2. Procedure of the test

4. Sample preparation
The total paeony glycoside, the licorice extract, the buckeye extract, the red deer blood extract and the traditional Chinese medicine composition are prepared into a sample containing 5g of Chinese herbal medicine per mL, and the results are shown in Table 1.

Example 5 Experimental study on acne in rats

1. Preparation of rat skin acne model
Except for the blank control group, the rats of the other groups were anesthetized with ether, and 50. 3. General lifestyle status observations in rats
During the administration period, the physiological and behavioral characteristics, wound morphology and animal death of each group of animals were observed and recorded before daily administration.
In the whole experimental process, the body hair of the rats in the normal control group is smooth, the diet is normal, the reaction is quick, and the activity is good. Except for the normal control group, after the rats in other groups are infected for 4 days continuously, auricle swelling parts are picked by using a sterile needle head and are plated on a flat plate for culture, and the strain growth proves that propionibacterium acnes, the area of hair follicles is enlarged, keratinized substances are in infundibulum parts, and sebaceous glands are proliferated, thereby indicating that the molding is successful.
In the administration process, the model control group can catch ears, reduce food intake, and have aggregation and overlapping phenomena. The positive control group and the experimental group of rats begin to recover food intake and increase activity along with the prolonging of treatment time, only a few rats have aggregation and overlapping phenomena, the quantity of keratinized substances is reduced from large to small, and the keratinocyte hyperplasia is not increased any more.
The results show that the positive control medicament and the experimental medicament can improve the living behavior of the model rat and can promote the acne-removing efficacy.
4. Observation of degree of swelling (%) of auricle of rat auricle acne model
Auricle thickness was measured before injection of propionibacterium acnes and after the 3 rd and 14 th days of administration using a micrometer thickness gauge. The auricle swelling rate was calculated as the auricle swelling rate (%) = (auricle thickness after injection-auricle thickness before injection)/auricle thickness before injection × 100%, and the experimental results are shown in table 3.

It can be seen that on day 3 of inoculation, there was a very significant difference in the degree of swelling of the auricle in rats compared to the model group (P < 0.001), and there was no difference in the degree of swelling of the auricle in rats compared to the other treatment groups and the model group, indicating that the acne model was successful and maintained for a long time. After 14 days of administration, compared with the model group, each treatment group obviously inhibits the auricle swelling degree of an acne model rat, and the traditional Chinese medicine composition has the best effect and better effect than a single extract.
Example 6
Clinical trial
Evaluation target: 70 young people with the age of 15-30 years old, wherein 35 male and 35 female are randomly and averagely divided into 7 groups of 10 people;
evaluating the basic condition of the skin of a subject: acne rosacea, acne vulgaris;
the evaluation method comprises the following steps: the above-mentioned Chinese medicinal compositions for removing acne effects prepared in examples 1 to 3 and comparative examples 1 to 4 were used for each group, and the experiment was carried out for 28 days in total, and the effect of 28 days was examined. Wherein, the comparative example 1 is composed of single component peony total glycoside, the comparative example 2 is composed of single component licorice extract, the comparative example 3 is composed of single component buckeye extract, and the comparative example 4 is composed of single component red deer blood extract.
The test method comprises the following steps: after the basic skin care step, a proper amount of the acne-removing traditional Chinese medicine composition is smeared, the smearing amount is increased at the position of acne, the smearing amount is increased once every morning and evening, each group only uses one acne-removing traditional Chinese medicine composition, and comprehensive evaluation is carried out by referring to Chinese acne treatment guidelines.

and (4) invalidation: the skin damage relieving degree is less than 30 percent, and various clinical symptoms are not improved or have obvious aggravation trend.

As can be seen from the table, examples 1 to 3 exhibited better acne removing effects than comparative example 1. Acne heals more, with the acne removal effect of example 3 being most prominent. The results prove that the acne-removing traditional Chinese medicine composition prepared by the invention has a good inhibition effect on acne and pox, contains natural components, and is safe and non-irritating.
The embodiment shows that the Chinese medicinal composition with the acne removing effect provided by the invention has the synergistic effect of the total paeoniflorin, the licorice extract, the buckeye extract and the deer blood extract, has more obvious activity than a single extract, and has the effects of repairing skin and treating skin flushing, local swelling and acne.
