Abstract
The invention belongs to the technical field of hair care agents, and particularly relates to an anti-hair-loss hair-growing essence and a preparation method thereof. The anti-hair loss and hair-growth essence contains active ingredients such as lactobacillus fermentation lysate, two-split yeast fermentation product filtrate, plant exosomes and the like, wherein the plant exosomes are at least one of ginseng exosomes, raspberry exosomes and dendrobium exosomes; on the other hand, the alopecia is effectively reduced by effectively inhibiting the activity of 5 alpha reductase, and the effects of preventing alopecia and promoting hair growth are comprehensively achieved.
Description
Technical Field
The invention belongs to the technical field of hair care agents, and particularly relates to an anti-hair-loss hair-growing essence and a preparation method thereof.
Background
In recent years, the proportion of the population suffering from alopecia is higher and the population suffering from alopecia is in a trend of younger age, and research and study show that alopecia phenomena comprise alopecia areata, psychogenic alopecia and androgenic alopecia (seborrheic alopecia), wherein more than 90% of the alopecia population is androgenic alopecia, and the mechanism is mainly that testosterone is converted into Dihydrotestosterone (DHT) with higher biological activity under the action of 5 alpha-reductase, so that the binding of testosterone to a receptor is competitively inhibited, the metabolic process is inhibited, static hair follicles are prevented from entering a growing period, hair follicles are gradually contracted until the hair follicles disappear, and alopecia is caused.
In the alopecia treatment, the western medicines commonly used in clinic are minoxidil and finasteride, but the two medicines are easy to relapse after stopping taking, and can also cause side effects such as skin irritation, scalp itching, facial and hand hirsutism, dizziness, anorexia, tachycardia and the like. In addition, although hair transplantation has a certain therapeutic effect, the clinical application of the hair transplantation is limited due to the large surgical trauma and high cost and the need of multiple surgeries. The effect of non-drug treatment such as acupuncture is also not ideal.
At present, research reports on alopecia treatment by using plant extracts such as traditional Chinese medicines are more and more, and plant source components are relatively safe and effective and are convenient to use. For example, the single or mixed use of astragalus root, fleece-flower root, glossy privet fruit, dodder seed and the like has the effects of stimulating the growth of hair follicles of mice cultivated in vitro, increasing the number of hair follicles, accelerating the hair regeneration speed and the like, however, the simple treatment of the traditional Chinese medicine extract has long treatment period and slow effect, and often causes physiological alopecia when the hair regeneration is carried out, and the effect of comprehensively improving the alopecia is also poor when the speed of promoting the hair regeneration of the traditional Chinese medicine extract is lower than that of the physiological alopecia.
Disclosure of Invention
Aiming at the problems existing in the prior art, the invention aims to provide the hair loss preventing and hair growing essence and the preparation method thereof, which can effectively inhibit the activity of 5 alpha reductase to reduce physiological alopecia, and can promote the proliferation of hair follicle cells to promote regeneration of new hair to comprehensively achieve the hair loss preventing and hair growing effects.
Based on the above purpose, the technical scheme adopted by the invention is as follows:
In a first aspect, the present invention provides an anti-hair-growing composition comprising the following components: lactobacillus fermentation lysate, saccharomyces cerevisiae fermentation product filtrate and plant exosomes;
The plant exosome is at least one of ginseng exosome, raspberry exosome and dendrobium exosome.
The research shows that the hair growth and regeneration depend on the blood supply of the organism to hair follicles, the hair follicles are surrounded by rich vascular beds, when microcirculation is blocked, the blood flow rate in the microcirculation is reduced, the normal supply of nutrients required by the organism to the hair follicles is affected, the hair follicles lack of nutrients, and alopecia occurs, and the hair loss prevention hair care composition can effectively promote the blood circulation of scalp, increase the blood supply of hair papilla, improve the nutrition of hair follicles, slow down the miniaturization process of hair follicles, increase the number of hair follicles and promote hair growth. In addition, the hair loss prevention and hair growth promoting composition can effectively inhibit the activity of 5 alpha reductase, effectively reduce physiological alopecia and comprehensively achieve the effect of improving hair loss prevention and hair growth promoting.
Preferably, the plant exosomes consist of ginseng exosomes, raspberry exosomes and dendrobium exosomes in a weight ratio of 1 (0.2-0.6): 1-1.5.
Experiments show that when the ginseng exosomes, the raspberry exosomes and the dendrobium exosomes are compounded according to the proportion, the prepared hair-loss preventing and hair-growing composition shows better hair-loss preventing and hair-growing effects.
Preferably, the weight ratio of the ginseng exosomes, the raspberry exosomes and the dendrobium exosomes is 1 (0.4-0.5): 1.2-1.5.
Experiments show that when the ginseng exosomes, the raspberry exosomes and the dendrobium exosomes are compounded according to the proportion, the prepared hair-loss preventing and hair-growing composition has better hair-loss preventing and hair-growing effects.
Preferably, the weight ratio of lactobacillus fermentation lysate, the filtrate of the fermentation product of the two yeasts to the plant exosome is (10-17): 8-12): 1.
Experiments show that when lactobacillus fermentation lysate, yeast fermentation product filtrate and plant exosome are compounded according to the proportion, the prepared hair loss preventing hair growth promoting composition shows better hair loss preventing hair growth promoting effect.
Preferably, the weight ratio of lactobacillus fermentation lysate, the filtrate of the fermentation product of the two yeasts to the plant exosomes is (10-15): 10:1.
Experiments show that when lactobacillus fermentation lysate, yeast fermentation product filtrate and plant exosome are compounded according to the proportion, the prepared hair loss preventing hair growth promoting composition shows better hair loss preventing hair growth promoting effect.
In a second aspect, the invention provides an anti-hair loss and hair growth promoting essence, which contains the anti-hair loss and hair growth promoting composition.
Preferably, the anti-hair loss and hair-growing essence comprises the following components in parts by weight: 20 to 45 parts of hair-growing composition with anti-alopecia effect, 6 to 13 parts of humectant, 1 to 5 parts of skin conditioning agent, 3 to 7 parts of antioxidant, 0.1 to 0.5 part of anti-allergic agent, 1 to 3 parts of solubilizer and 80 to 120 parts of water.
Preferably, the anti-hair loss and hair-growing essence comprises the following components in parts by weight: 30-40 parts of hair-growing composition, 6-13 parts of humectant, 1-5 parts of skin conditioner, 3-7 parts of antioxidant, 0.1-0.5 part of anti-allergic agent, 1-3 parts of solubilizer and 80-120 parts of water.
Preferably, the humectant is at least one of 1, 2-hexanediol, glycerol, and sodium hyaluronate; the skin conditioner is at least one of nicotinamide, saccharomycete polypeptide and nicotinamide adenine dinucleotide; the antioxidant is p-hydroxyacetophenone and/or ascorbyl palmitate; the anti-allergic agent is hydroxyphenyl propionamide benzoic acid; the solubilizer is PEG-40 hydrogenated castor oil.
In a third aspect, the present invention provides a method for preparing the above anti-hair loss and hair growth promoting essence, comprising the steps of:
(1) Mixing humectant, solubilizer and water, heating to 90-100 ℃, stirring uniformly to obtain a basic solution, and cooling the basic solution to 60-65 ℃ for standby;
(2) Adding an antioxidant into the basic solution, homogenizing for 1-2 min, then adding an anti-allergic agent and a skin conditioning agent, homogenizing for 1-2 min, and then adding an anti-hair loss composition, homogenizing for 1-2 min to obtain a mixed solution; controlling the homogenizing temperature to be 60-65 ℃, preserving the temperature of the mixed solution at 60-65 ℃ for 0.5-1 h after the homogenizing is finished, and cooling to room temperature to prepare the anti-hair loss and hair-growing essence.

The invention provides an anti-hair-loss hair-growing composition and an essence containing the same, which are characterized in that on one hand, the anti-hair-loss hair-growing essence promotes scalp blood circulation, increases hair papilla blood supply, improves hair follicle nutrition, slows down hair follicle miniaturization process, increases hair follicle number and promotes hair growth; on the other hand, the alopecia is effectively reduced by effectively inhibiting the activity of 5 alpha reductase, and the effects of preventing alopecia and promoting hair growth are comprehensively achieved.
Detailed Description
For a better description of the objects, technical solutions and advantages of the present invention, the present invention will be further described with reference to the following specific examples. It will be appreciated by persons skilled in the art that the specific embodiments described herein are for purposes of illustration only and are not intended to be limiting.
The test methods used in the examples are conventional methods unless otherwise specified; the materials, reagents and the like used, unless otherwise specified, are all commercially available.
Example 1
The embodiment provides an anti-hair loss and hair-growing essence, which comprises the following components in parts by weight: 20 to 45 parts of hair-growing composition with anti-alopecia effect, 6 to 13 parts of humectant, 1 to 5 parts of skin conditioning agent, 3 to 7 parts of antioxidant, 0.1 to 0.5 part of anti-allergic agent, 1 to 3 parts of solubilizer and 80 to 120 parts of water.
Wherein the anti-hair loss hair-growing composition is formed by compounding lactobacillus fermentation lysate, two-split yeast fermentation product filtrate and plant exosome according to the weight ratio of (10-17) (8-12): 1; the plant exosome is compounded with ginseng exosome, raspberry exosome and dendrobium exosome in the weight ratio of 1 (0.2-0.6) to 1-1.5.
The humectant is at least one of 1, 2-hexanediol, glycerol and sodium hyaluronate, preferably 1, 2-hexanediol; the skin conditioner is at least one of nicotinamide, saccharomycete polypeptide and nicotinamide adenine dinucleotide; the antioxidant is p-hydroxyacetophenone and/or ascorbyl palmitate; the anti-allergic agent is hydroxyphenyl propionamide benzoic acid; the solubilizer is PEG-40 hydrogenated castor oil.
The preparation method of the hair loss prevention and hair growth promoting essence comprises the following steps:
(1) Mixing humectant, solubilizer and water, heating to 90-100 ℃, stirring uniformly to obtain a basic solution, and cooling the basic solution to 60-65 ℃ for standby;
(2) Adding an antioxidant into the basic solution, homogenizing for 1-2 min, then adding an anti-allergic agent and a skin conditioning agent, homogenizing for 1-2 min, and then adding an anti-hair loss composition, homogenizing for 1-2 min to obtain a mixed solution; controlling the homogenizing temperature to be 60-65 ℃, preserving the temperature of the mixed solution at 60-65 ℃ for 0.5-1 h after the homogenizing is finished, and cooling to room temperature to prepare the anti-hair loss and hair-growing essence.
In order to explore the proper adding amount of the anti-hair-loss and hair-growth composition in the anti-hair-loss and hair-growth essence, only the weight part of the anti-hair-loss and hair-growth composition is taken as a single variable, the weight parts of other components are kept unchanged, the proportion of each component in the anti-hair-loss and hair-growth composition is kept unchanged, and the anti-hair-loss and hair-growth essence used in a specific test comprises the following components in parts by weight:
10-50 parts of anti-hair loss hair-growing composition; 10 parts of humectant 1, 2-hexanediol; the skin conditioning agent comprises 2 parts of nicotinamide, 2 parts of saccharomycete polypeptide and 1 part of nicotinamide adenine dinucleotide, and the total weight is 5 parts; the antioxidant comprises 5 parts of p-hydroxyacetophenone and 1 part of ascorbyl palmitate, and the total amount is 6 parts; 0.3 parts of an anti-sensitizer, namely, hydroxyphenyl propionamide benzoic acid; 2 parts of PEG-40 hydrogenated castor oil serving as a solubilizer and 100 parts of water.
Wherein the weight ratio of lactobacillus fermentation lysate, yeast fermentation product filtrate and plant exosome in the hair-growth preventing composition is 15:10:1, and the weight ratio of ginseng exosome, raspberry exosome and dendrobium exosome in the plant exosome is 1:0.5:1.5.
The weight parts of the hair-loss preventing composition in the hair-loss preventing composition are shown in table 1, and the 5a reductase inhibition rate and the number of hair follicles on the back of the mice of each hair-loss preventing composition are counted according to the following effect test method, and the results are shown in table 1.
(1) 5 Alpha reductase inhibition effect test
300. Mu.L of PBS buffer (pH 7.4), 500. Mu.L of 5. Alpha. Reductase of 0.5mg/mL, 50. Mu.L of a sample to be tested, 50. Mu.L of 50% ethanol and 100. Mu.L of NADPH are sequentially added to a centrifuge tube, rapidly mixed uniformly for 3s in a vortex mixer, reacted in a constant temperature water bath at 37℃for 30min, rapidly sampled for 400. Mu.L at the beginning of the reaction (at time t 0) and for 30min (at time t 30) respectively, rapidly added with 800. Mu.L of methanol for inactivating the enzyme, and vortexed for 5s to terminate the reaction. And detecting the concentration of 5 alpha reductase at time t 0 and time t 30 of the sample to be detected respectively, and calculating the concentration difference value of the sample to be detected. Detecting the dutasteride serving as a standard substance to obtain a concentration difference value which is a standard concentration difference value; the concentration difference obtained by replacing the test sample with 50 μl of PBS buffer is the blank concentration difference. The inhibition rate of the test sample to the 5 alpha reductase activity is calculated as follows:
Inhibition ratio (%) of 5α reductase activity= (standard concentration difference-test group concentration difference)/(standard concentration difference-blank group concentration difference).
(2) Test for promoting proliferation of mouse hair follicle cells
C57BL/6 pure-bred mice with the weight of 15-20g are taken, the male and female mice are used together, the back hair of the mice is pulled out by using a mixture of rosin and paraffin under the mild anesthesia of diethyl ether, the area is about 3cm multiplied by 4cm, and the mice are grouped, namely a normal control group and each test group. The next day after dehairing, each sample is uniformly smeared on the skin of the dehairing area at the back of each group of mice, the dosage of each test group is 10ml/kg, the same volume of physiological saline group is given to the control group, 1 time/day is continued for 10 days, the skin of the dehairing area at the back of the mice is taken after the administration for 10 days, formalin is used for fixing, pathological sections are made, the hair follicle growth condition is observed, the hair follicle number of 10 mice is counted by each sample, the average value is shown in table 1, and the average value of the hair follicle number of the mice in the control group is 102 hair follicle number/visual field.
(3) Volunteers are recruited to carry out hair loss prevention and hair growth effect test
50 Patients with alopecia are recruited, wherein men account for 80%, women account for 20%, the disease course is 10 years at the longest, 2 years at the shortest, and the ages are between 20 and 60 years. The essence samples of this example were used as test agents, and the patients were applied to the affected parts with the above test agents by applying 2ml each time, 2 times a day (once a day in the morning and evening), and 4 weeks of administration were a single treatment course, followed by 4 continuous treatment courses with no interruption in the middle.
Measurement standard:
The effect is shown: the hair loss phenomenon is avoided, the hair is thick and blackened gradually at the position where the scalp is bald and sparse;
the method is effective: no alopecia phenomenon, and sparse hair grows at the scalp baldness and sparseness;
invalidation: has alopecia phenomenon, and no hair growth occurs at the bald part of the scalp.
The total effectiveness (including efficacy and effectiveness) of the clinical treatment on 50 patients is counted as shown in table 1.

The test results are shown in table 1, along with the increase of the addition amount of the anti-hair-loss composition in the anti-hair-loss essence, the inhibition rate of the anti-hair-loss essence to the 5 alpha reductase tends to be increased and then decreased, it is shown that the inhibition effect on 5α reductase is poor when the content of the hair-growth preventing composition is less than 20 parts by weight, and that the inhibition rate of 5α reductase is less changed when the content of the hair-growth preventing composition is more than 45 parts by weight.
With the increase of the addition amount of the anti-hair-loss and hair-growth combination in the anti-hair-growth essence, the number of the hair follicles of the mice shows a trend of increasing firstly and then stabilizing and then decreasing, and it is presumed that when the addition amount of the anti-hair-loss and hair-growth combination in the anti-hair-loss and hair-growth essence is between 20 parts and 45 parts, especially between 30 and 40 parts, the effect of promoting the hair follicle growth is better and more stable, and then the decrease of the number of the hair follicles is presumed to be due to the fact that the skin is difficult to absorb excessive essence due to the too high concentration of the essence, but is unfavorable for the proliferation of the hair follicles.
When the adding amount of the anti-hair-loss and hair-growth composition in the anti-hair-growth essence is 20-45 parts, the total effective rate of the anti-hair-loss and hair-growth composition for clinical treatment of patients suffering from alopecia reaches 100%.
Example 2
The purpose of this example is to analyze the effect of specific components of the hair-setting composition on hair-setting effect, and the test method is as follows:
In the embodiment, the weight ratio of lactobacillus fermentation lysate, yeast two fermentation product filtrate and plant exosome in the hair loss prevention hair growth combination is taken as a single factor variable, and the hair loss prevention hair growth essence comprises the following components in parts by weight:
40 parts of an anti-hair loss hair-growing composition; 10 parts of humectant 1, 2-hexanediol; the skin conditioning agent comprises 2 parts of nicotinamide, 2 parts of saccharomycete polypeptide and 1 part of nicotinamide adenine dinucleotide, and the total weight is 5 parts; the antioxidant comprises 5 parts of p-hydroxyacetophenone and 1 part of ascorbyl palmitate, and the total amount is 6 parts; 0.3 parts of an anti-sensitizer, namely, hydroxyphenyl propionamide benzoic acid; 2 parts of PEG-40 hydrogenated castor oil serving as a solubilizer and 100 parts of water. Wherein the weight ratio of the ginseng exosomes to the raspberry exosomes to the dendrobium exosomes in the plant exosomes is 1:0.5:1.5.
Wherein the weight ratio of lactobacillus fermentation lysate (A), yeast fermentation product filtrate (B) and plant exosome (C) in the hair loss preventing hair growth promoting composition is shown in Table 2.
Referring to the test method for the inhibition effect of 5α reductase on each sample and the test method for promoting proliferation of mouse hair follicle cells described in example 1, the inhibition rate of 5α reductase and the mean number of hair follicles of each sample described in this example were counted, as shown in table 2.
Meanwhile, referring to the clinical test method described in example 1, the test of the essence sample described in table 2 was carried out on the patients suffering from alopecia, and the total effective rate on the patients suffering from alopecia was counted, and the results are shown in table 2.

As is clear from the results shown in Table 2, when the ratio of the lactobacillus fermentation lysate, the yeast fermentation filtrate, and the plant exosome in the hair-growing composition is within the specific ratio range (10-17): 8-12): 1, the hair follicle proliferation promoting and 5. Alpha. Reductase activity inhibiting effects are more excellent, and when the ratio exceeds the above ratio range, the hair follicle proliferation promoting and 5. Alpha. Reductase activity inhibiting effects are reduced, and the total effective rate is also reduced.
Example 3
The present example aims at analyzing the influence of the composition of plant exosomes in the hair-growth composition on the hair-growth effect of hair-growth, and the test method is as follows:
in the embodiment, the weight ratio of ginseng exosomes, raspberry exosomes and dendrobium exosomes in plant exosomes is used as a single factor variable, and the anti-hair loss hair-growing essence comprises the following components in parts by weight:
40 parts of an anti-hair loss hair-growing composition; 10 parts of humectant 1, 2-hexanediol; the skin conditioning agent comprises 2 parts of nicotinamide, 2 parts of saccharomycete polypeptide and 1 part of nicotinamide adenine dinucleotide, and the total weight is 5 parts; the antioxidant comprises 5 parts of p-hydroxyacetophenone and 1 part of ascorbyl palmitate, and the total amount is 6 parts; 0.3 parts of an anti-sensitizer, namely, hydroxyphenyl propionamide benzoic acid; 2 parts of PEG-40 hydrogenated castor oil serving as a solubilizer and 100 parts of water. Wherein the weight ratio of lactobacillus fermentation lysate, yeast two fermentation product filtrate and plant exosome in the hair loss preventing hair growth promoting composition is 15:10:1. The weight ratios of the ginseng exosomes (D), the raspberry exosomes (E) and the dendrobium exosomes (F) in the plant exosomes are shown in table 3.
Referring to the test method for the inhibition effect of 5α reductase on each sample and the test method for promoting proliferation of mouse hair follicle cells described in example 1, the inhibition rate of 5α reductase and the mean number of hair follicles of each sample described in this example were counted, as shown in table 3.
Meanwhile, referring to the clinical test method described in example 1, the test of the essence sample described in table 3 was carried out on the patients suffering from alopecia, and the total effective rate on the patients suffering from alopecia was counted, and the results are shown in table 3.

The plant exosomes consist of ginseng exosomes, raspberry exosomes and dendrobium exosomes in a weight ratio of 1 (0.2-0.6): 1-1.5;
The weight ratio of the lactobacillus fermentation lysate to the filtrate of the two-split yeast fermentation product to the plant exosome is (10-17): 8-12): 1.
2. The hair-loss preventing and hair-growing composition according to claim 1, wherein the weight ratio of ginseng exosomes, raspberry exosomes and dendrobium exosomes is 1 (0.4-0.5): 1.2-1.5.
3. The hair-loss preventing composition according to claim 1, wherein the weight ratio of lactobacillus fermentation lysate, yeast fermentation filtrate to plant exosomes is (10-15): 10:1.
4. An anti-hair loss and hair growth promoting essence characterized in that the anti-hair loss and hair growth promoting essence contains the anti-hair loss and hair growth promoting composition according to any one of claims 1 to 3.

The anti-hair loss and hair growth promoting essence according to claim 4, wherein the anti-hair loss and hair growth promoting essence comprises the following components in parts by weight: 20 to 45 parts of hair-growing composition with anti-alopecia effect, 6 to 13 parts of humectant, 1 to 5 parts of skin conditioning agent, 3 to 7 parts of antioxidant, 0.1 to 0.5 part of anti-allergic agent, 1 to 3 parts of solubilizer and 80 to 120 parts of water.

The anti-hair loss and hair growth promoting essence according to claim 5, wherein the anti-hair loss and hair growth promoting essence comprises the following components in parts by weight: 30-40 parts of hair-growing composition, 6-13 parts of humectant, 1-5 parts of skin conditioner, 3-7 parts of antioxidant, 0.1-0.5 part of anti-allergic agent, 1-3 parts of solubilizer and 80-120 parts of water.

The anti-hair loss and hair growth promoting essence according to claim 5 or 6, wherein the humectant is at least one of 1, 2-hexanediol, glycerin, and sodium hyaluronate; the skin conditioner is at least one of nicotinamide, saccharomycete polypeptide and nicotinamide adenine dinucleotide; the antioxidant is p-hydroxyacetophenone and/or ascorbyl palmitate; the anti-allergic agent is hydroxyphenyl propionamide benzoic acid; the solubilizer is PEG-40 hydrogenated castor oil.
8. A method for preparing the hair loss preventing and growing essence according to any one of claims 4 to 7, which is characterized by comprising the following steps:
(1) Mixing humectant, solubilizer and water, heating to 90-100 ℃, stirring uniformly to obtain a basic solution, and cooling the basic solution to 60-65 ℃ for standby;
(2) Adding an antioxidant into the basic solution, homogenizing for 1-2 min, then adding an anti-allergic agent and a skin conditioning agent, homogenizing for 1-2 min, and then adding an anti-hair loss composition, homogenizing for 1-2 min to obtain a mixed solution; controlling the homogenizing temperature to be 60-65 ℃, preserving the temperature of the mixed solution at 60-65 ℃ for 0.5-1 h after the homogenizing is finished, and cooling to room temperature to prepare the anti-hair loss and hair-growing essence.
