Background
The acne is clinically said, the previous acnes are mainly caused by excessive secretion of androgens in puberty, the cutin becomes thick under the action of the androgens, and meanwhile, sebaceous glands secrete a large amount of sebum to cause pores to be gradually blocked and accumulated continuously, so that cutin blocking particles are commonly called acnes, and bacteria are continuously bred in the acnes to form acnes with abscess.
The existing acne removing product can solve the problems of excessive sebum secretion, keratosis of hair follicle, pore blockage, inflammation caused by acne-related bacteria infection, grease degradation and the like, and can achieve the effects of bacteriostasis and oil control in a short time. For example, CN109528622a announces an anti-acne repair essence comprising dendrobium extract, matrine, leuconostoc/radish root fermentation product filtrate, transdermal EGF and the like. However, the whole course care of the acne skin is safe and effective aiming at the physiological and pathological characteristics of the acne skin, and further optimized and rich formulas are needed.

The pharmacological actions of the raw materials in the composition are as follows:
aloe vera has the effects of moistening, caring skin, whitening skin, preventing sunburn, and repairing skin; secondly, has the functions of resisting aging and detoxification; furthermore, the composition has the functions of immunity, anti-tumor, sterilization and anti-inflammatory; finally has the effects of easing pain, calming and deodorizing.

Viola yedoensis makino: bitter and pungent taste, cold property, heart and liver meridian homing, and has the effects of clearing heat and detoxicating, cooling blood and detumescence, and the like, and is clinically used for furuncle and pyogenic infections, carbuncles and back, erysipelas, venomous snake bite, and the like.
Chinese angelica root: has effects of regulating blood, promoting blood circulation, relieving inflammation and inhibiting bacteria, and can be used for reducing vaccinia formation and inflammation.
Rhizoma smilacis glabrae: clearing heat and detoxicating, eliminating heat toxin in the body, and reducing the generation and development of acnes; induce diuresis to alleviate edema, remove dampness and edema in vivo, and relieve red and swelling symptoms of acne.
Fructus Gleditsiae Abnormalis thorn: contains flavonoid compounds, and has antibacterial, repercussive, toxic materials removing, pus expelling, and mite killing effects.
Radix paeoniae rubra: promoting blood circulation, effectively preventing acne and macula, whitening facial skin, lubricating and luster, and simultaneously having obvious scar removing effect.
Pseudo-ginseng: swelling relieving, blood stasis dispelling, pain relieving, skin topical blood circulation promoting, pigmentation and metabolism promoting effects, and can be used for eliminating acne marks and eliminating scar.
Ligusticum wallichii: it can be combined with Chi Shao to dispel liver and blood, promote blood circulation and remove blood stasis.
Fructus Cnidii: warming kidney, supporting yang, eliminating dampness, dispelling pathogenic wind, removing acarid, and relieving itching.
The second aim of the application is to provide a preparation method of the antibacterial acne-removing composition, which has the advantages of simple preparation and convenient operation, and comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, and sieving with 20-40 mesh sieve to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propylene glycol, 1, 3-butanediol and pure water, and uniformly stirring for later use;
(3) Adding the medicinal powder and the extraction solvent into an extraction tank according to the weight ratio of (5-10), uniformly stirring, and soaking for 100-150min to obtain a mixed material;
(4) Heating the mixture to 50-60deg.C, stirring, ultrasonic extracting for 30-45min, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, and sterilizing to obtain the antibacterial acne-removing composition.
Preferably, the extraction solvent in the step (2) consists of 1, 3-propanediol, 1, 3-butanediol and pure water according to the weight ratio of 15:15:70.
Preferably, the refrigeration temperature in step (5) is 5-8 ℃.
The third object of the application is to provide an application of the antibacterial acne-removing composition, in particular to an application of the antibacterial acne-removing composition to skin care products, wherein the skin care products are any one of water aqua, powder, cream and gel, and the antibacterial acne-removing composition has the advantage of wide application range.
The application has the following beneficial effects:
(1) The coptis chinensis, the chrysanthemum, the fructus kochiae, the patrinia villosa and the viola philippica are compounded to be used for clearing heat and cooling blood, purging pathogenic fire and removing toxin, maintaining endocrine balance and providing an environment which is unfavorable for the growth of acnes from the source; the Chinese rose, the pseudo-ginseng and the ligusticum wallichii are utilized to promote blood circulation and promote menstruation, quicken the renewal of the horny layer, avoid the thickening of horny layer, and meanwhile, the dandelion balances the secretion of grease, dredges pores and avoids the formation of horny blocking particles; the aloe vera, the angelica sinensis, the glabrous greenbrier rhizome and the gleditsia sinensis are compounded to relieve pain, relieve itching, resist inflammation and inhibit bacteria, so that the problem of inflammation caused by bacterial infection and grease degradation of skin is prevented; the red paeony root is utilized to promote blood circulation, whiten and lubricate the facial skin and repair scars; aiming at the physiological and pathological characteristics of the skin, the whole-course comprehensive care of the acne skin is realized by starting from the sources of the oil control and the cutin metabolism of the acne skin, balancing and controlling the oil, treating the cutin, sterilizing and diminishing inflammation, repairing and regenerating, improving the immunity and the like.
(2) The low-irritation plants are selected as main acne-removing raw materials, and an ultrasonic heating extraction method is adopted to efficiently quench active ingredients of the plants.
(3) The antibacterial acne-removing composition is added into the skin care product, so that the acne-removing effect of the skin care product is improved, and meanwhile, when the skin care product is used, the skin repairing and skin barrier function recovering effects can be enhanced, and the wide application of the antibacterial acne-removing composition is facilitated.
Detailed Description
The present application will be further described with reference to examples, but the scope of the application as claimed is not limited to the scope expressed by the examples.
Example 1
An antibacterial acne-removing composition comprises the following raw materials: 20g of aloe vera, 10g of coptis chinensis, 15g of chrysanthemum, 50g of dandelion, 15g of osmanthus fragrans, 20g of patrinia villosa, 20g of viola philippica, 15g of angelica sinensis, 20g of smilax glabra, 10g of spina gleditsiae, 20g of radix paeoniae rubra, 10g of pseudo-ginseng, 20g of ligusticum wallichii and 20g of fructus cnidii.
The preparation method of the antibacterial acne-removing composition comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, sieving with a sieve with fineness of 40 meshes, and continuously pulverizing the residue until the residue is completely sieved to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propanediol, 1, 3-butanediol and pure water according to the weight ratio of 15:15:70, and uniformly stirring for standby;
(3) Adding the medicinal powder and an extraction solvent into an extraction tank according to the weight ratio of 1:5, uniformly stirring, and soaking for 150min to obtain a mixed material;
(4) Heating the mixture to 50deg.C, stirring, ultrasonic extracting at 28KHZ and 1200W for 45min, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate at 5-8deg.C for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, pumping into a jacketed stainless steel pan, maintaining the temperature in water bath to 90deg.C+ -2deg.C, and sterilizing for 30min to obtain the antibacterial acne removing composition.
Example 2
An antibacterial acne-removing composition comprises the following raw materials: 15g of aloe vera, 7g of coptis chinensis, 12g of chrysanthemum, 45g of dandelion, 12g of osmanthus fragrans, 15g of patrinia villosa, 15g of viola philippica, 12g of angelica sinensis, 15g of smilax glabra, 7g of spina gleditsiae, 15g of radix paeoniae rubra, 7g of pseudo-ginseng, 15g of ligusticum wallichii and 15g of fructus cnidii.
The preparation method of the antibacterial acne-removing composition comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, sieving with a 20-mesh sieve, and continuously pulverizing the residue until the residue is completely sieved to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propanediol, 1, 3-butanediol and pure water according to the weight ratio of 15:15:70, and uniformly stirring for standby;
(3) Adding the medicinal powder and an extraction solvent into an extraction tank according to the weight ratio of 1:10, uniformly stirring, and soaking for 120min to obtain a mixed material;
(4) Heating the mixture to 60 ℃, starting stirring, ultrasonically extracting for 30min at the parameters of 28KHZ and 1200W, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate at 5-8deg.C for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, pumping into a jacketed stainless steel pan, maintaining the temperature in water bath to 90deg.C+ -2deg.C, and sterilizing for 30min to obtain the antibacterial acne removing composition.
Example 3
An antibacterial acne-removing composition comprises the following raw materials: 25g of aloe vera, 13g of coptis chinensis, 18g of chrysanthemum, 55g of dandelion, 18g of osmanthus fragrans, 25g of patrinia villosa, 25g of viola yedoensis, 18g of angelica sinensis, 25g of smilax glabra, 13g of spina gleditsiae, 25g of radix paeoniae rubra, 13g of pseudo-ginseng, 25g of ligusticum wallichii and 25g of fructus cnidii.
The preparation method of the antibacterial acne-removing composition comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, sieving with a sieve with fineness of 30 meshes, and continuously pulverizing the residue until the residue is completely sieved to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propanediol, 1, 3-butanediol and pure water according to the weight ratio of 15:15:70, and uniformly stirring for standby;
(3) Adding the medicinal powder and an extraction solvent into an extraction tank according to the weight ratio of 1:8, uniformly stirring, and soaking for 120min to obtain a mixed material;
(4) Heating the mixture to 55deg.C, stirring, ultrasonic extracting at 28KHZ and 1200W for 35min, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate at 5-8deg.C for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, pumping into a jacketed stainless steel pan, maintaining the temperature in water bath to 90deg.C+ -2deg.C, and sterilizing for 30min to obtain the antibacterial acne removing composition.
Example 4
An antibacterial acne-removing composition comprises the following raw materials: 25g of aloe vera, 7g of coptis chinensis, 12g of chrysanthemum, 50g of dandelion, 15g of osmanthus fragrans, 15g of patrinia villosa, 15g of viola philippica, 18g of angelica sinensis, 25g of smilax glabra, 13g of spina gleditsiae, 20g of radix paeoniae rubra, 10g of pseudo-ginseng, 20g of ligusticum wallichii and 15g of fructus cnidii.

The preparation method of the antibacterial acne-removing composition comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, sieving with a sieve with fineness of 30 meshes, and continuously pulverizing the residue until the residue is completely sieved to obtain medicinal powder;

An antibacterial acne-removing composition comprises the following raw materials: 18g of aloe vera, 14g of coptis chinensis, 16g of chrysanthemum, 48g of dandelion, 16g of osmanthus fragrans, 18g of patrinia villosa, 23g of viola philippica, 16g of angelica sinensis, 18g of smilax glabra, 11g of spina gleditsiae, 19g of radix paeoniae rubra, 12g of pseudo-ginseng, 18g of ligusticum wallichii and 22g of fructus cnidii.
The preparation method of the antibacterial acne-removing composition comprises the following steps:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, sieving with a sieve with fineness of 30 meshes, and continuously pulverizing the residue until the residue is completely sieved to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propanediol, 1, 3-butanediol and pure water according to the weight ratio of 15:15:70, and uniformly stirring for standby;
(3) Adding the medicinal powder and an extraction solvent into an extraction tank according to the weight ratio of 1:7, uniformly stirring, and soaking for 130min to obtain a mixed material;
(4) Heating the mixture to 55deg.C, stirring, ultrasonic extracting at 28KHZ and 1200W for 40min, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate at 5-8deg.C for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, pumping into a jacketed stainless steel pan, maintaining the temperature in water bath to 90deg.C+ -2deg.C, and sterilizing for 30min to obtain the antibacterial acne removing composition.
Test item
A subject: 120 vaccinia muscle individuals, 60 young subjects between 15 and 30 years old, men and women, and many of them have acne caused by hormonal or endocrine dyscrasia. 60 middle-aged office workers with ages of 30-45 years old, men and women, and the like, often stay up and overtime due to irregular life and work, and often have acne; the animals were randomly divided into 6 groups of 20 persons each.
(1) Irritation test: the antibacterial acne-removing composition prepared in examples 1 to 6 was subjected to spot application experiments, and an equal amount of the sample was applied to the curved side of the arm of the subject, and was uniformly applied to the skin by gentle palm pressure for 30 minutes, 4 hours, 24 hours observation, and skin irritation change observation, and scoring was performed according to the criteria of table 1, and the results are shown in table 2 below:

The above results show that: the antibacterial acne-removing compositions prepared in examples 1-6 have no adverse reaction, low irritation and high safety.
(2) Acne removal test: the antibacterial acne-removing composition prepared in the embodiment 1-6 is prepared into gel according to the table 3, the gel is used for subjects in the corresponding group, the facial acne area is selected, the gel is smeared for 2 times a day, the instant acne-removing capability is observed for 1-5 days, and the recurrence condition of the acne is observed for 10 th and 20 th days, and the results are shown in the table 4:

The acne treatment effect evaluation criteria are as follows:
the method is effective: the acne is reduced, the red swelling is eliminated, and the pain and itch are obviously relieved.
Invalidation: the symptom sign changes were not apparent compared to pre-treatment.
As can be seen from table 4, the optimal acne treatment is example 3 and example 4, preferably example 3.
The number of people with acne marks after acne removal in each group of examples is observed, and the inhibition effect of the antibacterial acne-removing composition on the acne marks is evaluated, so that the following data are obtained:

(3) Antibacterial test: the antibacterial acne-removing compositions prepared in examples 1 to 6 all take 5g of propylene glycol with mass fraction of 20% and dissolve to 100ml for detection, and the blank group is propylene glycol with mass fraction of 20%; nutrient agar medium (36+ -1deg.C incubator for 48+ -2 h for Staphylococcus aureus), and meat vesicle medium (36+ -1deg.C incubator for 24-72 h for Propionibacterium acnes); the diameter of the inhibition zone was measured by the diffusion method (K-B method) and the results are shown in Table 6 below:

As can be seen from the data in Table 6, the composition of the present application has a good inhibitory effect on bacteria.
(4) Stability test: preparing essence water from the antibacterial acne-removing composition prepared in the embodiment 1-6 according to the table 7, directly placing a test sample at room temperature into a baking oven at 40+/-1 ℃ for 24 hours, and after the test sample is restored to the room temperature, observing whether the test sample has no obvious difference from the test sample or not, and taking the test sample as a heat resistance test; then placing the test sample into an oven at the temperature of minus 10+/-1 ℃ for 24 hours, and after the test sample is restored to the room temperature, observing whether the test sample has no obvious difference from the test sample or not, and taking the test sample as a cold property test; the results are shown in Table 8:

As can be seen from the data in table 8, the essence water prepared from the composition of the present application has no abnormal condition compared with the blank group after the heat resistance test and the cold resistance test are recovered to room temperature, which indicates that the essence water containing the composition of the present application has good stability and can be stored for a long period of time.
In addition, the antibacterial acne-removing composition can be prepared into skin care products in the form of gel and essence, and can also be prepared into cream or powder.

The freeze-dried powder is prepared by the following steps:
(1) Mixing water, glycerol, glycine, leucine, serine, trehalose and acid bean seed polysaccharide, heating to 40deg.C, maintaining the temperature for 50min, and stirring to obtain mixture A;
(2) Mixing mannitol, an antibacterial acne-removing composition and 1, 2-pentanediol, and stirring uniformly to obtain a mixture B;
(3) Cooling the mixture A to 5 ℃, then adding the mixture A into the mixture B, and stirring the mixture B at a slow speed for 2 hours to obtain a mixture C;
(4) The mixed solution C is placed in a container for low-temperature freeze-drying, and the specific method comprises the following steps: pre-freezing at-45deg.C for 5 hr, and vacuumizing at-35deg.C to 1Pa; then carrying out gradient heating to 30 ℃, controlling the temperature change gradient to be 5 ℃ each time, and freeze-drying for 2h each temperature gradient, so that the mixed liquid C in the container is freeze-dried to be in a freeze-dried powder form, then pressing the plug in a vacuum state, rolling the cap, and packaging to obtain the freeze-dried powder with the antibacterial acne-removing effect.

The antibacterial acne-removing composition according to claim 1, which is characterized by comprising the following raw materials in parts by weight: 20-25 parts of aloe vera, 7-10 parts of coptis chinensis, 12-15 parts of chrysanthemum, 45-50 parts of dandelion, 15-18 parts of sweet osmanthus, 15-20 parts of patrinia villosa, 20-25 parts of viola yedoensis, 15-18 parts of angelica sinensis, 20-25 parts of smilax glabra, 10-13 parts of spina gleditsiae, 15-20 parts of radix paeoniae rubra, 7-10 parts of pseudo-ginseng, 15-20 parts of ligusticum wallichii and 20-25 parts of fructus cnidii.

The antibacterial acne-removing composition according to claim 1, which is characterized by comprising the following raw materials in parts by weight: 20 parts of aloe vera, 10 parts of coptis chinensis, 15 parts of chrysanthemum, 50 parts of dandelion, 15 parts of osmanthus fragrans, 20 parts of patrinia villosa, 20 parts of viola philippica, 15 parts of angelica sinensis, 20 parts of smilax glabra, 10 parts of spina gleditsiae, 20 parts of radix paeoniae rubra, 10 parts of pseudo-ginseng, 20 parts of ligusticum wallichii and 20 parts of fructus cnidii.
4. A method for preparing the antibacterial acne-removing composition according to any one of claims 1 to 3, comprising the steps of:
(1) Accurately weighing the raw materials, pulverizing with a pulverizer, and sieving with 20-40 mesh sieve to obtain medicinal powder;
(2) Preparing an extraction solvent consisting of 1, 3-propylene glycol, 1, 3-butanediol and pure water, and uniformly stirring for later use;
(3) Adding the medicinal powder and the extraction solvent into an extraction tank according to the weight ratio of (5-10), uniformly stirring, and soaking for 100-150min to obtain a mixed material;
(4) Heating the mixture to 50-60deg.C, stirring, ultrasonic extracting for 30-45min, centrifuging for the first time to remove residues, and collecting primary filtrate;
(5) Refrigerating the filtrate for 24H, centrifuging again to remove residues, collecting the second-stage filtrate, and sterilizing to obtain the antibacterial acne-removing composition.
5. The method for preparing an antibacterial and acne-removing composition according to claim 4, wherein the extraction solvent in the step (2) consists of 1, 3-propanediol, 1, 3-butanediol and pure water in a weight ratio of 15:15:70.
6. The method for preparing an antibacterial and acne-removing composition according to claim 4, wherein the refrigerating temperature in the step (5) is 5-8 ℃.
7. Use of the antibacterial acne-removing composition according to any one of claims 1 to 3, characterized in that: the antibacterial acne-removing composition is added into skin care products.
8. The use of the antibacterial acne-removing composition according to claim 7, wherein the skin care product is any one of a water agent, a powder, a cream and a gel.
