Effect data
1. Bacteriostasis test
The oxford cup method is adopted to measure the bacteriostasis capability of two common bacteria of malassezia and staphylococcus aureus, and the experimental result judgment standard is as follows: the diameter of the inhibition zone is less than or equal to 10mm, which indicates that the inhibition effect is not obvious, the diameter of the inhibition zone is 10mm-15mm, which indicates moderate inhibition, and the diameter of the inhibition zone is more than or equal to 15mm, which indicates high inhibition, and the experimental results are shown in table 1.
TABLE 1
As can be seen from the detection results of table 1, the shampoo compositions prepared in examples 1-3 have good antibacterial performance, and show high antibacterial performance to malassezia and staphylococcus aureus, while omitting the cacumen biotae extract in comparative example 1 has obviously reduced antibacterial performance, which indicates that cacumen biotae can obviously influence the antibacterial performance of the product; comparative examples 2-3 the change of the kind or the ratio of the surfactant has little influence on the bacteriostasis performance; comparative examples 4-5 the change in the mass ratio of conditioner, chelating agent and water-oil balancing agent, when outside the scope of the present application, affects the absorption of the scalp to the ingredients in the shampoo composition to some extent, and thus affects the bacteriostatic properties, that is, only the shampoo composition protected by the present application has a good bacteriostatic property.
2. Antibacterial stability
The test method comprises the following steps: plate contact method
The operation method comprises the following steps: removing hair and fat meat from pigskin, cutting into square pieces of 4cm×4cm, and sterilizing by Co 60 -gamma ray irradiation.
1. Weighing 44.0g of Leemming-Notman culture medium foundation in 1L of deionized water, boiling until the solution is completely dissolved, adding 1.6mL of olive oil into each 80mL of the solution, cooling the solution to about 50 ℃ after autoclaving, adding 40mg of cycloheximide solution into each 80mL of the solution under aseptic operation, and preparing LN flat plates (90 mm multiplied by 15 mm) for later use. 100 mu L of 1X 10 5-3×105 CFU/mL of malassezia suspension are respectively dripped on LN plates, and the malassezia suspensions are uniformly coated for later use.
2. Uniformly wiping the surface of the pigskin with 0.5g of the shampoo samples prepared in examples 1-3 and comparative examples 1-5 for 1min, and rinsing the liquid soap on the pigskin with 100mL of sterile water for 1min for 2 times; after rinsing, placing the pigskin in a biological safety cabinet, ventilating and airing until no obvious water drops are generated on the surface of the pigskin, attaching the pigskin on a bacteria-dying plate for 15min, removing the pigskin after attaching, placing the plate in a (32+/-2) DEG C incubator for culturing for 72 hours, observing the growth condition of malassezia, and calculating the bacteriostasis rate, wherein the specific detection results are shown in the table 2 below.
TABLE 2
As can be seen from the detection data of the above Table 2, the shampoo compositions prepared in the examples 1-3 of the application have longer lasting antibacterial effect, the antibacterial performance is still more than 97% after 72 hours of use, while the antibacterial performance is obviously reduced after 1 hour of omitting the cacumen biotae extract in the comparative example 1, but the long-term antibacterial performance is better; comparative examples 2-3 changing the kind or ratio of the surfactant has a great influence on the long-term bacteriostasis performance; comparative examples 4-5 changing the mass ratio of conditioner, chelating agent and water-oil balancing agent would similarly affect the long-term bacteriostasis performance of the shampoo composition when not within the scope of the present application, that is, the shampoo composition protected by the present application has better bacteriostasis stability.
3.5 Alpha-reductase inhibition assay
The implementation principle is as follows: 5α -reductase is a protease produced in an amount that stimulates the conversion of testosterone (T) to Dihydrotestosterone (DHT) in the body, which causes androgenic alopecia when Dihydrotestosterone (DHT) accumulates to high levels in the skin. The pathogenesis of the hair follicle is that the Dihydrotestosterone (DHT) can inhibit the growth and development of hair follicles, the hair follicles are dysplasia and atrophy, the hair growth is naturally bad, in addition, the increase of the Dihydrotestosterone (DHT) level leads to the hypertrophy of sebaceous glands, the increase of grease secretion, and further the growth environment of the hair follicles is influenced, so that the higher the 5 alpha-reductase is, the heavier the hair loss is.
Materials and methods: 5α -reductase: extracting from rat prostate tissue, and storing at-80deg.C;
Positive control: finasteride 0.40 μm;
blank control: phosphate Buffered Saline (PBS);
other: testosterone, reduced coenzyme II, chloroform;
Sample preparation: taking 1g of shampoo compositions of the embodiments 1-3 and the comparative examples 1-5, adding 2ml of water to dissolve the shampoo compositions to obtain a dissolution liquid sample, and diluting the dissolution liquid sample with water to a test concentration of 1.00%;
The test process comprises the following steps: the 2mL reaction system contains: phosphate buffer (pH 6.5) 250. Mu.L, enzyme extract (1 mg/mL) 700. Mu.L, testosterone solution (0.5 mg/mL) 100. Mu.L, NADPH solution (1 mg/mL) 350. Mu.L; 600. Mu.L of 0.4. Mu.M finasteride was added to the positive control tube; 600. Mu.L of sample solution is added into the sample tube; 600. Mu.L of water was added to the complete reaction tube; the control tube replaced the enzyme extract with phosphate buffer (pH 6.5) and the reaction was stopped by adding 2.0mL of dichloromethane; the sample tube is reacted for 30min at 37 ℃, after the reaction is finished, 2.0mL of dichloromethane is added to stop the reaction, after shaking for 1min, the reaction is centrifuged for 10min at 3000 r/min. The upper aqueous phase was removed, about 1.0mL of the organic phase was removed and evaporated to dryness, and the residue was dissolved in 1.0mL of methanol and the testosterone peak area (T) was determined by high performance liquid chromatography. Each reaction was run in 3 parallel tubes, averaged, and the decimal place was retained, with the results shown in Table 3 below.
The calculation formula is as follows:
TABLE 3 Table 3
From the test results shown in Table 3, the shampoo compositions prepared in examples 1 to 3 of the application have a higher 5. Alpha. -reductase inhibition rate of 14% or more; whereas the omission of the biota orientalis extract 5α -reductase inhibition rate in comparative example 1 is significantly lower than that in the examples; the change of the types or the proportions of the surfactants in comparative examples 2 to 3 and the change of the mass ratio of the conditioner, the chelating agent and the water-oil balancing agent in comparative examples 4 to 5 can affect the inhibition rate of the shampoo composition to the 5α -reductase when the ratio is not within the protection scope of the application, so that the inhibition rate is obviously reduced, that is, only the shampoo composition protected by the application has better anti-falling effect.
The above examples are preferred embodiments of the present invention, but the embodiments of the present invention are not limited to the above examples, and any other changes, modifications, substitutions, combinations, and simplifications that do not depart from the spirit and principle of the present invention should be made in the equivalent manner, and the embodiments are included in the protection scope of the present invention.
