背景技术
硅在地壳中含量丰富,是土壤中仅次于氧的第二大元素,同时也是继N、P、K之后的第四大植物营养元素。由于大部分硅以硅酸盐矿物和石英矿的形式存在于自然界中,加之人们对硅元素的认识不足,因此没有把硅元素作为植物的必须元素进行专门的补充。在很长一段时间,硅能促进植物生长发育的重要作用没有得到发现和较好的利用。
1928年,美国的研究学者发现硅能使水稻等农作物增产。1955年,日本在肥料法中将硅肥列为一种新型的肥料,并开始硅肥的生产和销售,之后朝鲜、菲律宾、印度等亚洲国家对硅肥进行了引进和推广。随着对硅肥研究的深入,硅肥的研究的范畴逐渐扩大,硅肥研究的涵盖面越来越广,从硅肥促进作物茎叶的生长,抑制土壤板结与酸化,到硅肥对重金属的钝化,以及农业的可持续发展,硅肥的研究逐渐开始收到重视。有研究者利用具有解硅功能的微生物能够分解硅酸盐矿物的特性,将其应用于肥料生产中,如:将含解硅菌、固氮菌、溶磷菌、解钾菌、根瘤菌的复合菌剂作为发酵剂,使硅、钾、镁等离子转化为作物易吸收的活性态,施用于农田促进植物生长。
目前,具有解硅功能的微生物大多为硅酸盐细菌,部分已被开发成微生物肥料,田间或盆栽应用证明其可以促进作物生长、提高作物产量,但该类菌株主要作用是溶磷、解钾,具有一定的固氮能力,解硅能力不够强,因此,筛选高效稳定的解硅菌株,并确定其对矿物或土壤环境的适应性,对开发新型生物硅肥、促进农业可持续发展有重要意义。
发明内容
本发明的目的是提供一种既可高效释硅,又具有良好溶磷、解钾能力、耐酸碱、可生产铁载体和IAA的波兰青霉CFP-2及其应用。
本发明采用如下技术方案:
一种波兰青霉(Penicillium polonicum)CFP-2,其保藏编号为CGMCC No.40494,保藏于中国微生物菌种保藏管理委员会普通微生物中心,地址为北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,保藏日期为2023年2月8日。
所述的波兰青霉CFP-2在PDA培养基上生长良好,在PDA培养基上,菌落初期呈白色,后期产孢后呈青绿色。菌丝有横隔,分生孢子梗亦有横隔,基部无足细胞,顶端不形成膨大的顶囊,其分生孢子梗经过多次分枝,产生几轮对称或不对称的小梗,如扫帚状,孢子形态为圆形或椭圆形。
其中,该菌株具有释硅、溶磷和解钾能力。
其中,该菌株具有产铁载体和IAA能力。
其中,具有酸碱耐受能力,在初始培养基pH为4~10范围内均能很好生长,最适生长pH为4~8。
其中,该菌株具有盐耐受特性,在初始培养基NaCl浓度0~15%范围内均能很好生长。
其中,其可耐受生长温度为20~40℃,最适生长温度为30~35℃。
其中,其兼具释硅能力、解磷解钾和耐酸碱能力。
一种上述波兰青霉CFP-2在植物促生和/或土壤修复中的应用。
一种包含上述波兰青霉CFP-2的生物硅肥、磷肥、钾肥和/或土壤调理剂。
本发明的有益效果在于:本发明的波兰青霉CFP-2有释硅、溶磷、解钾、产铁载体和IAA功能,利用其应用于土壤,不仅可以促进植物生长,而且显著提高土壤中硅、磷、钾含量。
本发明的波兰青霉CFP-2在酸性或碱性土壤耐受性方面优势显著,定殖能力强,所以可以用于盐碱地和尾矿区土壤改良、生态复垦等。
本发明还可以为释硅、溶磷解钾功能微生物的开发提供优质菌种资源,为开发新的生物有机肥、土壤调理剂和微生物菌剂奠定基础,有广阔的推广应用前景。

具体实施方式
下面结合实施例和附图对本发明做进一步的说明。本发明保护范围不限于实施例,本领域技术人员在权利要求限定的范围内做出任何改动也属于本发明保护的范围。
实施例1波兰青霉CFP-2分离和保藏
本发明的波兰青霉CFP-2是从中国河北省唐山市迁安铁矿尾矿砂样品中,通过梯度稀释平板法初筛、生物浸矿复筛等方法分离获得。铁尾矿砂样品的化学成分如表1所示。

具体方法:将铁尾矿砂制备成10-3、10-4、10-5的3个稀释度悬液,均匀涂布于亚历山大罗夫培养基上,于30℃恒温培养箱中培养2~3天,挑取不同形态单菌落进一步分离纯化,直至获得单菌落,根据菌落形态初步判断为真菌,因此转接到PDA培养基斜面,4℃冰箱保存备用。以铁尾矿砂为底物,以发酵液中有效硅含量的增加为标准,对初筛得到的单菌落接种至浸矿脱硅培养基内进行释硅能力复筛。发酵第5d时进行取样,发酵液离心取上清,用硅钼蓝比色法测定有效硅含量,选出一株释硅效果较好的真菌CFP-2。
所用培养基配方:
亚历山大罗夫培养基(g/L):蔗糖5.0g、Na2HPO42.0g、MgSO4·7H2O 0.5g、FeCl30.005g、CaCO30.1g、三硅酸镁1.0g、琼脂15~20g,pH为7.0~7.4。
浸矿脱硅培养基(g/L):葡萄糖10g、KH2PO40.2g、MgSO4·7H2O 0.2g、NaCl 0.2g、CaCl2·2H2O 0.2g、CaCO35g、铁尾矿砂1.0g,pH为7.0~7.2。
PDA培养基(g/L): 马铃薯 200g、葡萄糖 20g、琼脂 15~20g、蒸馏水1L,pH 自然(固体培养基需加琼脂 15~20 g)。
结果如图1~图3所示,菌株CFP-2在PDA培养基上生长良好,菌落初期呈白色,后期产孢后呈青绿色。菌丝有横隔,分生孢子梗,孢子囊呈扫帚状,孢子形态为圆形或椭圆形。
取培养3d的菌丝体,利用真菌基因组DNA提取试剂盒提取菌株CFP-2基因组DNA,以其DNA为模板, ITS1(上游引物)5’-TCCGTAGGTGAACCFTGCGC-3’,ITS4(下游引物)5’-TCCFTCCGCFTTATTGATATGC-3’ 为引物,进行PCR扩增。经上海生工测序得基因序列。将其与GenBank数据库中的序列进行BLAST比对,并用MEGA 5.0做系统发育树(如图4所示)。
结果表明,该菌株CFP-2与Penicillium polonicum的同源性最高。结合形态学等特征,将其鉴定为波兰青霉Penicillium polonicum。该菌株于2023年2月8日在中国微生物菌种保藏管理委员会普通微生物中心进行了保藏,地址为北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,保藏编号为CGMCC No.40494。
实施例2波兰青霉CFP-2溶磷、解钾能力测定
制备菌株CFP-2种子平板,点接于NBRIP溶磷和解钾检测培养基上,于30℃恒温培养箱中培养3 d,观察其生长情况及是否有透明圈。计算方法为:溶解能力=透明圈直径D/菌落直径d。
溶磷活性检测培养基(NBRIP培养基):葡萄糖 10g、Ca3(PO4)25g、(NH4)2SO40.5g、MgSO4·7H2O 0.25g、KCl 0.2g、MgCl2·6H2O 5g、琼脂15g、0.4%溴酚蓝 6mL、超纯水1L,pH7.0~7.2。
解钾培养基:蔗糖10g、Na2HPO41g、(NH4)2SO40.5g、MgSO4·7H2O 1g、酵母粉 0.2g、NaCl 0.1g、CaCO30.1g、FeCl30.005g、钾长石 1g、水 1L、pH 7.0。
测定结果表明:在NBRIP培养基和解钾培养基中,菌株CFP-2的菌落周边均出现明显的透明圈,在NBRIP培养基D/d值大于3,解钾培养基D/d值大于1.5,说明其具有较强的溶磷、解钾能力。
实施例3波兰青霉CFP-2产铁载体和分泌IAA测定
分泌IAA性能:制备菌株CFP-2种子平板,将部分菌落接种于PDA液体培养基中(含100mg/L的L-色氨酸),置于30℃、180r/min的摇床上振荡培养3~5d,取经过8000r/min离心后的上清液50μL,加入50μL Salkowski比色液,滴在白瓷板上避光显色30min后,观察颜色变化,液体变为红色,表示菌株CFP-2能够分泌IAA,颜色越深表示分泌的强度越大。
铁载体产生能力:制备菌株CFP-2种子平板,接种针挑取部分菌落接于CAS检测培养基上,30℃培养3d,在菌落周边出现黄绿色晕圈,说明菌株CFP-2具有产铁载体能力。
结果表明,菌株CFP-2具有产IAA、铁载体的能力。IAA是一种植物生长激素,对植物抽枝或芽、苗等的顶部芽端形成有促进作用;而铁载体是细菌、真菌等分泌的一种化合物,用于传递到细胞外进行物质交换,与环境中的游离铁离子结合,从而将铁输送到细胞内部,满足植物的微量元素需求,促进植物生长。因此,间接说明菌株CBP-2有促进植物生长的能力。
实施例4波兰青霉CFP-2的抗逆性实验
制备菌株CFP-2种子平板,挑取部分菌苔接种于不同初始pH(4、5、6、7、8、9、10)、NaCl含量(0、0.5%、2%、4%、6%、8%、10%、12%、15%)的PDA液体培养基内,于25℃恒温养 2~3d。同时将菌株CFP-2菌落接入PDA液体培养基,于不同温度条件下(20℃、25℃、30℃、40℃、45℃、50℃)培养2~3 d,观察菌体生长情况,最后,过滤、对菌丝体进行称重。
结果表明(见图5~图7),该菌株CFP-2可在pH 4~10内生长,对酸碱均具有很好的耐受性,最适生长pH为6~8。在培养基NaCl含量0~15%均能正常生长,随着盐浓度增加,菌体干重缓慢增长,当在盐浓度为6~8%菌体干重最高,盐浓度从10%~15%,菌体干重呈下降趋势,说明其具有很强的耐盐能力。菌株CFP-2在20~40℃均能正常生长,最适生长温度为30~35℃,说明菌株CFP-2对逆境条件耐受能力强,具有高耐酸碱性和耐盐性。
实施例5波兰青霉CFP-2对土壤及植物生长的影响
菌株CFP-2孢子液制备:用接种环挑取部分青霉菌CFP-2孢子均匀接种于PDA固体平板培养基上,30℃,培养3~5d,长满青绿色孢子后,刮取培养基表面孢子置于20mL含有玻璃珠的无菌水中,25℃、200rpm摇床振荡 20~30min制成孢子悬液,血球计数板计数,将孢子浓度调整为1×108CFU/mL,按照20mL·kg-1拌入基质,基质是由部分园林土(40%)、铁尾矿砂(30%)和蛭石(30%)复合而成。对照则在基质中拌入相同体积的无菌水,进行玉米盆栽试验。置于25℃温室中,在种子萌发后保留每盆中生长较好的三株幼苗,40 d后对土壤和植株(玉米)的地上指标及根系生长状况进行测定。每组实验做3组平行处理。
结果显示(见表2和表3):与不接种CK处理相比,接种处理组对玉米植株的生长均有积极的促生效果,其中处理组的株高、根长、茎粗、叶绿素含量、植株的鲜重和干重分别为69.05 cm、34.59 cm、3.67 cm、32.51spad、6.58 g和0.58 g,比对照组提高了20.46%、47.32%、43.36%、36.60%、117.88%和48.72%。说明玉米在植株长势及根系方面都要显著优于对照处理。同时可以显著提高土壤中有效硅36.5%、速效钾53.7%和有效磷101.2%。

4.一种如权利要求1所述的波兰青霉CFP-2在制备人工基质中的应用。
5.一种如权利要求1所述的波兰青霉CFP-2在酸性、碱性或盐碱土壤修复中的应用。
