Abstract
The invention relates to a traditional Chinese medicine composition with acne removing effect, a traditional Chinese medicine preparation, a preparation method and application thereof, and relates to the field of daily cosmetics. The traditional Chinese medicine composition with the acne-removing effect comprises the following raw materials in parts by weight: 5-50 parts of paeoniflorin, 3-30 parts of licorice extract, 2-25 parts of semen aesculi extract and 1-10 parts of red deer blood extract. Technical Field
The invention relates to the field of daily cosmetics, and particularly relates to a traditional Chinese medicine composition and a traditional Chinese medicine preparation with acne removing effect, and a preparation method and application thereof.
Background
Acne is also known as "whelk", which is one of the typical skin problems, and 80% of people between 11 and 30 years of age will suffer from this condition. The components of the cosmetic for removing acnes mainly use hormone antagonists, tretinoin medicines, antibiotics and the like to treat the skin diseases, and the medicines have obvious curative effects, but the toxic effects are not very small, such as allergy, kidney function damage, even deformity and the like. Therefore, the plant extract components for safely and effectively treating acne become the development hot spot of acne-removing cosmetics. The cosmetic formula which can not only improve the natural tolerance threshold of skin, effectively treat acne and has good acne-removing and anti-inflammatory effects, but also can not generate other side effects is needed in the field.

In the invention, the traditional Chinese medicine composition with acne removing effect comprises 5-50 parts by weight of paeoniflorin, preferably 10-30 parts by weight, and more preferably 25 parts by weight.
In the invention, the paeonia lactiflora total glycoside is preferably obtained by extracting and purifying dried roots of paeonia lactiflora (Paeonia lactiflora PalL) belonging to the Ranunculaceae, and the total glycoside compound content in the extract is about 86%; the paeonia lactiflora is bitter in taste and slightly cold, enters liver meridian, and has the effects of clearing heat and cooling blood, and activating blood circulation to dissipate blood stasis. Modern researches have shown that radix Paeoniae Rubra contains terpenes and their glycosides, flavonoids, etc., and has various pharmacological effects and effects of protecting liver, protecting nerve, resisting thrombosis and oxidation, resisting endotoxin, etc.

In the invention, the traditional Chinese medicine preparation is preferably powder, plaster, ointment, toning lotion, emulsion, facial mask or gel.
The invention also provides an application of the traditional Chinese medicine composition with the acne-removing effect or the traditional Chinese medicine preparation with the acne-removing effect in cosmetics or medicines for treating skin diseases. The method of the present invention is not particularly limited, and may be applied as known to those skilled in the art.
The invention uses the composition of paeoniflorin, licorice extract, semen aesculi extract and red deer blood extract, and through antibiosis, skin exudation and inflammation resistance, damaged cell repair, skin epithelial tissue regeneration promotion, stratum corneum accumulation reduction, pore blockage, keratinocyte and fibroblast proliferation, subcutaneous tissue thickening, skin weakness and sensitivity prevention, skin elasticity and toughness increase.
The technical solutions provided by the present invention are described in detail below in conjunction with examples for further illustrating the present invention, but they should not be construed as limiting the scope of the present invention.
Example 1
1. Preparation of paeoniflorin
Reflux extracting radix Paeoniae Rubra with 40% ethanol for 2 times each for 2 hr, filtering, mixing filtrates, adding 1.0% medicinal active carbon, stirring, filtering, recovering ethanol from filtrate, concentrating, filtering, drying filter cake, and pulverizing into 60 mesh fine powder.
2. Preparation of licorice extract
Slicing Glycyrrhrizae radix, decocting with 6 times of water for 3 times each for 2 hr, filtering, mixing filtrates, concentrating, drying, and pulverizing to obtain Glycyrrhrizae radix extract.
3. Preparation of semen Aesculi extract
Pulverizing semen Aesculi, extracting with 50% ethanol for 3 times each for 2 hr, filtering, mixing filtrates, concentrating, drying, and pulverizing into 60 mesh fine powder to obtain semen Aesculi extract.
4. Preparation of red deer blood extract
At a substrate concentration (deer blood: distilled water) of 1:2 (m/m), 0.1mol/L H was used 2 SO 4 Or NaOH is regulated to a proper pH, the concentration of protease is 8%, papain is added at 50 ℃ for enzymolysis for 5h, enzyme is inactivated at 95 ℃ for 10min, centrifugation is carried out for 20min at 3500r/min, supernatant and precipitate are collected, and low-temperature drying is carried out.

Pulverizing semen Aesculi, extracting with 70% ethanol for 3 times each for 2 hr, filtering, mixing filtrates, concentrating, drying, and pulverizing into fine powder to obtain semen Aesculi extract.
4. Preparation of red deer blood extract

The paeoniflorin mentioned in the pharmacodynamic test is the sample extracted in the step 1 of the example 2; the licorice extract is the sample extracted in the step 2 of the example 2; the buckeye extract is a sample extracted in the step 3 of the example 2; the red deer blood extract is a sample obtained by enzymolysis in the step 4 of the example 2; the traditional Chinese medicine composition provided by the invention is the traditional Chinese medicine composition with the acne-removing effect, which is prepared in the example 2.
1. Preparation of bacterial suspension
Propionibacterium acnes and staphylococcus aureus were selected as test species.
The staphylococcus aureus ATCC6538 and propionibacterium acnes ATCC11827 frozen stock tubes were taken out from the-80 ℃ refrigerator and placed in a biosafety cabinet until they were returned to normal temperature. Then, 1 loop (10. Mu.L) of Staphylococcus aureus broth was inoculated in 10mL of the liquid medium and incubated at 37℃for 48 hours. Taking 1mL out to 9mL of 0.85% physiological saline added with 1%o peptone, diluting twice and 100 times to prepare into about 10 7 The CFU/mL bacterial suspension is used. Propionibacterium acnes was cultured in an anaerobic incubator at 37℃for 48h using a modified GAM broth with 5% defibrinated sheep blood, and the other steps were performed as in Staphylococcus aureus.
2. Test procedure
The BHI solid medium was dissolved, returned to 46℃and 15-20mL was poured into the plate until it solidified. 100. Mu.L of the bacterial suspension was pipetted and spread evenly on a plate with an L-shaped spreading bar, waiting for about 10min for drying. Two oxford cups were held with sterile forceps and placed on a plate with slight pressure to allow insertion and fixation on the medium. 200 mu L of the sample to be detected is respectively sucked and added into two oxford cups, two plates of the same sample are subjected to parallel control, and a Blank control plate Blank is arranged at the same time. The loaded plates were incubated at 37℃for 48h and the results read. Propionibacterium acnes was cultured in an anaerobic incubator at 37℃for 48h using a modified GAM solid medium with 5% defibrinated sheep blood, the other steps being the same as above.
3. Reading the results
The diameter of the inhibition zone is measured by a vernier caliper, two effective digits are reserved by taking millimeter mm as a unit, and the result is recorded.
The evaluation of the antibacterial performance is as follows: the bacteriostasis zone is less than 10mm, has no obvious bacteriostasis effect, the bacteriostasis zone is more than 10mm and less than 15mm, and the bacteriostasis zone is more than 15mm, and is highly bacteriostasis.
4. Sample preparation
Taking paeoniflorin, licorice extract, buckeye extract, red deer blood extract and the traditional Chinese medicine composition, preparing 5g samples per mL, and performing an anti-acne and antibacterial experiment, wherein the results are shown in Table 1.

The paeoniflorin, the semen aesculi extract and the traditional Chinese medicine composition have obvious antibacterial activity, wherein the traditional Chinese medicine composition has stronger antibacterial activity than each single component.
EXAMPLE 5 Experimental study on acne in rats
90 healthy Wistar rats weighing 180 g.+ -.20 g were collected and divided into 9 groups of 10 rats each. The specific arrangement is shown in Table 2.

1. Preparation of rat skin acne model
In addition to the blank group, the other groups of rats were anesthetized with diethyl ether and 50. Mu.L of Propionibacterium acnes suspension (6X 10) was injected with a syringe 8 Cfu.ml-1) was injected into the auricle skin of the right ear, and after injection, the auricle thickness of the rat was measured with a micrometer every 24 hours, and 3 days were continued. Normal control group rat skin was not treated.
2. Administration method
The skin of the rats in the normal control group and the model control group is not dosed, and other auxiliary treatments are not carried out; the positive control is smeared with clindamycin and metronidazole liniments, and the other parts are smeared with 50mg/mL of paeonia total glycoside, licorice extract, buckeye extract and red deer blood extract respectively. On day 14 of continuous dosing, rats were sacrificed after 24 hours and serum and rat auricles were collected.
3. General living behavior state observation of rats
During the administration period, the physiological and behavioral characteristics, wound morphology and animal death of each group of animals are observed and recorded before daily administration.
In the whole experimental process, the normal control group rat body Mao Guanghua has normal diet, quick response and good activity. After continuous 4d infection, other groups of rats except the normal control group were subjected to pinna swelling part picking by a sterile needle, and were plated for culture, and strain growth proved that propionibacterium acnes, hair follicle areas were enlarged, funnel parts were keratinized materials, sebaceous glands were proliferated, indicating successful molding.
In the administration process, the model control group can see the phenomena of ear grasping, reduced food consumption, aggregation and lamination. The positive control group and the experimental group rats gradually recover from food intake and increase in activity along with the extension of treatment time, only a few rats have aggregation and lamination phenomena, the hair follicle area is changed from large to small, the keratinized substances are changeable, and the keratosis is not increased any more.
The results show that the positive control medicament and the experimental medicament can improve the life behavior of the model rat and can promote the acne removing effect.
4. Auricle swelling (%) of the rat auricle acne model was observed
Auricle thickness was measured using a micrometer gauge before injection of propionibacterium acnes in rats, and after day 3 and 14 of dosing. Auricle swelling rate was calculated as auricle swelling rate (%) = (auricle thickness after injection-auricle thickness before injection)/auricle thickness before injection×100%, and experimental results are shown in table 3.

It can be seen that on day 3 of inoculation, the normal group had a very significant difference in auricle swelling levels in rats compared to the model group (P < 0.001), and the other treatment groups had no difference in auricle swelling levels in rats compared to the model group, indicating that the acne model was successful and the retention time was long. After 14 days of administration, each treatment group significantly inhibited the degree of auricle swelling in acne model rats compared with the model group, and the traditional Chinese medicine composition of the present invention had the best effect, and was better than that of a single extract.
Example 6
Clinical trial
Evaluation object: 70 young people with ages of 15-30 years old, wherein 35 men and 35 women are divided into 7 groups of 10 people at random;
evaluating a subject's skin basic condition: redness, swelling, acne, and vaccinia caused by various factors;
the evaluation method comprises the following steps: each group was tested for 28 days using the traditional Chinese medicine compositions with acne-removing efficacy prepared in examples 1 to 3 and comparative examples 1 to 4, respectively, and the effect was examined for 28 days. Wherein, the comparative example 1 consists of paeonia total glycosides with single component, the comparative example 2 consists of licorice extract with single component, the comparative example 3 consists of buckeye extract with single component, and the comparative example 4 consists of red deer blood extract with single component.
The test method comprises the following steps: after the basic skin care step, a proper amount of the acne-removing traditional Chinese medicine composition is smeared, the smearing amount is increased at the acne position, each time in the morning and evening, each group only uses one acne-removing traditional Chinese medicine composition, and comprehensive evaluation is carried out by referring to Chinese acne treatment guidelines.
And (3) healing: the skin lesions completely subside, with only a small amount of pigmentation and slight scarring;
the effect is shown: the skin injury relieving degree is more than or equal to 70%, and various clinical symptoms are obviously improved;
the method is effective: the skin injury relieving degree exceeds more than or equal to 30 percent, and various clinical symptoms have obvious improvement trend but do not subside;
invalidation: the skin injury relieving degree is less than 30 percent, and various clinical symptoms are not improved or have obvious aggravation trend.

As can be seen from the table, examples 1 to 3 exhibited better acne-removing effects than comparative example 1. The number of people who had cured from acne was greater, with the acne removal effect of example 3 being most pronounced. The results prove that the acne-removing traditional Chinese medicine composition prepared by the invention has good acne-removing effect on acnes, and is safe and non-irritating due to natural components.
From the above examples, it can be seen that the paeoniflorin, the licorice extract, the buckeye extract and the red deer blood extract in the traditional Chinese medicine composition with acne removing effect provided by the invention can act synergistically, have more obvious activity than a single extract, and have the effects of repairing skin, treating skin flushing, local swelling and acne.

Claims (7)
1. The traditional Chinese medicine composition with the acne-removing effect is characterized by comprising the following raw materials in parts by weight: 5-50 parts of paeoniflorin, 3-30 parts of licorice extract, 2-25 parts of semen aesculi extract and 1-10 parts of red deer blood extract;
the red deer blood extract is an enzymatic hydrolysate obtained by enzymatic hydrolysis of red deer blood by papain;
the licorice extract is an aqueous licorice extract; the semen aesculi extract is ethanol extract of semen aesculi.
2. The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following raw materials in parts by weight: 10-30 parts of paeoniflorin, 10-20 parts of licorice extract, 5-20 parts of semen aesculi extract and 3-8 parts of red deer blood extract.
3. The traditional Chinese medicine composition according to claim 1 or 2, wherein the paeoniflorin is obtained by adsorbing paeoniflorin ethanol extract with activated carbon.
4. The method for preparing the traditional Chinese medicine composition according to any one of claims 1 to 3, which is characterized by comprising the following steps:
drying and crushing paeoniflorin, licorice extract, semen aesculi extract and red deer blood extract, and uniformly mixing to obtain the traditional Chinese medicine composition with acne removing effect.
5. A traditional Chinese medicine preparation with acne-removing effect, which is characterized by being prepared from the traditional Chinese medicine composition with acne-removing effect according to any one of claims 1-4.
6. The traditional Chinese medicine preparation with acne removing effect according to claim 5, wherein the preparation is powder, plaster, ointment, toning lotion, emulsion, mask or gel.
