Table 1: composition of test group face cream
Continuing with table 1: composition of test group face cream
The preparation method of the test group face cream comprises the following steps: (1) Heating water to 70 ℃, adding an emollient, polyglycerol-3 methyl glucose distearate and/or sodium bis (lauramide glutamine) lysine, stirring for 1.5 hours at a rotating speed of 1500 rpm, and then adding a humectant and a thickener to obtain a mixed solution A; (2) Cooling the mixed solution A to 32 ℃, then adding sodium surfactin, cholesterol, ceramide 1, tocopherol and allantoin, and continuously stirring for 0.5 hour at a rotating speed of 1000 revolutions per minute to obtain a mixture B; (3) Continuously cooling to 25 ℃, then adding the falcate elemene fruit extract and/or the coastal cress extract and/or the dog rose fruit extract and/or the prickly pear stem extract, the pH regulator, the preservative, the chelating agent and the essence, and continuously stirring for 0.5 hour at the rotation speed of 1000 revolutions per minute to obtain the soothing and anti-aging face cream.
Anti-aging experimental test:
the invention characterizes its anti-aging properties by analyzing the free radical scavenging ability. 0.1mmol/L DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) solution is prepared by 60 percent ethanol, and the solution is preserved in a dark place. Measuring the absorbance A0 of DPPH at 520nm, taking 0.1-5g of test sample (face cream of the test group) and 1mL of DPPH solution, adding into the same test tube, shaking uniformly, standing in dark at room temperature for 30min, and measuring the absorbance A. The percent DPPH clearance (%) = (A0-a)/A0 x 100% was calculated and then the amount of sample required to reduce the percent DPPH clearance to 50% was used as an index for investigation. The amount of sample (cream of the test group of the present invention) required is shown in table 2.
Table 2: anti-aging test for test group of creams
As can be seen from the antioxidant test of the test group in Table 2 of the present invention, the cream of the present invention has excellent antioxidant properties, and the required amount of the sample for which DPPH clearance percentage is reduced to 50% can be reduced to 0.29g/mL.
As can be seen from a comparison of test group 1 and test groups 7-8, the combination of the polyglycerol-3 methyl glucose distearate and sodium bis (lauramide glutamine) lysine according to the present invention has a synergistic effect, which can provide the cream with excellent anti-oxidation properties, probably because the combination of the two can provide the active ingredients of the cream with a more uniform distribution, thereby providing better effects of the active ingredients, and the active ingredients of the cream can be more easily distributed and absorbed by the skin to provide better effects. As can be seen from a comparison of test groups 1, 4 and test groups 5-6, the mass ratio of polyglycerol-3 methylglucose distearate to sodium di (lauramide glutamine) lysine of the present invention is 1: the range of (0.8-3) enables the cream of the present invention to have a better effect.
As can be seen from the comparison of the test group 1 and the test groups 9-10, the combination of the inventive olive kernel extract and the coastal cress extract has a synergistic effect, so that the inventive facial cream has unexpected oxidation resistance increase.
Anti-irritation Property test: including erythrocyte hemolysis tests and protein denaturation tests.
(1) Erythrocyte hemolysis assay: preparation of erythrocyte suspensions: healthy rabbits are selected, 9mL of heart blood is taken, 1mL of 2% potassium oxalate solution is added, centrifugation is carried out, supernatant fluid is removed, 20mmol/L PBS solution is used for diluting sediment to 20mL, and the sediment is preserved at 4 ℃ for standby. The face cream of the test set of the invention was selected as a sample to be diluted with PBS solution to different concentrations, each sample being set with 5 concentration gradients. Taking 10mL of the dilution of the sample to be tested, adding 200 μl of the above erythrocyte suspension (controlling the final concentration of the sample to be 5, 10, 20, 50, 100mg/mL respectively), taking distilled water as total hemolysis control, taking PBS solution as negative control, gently mixing, incubating at 37deg.C for 30min, centrifuging at 2000r/min for 10min, collecting supernatant, and measuring its absorbance at 560nm with spectrophotometer (A 560 ) Calculating the hemolysis rate according to the following formula; hemolysis rate=100% (experimental group a 560 Negative control group A 560 ) (Whole hemolysis control group A) 560 Negative control group A 560 ). Then, a standard curve of the hemolysis ratio-sample concentration was drawn, and the sample concentration HD50 at which 50% of the erythrocytes were hemolyzed was calculated.
(2) Protein denaturation experiment: the face cream of the test group is selected as a sample, diluted to 10g/L by PBS solution, 10mL of the diluted solution of the sample to be tested is taken, 200 mu L of the erythrocyte suspension is added, distilled water is used as a blank control, 1mg/mL Sodium Dodecyl Sulfate (SDS) solution is used as a positive control, the mixture is gently mixed, incubated for 30min at 37 ℃, centrifuged for 10min at 2000r/min, the supernatant is taken, and the absorbance A of the supernatant at 540nm and 575nm is respectively tested by a spectrophotometer 540 And A 575 Calculating a protein Denaturation Index (DI) according to the following formula; di=100% (R1-R2)/(R1-R3). Wherein r1=blank control group a 575 Blank group A 540 R2=experimental group a 575 Experimental group A 540 R3=positive control group a 575 Positive control group A 540 。
The test results of the above-described erythrocyte hemolysis test and protein denaturation test are shown in table 3 below:
