Abstract

Where numerical ranges are provided in the examples, it is understood that unless otherwise stated herein, both endpoints of each numerical range and any number between the two endpoints are significant both in the numerical range. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this application belongs. In addition to the specific methods, devices, materials used in the embodiments, any methods, devices, and materials of the prior art similar or equivalent to those described in the embodiments of the present application may be used to practice the present application according to the knowledge of one skilled in the art and the description of the present application.
In the following embodiments of the present application, the mass ratio is 7:1, mixing 25wt% of hydrochloric acid with citrus fruit powder, and filtering to obtain filtrate and filter residue; concentrating the filtrate at 60deg.C to relative density of 1.10 to obtain concentrated solution A; the mass ratio is 6:1 mixing filter residues with 40wt% butanediol, filtering, concentrating at 60 ℃ until the relative density is 1.10, and obtaining a concentrated solution B; mixing the concentrated solution A and the concentrated solution B to obtain the citrus fruit extract.
In the following examples of the present application, 100g of Ganoderma sinense was taken, pulverized to a particle size of about 50 mesh, and put into an extraction vessel, and supercritical CO was used 2 As solvent, 75wt% methanol was used as entrainer, wherein the extraction conditions were: the extraction pressure is 34MPa, and the temperature is 50 ℃; the separation conditions are as follows: the pressure of the primary separator is 8MPa, the temperature is 50 ℃, the pressure of the secondary separator is 6MPa, and the temperature is 45 ℃; and (3) dynamically operating for 5 hours, obtaining a ganoderma lucidum extract from the primary separator and the secondary separator, and then drying to obtain the ganoderma lucidum extract. Examples 1 to 5
Referring to table 2, the ganoderma lucidum extract, vitamin B3, vitamin B5, vitamin B6, vitamin B7, acetyl tetrapeptide-3, diaminopyrimidine oxide, and green tangerine peel extract were mixed to obtain a hair care composition containing the ganoderma lucidum extract.

Referring to table 3, the hair care composition of example 5 was mixed with water, pentanediol, and hydroxyethyl cellulose, respectively, to obtain a essence containing ganoderma sinense.
Comparative examples 1-4 were also set up and the formulation is shown in Table 3.
Comparative example 1 differs from example 5 in that: commercial adenosine was used in place of Ganoderma sinense extract, and the commercial adenosine was added in an amount of 0.04wt% with the other components as in example 5 to obtain an essence.
Comparative example 2 differs from example 5 in that: the essence was obtained in the same manner as in example 5, except that the green tangerine peel extract was not added.
Comparative example 3 differs from example 5 in that: the essence was obtained in the same manner as in example 5, except that the purple ganoderma lucidum extract was not added.
Comparative example 4 differs from example 5 in that: the essence is obtained in the same manner as in example 5 without adding the extracts of Ganoderma sinense and pericarpium Citri Reticulatae viride.

Application example
In this application example, the essential liquids obtained in example 5 and comparative examples 1 to 4 were used for the investigation of the oil content, hair loss and sensory investigation, and the bacteriostasis test.
Experimental design based on the principle of parallel control, a test was performed for 4 weeks to verify the efficacy of the hair care compositions of the present application. The subjects maintained their normal washing procedure and applied the test essence formulation once a day, uniformly applying the test formulation (about 6 mL) to the scalp, massaging thoroughly for 2-5 minutes to absorption without rinsing.
Subjects were instrumentally tested and filled in a self-assessment questionnaire before the start of the experiment and at the end of the experiment (after 4 weeks), respectively.
Experimental data were statistically analyzed using SPSS. P <0.05 was considered to have significant differences and P <0.01 was considered to have very significant differences.
And comparing the same formula before and after use, and adopting paired t test or related sample rank sum test according to whether the net change value is subjected to normal distribution.

Subject information:
the experiments recruited 5 groups of subjects, 30 or more, aged 34 to 65 years, meeting the volunteer selection criteria of the subjects. The experiment is completed by 165 subjects (112 men and 53 women), and the number of each group is not less than 30, so that the statistical analysis requirement is met. The experimental group is shown in table 4.

From table 5, it can be seen that the subjects were able to significantly reduce the amount of scalp fat after using the example formulation for 4 weeks.
1.1.2 alopecia counts
Before the start of the experiment and at the end of the experiment, each subject was subjected to hair loss counting by TrichoScan software analysis using MoleMax HD dermoscope instrument, and the hair loss counting results are shown in table 5.

As can be seen from table 6, the number of hair loss in the subjects was extremely significantly reduced (P < 0.01) after 4 weeks of use of the hair care composition of the present application.
1.1.3 sensory evaluation
Subjects were scored for improvement in hair problems after use of the serum using a questionnaire format. Each sensory index is fully divided into 5 points, obviously improved into 5 points, and not improved into 0 point. The statistical results are shown in Table 7.

From Table 7, it is clear that the hair care composition of the present application has the best effect of improving the scalp discomfort symptoms of the subject.
From tables 5, 6 and 7, it is understood that the hair care composition of the present application has multiple effects of controlling lipid secretion, promoting hair growth, removing dandruff, relieving itching, and preventing the occurrence of acne on the scalp.
1.2 minimum inhibitory concentration test
Excessive secretion of grease brings enough nutrients for malassezia, causes blockage of scalp hair follicles, and finally leads to alopecia.
The minimum inhibitory concentration test (agar dilution) of the essence on malassezia furfur (Malassezia furfur) was determined by the method described in the "disinfection technical Specification" (2002 edition of Ministry of health).

As is clear from Table 3, each of comparative examples 1, 2, 3 and 4 and example 5 inhibited malassezia furfur, and the effect of example 5 was evaluated to be more excellent in combination, and the minimum inhibitory concentration was 2.5X10 4 . The composition of the application has synergistic effect, can effectively inhibit the propagation of malassezia furfur, and solves the problems of abnormal increase of dandruff, alopecia and the like caused by malassezia furfur infection.
1.3 5 alpha-reductase Activity inhibition assay
The experiment uses NADPH test method. The 5 alpha-reductase catalyzes the conversion of testosterone to dihydrotestosterone in the presence of reduced coenzyme II, and if a 5 alpha-reductase inhibitor is present, this reaction process is inhibited to some extent. Since reduced coenzyme II has characteristic absorption at 340nn wavelength, the characteristic absorption at 340nm wavelength of oxidized coenzyme II generated after participating in the reaction disappears. The measurement by an ultraviolet spectrophotometer can reflect whether a certain substance has an inhibition effect and the inhibition effect on 5 alpha-reductase.

As is clear from Table 9, all of comparative examples 1, 2, 3 and 4 and example 5 inhibit 5α -reductase, and the effect of example 5 was evaluated to be more excellent, and the 5α -reductase inhibition rate was 39.90. The composition provided by the application has a synergistic effect, and can effectively inhibit 5 alpha-reductase activity, so that testosterone is inhibited from being converted into dihydrotestosterone, and the problem of androgenetic alopecia is solved in a targeted manner.
In the composition provided by the application, the diaminopyrimidine oxide has a good inhibition effect on 5 alpha-reductase, and ganoderic acid DM in the ganoderma lucidum extract can inhibit the activity of 5 alpha-reductase, reduce the activity of sebaceous glands, limit sebum secretion and reduce bacterial proliferation; the citrus fruit extract has strong broad-spectrum antibacterial effect, has good inhibition effect on pityrosporum, and is beneficial to reducing dandruff; acetyl tetrapeptide-3 stimulates cell proliferation and controls cell differentiation, participates in intercellular communication and stimulates hair follicle development. The composition organically combines natural plants and non-plant-derived raw materials through a scientific formula of diaminopyrimidine oxide, citrus fruit extract, ganoderma lucidum extract and acetyl tetrapeptide-3, inhibits the activity of 5 alpha-reductase, reduces the proliferation of pityrosporum, controls the secretion of sebaceous glands and the like, achieves the synergistic effect respectively, improves alopecia in multiple ways, remarkably reduces the quantity of alopecia of subjects, and has no uncomfortable symptoms after use.
The addition of the ganoderma lucidum extract in the composition provided by the application can further enhance the activity of the composition for inhibiting 5 alpha-reductase, thereby reducing the combination of dihydrotestosterone and hair follicle receptors and increasing the hair growth cycle; the inhibiting effect of the citrus fruit extract on malassezia is also improved, and the composition can effectively remove dandruff and has a certain antipruritic effect; the composition of the application can be applied to washing and caring products to effectively improve seborrheic alopecia and promote hair growth.

Claims (7)
1. A hair care composition containing ganoderma lucidum extract is characterized by comprising the following raw materials in parts by weight:
the vitamin B is a mixture of vitamin B3, vitamin B5, vitamin B6 and vitamin B7, and the mass ratio of the vitamin B3 to the vitamin B5 to the vitamin B6 to the vitamin B7 is (0.1-5): (0.1-1): (0.005-1): (0.03-1);
the preparation method of the ganoderma lucidum extract comprises the following steps:
placing Ganoderma powder in extraction kettle, introducing supercritical CO 2 And entrainer, extract, get extract;
and (3) carrying out two-stage decompression separation on the extract, and drying to obtain the ganoderma sinense extract.
2. A composition according to claim 1, wherein the citrus fruit extract is prepared by a process comprising:
mixing the acid with the citrus fruit powder, and carrying out solid-liquid separation to obtain filtrate and filter residues;
concentrating the filtrate to obtain a concentrated solution A; mixing the filter residues with polyalcohol, carrying out solid-liquid separation, and concentrating to obtain a concentrated solution B;
mixing the concentrated solution A and the concentrated solution B to obtain the citrus fruit extract.
3. The composition of claim 2 wherein the acid is selected from the group consisting of aqueous hydrochloric acid; the concentration of the hydrochloric acid aqueous solution is 10-30wt%;
and/or the pH value of the mixed solution formed by mixing the acid and the citrus fruit powder is 1-4;
and/or the polyol is selected from butanediol aqueous solution; the concentration of the butanediol aqueous solution is 30-50wt%;
and/or, the mass ratio of the acid to the citrus fruit powder is (5-10): 1, a step of;
and/or the mass ratio of the filter residue to the polyol is (5-10): 1, a step of;
and/or concentrating at 60-65 deg.c to alcohol content not higher than 30% and relative density of 1.05-1.10.
4. The composition of claim 1, wherein the entrainer is ethanol or methanol;
and/or the extraction pressure is 20-50 MPa;
and/or the extraction temperature is 30-50 ℃;
and/or the primary separation pressure is 8-10 MPa;
and/or, the primary separation temperature is 35-50 ℃;
and/or the secondary separation pressure is 4-6 MPa;
and/or the secondary separation temperature is 30-45 ℃.
5. A method of preparing a composition according to any one of claims 1 to 4, comprising the steps of:
mixing the raw materials according to parts by weight to obtain the composition.
6. Use of a composition according to any one of claims 1 to 4 in the preparation of a hair care product.
7. A hair care product comprising the composition according to any one of claims 1 to 4.
