Description

Cosmetic composition capable of resisting wrinkle and compacting through mitochondria and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a cosmetic composition capable of resisting wrinkle and compacting through mitochondria and application thereof. The invention particularly relates to a cosmetic composition with the functions of optimizing skin energy regulation, establishing an autonomous anti-aging barrier of skin, improving youthful texture of skin and improving overall skin state and application thereof.
Background
Skin cells require energy to cope with external environmental pressure and repair skin damage, such as collagen synthesis in skin, DNA repair, regulation of cell proliferation, activation of apoptosis, etc., all of which require energy consumption.
About 95% of the energy required for vital cell activity is produced in the mitochondria within the cell. Mitochondria are used as an energy factory of cells, and nutrient substances are converted into chemical energy Adenosine Triphosphate (ATP) required for driving cell metabolism mainly through three stages of glycolysis, pyruvic acid circulation, tricarboxylic acid circulation and oxidative phosphorylation by aerobic respiration, and meanwhile, the growth and apoptosis of the cells are regulated and controlled.
Skin aging is triggered by internal and external factors, the accumulation of external pressure and internal dysfunction (such as cell aging, insufficient sleep, physiological pressure, external environment injury and the like caused by aging) changes the bioenergy circulation mechanism of skin cells, aggravates the loss of skin functions, and simultaneously affects the reduction of mitochondrial functions, so that ATP (adenosine triphosphate) produced by mitochondria is reduced, skin metabolism is reduced, and the 'electric power' required by collagen synthesis and damage repair of skin cells is insufficient; meanwhile, more Reactive Oxygen Species (ROS) and fewer endogenous antioxidants are generated in the process of generating ATP by mitochondria, so that unbalance of free radical metabolic capacity occurs, thereby increasing damage of mitochondrial DNA and further aggravating mitochondrial dysfunction. Cell aging is a core driving force for individual aging, and many studies in academia show that mitochondria are important triggers for cell aging, and accumulated damage of the mitochondria accelerates skin aging in the long term, so that skin loses the resistance and self-regulating adaptation stress which are needed by the skin, elasticity is reduced, tightness is lost, pigmentation, wrinkles and relaxation problems are more obvious, and obvious fatigue and aging signs appear.
The skin care agent acts on mitochondria of a cell energy station, is beneficial to restoring skin activity and solves the deep anti-aging problem. In view of this, the present invention provides a cosmetic composition for anti-wrinkle tightening through mitochondria.

Detailed Description
In order to better understand the present invention, the action mechanism of the cosmetic composition of the present invention by mitochondrial wrinkle resistance and the action of the main active ingredient in the cosmetic composition of the present invention are further explained below.
Mitochondrial production ATP aerobic respiratory link corresponds to component mechanism:
the main energy supply material in the human body is sugar, and the way in which ATP energy is generated by decomposing sugar by the human body: aerobic oxidation and anaerobic oxidation, wherein aerobic oxidation is the primary energy supply.
The process of generating energy by aerobic oxidation is divided into three stages:
1. the first stage, glycolysis;
2. the second phase, pyruvate enters the mitochondria, producing acetyl CoA;
3. the third stage, acetyl CoA, is energized by tricarboxylic acid cycle and oxidative phosphorylation.
(II) the action of the main active ingredients in the cosmetic composition of the present invention:
the cosmetic composition of the invention is effective for stimulating mitochondrial energy circulation, increasing energy reserve in skin, promoting cell metabolism and renewal, and providing skin cells with more energy to resist stimulation of the surrounding environment.
1. Glycolysis-sodium mannose phosphate
The production of cellular energy requires carbohydrates, and glucose is decomposed in the cytoplasm by glycolysis to produce pyruvic acid and NADH during glycolysis, producing energy ATP. NADH produced by this process enters the mitochondria to participate in subsequent reactions to continue ATP production.
The mannose phosphate sodium feeds mannose-6-phosphate complex into cells for glycolysis, so that protein saccharification can not be caused, the activity of ATP synthase in aged cells can be increased by 3 times, and the metabolism of the cells can be promoted; and stimulates the expression of various proteins involved in the dermis structure, significantly increases the density of the dermis reticular layer, and sodium mannose phosphate has the effect of improving fish tail, neck and spot.
2. Tricarboxylic acid cycle-undaria pinnatifida extract
The tricarboxylic acid cycle has a key enzyme, namely mitochondrial aconitase, which is important for metabolism and can also protect the integrity of mitochondrial DNA; however, aconitase enzymes are specifically altered during aging by oxidative damage, resulting in loss of catalytic function. The Undaria pinnatifida extract has effects of activating aconitase, improving activity of mitochondrial aconitase, protecting mitochondrial DNA, regulating inflammatory reaction, and promoting collagen synthesis.
3. Undaria pinnatifida extract for improving cell respiration oxygen utilization rate
The "HIF respiratory signaling factor" was discovered by the young physiological or medical prize, 2019, revealing how cells perceived and adapted to the oxygen supply. Mitochondria are able to detect and respond to oxygen levels, and hypoxia is sensed to initiate a series of adaptive responses and activate Hypoxia Inducible Factor (HIF). Under the anoxic environment, mitochondria have a short-time energy synthesizing capacity, so that free radical release is reduced, undaria pinnatifida extract can wake up intracellular respiratory signal factors, up-regulate gene expression of HIF-1 alpha, convert aerobic metabolism into anaerobic metabolism, promote oxygenation capacity, promote cell life energy and improve metabolism of skin 4, matrix recombination facial extract-Cochinchinensis extract
The major biomechanical properties of skin, such as softness, firmness or elasticity, are related to some key extracellular matrix components: collagen, hyaluronic acid or elastin. As skin ages, one can observe an increase in the degradation rate of structural elements within the skin (collagen, hyaluronic acid, elastin, etc.), and a lack of regeneration, leading to skin sagging, lack of skin tension and firmness, and loss of clear facial contours.
The extract of Cochinotus nudus is a concentrated extract of Cochinotus nudus from sea, and is a unique effective component carrying strong algae bioactive substances, focusing on lower face relaxation, and used for resisting gravity effect on skin aging.
The preparation method of the Abies starburst algae extract comprises the steps of restarting the generation of key biomarkers in skin extracellular matrix, influencing the expression of a whole set of genes related to dermis structure, elastic fiber tissue and fiber degradation protection, restarting the production of collagen fibers, enhancing extracellular matrix and reconstructing dermis scaffolds (collagen and elastic fibers), improving dermis structure and skin elasticity, recovering softness, compactness and elasticity of skin, remodelling facial contours and solving the problem of skin relaxation.
The invention will be further illustrated with reference to specific examples. It should be understood that the detailed description and specific examples are intended for purposes of illustration only and are not intended to limit the scope of the invention.
The specific techniques or conditions are not identified in the examples and are described in the literature in this field or are carried out in accordance with the product specifications. The reagents or equipment used were conventional products available for purchase through regular channels, with no manufacturer noted.
The experimental methods in the following examples are conventional methods unless otherwise specified. The test materials used in the examples described below, unless otherwise specified, are all commercially available products.

Table 1: summarizing test results of cosmetic human skin patches
As is clear from table 1, the test results of the human skin closed patch showed that the skin adverse reaction occurred in 0 cases among 30 persons, and the test object did not cause skin adverse reaction to the subjects in the present lot according to the specification of the cosmetic safety technical Specification (2015 edition), so the cosmetic in example 1 was in accordance with the detection specification.
Effect example 2: free radical scavenging effect of the cosmetic compositions of the present invention
DPPH is a stable free radical with nitrogen as a center, and the determination of DPPH clearance is a simple, rapid and sensitive method for evaluating the antioxidant activity of natural active substances at present. The antioxidant activity of the creams prepared in examples 1-2 and comparative examples 1-5 was evaluated by constructing a DPPH radical scavenging reaction system.
Table 2: free radical scavenging effect of the cosmetic compositions of the present invention
As can be seen from the experimental results of Table 2, the creams prepared in examples 1-2 of the present invention were significantly superior to the creams prepared in comparative examples 1-5 and the blank cream base described in comparative example 6 in scavenging free radicals.
Also, as shown in the results of comparative examples 1 to 3 in table 2, when the cream contains only one of three active ingredients of the extract of huperzia starring (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) or sodium mannose phosphate, the effect in scavenging free radicals is remarkably inferior to the combined group of the three, although the amount thereof in the cosmetic composition is the same. The results indicate that the combination of the three active ingredients produces a significant synergistic effect in scavenging free radicals.
It can also be seen from the results of comparative examples 4 and 5 that three components, namely, the extract of astrospora sinensis (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) and sodium mannose phosphate, are special and irreplaceable, and the effect of scavenging free radicals of the corresponding face cream is remarkably reduced after being replaced with other components having similar effects in the cosmetic field.
In addition, as shown in comparative example 6, the conventional cosmetic base for preparing a face cream alone has substantially no effect of scavenging free radicals.
Effect example 3: antioxidant effect of the cosmetic composition of the present invention
1. Evaluation principle
Reactive Oxygen Species (ROS) are oxygen-containing chemically reactive chemicals. Including peroxides, superoxides, hydroxyl radicals, and the like. ROS are formed as natural byproducts of normal metabolism of oxygen and have important roles in cell signaling and homeostasis. However, ROS levels increase dramatically during uv exposure. This can cause serious damage to the cellular structure, which is known as oxidative stress. Oxidative stress occurs when the free radical production is greater than the body scavenging capacity. Menadione can generate active oxygen free radicals, and when the free radical generation amount is larger than the organism scavenging capacity, oxidation stress reaction occurs, and the antioxidation effect of the test sample is evaluated through the fluorescence intensity of the zebra fish yolk sac.
2. Experimental protocol
The tested zebra fish were divided into several groups, including normal control group, model control group, examples 1-2 and comparative examples 1-6. Wherein the normal control group was not treated with any treatment, the model control group and the groups of examples 1-2 and comparative examples 1-6 were all added with an equal amount of menaquinone. Examples 1-2 and comparative examples 1-6 samples prepared in examples 1-2 and comparative examples 1-6 were added at a concentration of menaquinone at the same time. After a period of time, the whole zebra fish is subjected to specific fluorescent staining, and the generation condition of active oxygen free radicals in the yolk sac of the zebra fish is observed.
3. Evaluation index
Yolk sac ROS fluorescence intensity
4. Experimental sample
Samples prepared in groups 1-2 and comparative examples 1-6.
5. Basis of judgment
And (3) statistically analyzing that p is less than 0.05, the efficacy value is more than or equal to 20%, and judging that the effect is effective.
6. Experimental results
The experimental results are shown in table 3. A representative antioxidant photograph of example 1 is shown in figure 1.
Table 3: results of antioxidant efficacy test of cosmetic compositions of the present invention
As can be seen from the experimental results of Table 3, the creams prepared in examples 1-2 of the present invention have significantly better antioxidant effect than the creams prepared in comparative examples 1-5 and the blank cream base described in comparative example 6.
Also, as shown in the results of comparative examples 1 to 3 in table 3, when the cream contains only one of three active ingredients of the extract of huperzia starring (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) or sodium mannose phosphate, the effect on antioxidation is remarkably inferior to the combined group of the three although the amount is the same in the cosmetic composition. The results show that the combination of the three active ingredients produces a significant synergistic effect in terms of antioxidation.
It can also be seen from the results of comparative examples 4 and 5 that three components, namely, the extract of astrospora sinensis (GIGARTINA STELLATA), the extract of undaria pinnatifida (Undaria Pinnatifida) and sodium mannose phosphate, are special and irreplaceable, and the oxidation resistance effect of the corresponding cream is remarkably reduced after being replaced with other components having similar effects in the cosmetic field.
In addition, as shown in comparative example 6, the conventional cosmetic base used alone for preparing a face cream has substantially no effect on antioxidation.
As can be seen from fig. 1, the yolk sac fluorescence intensity of example 1 is significantly reduced compared with the control group of the residual model, indicating that the sample has significant antioxidant effect.
Effect example 4: human efficacy testing
1. Purpose of test
Skin tightness test was performed by using an instrument by using 4-week samples continuously from adult female volunteers under normal conditions, and the skin roughness and skin fish tail area ratio were photographed and analyzed to evaluate the anti-wrinkle and tightening effects of the test subjects.
2. Test article
Example 1, method of use: after the face is cleaned and the skin is refreshed, a proper amount of the product is evenly smeared on the face skin, and the product is gently massaged until the product is absorbed. The used part and frequency of the test are as follows: the face sample side is used once each after cleaning the face in the morning and evening.
3. Subject completion
Qualified subjects were enrolled 34 persons, aged 29-55 years. Of these, 4 subjects had fallen off because of non-compliance in usage. All 30 subjects were tested on demand for a trial of 28 days with an average age of 44.8.+ -. 7.69 years.
4. Evaluation, test item description
4.1 skin roughness:
photographs were taken using VISA-CR and analyzed with IPP.
Parameter description: texture duty cycle in the skin selection area; the larger the duty cycle, the rougher the skin.
And (3) result judgment: the skin roughness on the sample side is obviously reduced (P is less than 0.05) or the skin roughness difference on the sample side is obviously improved (P is less than 0.05) compared with the skin roughness on the control side before and after the test product, which shows that the test object has the effect of improving the skin roughness.
4.2 skin fish tail area ratio:
photographs were taken using VISA-CR and analyzed with IPP.
Parameter description: the area ratio of fish tail in the skin selected area; the smaller the ratio, the fewer wrinkles.
And (3) result judgment: the skin fish tail area of the sample side is obviously reduced (P is less than 0.05) before and after the test product is tested, or the skin fish tail area of the sample side is obviously improved (P is less than 0.05) compared with the control side, so that the test object has anti-wrinkle effect.
4.3 skin firmness F4 value:
the elastic tester was used to measure the overall amplitude log curve area of the skin during ten consecutive negative pressure cycles.
Parameter description: reflecting the degree of skin firmness; the smaller the measured value, the tighter the skin.
And (3) result judgment: after the product is tried out, the skin compactness F4 value of the sample side is obviously improved (P is less than 0.05), or the skin compactness F4 value difference of the sample side is obviously improved compared with the control side (P is less than 0.05), which shows that the test object has the compacting effect.
5. Test environment
Environmental requirements: temperature: (21+ -1) deg.C; relative humidity: (50.+ -. 10)%.
6. Test design
The test object is used on one side of the left side surface part and the right side surface part of the test object at random, the test object is not used on the other side, any product cannot be smeared on the test object on the same day, the clean water is uniformly used for cleaning the face, and the test is carried out after the test object is statically sitting in a constant temperature chamber for half an hour. The sample side and control side skin firmness F4 was tested before trial (D0) and after trial 28 days (D28); VISIA-CR photographs were taken and analyzed for sample side and control side skin roughness and fish tail area ratios using IPP image analysis software. The results or differences before and after use of the test object itself are compared with the comparison of the difference on the control side for statistical differences.
7. Test results

/>
/>
Note that: effectiveness is as follows: ". V" indicates effective (improved result and p < 0.05); "×" indicates no effect (improved results but p > 0.05 or p <0.05 but worse results).
8. Conclusion of the test
Under the test condition, after the product is tested for 28 days, the difference of the front and rear skin roughness of the side face of the sample is obviously improved compared with that of the control side (P is less than 0.05), so that the test object has the effect of improving the skin roughness.
Under the test condition, after the product is tested for 28 days, the difference of the front and rear skin fish tail area ratio of the side face of the sample is obviously improved compared with the comparison side (P is less than 0.05), so that the test object has anti-wrinkle effect.
Under the test condition, after the product is tested for 28 days, the F4 value of the skin compactness of the side face of the sample is obviously reduced (P is less than 0.05), and the difference value between the front and back is obviously improved compared with the comparison side (P is less than 0.05), so that the test object has the effect of improving the skin compactness.
In conclusion, under the test conditions, the test object has anti-wrinkle and tightening effects.
