Description
Technical Field
The application relates to the technical field of skin care products, in particular to an anti-inflammatory moisturizing skin external composition, a preparation method and application thereof.
Background
Skin is the largest organ of the human body. The skin is used as a barrier for the human body to contact with the outside, and mainly consists of three layers of epidermis, dermis and subcutaneous tissue, thereby playing an important role in protecting the health of the human body. The skin barrier is composed of sebum membrane, keratin, lipid, etc., and has effects in resisting external harmful substances, irritants, ultraviolet rays, etc., keeping moisture, and regulating antiinflammatory effects. However, due to the current cosmetic and cosmetology mess, the skin of most people has problems of impaired barrier and skin sensitivity, thereby causing dry skin, skin aging, pigmentation, even skin allergy, etc.
There are many ways to prevent skin allergy, and the general one can be classified into the following categories.
Away from the allergen: allergy is an immune abnormal reaction and an immune regulation disorder induced by a person having an allergic constitution after exposure to an allergen. It is important to prevent skin allergies by avoiding contact with the allergy causing substances as much as possible. The allergen is a necessary condition for anaphylactic reaction, and the allergen is separated, so that the anaphylactic reaction can be avoided, but the allergens of different human bodies are quite different, so that the allergen does not have an effect on all allergic patients.
Antihistaminic substances are used: antihistamines are antagonizing the biological effects of histamine on the human body, i.e., the use of antihistamine substances. The antihistamine substance can inhibit the mast cells from releasing allergic substances such as histamine and inhibit the division of eosinophils and mast cells, thereby preventing the aggregation of the cells at inflammation sites, relieving inflammatory reaction and achieving the purpose of treatment. However, antihistamines are mostly pharmaceutical ingredients, which are not allowed to be used in cosmetics.
Scavenging free radicals: recent studies have shown that the formation of allergic constitution, as well as the onset of allergy, is associated with excessive accumulation of free radicals in the body. The serious environmental pollution, chemical abuse and radiation problems all cause the accumulation of free radicals in the body directly. The allergic constitution can be gradually improved by scavenging free radicals and regulating the immunity of the organism, but the required time is longer and the short-term effect is not obvious.
Antipruritic, anti-irritant: skin itch is a common complication of sensitive skin, and causes of skin itch are numerous, mainly due to dry skin. On the one hand, the treatment of skin itch depends on medicines, but more importantly, patients pay attention to the nursing of skin, and control scratching, scratching can lead to the malignant circulation that the scratching is more itchy and the scratching is more itchy, so that symptoms are further aggravated. Chemicals typically first damage the cell membrane, reducing cellular tolerance, causing the cell to secrete various inflammatory factors, which in turn trigger inflammation. Thus, by increasing cell tolerance, protecting cell membranes is one of the effective ways to prevent skin allergy.
Enhancing skin barrier function: abnormality in the lipid barrier of the skin not only reduces the water storage and moisturizing functions of the skin, but also directly affects the growth and differentiation regulation of keratinocytes, affecting the formation of healthy horny layer, which is the core part of the physical barrier structure of the skin, and repairing or enhancing the barrier function of the skin is an effective way of reducing skin sensitivity.
In addition, some people have intact facial skin barrier, and the problems of skin sensitivity, aging, dark yellow, water deficiency and the like are caused by ultraviolet irradiation, acne, vaccinia and the like.
The moisture of the skin horny layer has important significance for maintaining skin health, defending various stimuli received from the outside, preventing skin aging, moistening and beautifying skin. To make the skin smooth, soft and elastic, it is necessary to maintain the water content of the stratum corneum at an optimum level, and it is generally accepted that the water content is between 10% and 20% and less than 10% of the skin will dry, rough and even chapped. In addition, keeping the skin moist is also a prerequisite for improving the physiological environment of the skin and promoting skin metabolism.
The moisturizing composition is characterized in that the moisturizing composition contains moisturizing components, can keep a certain moisture content of skin horny layer, can increase skin moisture and humidity and restore skin luster and elasticity, and mainly has the effects of resisting inflammation, resisting cell differentiation and relieving itching by repairing the barrier function of epidermis and increasing the moisture content of epidermis, and can be used for health skin and also can be used for treating skin diseases.
The moisturizing composition functions in three ways: preventing excessive evaporation of water; the moisture retention is good; so that the skin breathing function is smooth. The moisturizing composition aims at keeping proper moisture in the skin, especially the outermost layer of the skin, and is characterized in that the moisturizing composition can not only keep the balance of the moisture of the skin, but also supplement important oily components, hydrophilic moisturizing components and moisture, and can be used as an active component and a carrier of a medicament, so that the moisturizing composition is easy to be absorbed by the skin, and the aims of conditioning and nourishing the skin are fulfilled, so that the skin is moisturized and healthy.
Because the natural moisturizing factors are contained in the human body, the skin has the moisture absorption and moisturizing capability, the main components of the natural moisturizing factors are the nutrients and metabolites of the human body such as urea, amino acid, lactate, pyrrolidone carboxylic acid and the like, the natural moisturizing factors have strong hydrophilicity, the moisture loss can be effectively prevented, the lipid and protein of the keratinocytes together form a cell membrane for protecting the natural moisturizing factors, the loss of the natural moisturizing factors is prevented, and the stratum corneum maintains a certain water content. If the integrity of the stratum corneum is damaged, the natural moisturizing factor is lost, and the moisturizing effect of the skin is reduced, so that the research on the natural moisturizing factor has important significance for the development of skin care products.
A great deal of research has been conducted by this related investigator in related skin care products, wherein:

CAM preparation: egg candling is performed by marking the air chamber locations on the eggshell surface, and the labeled eggshell portions are peeled off with tweezers to expose the white eggshell membrane, taking care not to disrupt the integrity of the eggshell membrane. 0.5mL of 0.9% NaCl solution is added dropwise to fully wet the egg membrane, surface liquid is gently sucked by a paper towel, and the inner membrane is carefully removed by forceps, so that the vascular membrane is not damaged.
0.3ml of transparent test substance is directly dripped on the surface of CAM, the condition of CAM reaction is observed, and the occurrence time of each toxic effect within 5min of action is recorded. Reaction end point method: and (3) uniformly dripping or smearing 0.3mL of a non-transparent test object on the surface of the CAM, washing the test object by using 0.9% NaCl solution after 3min of action, and observing the degree of each toxicity effect change of the CAM.
Stimulation scoring (irritation score IS): the stimulation score (IS) was calculated using a reaction time method, and the results remained two decimal places. Subject eye irritation IS classified according to table 1 based on the calculated IS values.
Irritation score is= (301-SecH) ×5/300+ (301-SecL) ×7/300+ (301-SecC) ×9/300
Wherein: sec H (bleeding time) -the average time in seconds(s) at which bleeding is observed to begin on the CAM membrane;
sec L (vascular thawing time vessel lysis time) -the average time in seconds(s) at which onset of vascular thawing is observed on the CAM membrane;
sec C (clotting time) -the average time in seconds(s) at which clotting begins to occur is observed on the CAM membrane.
TABLE 1 reaction time results criterion
Stimulation scoring | IS<1 | 1≤IS<5 | 5≤IS<9 | IS≥10 |
Irritation classification | No irritation | Light irritation | Moderate irritation | Strong irritation/corrosiveness |
Endpoint scoring method (end point score ES): the end point score (ES) should be calculated for the test performed using the reaction end point method, with the result remaining two decimal places. Score per chick embryo = sum of extent of bleeding, clotting and vascular thawing observed per chick embryo, ES = mathematical sum of 6 chick embryo scores. The eye irritation of the test subjects was classified according to the ES values as shown in table 2.
TABLE 2 determination criteria for end-of-reaction results
Stimulation scoring | ES≤12 | 12<ES<16 | ES≥16 |
Irritation classification | No/light irritation | Moderate irritation | Strong irritation/corrosiveness |

2.4 test results: the skin elasticity test results are detailed in table 6 below.
The rate of increase = [ (28 day elasticity value-0 day elasticity value)/0 day elasticity value ]. Times.100%
TABLE 6 detection results of application examples 1-16 and application comparative examples 1-2, 5-6
The difference between application examples 1 to 3, application examples 5 to 7 and application examples 12 to 14 is that: the skin care products prepared in application examples 1 to 3 do not contain a thickening agent and an emulsifying agent, application examples 5 to 7 contain an emulsifying agent, and the skin care products prepared in application examples 12 to 14 contain a thickening agent and an emulsifying agent;
the application examples 8-11 differ in that: the proportions of the plant soothing component, the skin conditioning agent and the emollient are different;
the difference between application example 15 and application example 16 is that: the types and proportions of the thickener and the emulsifier are different.
From the test results in Table 6 above, it can be seen that:
application examples 5-7 the addition of the emulsifier based on application examples 1-3 did not significantly increase the skin elasticity enhancing effect of the anti-inflammatory moisturizing skin care product prepared by the method, indicating that the addition of the emulsifier did not significantly affect skin elasticity.
Application examples 12-14 the addition of the thickener and emulsifier based on application examples 1-3 resulted in an increase in the skin elasticity enhancing effect of the anti-inflammatory moisturizing skin care product, but not significantly, indicating that the addition of the thickener and emulsifier does not significantly enhance skin elasticity.
Application examples 8-11 change the ratio of plant soothing components, skin conditioning agents and emollients, which can influence the effect of anti-inflammatory moisturizing skin care products on enhancing skin elasticity to a certain extent, and show that the ratio can influence the interaction among the components, so that the skin external composition has better effect on enhancing skin elasticity, and in example 11, the plant soothing components are controlled to be a mixture of chamomile extract, white tea extract, calendula extract, licorice extract, motherwort extract and avocado extract in a mass ratio of 4:1:4:2:2:1; the skin conditioner is a mixture of nicotinamide, panthenol, ceramide and allantoin in a mass ratio of 5:2:1:4; the humectant is a mixture of glycerin, glyceroglycosides and hyaluronic acid in a mass ratio of 5:2:3; the skin external composition has the best effect of increasing skin elasticity, namely the anti-inflammatory moisturizing skin care product has the best moisturizing effect, wherein the skin external composition is prepared from the mixture of jojoba oil, shea butter, isooctyl palmitate and isopropyl myristate in a mass ratio of 4:2:1:3.
Application example 15 and application example 16 although the kinds and proportions of the thickener and the emulsifier are different, it is seen from the detection result that the effect of the two skin external compositions on enhancing skin elasticity is not significantly different; however, the addition of a specific kind and specific ratio of the thickener and emulsifier to the application example 11 improves the effect of the anti-inflammatory moisturizing skin care product on enhancing skin elasticity to some extent.
The replacement of urea with a humectant in comparative example 1 reduced the skin elasticity enhancing effect of the composition to some extent; comparative example 2 the replacement of the plant soothing component with a mixed extract of ophiopogon root and nopal stem also affects the skin elasticity enhancing effect of the composition to some extent, but does not affect much; comparative examples 3-4 are prior art disclosed products having significantly lower skin elasticity enhancing effects than the present invention.
4. Antiallergic Performance test
Hyaluronidase is a participant of anaphylactic reaction, has strong correlation with inflammation and allergy, and various medicines for releasing histamine by fat large cells can regulate the activity of the hyaluronidase and inhibit the activity of the hyaluronidase as indexes for researching the antiallergic effect. Thus, the in vitro hyaluronidase inhibition assay Elson-Morgan method was used.
0.1mL of 0.25mmol/LCaCl was taken 2 Incubating the solution and 0.5mL hyaluronidase solution at 37deg.C for 20min; 0.5g of skin external composition sample is added respectively, and the incubation at 37 ℃ is continued for 20min; adding 0.5mL sodium hyaluronate solution, maintaining the temperature at 37 ℃ for 30min, and standing at normal temperature for 5min; 0.1mL of 0.4mol/LNaOH solution and 0.5mL of acetylacetone solution were added, and immediately after heating in a boiling water bath for 15min, cooling with ice water for 5min; 1.0mL of the Escherichia reagent was added and diluted with 3.0mL of absolute ethanol, and the mixture was left for 20 minutes to develop a color, and the absorbance was measured by a spectrophotometer.
The calculation formula is as follows: hyaluronidase inhibition ratio (%) = [ ((a-B) - (C-D))/a-B ] ×100%
Wherein: a represents the ABS value of the control solution, B represents the ABS value of the control blank solution, C represents the ABS value of the sample solution, and D represents the ABS value of the sample blank solution.
The results of the measurements are shown in Table 7 below.

Comparative example 1 | 73.32 |
Comparative example 2 | 71.24 |
Comparative example 5 | 69.53 |
Comparative example 6 | 70.56 |
According to the detection results of the table 7, the anti-inflammatory moisturizing skin care product prepared by the application examples 1-16 has better anti-sensitization effect, and the hyaluronidase inhibition rate can reach more than 75%; and according to the detection results, the anti-allergic effect of the thickening agent and the emulsifying agent on the compound is not obviously improved in the application example 15 and the application example 16; however, in comparative example 1, the replacement of urea with a humectant would reduce the hyaluronidase inhibition rate to some extent, and it is presumed that omitting urea would affect the permeation and absorption of components having anti-inflammatory action, thus affecting the antiallergic effect of the composition; comparative example 2 substitution of plant soothing ingredients with a mixed extract of ophiopogon root and nopal stem also affects the antiallergic effect of the composition to some extent, thereby reducing the hyaluronidase inhibition rate; comparative examples 3-4 are prior art disclosed products with a significantly lower hyaluronidase inhibition than the present invention.
5. Moisture retention test
And (3) moisture retention performance test by an external weighing method: the samples (0.5 g) were weighed and uniformly coated on 5 cm. Times.5 cm glass plates each having a microporous ventilation tape attached thereto, and the glass plates were placed in a constant temperature and humidity dryer, and the mass of each glass plate after being placed for 8 hours was weighed, and the moisture retention rate was calculated.
The formula for calculating the moisture retention rate is as follows: moisture retention/% = (M 2 -M 0 )/(M 1 -M 0 )×100%。
Wherein M is 0 For glass plate mass/g, M 1 For glass plate mass/g after sample addition, M 2 The glass plate mass/g after 8 hours of standing in the dryer.
The results of the measurements are shown in Table 8 below
TABLE 8 Water retention test results
From the test results of table 8 above, it can be seen that:
application examples 5-7 the addition of the emulsifier based on application examples 1-3 does not significantly increase the moisturizing effect of the anti-inflammatory moisturizing skin care product, indicating that the addition of a single emulsifier does not significantly affect the moisturizing effect.
Application examples 12-14 are added with a thickening agent and an emulsifying agent based on application examples 1-3, and the moisturizing effect of the anti-inflammatory moisturizing skin care product prepared by the method is not obviously increased, which indicates that the addition of part of the thickening agent and the emulsifying agent does not obviously influence the moisturizing performance of the product.
Application examples 8-11 change the ratio of plant soothing components, skin conditioning agents and emollients, which can affect the moisturizing performance of the anti-inflammatory moisturizing skin care product to a certain extent, and illustrate that the ratio can affect the interaction between the components, so that the skin external composition has better moisturizing performance, and the application example 11 controls the plant soothing components to be a mixture of chamomile extract, white tea extract, calendula extract, licorice extract, motherwort extract and avocado extract in a mass ratio of 4:1:4:2:2:1; the skin conditioner is a mixture of nicotinamide, panthenol, ceramide and allantoin in a mass ratio of 5:2:1:4; the humectant is a mixture of glycerin, glyceroglycosides and hyaluronic acid in a mass ratio of 5:2:3; the emollient is a mixture of jojoba oil, shea butter, isooctyl palmitate and isopropyl myristate with a mass ratio of 4:2:1:3, and the obtained anti-inflammatory moisturizing skin care product has the best moisturizing effect and is consistent with an elasticity detection result.
Application example 15 and application example 16 although the kinds and proportions of the thickener and the emulsifier are different, it is seen from the detection result that the moisturizing effect of the two skin external compositions is not significantly different; however, the addition of a specific kind and specific ratio of thickener and emulsifier to the application example 11 improved the moisturizing effect of the compound to some extent.
The replacement of urea with a humectant in comparative example 1 reduced the moisturizing effect of the composition to some extent, indicating that the replacement of the moisturizing component did not achieve a similar moisturizing effect; comparative example 2 the replacement of the plant soothing component with a mixed extract of ophiopogon root and nopal stem also affects the moisturizing effect of the composition to some extent, but not much; comparative examples 3-4 are prior art disclosed products with a significantly lower moisturizing effect than the present invention.
6. Stability test
The test method comprises the following steps: the skin external preparation compositions prepared in application examples 11, 15 to 16 and comparative examples 3 to 4 were stored at 40℃for 24 hours, at room temperature for 24 hours, and at-5℃for 24 hours, and after 3 cycles in this order, it was observed whether or not there was delamination, oil-slick, and color change after 24 hours at room temperature, and the results are shown in Table 9 below.

As can be seen from the detection results of the table 9, the anti-inflammatory moisturizing skin care products prepared in the application examples 15-16 have better stability, and the conditions of layering, oil slick and color change can not be generated after the cold and hot circulation; the anti-inflammatory moisturizing skin care product prepared in the application example 11 is not added with a thickening agent and an emulsifying agent, and can change color after cold and hot circulation, but the use is not affected; the stability of the obtained anti-inflammatory moisturizing skin care product is also reduced to a certain extent compared with examples 15-16 by adding specific components and proportions of the components in examples 13-14, and the conditions of layering or color change after cold and hot circulation are obviously affected by weakening interaction between the thickening agent and the emulsifying agent by changing the types of the emulsifying agent or changing the mass ratio of the emulsifying agent and the thickening agent in comparative examples 3-4, which are not in the protection scope of the invention.
It will be apparent that the described embodiments are only individual embodiments of the invention, and not all embodiments. All other implementations, which can be made by those skilled in the art without the benefit of the teachings of this invention, are intended to be within the scope of this invention.
