Abstract
The invention relates to a natural anti-hair loss composition and a preparation method thereof, wherein the active ingredients of the composition comprise: nine natural extract components of polygonum multiflorum, angelica sinensis, glossy privet fruit, rhizoma drynariae, ginger, biota orientalis, saussurea involucrata, chilli and citrus are extracted, and the nine natural extract components are prepared by respectively adopting different physical or chemical extraction and purification methods for polygonum multiflorum, angelica sinensis, glossy privet fruit, rhizoma drynariae, ginger, biota orientalis, saussurea involucrata, chilli and citrus; the natural anti-hair loss composition can be added into conventional scalp washing and caring components or products, and has the effects of inhibiting the proliferation of microorganisms, repairing damaged hair follicles and reducing hair loss.

Technical Field
The invention belongs to the technical field of washing and caring daily necessities, and particularly relates to a natural anti-hair loss composition, and a preparation method and application thereof.
Background
With the current living environment influence including the rising of living pressure, the problems of excessive scalp grease secretion, dandruff, scalp sensitivity and the like are more and more common, so that the efficacy requirement of consumers on the hair care products is gradually increased, wherein the imbalance of scalp grease secretion is realized, the functions of the hair care products are continuously extended and gradually refined in terms of product functions, the functions of initial hair cleaning are gradually extended into the functions of oil control, dandruff removal, nursing, moistening and the like, and the functions of hair blacking, hair loss prevention, hair growth and the like are further extended.
Aiming at scalp and hair washing and caring products with repairing effect, the components of the washing and caring products on the market at present mainly adopt chemical synthetic substances, and various side effects such as dry and yellow hair, broken hair, hair loss and the like are shown after long-term use by consumers. Moreover, with the popularity of environmental protection, low carbon, green and natural trends in recent years, plant anti-dandruff and antipruritic shampoo is a trend in the future, but the effect is not obvious although a plurality of herbaceous plants are used in the current plant shampoo, for example, chinese patent CN106619385A discloses a traditional Chinese medicine anti-hair loss shampoo and a preparation method thereof, the shampoo adopts arborvitae tops, medlar, old ginger, arborvitae seeds, rhizoma drynariae, red sage roots, rhizoma polygonati, clove, glossy privet fruits, safflower, radix rehmanniae, red paeony roots, peach kernels, fleece-flower roots, white mulberry barks and fructus psoraleae, and the traditional Chinese medicinal materials adopted by the shampoo are added by the conventional crushing means, and a great amount of macromolecules or high polymer components such as proteins are difficult to obtain better absorption and obstruct the permeation effect of active substances, so the anti-hair loss effect of the technical scheme is not ideal; for example, chinese patent CN105362954a discloses a traditional Chinese medicine for treating baldness and a preparation method thereof, wherein the traditional Chinese medicine comprises lotus plumule, cortex acanthopanacis, eclipta, sesame flower, cockscomb, achyranthes root, dried rehmannia root, fructus aurantii, semen cuscutae, cortex lycii radicis, locust bean, rhizoma polygonati, fructus cnidii, dried orange peel, radix curcumae, radix philippine flemingiae, angelica sinensis, fructus lycii, glossy privet fruit, rehmannia root, radix astragali, polygonum multiflorum, cacumen biotae, eclipta, ligusticum wallichii and spatholobus stem, and the traditional Chinese medicine is extracted and not purified, and still contains a large amount of macromolecular substances, and the macromolecular substances such as protein and the like have no effect on the cleaning effect of hair follicles. Based on the anti-drop product which has been developed in the prior art and takes Chinese medicinal materials as main active ingredients, the washing and caring product which is prepared from the high-efficiency anti-drop Chinese medicinal extract component and has no side effect has more significance for the utilization rate of the active ingredients of the Chinese medicinal materials and alopecia crowd.
Disclosure of Invention
Aiming at the problems in the prior art, the invention provides a natural anti-hair loss composition, a preparation method and application thereof, and the natural anti-hair loss composition prepared by the invention has abundant active ingredients of natural products, can promote the cleaning and repair of scalp hair follicles, increases the permeability of the active ingredients and improves the anti-hair loss effect.
In a first aspect, the present invention provides a natural anti-hair loss composition comprising polygonum multiflorum extract, angelica extract, glossy privet fruit extract, drynaria extract, ginger extract, biota orientalis extract, saussurea involucrata extract, capsicum extract and citrus extract.
Further, the natural anti-hair loss composition comprises, by weight, 1-5 parts of polygonum multiflorum extract, 0.1-3 parts of angelica sinensis extract, 0.5-5 parts of glossy privet fruit extract, 0.1-2 parts of drynaria rhizome extract, 0.5-5 parts of ginger extract, 0.1-3 parts of biota orientalis extract, 0.1-3 parts of saussurea involucrata extract, 0.1-2 parts of chilli extract and 0.1-2 parts of citrus extract.
In a second aspect, the present invention provides a method for preparing nine natural extract components in the natural anti-hair loss composition, which comprises the following specific preparation methods:
(1) The preparation method of the polygonum multiflorum extract comprises the following steps: cleaning tuber fleeceflower root, drying, crushing, soaking in solvent, reflux extracting, filtering and concentrating to obtain extractum, stepped extracting with solvent, filtering and concentrating to obtain coarse fraction, column chromatographic treatment of the coarse fraction, and concentrating the eluted fraction to obtain tuber fleeceflower root extract.
(2) The preparation method of the angelica sinensis extract comprises the following steps: soaking radix Angelicae sinensis powder in solvent, reflux extracting, filtering, concentrating to obtain extract, subjecting the extract to fractional extraction with solvent, filtering, concentrating to obtain crude fraction, subjecting the crude fraction to column chromatography, and concentrating the eluted fraction to obtain radix Angelicae sinensis extract.
(3) The preparation method of the glossy privet fruit extract comprises the following steps: pulverizing fructus Ligustri Lucidi, soaking in solvent, reflux extracting, filtering, concentrating to obtain extract, subjecting the extract to fractional extraction with solvent, filtering, concentrating to obtain crude fraction, subjecting the crude fraction to column chromatography, and concentrating the eluted fraction to obtain fructus Ligustri Lucidi extract.
(4) The preparation method of the drynaria extract comprises the following steps: soaking rhizoma Drynariae powder in solvent, reflux extracting, filtering, concentrating to obtain extract, subjecting the extract to fractional extraction with solvent, filtering, concentrating to obtain crude fraction, subjecting the crude fraction to column chromatography, and concentrating the eluted fraction to obtain rhizoma Drynariae extract.
(5) The preparation method of the ginger extract comprises the following steps: cleaning rhizoma Zingiberis recens, mashing, soaking in water, ultrasonic treating, filtering, soaking the residue in water of the same quality, ultrasonic treating, filtering, repeating the same steps for three times, mixing the three filtrates, and concentrating to obtain rhizoma Zingiberis recens extract.
(6) The preparation method of the biota orientalis leaf extract comprises the following steps: cleaning folium Platycladi, drying, pulverizing, soaking in hot water, ultrasonic treating, filtering, deproteinizing the filtrate, dialyzing, and concentrating to obtain folium Platycladi extract.
(7) The preparation method of the lotus seed extract comprises the following steps: cleaning herba Saussureae Involueratae, mashing, soaking in solvent, ultrasonic treating, filtering, reflux extracting, filtering, mixing filtrates, concentrating to obtain crude extract, respectively extracting with solvent, filtering, mixing filtrates, and concentrating to obtain herba Saussureae Involueratae extract.
(8) The preparation method of the chilli extract comprises the following steps: cleaning capsicum, drying, pulverizing, soaking in solvent, ultrasonic treating, filtering, repeating the steps for 3-5 times, mixing filtrates, and concentrating to obtain capsicum extract.
(9) The preparation method of the citrus extract comprises the following steps: cutting citrus peel, soaking in distilled water, heat extracting, filtering, and drying to obtain citrus extract.
The solvent used in the preparation process of the nine natural extract components is one or more selected from methanol, ethanol, petroleum ether, dichlorohexane, chloroform, ethyl acetate, n-butanol, isobutanol and n-hexane.
In a third aspect, the present invention also provides the use of the hair care composition in a scalp, hair or hair follicle repair care product.
Further, the scalp, hair or hair follicle repair care product is in any form of liquid, paste, emulsion, foam or spray, but the product is not limited to the form.
Compared with the prior art, the natural anti-hair loss composition prepared by the invention adopts nine natural extract components, the compatibility among the natural extract components is good, different extraction and purification means are adopted for different natural traditional Chinese medicinal material components, the extraction of active components is realized, and the finally formed composition has rich natural product active components, so that the cleaning and repairing of scalp hair follicles can be promoted, the permeability of the active components is increased, and the anti-hair loss effect and effect are improved.

The preparation method of the polygonum multiflorum extract comprises the following steps: washing, drying and crushing tuber fleeceflower root, taking methanol (70 wt%) of tuber fleeceflower root powder with 12 times of mass, soaking for 6h, then carrying out reflux extraction for 2h, concentrating the filtered filtrate to obtain extract, sequentially extracting the extract with petroleum ether (3 times of the mass of the extract) for 3 times, extracting the extract with ethyl acetate (3 times of the mass of the extract) for 3 times, extracting the extract with methanol (2 times of the mass of the extract) for 3 times, respectively carrying out filtration and concentration to obtain petroleum ether section crude fraction, ethyl acetate section crude fraction and methanol section crude fraction, carrying out column chromatography on the ethyl acetate section crude fraction, loading the column with ODS reverse phase silica gel packing, eluting with a methanol-water eluting system, eluting gradient of 30:70, 40:60, 50:50, 60:40, 70:30, 100:0, each gradient eluting for 2 column volumes, sequentially obtaining A-F6 fragments, collecting fraction A-D, merging and concentrating, further carrying out Sephax LH-20 column chromatography, eluting with methanol with a flow rate of 1-2 drops/second, collecting a tube, collecting 2ml of eluate with a thin layer chromatography (d=170 cm), and carrying out TLC analysis on the concentrated extract, obtaining the tuber fleeceflower root extract after the Thin Layer Chromatography (TLC).

10 hair loss patients with 25-45 years old, healthy head skin, no allergy or scar and no local systemic other diseases, 5 men and 5 women, were selected, and the natural hair loss prevention composition prepared in the example 12 was continuously used for 2 months, after every two days, after cleaning hair and scalp, 2-5 mL each time was used, and was uniformly applied to hair and scalp, and kept for 2-3 minutes, and after rinsing, the hair loss number (root) after three times of hair and scalp cleaning was recorded and averaged before use: the average hair loss numbers after 37.92 days, 15 days, 30 days, 45 days and 60 days were 32.88, 26.85, 19.9 and 13.08, respectively, and thus it was found that the natural anti-hair loss composition prepared in example 12 was poor in anti-hair loss effect.
The anti-hair loss compositions prepared in examples 1 to 12 of the present invention were subjected to related toxicology tests before evaluating the anti-hair loss effect thereof, and were rinsed by an acute eye irritation test (30 s), and as a result, were non-irritating; through an acute skin irritation test, the result shows that the skin irritation is free; the result of the skin phototoxicity test shows that no skin phototoxicity is seen; the skin allergy test shows that no skin allergy is found.
The anti-hair loss composition of the present invention was mixed with a general shampoo/shampoo product having only a cleansing effect in evaluating its anti-hair loss effect, wherein the anti-hair loss composition was added in an amount of 10% of the total mass of the mixture. The ingredients of conventional shampoos/shampoos may include the usual additives ingredients of lipids, humectants, surfactants, and water.

The natural anti-hair loss composition according to claim 1, wherein the composition comprises, in parts by weight, 1 to 5 parts of polygonum multiflorum extract, 0.1 to 3 parts of angelica extract, 0.5 to 5 parts of glossy privet fruit extract, 0.1 to 2 parts of drynaria extract, 0.5 to 5 parts of ginger extract, 0.1 to 3 parts of biota orientalis extract, 0.1 to 3 parts of saussurea involucrata extract, 0.1 to 2 parts of capsicum extract, and 0.1 to 2 parts of citrus extract.

The natural anti-hair loss composition according to claim 1, wherein the polygonum multiflorum extract is prepared by the following steps: cleaning tuber fleeceflower root, drying, crushing, soaking in solvent, reflux extracting, filtering and concentrating to obtain extractum, stepped extracting with solvent, filtering and concentrating to obtain coarse fraction, column chromatographic treatment of the coarse fraction, and concentrating the eluted fraction to obtain tuber fleeceflower root extract.

The natural anti-hair loss composition according to claim 1, wherein the angelica extract is prepared by the following steps: soaking radix Angelicae sinensis powder in solvent, reflux extracting, filtering, concentrating to obtain extract, subjecting the extract to fractional extraction with solvent, filtering, concentrating to obtain crude fraction, subjecting the crude fraction to column chromatography, and concentrating the eluted fraction to obtain radix Angelicae sinensis extract.
