本发明涉及医药技术领域,尤其涉及中药提取液、制备方法及其在口服液中的应用。本发明以枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻为原料制备口服液具有滋肾养肝,同时提高口服液中有效成分的含量且改善口服液的澄清度、稳定性并改善口感,操作方法简便、实用、安全、高效。

本发明涉及医药技术领域,尤其涉及中药提取液、制备方法及其在口服液中的应用。
背景技术
口服液吸收了中药注射剂的工艺特点,是将汤剂进一步精制、浓缩、灌封、灭菌而得到的。口服液具有服用剂量小、吸收较快、质量稳定、携带和服用方便、易保存等优点,尤其适合工业化生产。
其中中药提取液的制备方法包括水提法、有机溶剂提取法、微波法、超声法等手段,其中提取分离技术是中药领域的关键技术,提取液中含有很多杂质,达到提纯与精制的目的,通常采用醇沉、高速离心、膜分离或絮凝等去除杂质。提取分离过程尝尝存在有效成分含量可控性差、产品质量不稳定、贮藏和运输不便、选择性差、周期长、有效物质浸出率低等问题。
CN113456728A公开了一种治疗慢性乙型肝炎的中药口服液及其制备方法,制成中药口服液的原材料为:黄芪、归尾、柴胡、川芎、白芍、白术、败酱草、枝子、丹参、丹皮、枳实、厚朴、黄连、黄岑、茵陈、生军、甘草。本发明的中药口服液主要治疗治疗慢性乙型肝炎,有疏肝健脾、清肝利湿的功效,可促进肝功能恢复,修复受损肝组织,降低转氨酶,预防和治疗肝纤维化等作用,对慢性乙型肝炎具有良好临床疗效。但是该发明并未改善口服液的澄清度、稳定性。
发明内容
有鉴于现有技术的上述缺陷,本发明所要解决的技术问题是提供一种具有优于澄清度、稳定性的中药提取液同时改善其口感。
为实现上述目的,本发明提供了一种中药提取液,其特征在于:包括制备中药混合粉末、提取混合液、除杂、超滤。
一种中药提取液的制备方法,包括以下步骤:
步骤1:对1-5重量份枸杞、3-6重量份菊花、1-3重量份熟地黄、1-10重量份山茱萸、1-5重量份牡丹皮、2-4重量份山药、2-4重量份茯苓、1-3重量份泽泻清除杂质后粉碎过100-150目筛,得到中药混合粉末;
步骤2:将中药混合粉末加入100重量份水中混合均匀,然后加入3-5重量份葡萄糖、0.1-0.2重量份磷酸二氢钾、0.1-0.2重量份尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种3-5wt%发酵菌,在30-35℃发酵12-24h;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入50-60重量份水中进行微波处理3-5min,其中微波功率为250-400W,过滤取上清液,得到提取液B;
步骤4:将提取液A和提取液B混合均匀,得到提取混合液;
步骤5:将果胶酶加入提取混合液中加热至30-35℃酶解20-30min,然后加入除杂剂搅拌均匀再静置1-3h,过滤,得到澄清液,其中除杂剂的加入量为0.1-0.5g/L,果胶酶的加入量为提取混合液的3-5wt%;
步骤6:将上述澄清液进行超滤,其中超滤选用5000-8000Da的超滤膜,得到中药提取液。
所述发酵菌为植物乳杆菌、罗伊氏乳杆菌、动物双歧杆菌、嗜酸乳杆菌中的一种或多种混合物。
所述除杂剂的制备方法,包括以下步骤:
S1将双六甲基三胺加入环氧氯丙烷中混合均匀,加热至45-55℃反应1-3h,反应结束后,减压蒸馏、干燥,得到中间产物,其中双六甲基三胺与环氧氯丙烷的质量比为(1-2):1;将淀粉加入1-5wt%氢氧化钠水溶液中混合均匀,加热至45-55℃℃反应1-2h,然后加入中间产物,加热至60-70℃反应5-8h,反应结束后冷却至室温,采用0.05-0.1mol/L乙酸中和反应,最后离心取沉淀、洗涤、干燥,得到改性淀粉,其中淀粉与1-5wt%氢氧化钠水溶液的固液比为1g:(10-15)mL,淀粉与中间产物的质量为10:(1-3);
S2将海藻多糖、步骤S1得到的改性淀粉混合均匀,粉碎过100目筛,得到除杂剂,其中海藻多糖、改性淀粉的质量比为(0.5-1.2):1。
本发明以枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻为原料与葡萄糖、磷酸二氢钾、尿素混合均匀作为发酵基质,接种发酵菌进行发酵,提高中药原料中有效成分的溶出率,进一步提高其利用率及其提取率;进一步采用微波方法对经发酵处理后的滤渣进行处理,提高有效成分的溶出同时改善提取率;将发酵得到提取液与微波处理后的提取液混合得到提取混合液;虽然提取混合液中含有大量的有效成分,但是提取混合液中含有大量的淀粉、蛋白质、粘液质、靴质、色素、树脂、树胶等无效成分,在提取液中稳定性差、澄清度不好、贮藏和运输不便等问题的存在,进一步影响外观及其口感。
在上述基础上,添加果胶酶,果胶酶可以软化、酶解提取液中存在的大量淀粉、蛋白质、粘液质、靴质、色素、树脂、树胶等,使得物质分解为小分子,降低其黏度,使得原有固形物沉淀下来,增加澄清度,同时能保持中药原来的口感;将海藻多糖与改性淀粉加入酶解后的提取混合液进一步除去悬浮物,改善澄清度、及其稳定性、延长储存时间。本发明中采用生物酶解、电中和、凝聚、吸附等方式对提取混合液中的杂质进行除杂、澄清处理,改善澄清度、及其稳定性、延长储存时间。
海藻多糖由不同的单糖基通过糖苷键相连而成,含有大量的硫酸基,具有高黏度或凝固的能力。
本发明中以双六甲基三胺、环氧氯丙烷、淀粉为原料制备改性淀粉,具有良好的吸附和凝聚双重作用,使得提取液中的蛋白质、鞣质、色素、果胶等大分子物质快速凝聚沉降,制备的改性淀粉中含有胆量正电荷可与提取液中带负电荷的树胶、纤维素等杂质相互作用形成沉淀,海藻多糖与改性淀粉相互作用对蛋白质、糖类、鞣质、色素、细菌等具有很强的吸附能力,对中药提取液中的重金属具有捕集作用,而对微量元素钙、锰、锌等不产生影响,并保持良好的口感,改善提取液的除杂、澄清能力。
本发明还公开了中药提取液在口服液中的应用。

所述甜味剂为蔗糖、蜂蜜、低聚果糖、木糖醇中的一种或多种混合物。

菊花(拉丁学名:Dendranthema morifolium(Ramat.)Tzvel.):在植物分类学中是菊科、菊属的多年生宿根草本植物;甘苦、凉;功能主治:疏风,清热,明目,解毒;治头痛,眩晕,目赤,心胸烦热,疔疮,肿毒。
熟地黄为玄参科植物地黄或怀庆地黄的根茎,经加工蒸晒而成;甘、微温,具有滋阴、补血等功效。
山茱萸(学名:Cornus officinalis Sieb.et Zucc.)落叶乔木或灌木,果肉内含有16种氨基酸,同时含有大量人体所必需的元素。另外,含有生理活性较强的皂甙原糖、多糖、苹果酸、酒石酸、酚类、树脂、鞣质和维生素A、C等成分。其味酸涩,具有滋补、健胃、利尿、补肝肾,益气血等功效。
牡丹皮为毛茛科植物牡丹干燥根皮,苦、辛,微寒,归心、肝、肾经,具有清热凉血、活血化淤、退虚热等功效。
山药又称薯蓣,薯蓣(学名:Dioscorea oppositifolia L.)是薯蓣科薯蓣属植物,甘、温、平、无毒,即可食用又可药用入药能补脾胃亏损,治气虚衰弱、消化不良等功效。
茯苓(学名:Wolfiporia cocos(F.A.Wolf)Ryvarden&Gilb.)是多孔菌科、茯苓属真菌。菌丝体呈白色绒毛状,味甘、淡、性平无毒,入心、脾、肺,肾四经,具有渗湿利水,益脾胃保肾,安神生津等功效。
泽泻(学名:Alisma plantago-aquatica Linn.),多年生水生或沼生草本,甘、寒,功能主治:利水,渗湿,泄热。治小便不利,水肿胀满,呕吐,泻痢,痰饮,脚气,淋病,尿血。
本发明的有益效果:
本发明以枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻为原料制备口服液具有滋肾养肝,同时提高口服液中有效成分的含量且改善口服液的澄清度、稳定性并改善口感,操作方法简便、实用、安全、高效。

植物乳杆菌,Lactobacillus plantarum,CICC22698,购买自中国工业微生物菌种保藏管理中心。
罗伊氏乳杆菌,Lactobacillus reuteri,CICC6131,购买自中国工业微生物菌种保藏管理中心。
双六甲基三胺(Bis(hexamethylene)triamine),CAS:143-23-7。
果胶酶,酶活力:500u/mg,购买自上海源叶生物科技有限公司;
实施例中采用的淀粉为玉米淀粉。
实施例1
一种中药提取液的制备方法,包括以下步骤:
步骤1:对5重量份枸杞、6重量份菊花、3重量份熟地黄、5重量份山茱萸、3重量份牡丹皮、4重量份山药、2重量份茯苓、2重量份泽泻清除杂质后粉碎过100目筛,得到中药混合粉末;
步骤2:将中药混合粉末加入100重量份水中混合均匀,然后加入5重量份葡萄糖、0.2重量份磷酸二氢钾、0.2重量份尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种5wt%发酵菌,在35℃发酵20h;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入50重量份水中进行微波处理3min,其中微波功率为300W,过滤取上清液,得到提取液B;
步骤4:将提取液A和提取液B混合均匀,得到提取混合液;
步骤5:将上述提取混合液进行超滤,其中超滤选用8000Da的超滤膜,得到中药提取液。
步骤2中所述发酵菌由植物乳杆菌、罗伊氏乳杆菌按质量比1:1混合而成。
实施例2
一种中药提取液的制备方法,包括以下步骤:

所述除杂剂的制备方法,包括以下步骤:
S1将双六甲基三胺加入环氧氯丙烷中混合均匀,加热至50℃反应1h,反应结束后,减压蒸馏、干燥,得到中间产物,其中双六甲基三胺与环氧氯丙烷的质量比为2:1;将淀粉加入3wt%氢氧化钠水溶液中混合均匀,加热至50℃反应2h,然后加入中间产物,加热至70℃反应5h,反应结束后冷却至室温,采用0.1mol/L乙酸中和反应,最后离心取沉淀、洗涤、干燥,得到改性淀粉,其中淀粉与3wt%氢氧化钠水溶液的固液比为1g:15mL,淀粉与中间产物的质量为10:3;
S2将海藻多糖、步骤S1得到的改性淀粉混合均匀,粉碎过100目筛,得到除杂剂,其中海藻多糖、改性淀粉的质量比为0.5:1。
实施例4
一种中药提取液的制备方法,包括以下步骤:
步骤1:对5重量份枸杞、6重量份菊花、3重量份熟地黄、5重量份山茱萸、3重量份牡丹皮、4重量份山药、2重量份茯苓、2重量份泽泻清除杂质后粉碎过100目筛,得到中药混合粉末;
步骤2:将中药混合粉末加入100重量份水中混合均匀,然后加入5重量份葡萄糖、0.2重量份磷酸二氢钾、0.2重量份尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种5wt%发酵菌,在35℃发酵20h;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入50重量份水中进行微波处理3min,其中微波功率为300W,过滤取上清液,得到提取液B;

实施例5
一种中药提取液的制备方法,包括以下步骤:
步骤1:对5重量份枸杞、6重量份菊花、3重量份熟地黄、5重量份山茱萸、3重量份牡丹皮、4重量份山药、2重量份茯苓、2重量份泽泻清除杂质后粉碎过100目筛,得到中药混合粉末;
步骤2:将中药混合粉末加入100重量份水中混合均匀,然后加入5重量份葡萄糖、0.2重量份磷酸二氢钾、0.2重量份尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种5wt%发酵菌,在35℃发酵20h;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入50重量份水中进行微波处理3min,其中微波功率为300W,过滤取上清液,得到提取液B;
步骤4:将提取液A和提取液B混合均匀,得到提取混合液;
步骤5:将果胶酶加入提取混合液中加热至35℃酶解30min,然后加入除杂剂搅拌均匀再静置2h,过滤,得到澄清液,其中除杂剂的加入量为0.5g/L,果胶酶的加入量为提取混合液的3wt%;
步骤6:将上述澄清液进行超滤,其中超滤选用8000Da的超滤膜,得到中药提取液。

测试例1
透光率测试
采用分光光度法(UV-1600型紫外-可见分光光度计,上海美普达仪器有限公司)分别对实施例1-5制备得到的中药提取液进行测试,以蒸馏水作参比在625nm下测定透光率。

本发明采用果胶酶、除杂剂结合的手段,改善中药提取液的澄清度,除去中药提取液中无效成分如淀粉、蛋白质、粘液质、靴质、色素、树脂、树胶等,同时保留有效成分存在,避免有效成分流失。
测试例2
清除DPPH自由基活性测定:DPPH·法是一种简单快速的自由基分析方法,其原理是依据稳定自由基DPPH·在甲醇溶液中呈深紫色,在517nm波长处有一强吸收,自由基清除剂通过与其单电子配对使其颜色逐渐消失,褪色程度与接收电子数成化学计量关系。因此,通过吸光度的变化可以衡量试样清除自由基的能力。抗氧化剂清除自由基的清除率越高,其抗氧化性越强。
(1)试剂:0.1mM DPPH-甲醇溶液,0.05mol/L Tris-HCl缓冲液
(2)操作步骤:
样品管:2mL DPPH-甲醇溶液,0.9mL Tris-HCl缓冲液和0.5mL口服液。
对照管:2mL DPPH-甲醇溶液,0.9mL Tris-HCl缓冲液和0.5mL甲醇的混合液。
蒸馏水管调零。
加入样品后准确黑暗反应30min,在517nm处测吸光值。
(3)计算公式:清除率(%)=(对照管吸光度-样品管吸光度)/对照管吸光度)×100。
表2口服液的抗氧化性能测试结果
本发明以枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻为原料与葡萄糖、磷酸二氢钾、尿素混合均匀作为发酵基质,接种发酵菌进行发酵,提高中药原料中有效成分的溶出率,进一步提高其利用率及其提取率;进一步采用微波方法对经发酵处理后的滤渣进行处理,提高有效成分的溶出同时改善提取率;将发酵得到提取液与微波处理后的提取液混合得到提取混合液;虽然提取混合液中含有大量的有效成分,但是提取混合液中含有大量的淀粉、蛋白质、粘液质、靴质、色素、树脂、树胶等无效成分,在提取液中稳定性差、澄清度不好、贮藏和运输不便等问题的存在,进一步影响外观及其口感。
在上述基础上,添加果胶酶,果胶酶可以软化、酶解提取液中存在的大量淀粉、蛋白质、粘液质、靴质、色素、树脂、树胶等,使得物质分解为小分子,降低其黏度,使得原有固形物沉淀下来,增加澄清度,同时能保持中药原来的口感;将海藻多糖与改性淀粉加入酶解后的提取混合液进一步除去悬浮物,改善澄清度、及其稳定性、延长储存时间。本发明中采用生物酶解、电中和、凝聚、吸附等方式对提取混合液中的杂质进行除杂、澄清处理,改善澄清度、及其稳定性、延长储存时间。
海藻多糖由不同的单糖基通过糖苷键相连而成,含有大量的硫酸基,具有高黏度或凝固的能力。
本发明中以双六甲基三胺、环氧氯丙烷、淀粉为原料制备改性淀粉,具有良好的吸附和凝聚双重作用,使得提取液中的蛋白质、鞣质、色素、果胶等大分子物质快速凝聚沉降,制备的改性淀粉中含有胆量正电荷可与提取液中带负电荷的树胶、纤维素等杂质相互作用形成沉淀,海藻多糖与改性淀粉相互作用对蛋白质、糖类、鞣质、色素、细菌等具有很强的吸附能力,对中药提取液中的重金属具有捕集作用,而对微量元素钙、锰、锌等不产生影响,并保持良好的口感,改善提取液的除杂、澄清能力。
实施例6
一种口服液,由以下重量份原料组成:实施例1得到的中药提取液、10重量份蔗糖、0.15重量份苯甲酸钠。
实施例7
一种口服液,由以下重量份原料组成:实施例2得到的中药提取液、10重量份蔗糖、0.15重量份苯甲酸钠。
实施例8
一种口服液,由以下重量份原料组成:实施例3得到的中药提取液、10重量份蔗糖、0.15重量份苯甲酸钠。
实施例9
一种口服液,由以下重量份原料组成:实施例4得到的中药提取液、10重量份蔗糖、0.15重量份苯甲酸钠。
实施例10
一种口服液,由以下重量份原料组成:实施例5得到的中药提取液、10重量份蔗糖、0.15重量份苯甲酸钠。
测试例3
分别将实施例6-10制备得到的口服液置于25℃±2℃、相对湿度为60%±10%条件下静置24个月,考察口服液的形状。

1.一种中药提取液的制备方法,其特征在于,包括以下步骤:
步骤1:对枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻清除杂质后粉碎过筛,得到中药混合粉末;
步骤2:将中药混合粉末加入水中混合均匀,然后加入葡萄糖、磷酸二氢钾、尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种发酵菌,发酵;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入水中进行微波处理,得到提取液B;
步骤4:将提取液A和提取液B混合均匀,得到提取混合液;
步骤5:将提取混合液进行超滤,得到中药提取液。
2.如权利要求1所述的中药提取液的制备方法,其特征在于,包括以下步骤:
步骤1:对枸杞、菊花、熟地黄、山茱萸、牡丹皮、山药、茯苓、泽泻清除杂质后粉碎过筛,得到中药混合粉末;
步骤2:将中药混合粉末加入水中混合均匀,然后加入葡萄糖、磷酸二氢钾、尿素搅拌均匀后得到发酵基质;最后向发酵基质中接种发酵菌,发酵;发酵结束后,固液分离,得到滤渣A和提取液A;
步骤3:将滤渣A加入水中进行微波处理,得到提取液B;
步骤4:将提取液A和提取液B混合均匀,得到提取混合液;
步骤5:将果胶酶加入提取混合液中进行酶解处理,然后加入除杂剂搅拌均匀再静置,过滤,得到澄清液;
步骤6:将上述澄清液进行超滤,得到中药提取液。
3.如权利要求1或2所述的中药提取液的制备方法,其特征在于,所述发酵菌为植物乳杆菌、罗伊氏乳杆菌、动物双歧杆菌、嗜酸乳杆菌中的一种或多种混合物。
4.如权利要求1或2所述的中药提取液的制备方法,其特征在于,步骤1中各原料的用量为:1-5重量份枸杞、3-6重量份菊花、1-3重量份熟地黄、1-10重量份山茱萸、1-5重量份牡丹皮、2-4重量份山药、2-4重量份茯苓、1-3重量份泽泻。
5.如权利要求2所述的中药提取液的制备方法,其特征在于,步骤5中所述除杂剂的加入量为0.1-0.5g/L,果胶酶的加入量为提取混合液的3-5wt%。
6.一种中药提取液,其特征在于,由权利要求1-5中任一项所述的中药提取液的制备方法制备而成。
7.如权利要求6所述的中药提取液在口服液中的应用。
