口服液剂是以汤剂为基础,提取有效成分,加入矫味剂、抑菌剂等附加剂,并按注射剂安瓿灌封处理工艺,制成的一种无菌或半无菌的口服液体制剂。它是汤剂、糖浆剂和注射剂三种剂型相结合的一种新型制剂。如生脉饮口服液、四逆汤口服液、参茸蜂皇浆口服液等。其特点是服用剂量小,味道好、吸收快、奏效迅速,易为患者所接受。
牡蛎肉含有丰富的糖原、蛋白质、氨基酸、脂肪、微量元素和维生素等,其中糖原占18%~39%,蛋白质为45%~57%,脂肪为7%~11%。牡蛎氨基酸组成完善,据世界粮农组织评定,牡蛎肉中必需氨基酸完全程度和质量比例优于人乳和牛乳,其除了含人体必需的20种常见氨基酸外,还含有β-氨基丙酸、γ-氨基丁酸、鸟氨酸、牛磺酸等多种具有重要生理价值的氨基酸。特别是牛磺酸的含量尤其丰富,明显高于其他动物性食品,含量可以达到总氨基酸量的30.98%。
随着经济建设、科学技术的发展和人民生活水平的提高,人们对生命的价值和健康的重要性认识日益深刻,越来越重视,口服液受到人们的青睐,如中国专利申请号为CN102551067B的专利文件中公开了《调节免疫力的口服液及其制备方法》,上述专利文件所制备的牡蛎口服液虽然具有一定的提高免疫力,但是其提高免疫力的效果不佳,也缺少抗肿瘤及抗氧化作用,功能的单一,影响了所制备的口服液的性能及质量。
发明内容
针对现有技术所存在的上述缺点,本发明在于提供提高免疫力的牡蛎肽酵素口服液,本发明通过制备牡蛎酶解液、预处理和凝胶柱解析,实现原味提取牡蛎活性肽,去除了牡蛎中的非活性物质,高度保留了牡蛎的天然活性成分,使得提取的牡蛎活性肽酵素更易被肌体吸收,具有抗氧化能力,同时,蛎活性肽对癌细胞具有抑制作用,且引起细胞形态学改变,并能诱导其发生凋亡;使得所制备的提高免疫力的牡蛎肽酵素口服液的品质得到有效地提高与改善。
为实现上述目的,本发明提供了如下技术方案:
高免疫力的牡蛎肽酵素口服液,包括如下重量份的原料:牡蛎20-30份、枸苠4-10份、人参2-10份、海参2-10份、石斛2-8份、调味剂3-8份、水20-30份。
通过采用上述技术方案:本发明采用牡蛎为原料,提取牡蛎肽酵素,牡蛎肽酵素具有提高免疫力、提高男性血清睾水平、保肝、降血糖和抗肿瘤等多种潜在功能,提高男性血清睾酮水平、调节血脂、抑制血小板聚集、改善高血糖症状、提高人体、促进新陈代谢等功能,对和防止癌细胞扩散也有一定效果,同时加入枸苠、人参、海参、石斛和调味剂,枸苠对人体能够延缓衰老,人参能够治疗心血管系统疾病以及抗疲劳,石斛能够增强机体免疫力和促进消化,通过调味剂进行调味,在保证口味的同时,还实现了延缓衰老、增强人体免疫力等的效果。
本发明进一步设置为:所述调味剂为低聚果糖、低聚半乳糖中的一种或者两种按任意比例混合。
通过采用上述技术方案:本发明以调味剂来实现制备后的口服液的甜味口感,甜味清爽,具有非消化性、胰岛素非依赖性,摄食后能有效降低空腹时的血糖水平。
提高免疫力的牡蛎肽酵素口服液的制备方法,包括以下步骤:
S1、制备牡蛎酶解液。
S2、牡蛎酶解液的预处理。
S3、将预处理后的牡蛎酶解液,用凝胶柱层析后,制得牡蛎肽酵素溶液。
S4、将枸苠、人参、海参、石斛放入粉碎机中粉碎,得粉碎混合物。
S5、将粉碎混合物、调味剂和牡蛎肽酵素溶液放置到搅拌中,并将水加入到搅拌机中,搅拌混合均匀。
S6、对S5中的混合溶液过滤,并水浴灭菌,冷却至室温,装瓶即可。

取新鲜牡蛎去壳、清洗,得新鲜牡蛎肉;将牡蛎肉加入到搅拌机中,并向搅拌机中加入预冷的双蒸水,在搅拌转速为200-300r/min的条件下,高速均浆5-10min;向浆液中加入菠萝蛋白酶,在PH8.0,温度为55℃的条件下,酶解5h;沸水浴灭活8min,冷却至室温;用高速离心机在转速为6000-7000r/min,温度为4℃的条件下高速离心30min;取上清液,制得牡蛎酶解液。
通过采用上述技术方案:本发明以新鲜牡蛎为原料,保障了原材料的质量,通过搅拌搅拌、离心、酶解,能够从牡蛎肉内提取到牡蛎活性肽,去除了牡蛎中的非活性物质,高度保留了牡蛎的天然活性成分,还采用了菠萝蛋白酶提升了酶解率。
本发明进一步设置为:在所述步骤S2中,牡蛎酶解液的预处理为:
将牡蛎酶解液放置于4℃冰箱12h;采用0.45um的微孔滤膜过滤,收集滤液。
通过采用上述技术方案:本发明对牡蛎酶解液进行预处理,可以有效的避免对凝堵塞、流速降低等影响,保障了后续的凝胶柱的正常使用,提高了制备的口服液的口感。
本发明进一步设置为:在所述步骤S4中,在枸苠、人参、海参、石斛放入粉碎机前,对枸苠、人参、海参、石斛清洗、风干和切块。
通过采用上述技术方案:本发明通过加入枸苠、人参、海参、石斛,实现了延缓衰老、增强人体免疫力的效果。
本发明进一步设置为:在所述步骤S5中,所述搅拌机的转速为200-300r/min,搅拌时间为30min。
通过采用上述技术方案:本发明以粉碎机对枸苠、人参、海参、石斛进行粉碎,得到混合物,用于后续的混合制备口服液。
本发明进一步设置为:在所述步骤S5中,采用0.45um的微孔滤膜对混合溶液进行过滤。
通过采用上述技术方案:本发明以0.45um的微孔滤膜对混合溶液进行过滤,能够使得制备的口服液的细腻。

采用本发明提供的技术方案,与已知的公有技术相比,具有如下有益效果:

实施例1
请参照图1所示,提高免疫力的牡蛎肽酵素口服液,包括如下重量份的原料:牡蛎20份、枸苠4份、人参2份、海参2份、石斛2份、调味剂3份、水20份。
调味剂为低聚果糖、低聚半乳糖中的一种或者两种按任意比例混合。
提高免疫力的牡蛎肽酵素口服液的制备方法,包括以下步骤:
步骤一、制备牡蛎酶解液。
步骤二、牡蛎酶解液的预处理。
步骤三、将预处理后的牡蛎酶解液,用凝胶柱层析后,制得牡蛎肽酵素溶液。
步骤四、将枸苠、人参、海参、石斛放入粉碎机中粉碎,得粉碎混合物。
步骤五、将粉碎混合物、调味剂和牡蛎肽酵素溶液放置到搅拌中,并将水加入到搅拌机中,搅拌混合均匀。
步骤六、对步骤五中的混合溶液过滤,并水浴灭菌,冷却至室温,装瓶即可。
制备牡蛎酶解液的方法为:

实施例2
本实施例所提供的提高免疫力的牡蛎肽酵素口服液大致和实施例1相同,其主要区别在于:包括如下重量份的原料:牡蛎30份、枸苠10份、人参10份、海参10份、石斛8份、调味剂8份、水30份;
制备牡蛎酶解液的过程中,搅拌转速为300r/min,高速均浆10mim,用高速离心机在转速为7000r/min的条件下高速离心;
对比例1
本实施例所提供的提高免疫力的牡蛎肽酵素口服液大致和实施例1相同,其主要区别在:未添加牡蛎;
对比例2
本实施例所提供的提高免疫力的牡蛎肽酵素口服液大致和实施例1相同,其主要区别在:未保留牡蛎的活性成分。
性能测试
取实施例1~2和对比例1~2所制备口服液,并对所制备出的口服液的相关性能进行检测,其检测方法如下:
1、按照检测标准GT/5009.194-2003分别对上述各口服液的提高免疫力(免疫球蛋白IgG)性能进行检测,将所得测试结果记录于表1;

2、按照检测标准GT/5009.7-2003分别对上述各口服液的营养(糖原)性能进行检测,将所得测试结果记录于表2;

通过分析上述表中的相关数据可知,通过本发明所制备的提高免疫力的牡蛎肽酵素口服液不仅具有良好抗免疫力,还具有丰富的营养价值,使得所制备的提高免疫力的牡蛎肽酵素口服液的品质得到有效地提高与改善。
3、抗肿瘤性能进行检测:
收集对数生长期的Tca8113细胞,以1.0×106个细胞种入25mL的培养屏瓶中,待细胞贴壁后,换新鲜培养液以除去未贴壁细胞,加入本发明提取的牡蛎肽酵素液,对照组不加入牡蛎肽酵素液,置于37℃、5%CO2饱和湿度培养箱中连续培养2d,消化细胞并离心收集,沿管壁缓慢加入戊二醛固定液,于4℃下固定2h以上,经梯度乙醇脱水,环氧树脂包埋,超薄切片。醋酸铀、柠檬酸铅双重染色后,透射电镜下观察,通过观察获得:未本发明提取的牡蛎肽酵素液组细胞核形态规则,细胞膜完整,核仁清晰,染色质均匀分布于细胞核内,胞浆内线粒体等细胞器清晰可见,微绒毛丰富,处理48h后Tca8113细胞呈现凋亡的典型形态,细胞微绒毛减少,细胞体积缩小,可见部分细胞核仁消失,部分细胞核解体,已形成由质膜包裹内含完整的细胞器和核碎片等细胞内容物的凋亡,对照组细胞并未凋亡小,可知本发明提取的牡蛎肽酵素液具有抑制作用,且引起细胞形态学改变,并能诱导其发生凋亡的效果。
4、抗氧化能力进行检测:
取0.15mol/L,PH7.4的磷酸盐缓冲液1.5L,110ug/mL的番红溶液0.2mL,2mmol/LdeEDTA-Na2-Fe(Ⅱ)0.7mL,再加本发明提取的牡蛎肽酵素液0.8mL,最后加入体积分数为1.5%的H2O20.8mL以启动反应,混合后于37℃水浴保温30min,在波长为520mm处测吸光度,以未加入本发明提取的牡蛎肽酵素液作为对照组,可知,本发明提取的牡蛎肽酵素液,具有良好的抗氧化能力。
本发明通过制备牡蛎酶解液、预处理和凝胶柱解析,实现原味提取牡蛎活性肽,去除了牡蛎中的非活性物质,高度保留了牡蛎的天然活性成分,使得提取的牡蛎活性肽酵素更易被肌体吸收,具有抗氧化能力,同时,蛎活性肽对癌细胞具有抑制作用,且引起细胞形态学改变,并能诱导其发生凋亡,同时加入枸苠、人参、海参、石斛和调味剂,枸苠对人体能够延缓衰老,人参能够治疗心血管系统疾病以及抗疲劳,石斛能够增强机体免疫力和促进消化,通过调味剂进行调味,还实现了延缓衰老、增强人体免疫力等的效果,使得所制备的提高免疫力的牡蛎肽酵素口服液的品质得到有效地提高与改善。

Claims (8)
1.提高免疫力的牡蛎肽酵素口服液,其特征在于,包括如下重量份的原料:牡蛎20-30份、枸苠4-10份、人参2-10份、海参2-10份、石斛2-8份、调味剂3-8份、水20-30份。
2.根据权利要求2所述的提高免疫力的牡蛎肽酵素口服液,其特征在于,所述调味剂为低聚果糖、低聚半乳糖中的一种或者两种按任意比例混合。
3.提高免疫力的牡蛎肽酵素口服液的制备方法,其特征在于,使用了根据权利要求1-2任一项所述的提高免疫力的牡蛎肽酵素口服液,包括以下步骤:
S1、制备牡蛎酶解液;
S2、牡蛎酶解液的预处理;
S3、将预处理后的牡蛎酶解液,用凝胶柱层析后,制得牡蛎肽酵素溶液;
S4、将枸苠、人参、海参、石斛放入粉碎机中粉碎,得粉碎混合物;
S5、将粉碎混合物、调味剂和牡蛎肽酵素溶液放置到搅拌中,并将水加入到搅拌机中,搅拌混合均匀;
S6、对S5中的混合溶液过滤,并水浴灭菌,冷却至室温,装瓶即可。
4.根据权利要求3所述的提高免疫力的牡蛎肽酵素口服液,其特征在于,在所述步骤S1中,制备牡蛎酶解液的方法为:

8.根据权利要求3所述的提高免疫力的牡蛎肽酵素口服液,其特征在于,在所述步骤S5中,采用0.45um的微孔滤膜对混合溶液进行过滤。
