Abstract
本发明涉及一种用于饲料的防霉保水瘤胃调控剂、制备方法及应用,涉及饲料领域,包括丙酸、丙酸钙、乳酸、聚乙二醇甘油蓖麻酸酯、丝兰皂苷、植物乳杆菌、枯草芽孢杆菌。本发明中各组分复配能够协同起到防霉保水调控瘤胃发酵的作用,其中丙酸与丙酸盐形成缓冲体系,延长杀菌时间,改善瘤胃微生物区系;乳酸起到脱毒、提高谷物淀粉得率、增加瘤胃抗性淀粉含量,从而改善反刍动物瘤胃发酵;丝兰皂苷和聚乙二醇甘油蓖麻酸酯能够保水且够控制水活度防止霉菌生长,丝兰皂苷改善反刍动物甲烷的排放量,聚乙二醇甘油蓖麻酸酯能够调控瘤胃发酵,提高反刍动物表观消化率及生长性能;植物乳杆菌及枯草芽孢杆菌能够参与调控瘤胃发酵。

技术领域
本发明涉及饲料领域,具体涉及一种用于饲料的防霉保水瘤胃调控剂、制备方法及应用。
背景技术
当前,随着农业发展速度的不断加快,畜牧养殖产业进入全新的发展阶段,与此同时饲料安全卫生越来越受到人们的重视。这加剧了饲料行业的竞争,除了满足饲料的品质外,对其储存要求也越来越高。饲料在生产加工运输及存储等各个环节均难以完全避免霉菌毒素的混入,饲料霉变是导致饲料品质下降,安全性受影响的重要原因。饲料霉变既影响饲料品质,又影响适口性,饲料的霉菌毒素超标严重还会导致动物出现免疫力降低,生产性能下降等情况,甚至会在食物链中逐步积累,威胁人类身体健康。饲料中常见的毒素有黄曲霉毒素、赭曲霉毒素、玉米赤霉烯酮、T-2毒素和呕吐毒素等,这些毒素均能引起动物生长受阻,繁殖机能下降、组织坏死和免疫抑制等。因此饲料的防霉工作是非常重要的。
目前饲料防霉主要通过控制饲料原料质量、加强饲料生产及储存管理、使用防霉包装袋以及合理添加防霉剂为主要解决手段,通过在饲料中添加相应的防霉物质为主要方向,根据我国农业部公告第2045号《饲料添加剂目录(2013)》“第8部分:防腐剂、防霉剂和酸度调节剂”和GB/T 36863-2018《混合型饲料添加剂防霉剂通用要求》“附录A允许使用的防霉剂名单”,我国允许在饲料加工过程中添加的防霉剂有丙酸及丙酸盐类、甲酸、乙酸及乙酸盐类、苯甲酸及苯甲酸钠、山梨酸及山梨酸盐类。目前应用的防霉剂主要分为有机酸防霉剂、有机酸盐防霉剂和复合防霉剂,其中有机酸防霉剂防霉效果好,但其腐蚀性较大,主要包括苯甲酸、丙酸、乙酸、脱氢醋酸、山梨酸、富马酸等,有机酸盐防霉剂防霉效果比有机酸防霉剂效果差,但它的腐蚀性较小,该类防霉剂主要包括苯甲酸钠、丙酸盐、山梨酸钠等,其特点是抗菌谱广,对酵母菌、霉菌、肠道细菌、球菌等都具有良好的抑制作用。当前饲料中以丙酸及丙酸铵的添加形式居多,但丙酸铵热稳定性较低,分解后易产生氨气影响饲料适口性。
当前,国内的日粮大多为玉米-豆粕型日粮,一般在生产制粒后水分含量会控制在13%以内,但随着米糠粕、DDGS等农副产物(水分含量约9%-12%左右)的大量使用,使成品水分降到12%左右,且在不同的季节日粮中的水分是不同的,夏秋季节饲料水分一般在11%-13%,但由于夏季高温,饲料中的水分会有所降低,在粉碎和制粒过程中又会有部分损失,导致成品的水分降至9%-11%。水分过低会造成饲料成品损耗增加,制粒过程能耗增加,制粒过程糊化程度不理想,适口性下降等,这直接影响饲料产品品质及企业经济效益,因此饲料的保水工作是非常重要的。
关于饲料保水现有技术采用在生产过程中控制粉碎粒度、在混合时物化喷水、制粒过程合理控制水分、增加物料在调质器中的调质时间、关闭或调整供气管道中的疏水阀、选择适宜规格的制粒环模、饲料冷却时选择适宜的冷却风量等常规措施。目前饲料行业尚无专业的饲料水分调节剂来解决这个问题,大多数企业还是选择直接添加自来水的方式来恢复饲料正常水分,但保水率不到30%,而且极易霉变。部分饲料企业选择采用液体防霉剂,在饲料防霉的同时,顺便恢复一部分饲料的正常水分,但保水率仍然很低(颗粒饲料一般仅50%左右,粉料仅85%左右),且饲料霉变的风险较高。
反刍动物就是有反刍现象的动物,反刍是指进食经过一段时间以后将在胃中半消化的食物返回嘴里再次咀嚼。反刍动物最大的消化生理特点是具有瘤胃,反刍动物同瘤胃内细菌、真菌、甲烷菌和原虫等厌氧微生物存在一种共生关系,宿主动物为瘤胃微生物发酵提供营养及适宜的生存条件,而瘤胃微生物通过降解和发酵饲料,为宿主提供能量(挥发性脂肪酸,VFA)和蛋白质(微生物蛋白质,MCP)。然而这种共生关系也造成能量(甲烷释放)与蛋白质(氨氮的流失)的损失。这不仅降低生产性能和饲料利用率,而且增加了环境压力,由于瘤胃对反刍动物营养具有不可代替的重要性,加之为改善瘤胃能量和蛋白质的利用效率,故调控瘤胃发酵对反刍动物具有重要意义。目前常用的瘤胃调控剂种类繁多,如微生态制剂、植物提取物、中草药提取物、酶制剂、离子载体饲料添加剂等,现有技术对此类的相关研究较多,其中微生态制剂植物乳杆菌、枯草芽孢杆菌对反刍动物瘤胃有一定的调节作用,均能够改善瘤胃微生物区系进而影响瘤胃发酵,还可以产生多种酶类降低有害气体的排放,改善反刍动物生活环境。当前有研究表明乳化剂也可添加至畜禽饲料中并能够提高生产性能,但将乳化剂应用于羊饲料中的研究仍处于空白阶段。
鉴于上述的分析,本发明提供了一种用于饲料的防霉保水瘤胃调控剂、制备方法及应用,这对饲料工业及畜牧行业具有深远影响。
发明内容
本发明所要解决的技术问题是提供一种用于饲料的防霉保水瘤胃调控剂、制备方法及应用。其目的是针对防霉剂中丙酸铵热稳定性低,分解后易产生氨气影响饲料适口性,液体防霉剂保水效率较低,及缺乏调控反刍动物瘤胃发酵的问题,提供包含丙酸、丙酸钙、乳酸、聚乙二醇甘油蓖麻酸酯、丝兰皂苷、植物乳杆菌和枯草芽孢杆菌,具有协同作用且适用于反刍动物的具有防霉、保水、瘤胃调控的饲料添加剂。植物乳杆菌均为耐高温型植物乳杆菌。
本发明为了解决上述技术问题,第一个目的是提供:一种用于饲料的防霉保水瘤胃调控剂,其由以下重量含量的组分组成:丙酸24~28%、丙酸钙24~30%、乳酸10~14%、聚乙二醇甘油蓖麻酸酯2~6%、丝兰皂苷3~7%、植物乳杆菌1~6%、枯草芽孢杆菌1~6%及水3~35%。
本发明的有益效果是:本发明中各组分复配能够协同起到防霉保水调控瘤胃的作用,其中丙酸通过在霉菌细胞外形成高渗透压从而使霉菌细胞脱水失去繁殖能力,同时丙酸可以穿透霉菌细胞壁抑制胞内酶活性,对霉菌毒素有很强的杀灭作用;丙酸与丙酸盐形成强大的缓冲体系,能够延长丙酸的杀菌时间,丙酸盐的有效成分是未解离的丙酸分子,其以耗能的形式除去质子,改变瘤胃pH,抑制微生物细胞膜上的酶合成,阻碍其代谢过程,在一定程度上改善瘤胃微生物区系;乳酸通过吸附霉菌毒素进而起到脱毒的效果,同时添加乳酸能够提高谷物淀粉得率,增加瘤胃抗性淀粉含量,改善反刍动物瘤胃发酵情况;丝兰皂苷、聚乙二醇甘油蓖麻酸酯,它们均属表面活性剂,能够降低表面张力,进而使其维持相对稳定的状态,达到保水效果的同时能够控制水活度,限制霉菌生长,丝兰皂苷作为植物提取物能够通过降低瘤胃内原虫数量改善反刍动物甲烷的排放量,聚乙二醇甘油蓖麻酸酯作为一种非离子型乳化剂能够调控瘤胃发酵,提高反刍动物表观消化率及生长性能;植物乳杆菌及枯草芽孢杆菌均属微生态制剂,能够参与调控瘤胃发酵。
在上述技术方案的基础上,本发明还可以做如下改进。
进一步,其由以下重量含量的组分组成:丙酸25~27%、丙酸钙25~29%、乳酸11~13%、聚乙二醇甘油蓖麻酸酯3~5%、丝兰皂苷4~6%、植物乳杆菌1~5%、枯草芽孢杆菌1~5%及水10~30%。
进一步,其由以下重量含量的组分组成:丙酸25%、丙酸钙25%、乳酸11%、聚乙二醇甘油蓖麻酸酯3%、丝兰皂苷4%、植物乳杆菌1%、枯草芽孢杆菌1%及水30%。
进一步,其由以下重量含量的组分组成:丙酸26%、丙酸钙28%、乳酸11%、聚乙二醇甘油蓖麻酸酯4%、丝兰皂苷5%、植物乳杆菌4%、枯草芽孢杆菌4%及水18%。

本发明的第二个目的是提供一种用于饲料的防霉保水瘤胃调控剂的制备方法,其特征在于,包括如下步骤:取上述重量含量的丙酸、丙酸钙、乳酸、聚乙二醇甘油蓖麻酸酯、丝兰皂苷、植物乳杆菌及枯草芽孢杆菌及水,混合均匀,得到用于饲料的防霉保水瘤胃调控剂。本发明制备得到的用于饲料的防霉保水瘤胃调控剂需与水以1:24的质量比混合稀释后再使用。
采用上述方案的有益效果是:本发明的用于饲料的防霉保水瘤胃调控剂制备方法简单,实用性和经济性较高。
本发明的第三个目的是提供一种用于反刍动物的饲料,所述用于反刍动物的饲料包括如由上述所述一种用于饲料的防霉保水瘤胃调控剂。
进一步,所述用于反刍动物的饲料中含有重量含量为0.05~0.2%所述的用于饲料的防霉保水瘤胃调控剂。
采用上述方案的有益效果是:本发明的用于饲料的防霉保水瘤胃调控剂添加的重量含量为0.05~0.2%,就可达到较好的调控瘤胃发酵,提高反刍动物表观消化率及生长性能,与现有的饲料的防霉保水添加剂相比,添加量减少能够节约成本。
进一步,所述用于反刍动物的饲料中含有重量含量为0.1%所述的用于饲料的防霉保水瘤胃调控剂。

实施例5:试验功效验证
1.试验材料
饲料的基础日粮配方:玉米36%、豆粕(43%)4%、玉米蛋白粉6%、DDGS(玉米酒糟蛋白)14%、葵皮35%、预混料4%,石粉1%。
目前市场上常使用的液体防霉保水剂样品A,其含水量在10-40%之间,液体防霉保水剂样品A含有丙酸、丙酸铵、乳酸、水,有效成分:总酸≥52%,丙酸51.5%-59.0%,乳酸0.5-1.0%。

本试验设置6个试验组,分别为对照组、试验1组、试验2组、试验3组、试验4组、试验5组,各实验组的样品的具体处理详见表1。

2.2防霉功效测定方法
准备1L的烧杯和200mL的小塑料杯子,将各试验组样品置于大杯子中,将小杯子置于样品中央位置。在小杯子中注入60mL蒸馏水,不要将水散落在样品中,用两层保鲜膜将大杯子封口以保证整个大杯子密封。后将整个大杯子置于30℃的恒温箱中或其他类似的温暖条件下。每天观察样品1次,观察是否有霉菌(丝)长出,并对试验进行记录,防霉功效试验结果如表2。

由表2数据可知,在30℃高温且高湿的环境下,通过15天的观察发现对照组的样品第7天开始发霉,试验1组的样品第10天发霉、试验2组的样品第13天发霉,试验4组的样品第15天发霉,试验5组的样品第14天开始发霉,试验组3组无发霉情况,防霉效果最好。
3.防霉保水功效测定
3.1试验组别设计及防霉保水功效测定方法
本试验设置6个试验组,分别为对照组、试验1组、试验2组、试验3组、试验4组、试验5组,各组的样品详见表1的记载。按照表1中样品的记载,将每个实验组的样品连续生产3批,分别对同一样品的每批样品取样,取样时避开头尾料,等量抽取每批中的3个不同袋的样品,混匀后作为实验组,每个实验组重复6次,取样完成后用饲料厂常规包装袋装好,封口用缝包机缝好,做好标记(对照组、试验1组、试验2组、试验3组、试验4组、试验5组),置于类似饲料厂仓储环境的室内储藏。
在贮存期内,分别对5个试验组的样品每10d测定一次水分值和霉菌数,每个样品测定3次,结果取平均值。测定时每次取完样品后立即用缝包机缝好袋口,再放回原处。
饲料水分测定方法:参照GB 6435。饲料霉菌数的测定方法:参照GB 13092。防霉保水功效测定结果如表3和4。

由表3可见,试验1-5组初始水分值明显增加,其中试验3组的起始水分最高,证明其保水性较好。与0d相比,40天后试验1-5组水分分别降低0.016%,0.007%,0.003%,0.004%,0.005%;对照组水分降低0.114%,这说明本发明的防霉保水瘤胃调控剂(试验2-5组)其保水效果较理想,其中试验3组保水效果最好。
表4储存期饲料霉菌数试验结果
由表4可知,由于所用饲料样品水分含量低,各试验组初始(0天)霉菌数较低,随着贮存时间增加,霉菌数增多;贮存到第10天时,对照组及试验1-5组的霉菌数增加量分别为2.4×103/g,1.2×103/g,1.5×103/g,0.9×103/g,1.1×103/g,1.6×103/g。贮存到第20天时,对照组及试验1-5组的霉菌数增加量分别为16.9×103/g,6.1×103/g,3×103/g,1.4×103/g,1.8×103/g,3.1×103/g;贮存到第30天时,对照组及试验1-5组的霉菌数增加量分别为31×103/g,19.7×103/g,14.5×103/g,2.9×103/g,4.5×103/g,9.6×103/g;贮存到第40天时,对照组及试验1-5组的霉菌数增加量分别为62.3×103/g,31.5×103/g,19.3×103/g,8.5×103/g,10.9×103/g,13.3×103/g。由此可知,在0-30d,霉菌数增加较慢,30-40d后期霉菌数增多较快,0-40d期间试验2-5组的防霉效果优于对照组和试验1组,且试验3组的防霉效果较强。
4.瘤胃调控功效
4.1分组及饲养管理
本试验选择体重相近、健康的小尾寒羊300只,随机分为6组,分别为对照组、试验组1、试验组2、试验组3、试验组4及试验组5,详见表5,每组50只。各组饲喂分别按照表5进行分栏。试验期共97d,其中前7d为预试期。采用全自动采食料槽,每3天喂一次,记录投料量与剩料量,确保剩料量为投料量的0.5%,自由饮水,试验中途不添加任何外用药物及饲料原料。试验结束后测定小尾寒羊的生长性能、养分表观消化率及瘤胃发酵指标。

4.2生长性能
试验开始及结束后对各组小尾寒羊进行空腹称重,试验过程准确记录采食量用于统计各组小尾寒羊的平均日增重、平均日采食量及料肉比。
平均日增重=(试验末重-试验初始重)/试验天数;
平均日采食量=总采食量/试验天数;
料肉比=平均日采食量/平均日增重。
4.3养分表观消化率
试验结束前5d,每天16:00采集同一只小尾寒羊的粪便约100g左右,每组选10只进行粪便的采集,采集后用10%的硫酸进行固氮。养分表观消化率的计算如下:
养分表观消化率=(食入的营养物的质量-粪中营养物质排出的质量)/食入营养物质量×100%。
4.4瘤胃发酵参数
参考柏雪[柏雪,柏凡,方思敏,等.反相高效液相色谱法测定瘤胃内容物中挥发性脂肪酸的方法研究[J].中国饲料,2014(13):28-29.]测定挥发性脂肪酸(VFA)浓度;参考冯宗慈[冯宗慈,高民.通过比色测定瘤胃液氨氮含量方法的改进[J].畜牧与饲料科学,2010(6):37.]方法测定氨态氮浓度;采用文献[Cotta M A,Russell J B.Effect of peptides and amino acids on efficiency of rumen bacterial protein synthesis in continuous culture[J].Journal of Dairy Science,1982,65(2):226-234.]方法测定微生物蛋白(MCP)含量。
4.5数据统计及结果分析
所有试验数据均采用Excel2007进行数据统计及分析。
4.5.1对小尾寒羊生长性能的影响
表6对小尾寒羊生长性能的影响
由表6可知,与对照组相比,试验组1、试验组2、试验组3、试验组4及试验组5的小尾寒羊的末重及平均日增重均有提高,且各试验组1-5均能够降低料肉比,其中试验组2-5组表现较为突出,证明试验组2-5组能够更好的提高小尾寒羊生长性能,且试验组3效果最佳。
4.5.2对小尾寒羊表观消化率的影响
表7对小尾寒羊表观消化率的影响
由表7可知,与对照组相比,试验组2、试验组3、试验组4、试验组5的小尾寒羊对粗蛋白、粗脂肪、酸性洗涤纤维、中性洗涤纤维的表观消化率均增加。其中试验组2-5组表现较为突出,证明试验组2-5组能够更好的提高小尾寒羊表观消化率,且试验组3效果最佳。
4.5.3对小尾寒羊瘤胃发酵的影响

由表8可知,与对照组相比,试验组1、试验组2、试验组3、试验组4、试验组5的小尾寒羊瘤胃氨态氮含量降低,瘤胃微生物蛋白含量增加,丙酸含量增加,总挥发性脂肪酸含量增加,其中试验组2-5组表现较为突出,证明试验组2-5组能够更好的提高小尾寒羊的瘤胃调控作用,且试验组3最佳。
综上可知,本发明提供的一种用于饲料的防霉保水瘤胃调控剂具有优良特性,且添加丝兰皂苷及聚乙二醇甘油蓖麻酸酯既能够控制水活度限制霉菌生长,起到防霉保水作用还能够调控小尾寒羊瘤胃发酵,添加丙酸、丙酸钙、乳酸在起到防霉保水作用的同时还能够调控瘤胃微生物区系,添加植物乳杆菌及枯草芽孢杆菌也能够起到瘤胃调控作用,各组分相互协同发挥作用,且本发明的添加量较一般产品低,可提高经济效益。

Claims (10)
1.一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸24~28%、丙酸钙24~30%、乳酸10~14%、聚乙二醇甘油蓖麻酸酯2~6%、丝兰皂苷3~7%、植物乳杆菌1~6%、枯草芽孢杆菌1~6%及水3-35%。
2.根据权利要求1所述一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸25~27%、丙酸钙25~29%、乳酸11~13%、聚乙二醇甘油蓖麻酸酯3~5%、丝兰皂苷4~6%、植物乳杆菌1~5%、枯草芽孢杆菌1~5%及水10~30%。
3.根据权利要求1所述一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸25%、丙酸钙25%、乳酸11%、聚乙二醇甘油蓖麻酸酯3%、丝兰皂苷4%、植物乳杆菌1%、枯草芽孢杆菌1%及水30%。
4.根据权利要求1所述一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸26%、丙酸钙28%、乳酸11%、聚乙二醇甘油蓖麻酸酯4%、丝兰皂苷5%、植物乳杆菌4%、枯草芽孢杆菌4%及水18%。
5.根据权利要求1所述一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸26%、丙酸钙27%、乳酸12%、聚乙二醇甘油蓖麻酸酯4%、丝兰皂苷5%、植物乳杆菌3%、枯草芽孢杆菌3%及水20%。
6.根据权利要求1所述一种用于饲料的防霉保水瘤胃调控剂,其特征在于,其由以下重量含量的组分组成:丙酸27%、丙酸钙29%、乳酸13%、聚乙二醇甘油蓖麻酸酯5%、丝兰皂苷6%、植物乳杆菌5%、枯草芽孢杆菌5%及水10%。
7.基于权利要求1-6任一项所述的一种用于饲料的防霉保水瘤胃调控剂的制备方法,其特征在于,包括如下步骤:取上述重量含量的各组分,将各组分混合均匀,得到用于饲料的防霉保水瘤胃调控剂。
8.一种用于反刍动物的饲料,其特征在于,所述用于反刍动物的饲料包括如由权利要求1-6任一项所述的于饲料的防霉保水瘤胃调控剂。
9.根据权利要求8所述一种用于反刍动物的饲料,其特征在于,所述用于反刍动物的饲料中含有重量含量为0.05~0.2%所述的用于饲料的防霉保水瘤胃调控剂。
10.根据权利要求8所述一种用于反刍动物的饲料,其特征在于,所述用于反刍动物的饲料中含有重量含量为0.1%所述的用于饲料的防霉保水瘤胃调控剂。
