Culturing T25 cell containing human epidermal melanocyteAt 37 ℃ C, 5% CO 2 And standing in a cell culture box with saturated humidity for 3-4 h. Sucking out the culture medium in the culture bottle, and washing the cells once by using the PBS; adding 1mL of 0.25% trypsin digestion solution into the culture bottle, slightly rotating the culture bottle until the digestion solution covers the whole bottom of the culture bottle, sucking out the redundant trypsin digestion solution, and carrying out warm bath at 37 ℃ for 1-3 min; after cell retraction rounding off was observed microscopically, digestion was stopped by adding 5mL of complete medium.
Adjusting the density of human epidermal melanocytes to 1 × 10 5 One cell/mL, 0.5mL per well was inoculated into a 24-well plate, and the medium was added to make the cell density 5X 10 4 Cell/well, at 37 ℃ and 5% CO 2 And culturing for 24 hours in a cell culture box with saturated humidity. The 24-well plate was removed, the medium was aspirated, 1mL of 10 μ M eriodictyol solution prepared from the complete medium (group 01), 10 μ M7-O-methyl eriodictyol solution (group 02), 10 μ M eriodictyol-7-O-glucoside solution (group 03), 10 μ M high eriodictyol solution (group 04), 10 μ M high eriodictyol-7-O-glucoside solution (group 05), 10 μ M7, 4 '-di-O-methyl eriodictyol solution (group 06), 10 μ M eriodictyol-7, 3' -dimethyl ether solution (group 07), and 10 μ M trimethyl eriodictyol solution (group 08) were added to each well of the experimental group, and 1mL of complete medium containing no sample to be tested (group 00) was added to the control group, and the control group was placed in an incubator for culture for 36 hours. After aspirating the medium and washing the cells twice with 1mL of PBS, 100 μ L of PBS solution containing 10% dmso, 1M NaOH was added per well, placed in a water bath at 80 ℃ for 1h in a water bath, the cell eluate from each well of the 24-well plate was transferred to a 96-well plate, the absorbance value of each well was measured at 475nm using a microplate reader, and the relative melanin content of each group was calculated (relative melanin content = absorbance value of cell eluate from each group/absorbance value of cell eluate from control group ×. 100%).
