Abstract
本发明公开了一种防治鸡球虫病的中药组合物及其制备方法与应用。属于中药领域,所述中药组合物含有活性成分,所述活性成分由青蒿、柴胡、苦参、常山、使君子、仙鹤草、马齿苋、槟榔、陈皮、神曲制成。本发明中药组方以中兽医理论为指导,遵守方剂组方原则,对患球虫病的病鸡进行辨证论治,可有效用于防治鸡球虫病,具有原料来源广泛、安全、环保、制备工艺简单、生产成本低等诸多优点。

技术领域
本发明涉及兽用中药领域,特别涉及一种防治鸡球虫病的中药组合物及其制备方法与应用。
背景技术
鸡球虫病是由艾美耳科艾美耳属球虫引起的一类发病率较高的寄生虫病,主要危害雏鸡,通常是小于3月龄的鸡,其中以15~45日龄的雏鸡最易发病,成年鸡也能够感染,但往往呈慢性经过,无任何症状或仅出现轻微的症状。病鸡感染后可见肠道发炎和出血,影响肠道消化吸收功能,使机体消瘦、发育不良、排出血便,对其他疾病的抵抗力降低,严重时由于极度衰竭而死,尤其是急性发病往往会导致大批死亡,严重损害鸡生产的经济效益。该病一般呈地方性流行,尤其在春夏季节,气温升高,土壤湿度大,鸡群爆发鸡球虫病的可能性增加。
目前用于鸡球虫病防治的药物主要为抗生素以及化学药物,例如氨丙啉、盐霉素等,化学药物和抗生素的滥用可能产生各种耐药性,在禽产品中造成药物残留,并通过食品将耐药性传给人,对人的身体健康造成威胁。使用中药具有疗效确切、双向调节作用、低毒低残留、功能多样性等特点,得到了国内外专家的认可和支持。与抗生素直接作用于细菌等病原体的机制不同,中药是通过扶正祛邪的调理作用,帮助机体恢复到平衡状态并增强非特异性免疫功能,所以中药较难产生耐药性;随着时间的推移,化学药物及抗生素的弊端已逐渐显现,引起的毒副作用和动物药源性疾病已经严重威胁了动物安全,中药在长期的临床验证中,并未发现其具有明显的毒副作用;化药成分单一,只能单向作用于机体,使其兴奋或抑制,其作用方式具有局限性,中药由于其成分多样性,可针对不同机体状态双向发挥作用,将机体调整为平衡状态。且其成分多样性同样决定了中药拥有较成分单一的化药更多样的功能,弥补了化药因作用单一需数药并用的缺点,使用中药组合物,不仅可以直达病灶,驱除球虫,还可在此基础上,调理脾胃,促进饲料吸收,进一步增强机体免疫力,促进机体恢复。
发明内容
为了解决上述技术问题,本发明提供了一种防治鸡球虫病的中药组合物。
一种防治鸡球虫病的中药组合物,含有活性成分,所述活性成分由青蒿、柴胡、苦参、常山、使君子、仙鹤草、马齿苋、槟榔、陈皮、神曲制成。
进一步,所述活性成分由青蒿220-380份、柴胡50-130份、苦参320-480份、常山120-300份、使君子50-130份、仙鹤草80-160份、马齿苋80-120份、槟榔60-140份、陈皮60-140份、神曲40-120份制成。
进一步,所述活性成分由青蒿260-340份、柴胡70-110份、苦参360-440份、常山160-240份、使君子70-110份、仙鹤草100-140份、马齿苋100-140份、槟榔80-120份、陈皮80-120份、神曲60-100份制成。
进一步,所述活性成分由青蒿300-320份、柴胡90-100份、苦参400-420份、常山200-220份、使君子90-100份、仙鹤草120-130份、马齿苋120-130份、槟榔100-110份、陈皮100-110份、神曲80-90份制成。
进一步,所述活性成分由青蒿300份、柴胡90份、苦参400份、常山200份、使君子90份、仙鹤草120份、马齿苋120份、槟榔100份、陈皮100份、神曲80份制成。
本发明还提供了所述的防治鸡球虫病的中药组合物的制备方法,包括以下步骤:
(1)按重量配比称取各药物原料;
(2)将步骤(1)所述的药物原料加水后进行高温提取,过滤后合并滤液;
(3)将步骤(2)所述的滤液在60℃减压条件下浓缩成浸膏;
(4)将步骤(3)所述的浓缩浸膏与无水葡萄糖混合均匀制备成药,即得。
进一步,上述制备方法其制得的剂型包括可溶性粉剂、颗粒剂或软膏。
本发明还提供了所述的防治鸡球虫病的中药组合物在制备防治鸡球虫病药物方面的应用。
本发明还提供了所述的防治鸡球虫病的中药组合物在制备防治鸡球虫病饲料方面的应用。
本发明还提供了含有所述的防治鸡球虫病的中药组合物的饲料。
本发明中药颗粒的各药物组分及其用量是以中兽医理论为指导,遵守方剂组方原则,对患球虫病的病鸡进行辨证论治,并进行大量试验反复验证。中医认为虫证是指寄生在肠道内的虫类引起的病证,一般具有毛焦浮吊、饱食不长或粪便失调等症状。且寄生虫寄生在体内常伴有体弱脾虚,正气损耗,因此药物治疗以杀虫消积、理气健脾、攻补兼施为治疗原则进行内外兼治,双管齐下。在本发明组方中,青蒿、常山作为君药,起清热祛湿、截疟杀虫的作用;苦参、柴胡、使君子作为臣药,起清热燥湿、杀虫消积、升阳举陷的功效,其中苦参加强君药清热除湿、杀虫的作用,并在此基础上使用柴胡和使君子消积、疏肝解郁、升举阳气,攻补兼顾;仙鹤草、马齿苋、槟榔、陈皮、神曲作为佐使药,起截疟止痢、消食化积、健脾和胃的功效,槟榔、仙鹤草、马齿苋加强截疟止痢杀虫的主功效,陈皮、神曲则在健脾和胃、升阳举陷的基础上进一步消食化积、健胃行脾、燥湿化痰。诸药合用,共奏清热燥湿、杀虫消积、健脾和胃、升举阳气之功效,驱除球虫的同时,又增强机体免疫力,改善病鸡进食状况,提高饲料转化率。
本发明中药组合物相对于现有技术的有益效果在于:
本发明中药组方以中兽医理论为指导,遵循配伍原则,经过辨证论治,对鸡球虫病有着显著的疗效;组方中的原料药来源广泛且成本较低,绿色健康,不会给动物机体造成过多的负担,不会产生耐药性,更不会造成药物残留从而对人的身体健康产生影响;本发明中药组方采用现代制药技术进行制备,可被动物机体快速吸收、同时采取不同给药方式集中给药,可以有效集中防治鸡球虫病。本发明的药物扩展了治疗鸡球虫病的原料使用,跟现有的药物相比,本发明使用的大多数中药原料成本较低,可大幅降低成本,且效果要优于现有的化学药。
本发明中药组合物经过试验验证,可显著提高鸡相对增重率和存活率,改善其盲肠病变程度,降低其粪便中的卵囊数量,并且其效果要优于现有的一些中药组方。
具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。
实施例1
按重量配比称取各药物原料:青蒿300份、柴胡90份、苦参400份、常山200份、使君子90份、仙鹤草120份、马齿苋120份、槟榔100份、陈皮100份、神曲80份,按如下方法进行制备和使用
将所称取药材按以下步骤进行处理:
(1)加12倍量水,100℃回流提取2次,每次1h,过滤,合并滤液;
(2)滤液75℃下减压浓缩至相对密度1.18,即1ml浸膏相当于1.91g原生药。
(3)将浸膏与无水葡萄糖按照0.7~1.1:1的比例进行混匀,制成合适的软材过筛制粒;45℃~55℃之间烘干,整粒即得。
本发明药物使用方法为:用于感染鸡球虫的预防与治疗,混饮:每1吨水添加本发明药物2kg,连用5-7d。
实施例2
按重量配比称取各药物原料:青蒿260份、柴胡70份、苦参360份、常山160份、使君子70份、仙鹤草100份、马齿苋100份、槟榔80份、陈皮80份、神曲60份,按实施例1的方法制备和使用。
实施例3
按重量配比称取各药物原料:青蒿340份、柴胡110份、苦参440份、常山240份、使君子110份、仙鹤草140份、马齿苋140份、槟榔120份、陈皮120份、神曲100份,按实施例1的方法制备和使用。
实施例4
按重量配比称取各药物原料:青蒿320份、柴胡100份、苦参420份、常山220份、使君子100份、仙鹤草130份、马齿苋130份、槟榔110份、陈皮110份、神曲90份,按实施例1的方法制备和使用。
对比例1
按重量配比称取各药物原料:青蒿30份、常山25份、仙鹤草10份、苦参10份、柴胡5份、黄芪5份、山楂5份、木香5份,按实施例1的方法制备和使用。
对比例2
按重量配比称取各药物原料:称取常山12份、仙鹤草10份、青蒿8份、地锦草8份、苦参8份、马齿苋10份、五味子10份,按实施例1的方法制备和使用。
对比例3
按重量配比称取各药物原料:青蒿5份、常山10份、柴胡15份、苦参10份、连翘10份、仙鹤草5份、白头翁5份、白术15份,按实施例1的方法制备和使用。
对比例4
按重量配比称取各药物原料:青蒿15份、常山15份、马鞭草10份、黄柏10份、苦参8份、郁金8份、栀子8份、当归8份、陈皮6份、金银花6份、仙鹤草6份,按实施例1的方法制备和使用。
对比例5
按重量配比称取各药物原料:使君子25份、牵牛子20份、槟榔35份、大黄25份、黄连10份、玄明粉5份、白术18份、山药2份、鸡内金15份、神曲15份,按实施例1的方法制备和使用。
试验例1
为验证本发明药物疗效,2021年7月,在大北农集团动物保健技术研究中心试验动物房对本发明药物进行了人工感染柔嫩艾美耳球虫的疗效试验,结果如下:
1材料与动物
1.1试验材料本发明实施例1~4制备的中药颗粒;磺胺氯丙嗪钠可溶性粉,购自北京大北农动保;无抗球虫药物添加饲料,购自大北农集团;氯化钠;重铬酸钾;60目铜筛网;100目铜筛网;麦氏计数板;一次性吸管;离心机;摇床;显微镜;天平等。
1.2试验动物白羽肉鸡,购自北京家禽育种有限公司。
2方法
2.1分组及给药
将70只20日龄白羽肉鸡随机分为7组,每组10只,分别为空白对照组、模型组、阳性药物对照组、试验1~4组,除空白对照组外,其余6组试验鸡人工感染柔嫩艾美耳球虫,灌服5×104个孢子化卵囊。造模24h后开始给药,其中,阳性药物对照组选用磺胺氯丙嗪钠可溶性粉,按1g/kg剂量饮水给药,连用7d;试验1~4组分别为本发明实施例1~4所制备的药物,按照2kg/吨水剂量饮水给药,连用7d;空白组和模型组喂食同等量生理盐水。
2.2观察指标
观察并记录每组试验鸡的精神状态、粪便情况、发病情况及临床表现,剖杀时记录盲肠病变情况,并对盲肠进行病变计分,计算平均增重、相对增重率、死亡率、抗球虫指数(ACI)、卵囊值、卵囊比数(%)
其中,抗球虫指数:ACI=(相对增重率+存活率)—(病变值+卵囊值),ACI<120为无效,120~160为低效,160~180为中效,>180为高效。
2.3数据处理
采用SPSS 17.0软件对数据的显著性检验进行分析。
3结果
3.1临床症状
在灌服卵囊3~4天后空白组试验鸡采食、饮水、活动量、精神状态及粪便情况正常,模型组及各给药组均出现精神状态异常、采食量下降、聚堆不动、羽毛杂乱、排血便等情况,阳性药物对照组及实施例给药组,血便情况有所好转,实施例组采食量出现明显好转。
3.2病理学剖检状况
通过对试验鸡的剖检发现,空白对照组各脏器正常、无明显病变;模型组盲肠中出现大量出血点、肿胀、含有大量含血内容物;给药组同样出现肿胀、出血等症状,程度较模型组相比明显减轻,且其他脏器并未发现明显异常。
3.3各指标结果
对各组试验鸡的平均增重、相对增重率、存活率、盲肠病变值打分及病变值、卵囊比数及卵囊值、ACI等数据进行记录并处理,试验结果见表1.

由表1试验结果可以看出,模型组较空白组相比,平均增重较低,且存活率较低,盲肠病变严重,卵囊数高,表明造模成功;与模型组相比,药物对照组与试验各组均可显著提高试验鸡相对增重率和存活率,改善盲肠病变程度,降低卵囊数量,具有治疗效果,且各给药组间相比,本发明药物治疗效果优于对照药物。综合来看,试验1组的试验效果最好,为最优处方。
试验例2
为进一步验证本发明药物对鸡球虫的防治的临床效果,对本发明药物进行临床疗效观察试验,以下通过试验例来进一步阐述本发明药物的有益效果。
河北省保定市张某饲养的5000只肉鸡中,有部分鸡出现羽毛蓬松、杂乱,聚堆,精神状态不佳、采食量减少、嗜睡且伴有血便排出,剖检后发现盲肠和十二指肠中含有大量血样内容物,肠壁有多处出血点,盲肠显著肿大,上皮变厚,严重者甚至伴有糜烂,根据流行病学、临床症状诊断及实验室诊断,确诊为球虫感染。技术员采用本发明药物实施例1制备的药物饮水给药,按照每1L水添加本发明药物2g,自由饮用,连用7-10日,并按照指导对环境进行消杀。
一周后对张某进行回访,反映球虫感染已经得到有效控制,在用药3天左右即可看到显著效果,用药结束后已痊愈,且有效提高了病鸡的采食量,提高病鸡自身免疫力。
试验例3
为进一步验证本发明药物的突出疗效,于2022年4月在辽宁省某肉鸡养殖场选取临床患有鸡球虫病的600只病鸡分别采用本发明药物及对比药物进行治疗,所选病鸡精神萎靡、聚堆嗜睡,采食量明显下降,有血便排出,因耐药性等问题,地克珠利溶液及磺胺类化药使用效果不佳。将选取的600只病鸡随机分为6组,每组100只,分别为为本发明实施例1、对比例1~5所制备的药物组。实施例1组和对比例1~5组均按照2kg/吨水剂量饮水给药,连用7d,其他饲养环境保持一致。
治疗效果判定标准:(1)有效:病鸡精神和食欲恢复正常,血便消失,粪便中卵囊数量显著降低且不复发;(2)无效:用药后,病鸡症状没有缓解,粪便中卵囊值并未降低,仍有血便且还有可能加重或死亡。试验结果见表2。

由表2试验结果可以看出,本发明药物实施例1制备的中药颗粒对抗鸡球虫疗效显著,有效率达到95%,而选用对比例1、对比例2、对比例3、对比例4、对比例5的药物其有效率都在90%以下。
本试验结果表明,本发明药物对治疗鸡球虫效果显著,并且目前在同类产品中效果也是较显著的。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明技术原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.一种防治鸡球虫病的中药组合物,含有活性成分,其特征在于,所述活性成分由青蒿260~340份、柴胡70~110份、苦参360~440份、常山160~240份、使君子70~110份、仙鹤草100~140份、马齿苋100~140份、槟榔80~120份、陈皮80~120份、神曲60~100份制成。
2.根据权利要求1所述的防治鸡球虫病的中药组合物,其特征在于,所述活性成分由青蒿300~320份、柴胡90~100份、苦参400~420份、常山200~220份、使君子90~100份、仙鹤草120~130份、马齿苋120~130份、槟榔100~110份、陈皮100~110份、神曲80~90份制成。
3.根据权利要求2所述的防治鸡球虫病的中药组合物,其特征在于,所述活性成分由青蒿300份、柴胡90份、苦参400份、常山200份、使君子90份、仙鹤草120份、马齿苋120份、槟榔100份、陈皮100份、神曲80份制成。
4.一种权利要求1~3任一项所述的防治鸡球虫病的中药组合物的制备方法,包括以下步骤:
(1)按重量配比称取各药物原料;
(2)加12倍量水,100℃回流提取2次,每次1h,过滤,合并滤液;
(3)滤液75℃下减压浓缩至相对密度1.18,即1ml浸膏相当于1.91g原生药;
(4)将浸膏与无水葡萄糖按照0.7~1.1:1的比例进行混匀,制成合适的软材过筛制粒,45℃~55℃之间烘干,整粒即得。
