Abstract

The preparation method of the mixed extract of other natural plants comprises the following steps: adding rhizoma Gastrodiae, radix astragali, radix Angelicae sinensis, rhizoma Ligustici Chuanxiong, cortex Magnolia officinalis, poria, and radix Glycyrrhizae Preparata into an extraction tank, adding 5-10 times of water, soaking for 0.5-1.0 hr, boiling, timing, decocting for 2 hr, and discharging decoction; adding 5-10 times of food water, decocting for 90min, and discharging decoction. Mixing the decoctions, filtering with gauze, and concentrating the filtrate under reduced pressure to obtain extract with density of 1.1-1.3.
Mixing broad bean (with pod) extract and other natural plant mixed extract at a certain ratio (1-8): 1, adding acesulfame potassium 0.05% -0.1%, and packaging after instant killing to obtain the health product beverage.
The parkinsonism belongs to the category of 'tremor' in traditional Chinese medicine, and the disease is located in the brain and relates to liver, kidney and spleen. The pathogenesis of the disease is the deficiency of liver and kidney and the deficiency of qi and blood, and generally adopts the methods of nourishing liver and kidney, tonifying qi and nourishing blood, calming endogenous wind and stopping fibrillation.

The preparation method of the broad bean (with pod) extract comprises the following steps: screening fresh semen Viciae Fabae (with pod), weighing 16.3kg, cleaning, boiling with 1-2 times of water for inactivating enzyme for 1min, filtering to remove water, squeezing, and filtering with 400 mesh filter cloth. Adding 0.01% sodium metabisulfite and 0.2% edetate disodium into the filtrate to obtain broad bean (containing pod) extract.
The preparation method of the mixed extract of other natural plants comprises the following steps: adding 12 parts of gastrodia elata, 12 parts of astragalus membranaceus, 8 parts of angelica sinensis, 2 parts of ligusticum wallichii, 3 parts of magnolia officinalis, 5 parts of poria cocos and 1 part of honey-fried licorice root into an extraction tank, adding 8 times of water of the total weight of the plants, soaking for 0.5 hour, boiling the soaked plants, starting timing, decocting for 2 hours, and discharging a decoction; adding 5 times of food water, decocting for 90min, and discharging decoction. Mixing the decoctions, filtering with gauze, and concentrating the filtrate under reduced pressure to obtain extract with relative density of 1.17 at 25deg.C.
Taking 4 parts of broad bean (with pod) extract and 1 part of other natural plant mixed extract, adding 0.05% acesulfame potassium and 0.05% potassium sorbate, and packaging into 50 ml/bottle after 30s of instant killing at 130 ℃ to obtain the health-care product beverage containing broad bean (with pod) extract.
Example 2: pharmacodynamic experiments
Adult healthy mice were taken in 30 groups, which were divided into blank groups, model groups, test groups 3, and 10 groups each.
Experimental grouping: adult mice were taken 30, 3 groups of 10, and a blank group (normal saline lavage 1-21d, normal saline intraperitoneal injection 22-29 d), a model group (normal saline lavage 1-21d, MPTP (injection: MPTP is 1-methyl-4-phenyl-1, 2,3, 6-tetrahydropyridine, the same applies hereinafter)) 22-29d, and a dosing group (health drink 1-21d of the present invention, MPTP 22-29 d)
Dosage and mode of administration: normal saline (lavage/intraperitoneal injection): 30mg/kg/d
Health beverage (stomach irrigation): 1ml/kg of beverage was prepared as in example 1.
MPTP (intraperitoneal injection): 30mg/kg/d
Frequency of administration: once a day
The behavioral method is used for detecting:
pole climbing experiment: a smooth wood rod with a diameter of 1cm and a height of 50cm and a wood ball with a diameter of 3cm at the top is taken. The mice to be tested were placed on cork pellets during the test, and the time required for the mice to climb to the bottom of the pole was recorded and statistically analyzed. All mice were assayed for pole-climbing time on day 21 of dosing and again after 1h of dosing on day 29.
Rotating shaft experiment: the mice were placed on the rotating stick and the time from the beginning of rotation of the test mice to the falling off the rotating stick was measured. Spindle time was measured for all mice on day 21 of dosing and for 1 hour after dosing on day 29.
Grip strength experiment: the tested mice are placed on a tensile tester, the tail parts of the mice are grasped, the mice are pulled backwards in a straight line, and the grabbing force data are recorded. All mice were measured for grip size on day 21 of dosing and their tension again after dosing on day 29.
Efficacy results:
from the data, it can be seen that: through the Parkinson model of the mice, the administration group can obviously increase the pole climbing time and the rotating shaft time of the mice in the model group and increase the pulling force of the mice. The health-care product beverage has obvious treatment effect on Parkinson model mice.
Example 3: pharmacodynamic experiments
Adult healthy mice were taken in 30 groups, which were divided into blank groups, model groups, test groups 3, and 10 groups each.
Experimental grouping: taking 30 adult mice, dividing the mice into 3 groups, 10 blank control groups (physiological saline lavage 1-21d, physiological saline intraperitoneal injection 22-21 d), model group 1 (physiological saline lavage 1-21d, arecoline modeling), model group 2 (physiological saline lavage 1-21d, oxidation tremorine modeling), and administration group (health care beverage 1-21d, arecoline modeling).
Dosage and mode of administration: normal saline (lavage/intraperitoneal injection): 30mg/kg/d
Health beverage (stomach irrigation): 1ml/kg of beverage was prepared as in example 1.
Betulin (intraperitoneal injection): 30mg/kg/d
Oxidative tremorine (intraperitoneal injection): 0.30mg/kg/d
Frequency of administration: once a day
The behavioral method is used for detecting: the time required for each mouse to develop until disappearance of the visually apparent muscle tremor was recorded as the tremor duration.
Efficacy results:
from the data, it can be seen that: obvious tremors appear after the mice are intraperitoneally injected with arecoline and oxitremors, and the tremors of arecoline groups are longer. However, the administration groups given in advance with the health drink prepared by the invention have obvious shortened duration of tremors, which indicates that the health drink of the invention has obvious improving effect on tremors simulating parkinsonism. The betulin beverage has the effect of relieving tremors formed by arecoline modeling and tremors formed by oxytremors modeling, and has obvious treatment effect on Parkinson model mice.
