技术领域
本发明属于生物医药领域,具体是一种慢病毒高效感染人NK细胞的方法。
背景技术
作为固有免疫系统的重要成员,自然杀伤(natural killer,NK)细胞是机体抗击病毒感染及肿瘤的第一道防线。基于NK细胞MHC非限制性、泛特异性识别和杀伤靶细胞及快速应答的特点,嵌合抗原受体自然杀伤细胞(chimeric antigen receptor naturalkiller cell,CAR-NK)在血液肿瘤免疫治疗的应用方面被寄予越来越多的关注和期待。嵌合抗原受体(CAR)是一种受体蛋白,它赋予免疫细胞新的能力,以靶向特定的抗原蛋白。新近的临床研究及临床前研究结果均提示CAR-NK细胞对于AML、淋巴瘤、MM等恶性血液肿瘤具有较强的抗肿瘤效应且不良反应较小,未来作为细胞免疫治疗拥有良好的前景及研究价值。
肿瘤细胞可通过免疫逃逸机制逃避NK细胞攻击而限制了NK细胞的临床应用,CAR-NK细胞可通过反转录病毒及慢病毒转染表达胞外抗原识别区而克服其免疫逃逸缺陷。但因NK细胞仅占外周血淋巴细胞的15%左右、体外扩增培养技术仍不成熟、DNA转染效率低等技术瓶颈,临床应用方案等研究仍远远落后于T细胞。
目前CAR基因转染的两大系统主要是无病毒的转座子基因转染系统和基于慢病毒、反转录病毒的转染体系。原代NK细胞利用病毒转染体系的效率仅约15%,且反转录病毒转染可能会引起插入突变,外周血来源的NK细胞采用慢病毒转染效率亦很低。慢病毒与逆转录病毒不同,它不需要主动分裂细胞,因此能够转导更多类型的NK细胞,因此比逆转录病毒转导具有一些潜在的优势。
原代NK细胞的慢病毒转导效率通常依赖于促进病毒进入的化学试剂,如阳离子聚丙稀烯和鱼精蛋白硫酸盐。然而,这些试剂已经显示出对NK-92细胞的毒性,因此替代试剂,如DEAE-葡聚糖和多聚-L-赖氨酸,近年来也被使用。此外,无毒的阳离子多肽,如维甲酸,已被用于转染造血干细胞来源的NK细胞,并显示出比化学试剂更高的转染效率。
发明内容
为解决现有的病毒感染NK细胞效率很低,且感染后细胞活率低的技术问题,本发明提供一种高效的慢病毒高效感染人NK细胞的方法。
本发明的技术方案包括:
第一方面,本发明提供了一种将目标基因导入NK细胞的方法,所述方法包括两次病毒感染的步骤,第一次病毒感染时加入polybrene,第二次病毒感染时加入PMA、IONOMYCIN、polybrene、IL-2,所述病毒携带目标基因。
优选地,所述polybrene的工作浓度是8μg/ml。
优选地,所述PMA的工作浓度是1μg/ml。
优选地,所述IONOMYCIN的工作浓度是0.25μg/ml。
优选地,所述IL-2的工作浓度是150ng/ml。
优选地,所述第一次病毒感染时的病毒复数是3-5MOI;最优选地,所述第一次病毒感染时的病毒复数是4MOI。
优选地,所述第二次病毒感染时的病毒复数是5-7MOI;最优选地,所述第二次病毒感染时的病毒复数是6MOI。
优选地,所述第一次病毒感染的持续时间是1-3小时;最优选地,2小时。

本发明所述“导入”还可以称为感染、转导、转染等,是指使NK细胞表达目标基因的步骤,更具体地,在本发明中是指病毒介导的DNA转移的过程。
本发明所述polybrene(聚凝胺),也称Hexadimethrine Bromide(海美溴铵)是一种多聚阳离子聚合物。
本发明所述PMA(phorbol 12-myristate 13-acetate),也称12-O-十四烷酰基佛波醇13-乙酸酯、佛波肉荳蔻醋酸、佛波酯,是一种广泛用于体内外实验的佛波酯类PKC激活剂。
本发明所述,本发明所述IONOMYCIN(离子霉素)是一种来源于Streptomycesconglobatus的聚醚类窄谱抗生素,是一种有效的钙离子载体,高度选择性结合钙离子,也能转运其他离子,但亲和力相对较低。
本发明所述CAR(嵌合抗原受体)是一种受体蛋白,它赋予免疫细胞新的能力,以靶向特定的抗原蛋白。CAR通常包括三个主要部分:胞外域(抗原结合域)、跨膜域和胞内域。胞外域负责识别抗原,其亲和力特征是一个特别重要的抗原结合结构域参数,它解释了多种受体-配体相互作用,从根本上决定了CAR分子的功能,胞外域通常还可以包含铰链区(hinge)。跨膜域的主要功能在于将CAR分子锚定在细胞膜上,将CAR分子的细胞外结构域与细胞内结构域连接起来,从而将配体识别信号转导至细胞内,其对CAR分子表达的稳定性具有重要作用。胞内域可以包括信号转导结构域和共刺激结构域。本领域技术人员根据所要靶向的特定抗原蛋白可以根据需求选择不同结构域的类型,组装出靶向不同治疗靶点的CAR,例如CD7、CD19、CD33、CD22、BCMA、ROBO1、PSMA、Mesothelin、HER2、MUC1等。本发明具体实施例以靶向CD19的CAR为例进行了实验验证。
另一方面,本发明还提供了一种病毒感染NK细胞的培养系统,所述培养系统包括第一病毒感染单元和第二病毒感染单元,所述第一病毒感染单元中向病毒感染体系中加入polybrene,所述第二病毒感染单元中向病毒感染体系中加入PMA、IONOMYCIN、polybrene、IL-2。
优选地,所述polybrene的工作浓度是8μg/ml。

优选地,所述对慢病毒进行包装可以使用人类细胞株或非人细胞株。
本发明所述慢病毒可以选用任意包装系统,目前有四质粒系统(编码gag-pol基因、Rev基因、VSVG基因、SIN转移基因分别位于四个不同的质粒)、三质粒系统(去掉了编码Rev基因的质粒,在gag-pol质粒中gag-pol基因采用了在人细胞中偏爱性的密码子)和二质粒系统(慢病毒载体包装所必需的辅助基因位于同一个质粒上,这些辅助基因是单一的基因序列;另一个则是转基因质粒)。也有超过四质粒系统的慢病毒包装系统在使用。
另一方面,本发明还提供了上述培养系统在制备含有目标基因的NK细胞中的用途。
另一方面,本发明还提供了上述培养系统在将目标基因高效转入NK细胞中的用途。
优选地,所述目标基因可以是CAR(嵌合抗原受体)。
另一方面,本发明还提供了一种促进病毒转导的试剂组合,所述试剂组合包括PMA、IONOMYCIN、polybrene、IL-2。
优选地,所述试剂组合中还可以包括病毒制备所需要的试剂、制备NK细胞所需要的试剂。
另一方面,本发明还提供了上述方法制备得到的NK细胞以及在制备治疗癌症的药物、在制备细胞移植治疗所用药物中的应用。

具体实施方式
下面结合实施例对本发明做进一步的说明,以下所述,仅是对本发明的较佳实施例而已,并非对本发明做其他形式的限制,任何熟悉本专业的技术人员可能利用上述揭示的技术内容加以变更为同等变化的等效实施例。凡是未脱离本发明方案内容,依据本发明的技术实质对以下实施例所做的任何简单修改或等同变化,均落在本发明的保护范围内。
实施例一、慢病毒高效感染人NK细胞
1、慢病毒的制备
示例性地以CD19CAR为目标基因进行NK细胞转染,所述CD19CAR的组成如图1所示。慢病毒购买于谱新生物(货号LVHCNV01),按照常规方法制备为携带CD19CAR的慢病毒。
2、PBMC的培养-NK细胞诱导
使用依科赛NE000-N022 NK培养套装培养瓶预处理:室温下融化细胞因子I,取50mL离心管,加入15mL DPBS,吸取45μL细胞因子I至DPBS中(若细胞因子I一次性用完,建议吸取50mL离心管内1mL DPBS将细胞因子I管冲洗1次并加回离心管内),上下颠倒混匀,加入底面积75cm2的细胞培养瓶(T75)中,前后左右晃动,使液体分散在瓶底,4℃活化过夜。若不马上使用,可置于4℃冰箱储存,建议3天内使用。第0天,取出4℃活化过夜的培养瓶,弃掉包被液,使用15mL NK培养基(可添加10%的热灭活自体血浆)重悬PBMC接种于T75瓶中(推荐接种密度2×106cells/mL),添加细胞因子Ⅱ150μL,前后左右晃动,放入37℃,5%CO2培养箱中培养。第3天,沿培养瓶侧壁缓慢补加15mL的新鲜NK培养基添加10%的热灭活自体血浆,第5天,取样计数,补加新鲜NK培养基,调整细胞密度1.0-1.5E6 cells/ml第7天,取样计数,补加新鲜NK培养基,调整细胞密度1.0-1.5E6 cells/mL,第9天,取样计数,补加新鲜NK培养基,调整细胞密度1.0-1.5E6 cells/mL第10天进行慢病毒感染。
3、慢病毒感染NK细胞
实验组:
第一次感染:加入8μg/ml polybrene,然后加入4MOI lentivirus。
第二次感染:放回培养箱(37℃、5%CO2)培养2小时后进行第二次感染,加入1μg/ml的PMA(陶术化学TQ0198)、0.25μg/ml的IONOMYCIN(陶术化学T11665)、8μg/ml的polybrene(Merck TR-1003)、150ng/ml的IL-2(同立海源GMP-TL777)混合物,加入6MOIlentivirus。
以上浓度皆为对应物质的工作浓度。
放回培养箱培养4小时,自第一次感染6小时后,取出全部细胞悬液,离心400g5min,DPBS 40ml重悬细胞,离心400g 5min,去上清,重悬于扩增培养基待测。所述扩增培养基是购买自依科赛(NE000-N022)。
另设置空白组和对照组,其中空白组不进行感染,对照组仅进行上述第一次感染,其他操作同实验组一致。
4、NK细胞活率、阳性表达率对比
以进行上述第一次感染为0h开始计算,在感染到6h、12h、24h时分别检测CAR阳性率和细胞活率,结果如图2所示。在连续培养4天后再次检测阳性率,结果如图3所示。
各个时间的检测皆显示,进行两次感染的NK细胞的阳性率显著高于只进行一次感染的NK细胞。
5、细胞杀伤效果对比
以杀伤NALM-6细胞(人急性淋巴细胞白血病细胞)为例检测NK细胞的杀伤效果。
NK细胞感染后第二天,和Nalm6-GFP细胞按照NK:Nalm比例1:2和1:1混合共培养24小时,流式细胞仪检测GFP阳性细胞比率,代表Nalm6的杀伤效果,结果如图4A所示。
使用DELFIA EuTDA试剂盒检测细胞毒性效果:Nalm6细胞与BATDA reagent浓度2μl/ml预先混合孵育30min在37度,经过5次洗涤彻底洗脱细胞外残留reagent,按照比例Nalm6:NK=1:1 1:2 1:4 1:8混合孵育2小时后,Nalm6细胞被杀伤后,reagent从Nalm6中释放到细胞培养基中,由于不同杀伤效果,reagent释放的量不同,收集细胞上清液,包含reagent,混合ES溶液室温孵育15分钟,每3分钟,轻轻震动混匀。使用酶标仪TRF通道读取荧光数据(315nm激发640nm接收)。计算公式为:%=(上清读数-自发光读数)/(细胞裂解最大读数-自发光读数),获得杀伤率,结果如图4B所示。
实验结果均表明进行两次感染的NK细胞的杀伤效果显著优于只进行一次感染的NK细胞。
Claims (49)
1.一种将目标基因导入NK细胞的方法,所述方法包括使用携带目标基因的病毒对NK细胞进行两次病毒感染的步骤,第一次病毒感染时加入8μg/ml的polybrene,第一次感染2小时后进行第二次感染,第二次病毒感染时加入1μg/ml 的PMA、0.25μg/ml 的IONOMYCIN、8μg/ml的polybrene、150ng/ml 的IL-2,第二次感染持续4小时后分离细胞进行下一步操作,所述病毒是慢病毒。
2.如权利要求1所述方法,所述第一次病毒感染时的病毒复数是3-5MOI。
3.如权利要求2所述方法,所述第一次病毒感染时的病毒复数是4MOI。

15.一种病毒感染NK细胞的培养系统,所述培养系统包括第一病毒感染单元和第二病毒感染单元,所述第一病毒感染单元中向病毒感染体系中加入polybrene,所述第二病毒感染单元中向病毒感染体系中加入PMA、IONOMYCIN、polybrene、IL-2;第一病毒感染单元的病毒感染持续2小时,第二病毒感染单元的病毒感染持续4小时,所述病毒是慢病毒;
所述polybrene的工作浓度是8μg/ml;
所述PMA的工作浓度是1μg/ml;
所述IONOMYCIN的工作浓度是0.25μg/ml;
所述IL-2的工作浓度是150ng/ml。
16.如权利要求15所述的培养系统,所述培养系统还包括以下单元:
1)慢病毒制备单元,
2)NK细胞收集或制备单元,
3)第一病毒感染单元,和,
4)第二病毒感染单元。
17.如权利要求15所述的培养系统,所述培养系统还包括NK细胞扩增培养单元。
18.如权利要求15所述的培养系统,所述培养系统还包括NK细胞阳性率质检单元。
19.如权利要求15所述的培养系统,所述培养系统还包括NK细胞杀伤活力质检单元。
20.如权利要求15所述的培养系统,所述慢病毒的载体包括pLK0.1-puro、pLK0.1-CMV-tGFP、pLKO.1-puro-CMV-tGFP、pLK0.1-CMV-Neo、pLK0.1-Neo、pLKO.1-Neo-CMV-tGFP、pLKO.1-puro-CMV-TagCFP、pLKO.1-puro-CMV-TagYFP、pLKO.l-puro-CMV-TagRFP、pLKO.1-puro-CMV-TagFP635、pLKO.-puro-UbC-TurboGFP、pLKO.l-puro-UbC-TagFP635、pLKO-puro-IPTG-1xLacO、pLKO-puro-IPTG-3xLacO、pLPl、pLP2、pLP/VSV-G、pENTR/U6或pLenti6/BLOCK-iT-DEST。
