗对照样品管加入200 µl结合缓冲液,待测样品管分别加入特异抗体18F5-A,18F5-B,18F5-C,18F5-D(scFv-Fc形式)至终浓度为0.125 µg/ml,冰上孵育30 min。如上漂洗细胞3次后用100 µl结合缓冲液重悬细胞。所有样品管加入5 µl荧光标记二抗抗体(Goat anti-human IgG Fc PE Antibody, Ebioscience)。如上漂洗细胞一次。用200 µl PBS重悬细胞后,上机(Cytoflex,Beckman)检测。对Jurkat细胞系的流式分析图如图3所示。FACS结果证实了这些人源化改造的克隆18F5-A,18F5-B,18F5-C,18F5-D都能特异性结合人T细胞(见图3),验证了这些单链抗体与人T细胞表面抗原CD3的结合能力。
实施例3 TROP2×CD3双特异性抗体设计
本实施例以肿瘤细胞表面抗原TROP2和免疫细胞表面抗原CD3作为靶点,设计一种独特的双特异性抗体,同时识别TROP2表达细胞和表达CD3的T细胞,用于TROP2高表达的临床肿瘤的免疫治疗。
结合蛋白质结构设计软件和大量的人工实验筛选,本发明在多种结合TROP2和CD3的双特异性抗体结构中筛选确定了由两个结合单位组成的具有对称结构的双特异性抗体结构,本发明的这种技术平台称之为FIST(fusion of IgG and scFv technology)。具体而言,结合CD3的单链抗体功能单位插入在结合TROP2的第一功能单位(Fab)和Fc之间,这种双抗组建方式命名为mFIST。其完整分子包括抗TROP2单克隆抗体的Fab、铰链区和Fc。两条CD3特异性单链抗体的N端分别通过连接肽与抗TROP2抗体的2条重链的第一恒定区(CH1)的C端连接,两条CD3特异性单链抗体的C端分别通过连接肽与抗TROP2抗体的铰链区(hinge)的2条铰链的N端连接。由此获得对称结构的mFIST双特异性抗体,其结构示意图如图4所示。
本发明mFIST设计的双抗让两个抗体结合单元具有适中的空间距离,同时具有对称型结构,而且两个结合单元的功能都能得到完全体现。Fc的效应功能、长半衰期功能赋予了所产生双抗的优异生物学性质和药学性质,是研发双抗药物的最优选择之一。
经典的抗体Fc介导的效应机制如补体依赖的细胞毒作用(CDC)、抗体依赖性细胞介导的细胞毒作用(ADCC)和抗体依赖性细胞吞噬作用(ADCP)是消除病原体或病变细胞(如感染或癌细胞)的关键机制。IgG抗体有4种亚型:IgG1、IgG2、IgG3、IgG4,尽管二硫键的位置和数目不同,但4种亚型抗体的空间结构很相似,含量依次减少。IgG1是血浆中含量最多的一种亚型。它是肿瘤免疫治疗中最具潜力的亚型,是一种比较理想的可工业化生产(GMP)的抗体。IgG1是现阶段最常被使用的Fc亚型,科学家在此基础上通过Fc工程化策略来优化其功能特性,稳定性和药代动力等特征。IgG2主要用来中和抗原或阻断受体配体的结合,其CDC和ADCC效应表现非常弱。IgG3有一个延长的铰链区域,其核心铰链区有11对二硫键,因此对于蛋白酶切割不稳定。IgG3与FcγRs的结合能力最强,能引发ADCC和ADCP,且CDC效应比IgG1更强。IgG4分子的铰链区较短,且其与FcγRI(CD64)之外的FcγRs结合较弱。IgG4分子不能引起CDC和NK细胞介导的ADCC。
Fc类别因不同亚型而具有不同强度的体内效应功能。本发明中的mFIST形式的CD3免疫双抗交联T细胞和癌细胞形成复合体。为降低/消除其中的Fc效应功能对T细胞的损伤,有必要选择天然效应功能微弱的Fc(如IgG4的Fc),或者通过基因工程改造、效应功能进一步降低的工程Fc(沉默Fc,SL-Fc)。然而,优化的Fc可能具有预期之外的理化性质,如稳定性降低等,需要在开发研究中进行验证。
本发明FIST平台中双抗分子中单链抗体(scFv)对相应抗原的结合是活性可调的,这个可调性是通过两个手段实现的:亲和力调节及距离调节。在亲和力上,单个或一对单链抗体(scFv)对抗原(如本发明中对CD3亚基)的结合比较弱,其亲和力弱于10-7M,而Fab端对相应抗原(结合本发明中的TROP2)的结合,即使是单价,内在亲和力(affinity)也达到10- 9M。因此双抗分子在血流中将首先结合表达TROP2的癌细胞,并在癌细胞表面形成簇状集束(cluster),此时通过表观亲和力(avidity)才与附近巡游的T细胞结合,从而局部活化T细胞并介导T细胞增殖及杀伤。
FIST双抗调节结合活性的第二个机制是改变单链抗体单元(scFv)和IgG单元的Fab之间距离来实现的。结构上,单链抗体可以插入/融合到IgG的多个位置,包括轻链的N端、C端,重链可变区的N端,Fc的C端,CH1的C端及铰链区N端之间等。不同的融合方法对双抗的功能有不同的影响,包括能否同时结合两个抗原,以及能否保持原有的结合力。本发明中选择mFIST形式,是在综合内部其它工作的基础上选择的最好形式。考虑了不同的TROP2靶标表达水平,综合双抗活性/安全性各个因素,选择了亲和力不同的结合CD3的单链抗体与结合TROP2的IgG进行了融合。
实施例4 TROP2×CD3双特异性抗体的制备
将实施例1中hRS7抗体可变区基因(SEQ ID NO. 3和SEQ ID NO. 4)分别与实施例2中的18F5的四株人源化克隆基因(SEQ ID NO. 7-10),以及两种不同类型的Fc(IgG4的Fc和效应功能沉默突变的Fc,SEQ ID NO. 11-12)进行组合,获得根据实施例3中设计的mFIST形式的8种双特异性抗体。将它们的抗体编码基因克隆至表达载体pCDNA3.1,得到的8个双特异性抗体克隆YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7。将这些mFIST双抗表达质粒转染CHO-K1,表达制备双抗蛋白。YT03-A-hRS7的重链(HC)序列如SEQ ID NO.13所示,YT03-A-hRS7的轻链(LC)序列如SEQ ID NO.6所示,YT03-B-hRS7的HC序列如SEQ ID NO.14所示,YT03-B-hRS7的LC序列如SEQ ID NO.6所示,YT03-C-hRS7的HC序列如SEQ ID NO.15所示,YT03-C-hRS7的LC序列如SEQ ID NO.6所示,YT03-D-hRS7的HC序列如SEQ ID NO.16所示,YT03-D-hRS7的LC序列如SEQ ID NO.6所示。SL YT03-A-hRS7重链序列如SEQ ID NO.17所示,SLYT03-A-hRS7的轻链序列如SEQ ID NO.6所示。SL YT03-B-hRS7重链序列如SEQ ID NO.18所示,SL YT03-B-hRS7的轻链序列如SEQ ID NO.6所示。SL YT03-C-hRS7重链序列如SEQ IDNO.19所示,SL YT03-C-hRS7的轻链序列如SEQ ID NO.6所示。SL YT03-D-hRS7重链序列如SEQ ID NO.20所示,SL YT03-D-hRS7的轻链序列如SEQ ID NO.6所示。这些分子统称为YT03序列双抗。
为获得高纯度的双特异性抗体,按照以下步骤进行纯化。(1)料液预处理:发酵培养的上清液通过2000 rpm离心,10 min,然后用0.22 μM滤膜过滤处理。(2)亲和层析:用MabselectSuRe亲和层析柱(购自GE公司)捕获经过预处理的发酵液中的抗体,用平衡缓冲液(10 mM PB,0.1 M NaCl,pH7.0)充分平衡层析柱后,过亲和层析柱,用洗脱缓冲液(0.1 M柠檬酸酸,pH 3.0)洗脱。(3)阳离子交换层析:经亲和层析制备的样品,进一步通过分子筛交换层析纯化,缓冲液(50 mM PBS,0.2 M Na2SO4,pH 6.7)。
将YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SLYT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7经表达和纯化成功制备得到了8个双特异性抗体,SDS-PAGE分析结果如图5所示,其中以抗体YK012作为参考,该抗体可购自益科思特(北京)医药科技发展有限公司。
实施例5 流式分析法检测双特异性抗体与TROP2阳性细胞及人T细胞的结合
为验证YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7系列抗体(YT03系列双抗分子)对TROP2细胞系及T细胞的结合,以FACS方法分析它们对表达CD3的人T细胞及表达TROP2的乳癌细胞系的结合。
取乳腺癌细胞系MDA-MB-468和CD3+T(Jurkat E6.1)细胞,按1 × 106细胞/反应管重悬于 1%BSA的PBS中,以1 ml 结合缓冲液(含有0.5% w/v BSA+2 mM EDTA的PBS)漂洗细胞一次,离心(350 ×g,4℃,5 min),后用100 µl 结合缓冲液重悬细胞。按细胞管分别加入各个双特异抗体YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7至两种细胞中,其终浓度在MDA-MB-468细胞中为2.5 µg/ml,在人T细胞中为20µg/ml,均4°C孵育30 min。如上漂洗细胞3次后用100µ1以 1:100稀释的荧光标记二抗抗体(Goat anti-human IgG Fc PE Antibody,Ebioscience)稀释液重悬细胞,4°C孵育30 min。如上漂洗细胞3次。用200 µl PBS重悬细胞后,上机(Cytoflex,Beckman)检测。对MDA-MB-468细胞系的流式分析图如图6所示,结合阳性率及MFI分析结果见表2;对Jurkat E6.1细胞系的流式分析图见图7所示,结合阳性率及MFI分析结果见表3。

经FACS检测证明,上述8个双抗对表达TROP2的靶细胞MDA-MB-468的结合率及MFI相当,功能结合正常。同时,这8个双抗分子都能与表达人CD3分子Jurkat E6.1结合,结合强度彼此间有差异。
实施例6 TROP2/CD3双特异性抗体介导的T细胞活化检测
双特异靶向TROP2和CD3的双抗能同时结合、交联人T细胞和表达TROP2的肿瘤细胞,形成肿瘤细胞-双抗-T细胞的复合体。在此时的复合体中,由于肿瘤抗原的高密度表达,T细胞的CD3受体复合体被肿瘤细胞表达的“集束”TROP2通过双抗分子而交联(crosslinking),从而导致TCR寡聚并触发信号通道活化,激活一系列下游级联反应(例如NFAT信号通道),产生包括T细胞增殖、细胞因子释放、毒性分子释放等多种生物学效应。
为验证本发明中的系列TROP2/CD3双抗的激活T细胞的功能,构建了报告细胞系Jurkat E6.1 NFAT Luc。Jurkat E6.1 NFAT Luc细胞系的构建过程是:含NFAT启动子及荧光素酶编码基因(NFAT-Luc)克隆至pLVX-Puro( pLVX-Puro购自Takarabio),制备包装plvx-NFAT*3-luciferase-puromycin的慢病毒,收病毒上清液;用病毒上清感染JurkatE6.1细胞,48h后加puromycin,1周后进行单克隆化。验证TROP2/CD3双抗的激活活性时,以MDA-MB-468为靶细胞,Jurkat E6.1 NFAT Luc作为免疫效应细胞,分别加入双特异性抗体YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7,孵育后检测。具体实验步骤如下:
分别取30×106的Jurkat E6.1 NFAT Luc细胞,和3× 106的MDA-MB-468细胞,300g离心10min,弃上清;再用10 ml PBS重悬,300g 离心10 min,弃上清;分别用15 ml 10%FBS-1640重悬,Jurkat E6.1 NFAT Luc重悬为2× 106/ml,MDA-MB-468重悬为0.2× 106/ml;Jurkat E6.1 NFAT Luc和MDA-MB-468均取50 μl/孔,加入到96孔板中。测试双抗抗体均稀释至4nM,然后按1:3稀释,制备10个梯度:2-12加入100µl 10% FBS-1640,取133.3µl 起始浓度(8nM),加入到第1列中,混匀;从第1列中取33.3µl到第2列中,混匀;再从第2列中取33.3µl到第3列中,依次稀释至第10列,混匀后弃去33.3 μl。稀释的各抗体按100 μl /孔加入到细胞板里。将抗体和细胞混匀,37℃,二氧化碳培养箱中,培养24h;混匀后,取60 μl细胞悬液,加入60 μl底物,轻拍混匀,用酶标仪检测荧光值,并根据荧光信号-抗体浓度做曲线及各双抗分子的EC50值。
将所有检测孔对应的抗体浓度转变为log10,以此作为横坐标、RLU作为纵坐标制作曲线。YT03系列分子(YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7)对免疫效应细胞Jurkat E6.1NFAT Luc的激活浓度-梯度曲线如图8所示;YT03不同分子对Jurkat E6.1 NFAT Luc表现出不同的激活能力,它们介导激活的EC50如表4所示。

实施例7 TROP2/CD3双特异性抗体介导的T细胞杀伤检测
双特异靶向TROP2和CD3的双抗同时结合人T细胞和表达TROP2的肿瘤细胞,形成肿瘤细胞-双抗-T细胞的复合体。在此时的复合体中,由于TROP2肿瘤抗原的高密度表达,T细胞的CD3受体被肿瘤细胞表达的“集束”TROP2通过双抗分子而交联(crosslinking)TCR,导致T细胞的CD3信号通道活化,T细胞活化后释放毒性分子(颗粒酶B和穿孔素)。毒性分子扩散至T细胞附近的肿瘤靶细胞并发挥杀伤功能。
本实施例以MDA-MB-468-lp为靶细胞验证YT03系列双抗介导的杀伤功能实验。MDA-MB-468-lp细胞系为益科思特公司构建,其简要步骤如下:编码启动子EF1α和萤光素酶报告基因(Luc)的DNA片段(EF1α-Luc)克隆至pLVX-Puro( pLVX-Puro购自Takarabio),包装plvx-EF1α-luc-puromycin慢病毒,收上清;用上清感染MDA-MB-468细胞,48h后加puromycin,1周后进行细胞的单克隆化,获得MDA-MB-468-lp报告细胞系。用anti-CD3/CD28磁珠激活PBMC,得到活化的T细胞作为免疫效应细胞,检测YT03系列双特异性抗体YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SL YT03-A-hRS7,SL YT03-B-hRS7,SLYT03-C-hRS7,SL YT03-D-hRS7介导的T细胞的对表达TROP2的MDA-MB-468-lp的杀伤作用。杀伤实验具体实验步骤如下:
分别取30×106按上述方法活化的T细胞,和3× 106的MDA-MB-468-lp细胞,300g离心10min,弃上清;细胞沉淀再用10ml PBS重悬,300g 离心10min,弃上清;细胞沉淀分别用15 ml X-VIVOTM15重悬。其中T细胞重悬为2× 106/ml,MDA-MB-468-lp重悬为0.2× 106/ml。重悬的T细胞和MDA-MB-468-lp均取50 μl/孔,加入到96孔板中。测试的8个双抗均稀释至0.125 nM,然后按1:3稀释下去,制备10个梯度方法如下:2-12列首先加入100 µl X-VIVOTM15。取133.3µl 起始浓度(0.125nM)双抗样品,加入到第1列中;从第1列中取33.3µl到第2列中,混匀;再从第2列中取33.3µl 到第3列中,依次稀释至第10列,混匀后弃去33.3 μl。稀释的各抗体按100 μl /孔加入到细胞板里。将抗体和细胞混匀,37℃,二氧化碳培养箱中,培养24h;混匀后,取60 μl细胞悬液,加入60 μl底物,轻拍混匀,用酶标仪检测荧光值。
将所有检测孔的靶细胞杀伤比例对应的抗体浓度转变为log10,以此作为横坐标、RLU作为纵坐标制作曲线。YT03-A-hRS7,YT03-B-hRS7,YT03-C-hRS7,YT03-D-hRS7,SLYT03-A-hRS7,SL YT03-B-hRS7,SL YT03-C-hRS7,SL YT03-D-hRS7对敏感细胞株MDA-MB-468-lp的杀伤浓度-梯度曲线如图9所示;YT03不同分子对TROP2+细胞表现出不同的杀伤能力,它们各自介导杀伤的EC50如表5所示。

实施例8 FACS比较不同Fc组成的双抗组对PBMC的结合
人外周血单个核细胞(peripheral blood mononuclear cell,PBMC),其细胞类型为血液里具有单个核的细胞,主要包括淋巴细胞(T细胞、B细胞和NK细胞),单核细胞,吞噬细胞,树突状细胞和其他少量细胞类型。其中淋巴细胞占很大一部分。人的IgG Fc受体有三大类:FcγRI(CD64),FcyRII(CD32)和FcγRIII(CD16)。其中第二类包含IIA/IIB/IIC三个亚类,第三类包含IIIA/IIIB两个亚类。与激活具有杀伤功能的先天免疫细胞(如NK细胞)最相关的受体是IIIA,也被称为CD16A。CD16A表达于NK细胞、单核细胞和巨噬细胞表面,也少量表达于DC细胞表面,属于激活型受体。表达IgG受体的多种血液细胞有可能会代替靶标抗原细胞而寡聚含有Fc的IgG单抗、双抗、ADC等分子。在T细胞免疫双抗项目中有可能直接造成T细胞的活化,从而带来非特异性的细胞毒性释放和免疫系统的(低水平)活化。为考察本发明中两种不同Fc的TROP2/CD3双抗在和PBMC孵育时是否会由于Fc受体结合而导致T细胞的活化,以YT03-C-hRS7和 SL YT03-C-hRS7为例,检测了PBMC和双抗孵育后T细胞活化标志分子CD69的水平。具体的研究方法如下:
SL YT03-C-hRS7和YT03-C-hRS7的浓度从1000 ng/ml起始进行4倍稀释,总共6个梯度。稀释方法是,除首个浓度外,后续5个浓度,按1:3稀释(167 µl+500 µl,混匀后,取167µl到下一个梯度),每孔加入抗体0.5 ml;然后加入0.5×106/孔的PBMC,0.05×106/孔MDA-MB-468细胞,总体积1 ml/孔,将抗体与细胞混匀。
平行于上述操作,设置仅使用稀释的双抗和PBMC,不添加MDA-MB-468的组别。设置对照孔如下:PBMC,PBMC+ MDA-MB-468。均放置在37℃ CO2培养箱中,培养120小时。
收集细胞进行流式检测,将样品混匀后收集到1.5ml EP管中,离心去上清,加入1ml 1% PBSB重悬,洗一次,离心去上清;检测抗体均为3 µl /test,配制: APC anti-humanCD69(Biolegend),FITC anti-human CD4(Biolegend)。抗体预混液的1% PBSB溶液,混匀后,100 µl /test重悬样品;需要单染的样品单独加抗体;细胞标记分组如下:
PBMC+ MDA-MB-468 +YT03-C-hRS7不染色,PBMC+MDA-MB-468+ YT03-C-hRS7 染CD4,PBMC+ MDA-MB-468+ YT03-C-hRS7 染CD69,PBMC+ MDA-MB-468+ YT03-C-hRS7 染CD4+CD69,PBMC+ MDA-MB-468+ SL YT03-C-hRS7不染色,PBMC+ MDA-MB-468+ SL YT03-C-hRS7染CD4,PBMC+ MDA-MB-468+ SL YT03-C-hRS7 染CD69,PBMC+ MDA-MB-468+ SL YT03-C-hRS7 染CD4+CD69。其它样品染CD4+CD69。4℃孵育30 min,1ml 1% PBSB洗三次,用250 µl1% PBSB重悬后,上机检测,获得各组CD69 T细胞阳性群百分数曲线图(图10)。
本发明的FIST平台显然不是简单的IgG+scFv形式,而是通过多个设计细节组成,包括融合形式、抗体选择、Fc选择、链接肽组成和长度、抗体亲和力等等,这些技术细节是决定本发明特定双特异性抗体能否成功开发的关键所在。此外,尽管FIST的2+2结构可以通过传统的化学交联法实现,但化学交联法难以固定交联位点和交联分子比例,在工艺上、成本上很难过关,因而本发明采用了如上制备方式。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (10)
1.靶向TROP2和CD3的双特异性抗体,其特征在于,所述双特异性抗体具有对称形式的类IgG结构,所述双特异性抗体包括:一对结合TROP2的Fab分子、结合人T细胞表面抗原CD3的2条序列相同的单链抗体、铰链区以及Fc;2条所述单链抗体的N端分别通过连接肽与Fab分子的两个CH1的C端连接,2条所述单链抗体的C端分别通过连接肽与铰链区中2条铰链的N端连接。
2.根据权利要求1所述的双特异性抗体,其特征在于,所述Fab分子包括重链可变区和轻链可变区,所述重链可变区具有如SEQ ID NO.3所示的氨基酸序列,所述轻链可变区具有如SEQ ID NO.4所示的氨基酸序列;所述单链抗体具有如SEQ ID NO. 7-10任一条所示的氨基酸序列;
和/或,所述双特异性抗体的轻链具有如SEQ ID NO.6所示的氨基酸序列。
3.根据权利要求2所述的双特异性抗体,其特征在于,所述Fc具有如SEQ ID NO.11或SEQ ID NO.12所示的氨基酸序列。
4.根据权利要求1-3任一项所述的双特异性抗体,其特征在于,所述双特异性抗体具有SEQ ID NO. 13-20任一条所示的重链氨基酸序列和SEQ ID NO. 6所示的轻链氨基酸序列。
5.根据权利要求1所述的双特异性抗体,其特征在于,所述连接肽的氨基酸序列为(GGGGX)n,其中,X为Gly或Ser,n为1-5的自然数。
6.一种核酸分子,其特征在于,其编码权利要求1-5任一项所述的双特异性抗体。
7.含有权利要求6所述的核酸分子的生物材料,所述生物材料包括重组DNA、表达盒、载体、宿主细胞、工程菌或细胞系。
8.一种制备权利要求1-5任一项所述的双特异性抗体的方法,其特征在于,包括:将编码所述双特异性抗体的核酸导入宿主细胞中,获得稳定表达所述双特异性抗体的宿主细胞;培养所述宿主细胞,经分离纯化获得所述双特异性抗体。
9.一种产品,其特征在于,包含权利要求1-5任一项所述的双特异性抗体,所述产品为多特异性抗体、融合蛋白、免疫毒素、药物、检测试剂或检测试剂盒。
10.权利要求1-5任一项所述的双特异性抗体或权利要求6所述的核酸分子或权利要求7所述的生物材料的如下任一种应用:
(1)在制备用于诊断、预防或治疗TROP2表达的相关疾病的药物中的应用;
(2)在制备用于诊断、预防或治疗以TROP2为靶标的疾病的药物中的应用;
(3)在制备用于杀伤TROP2表达的细胞的药物中的应用;
(4)在制备TROP2和/或CD3的检测试剂或试剂盒中的应用;
(5)在制备适用于CAR-T疗法的相关试剂中的应用;
(6)在制备免疫毒素或标记抗体中的应用;
(7)在制备抗体-药物偶联物中的应用。
