(1) Sample treatment: the compositions of examples 1 to 16 and comparative examples 1 to 6, which were just prepared, were designated as A 0 The method comprises the steps of carrying out a first treatment on the surface of the The compositions of examples 1 to 16 and comparative examples 1 to 6 were placed in an oven at 50℃for 30 days in the absence of light, and the data obtained were designated A 50 . The compositions of examples 1 to 16 and comparative examples 1 to 6 were each prepared as an aqueous solution having a mass concentration of 5%.
(2) And (3) data processing: rate of change = 1-a 0 /A 50 The results are shown in Table 1.
TABLE 1 stability test results
As can be seen from Table 1, the stability of the formulation was divided into four equal runs, examples 10-16, examples 6-9, examples 1-5 and comparative examples 1-6, respectively, with the stability being high to low. The result is due to the different components of the formulation. All examples are in a more stable state, example 16 is the highest concentration ratio of all components, and the system is the most stable at the ratio, and the stability rate reaches 95% and above. While the comparative examples 5 and 6 are clearly higher in stability than the other comparative examples due to their inclusion of the lacquer wax component, which also demonstrates the stability. However, overall, all examples were more stable than all comparative examples, sufficient to demonstrate formulation stability.
2. Repair efficacy test
The cosmetic effect of reference T/GDCA 009-2022 was tested by the transepidermal water loss test method.
(1) Test sample: the compositions of examples 1 to 16 and comparative examples 1 to 6 were prepared as 5% aqueous solutions, respectively. Meanwhile, water is used as a control group.
(2) Subject selection
The selection standard is that the age is between 18 years old and 60 years old, and healthy men or women; lactic acid stinging positive; subjective skin has one of symptoms of impaired skin barrier function, such as dryness, desquamation, redness, etc.; the test process can be understood, and the test is voluntarily participated and the informed consent person is signed; the product can be used in a proper amount according to the requirement.
Exclusion criteria are pregnant or lactating women, or those who have a pregnancy-preparing program recently; patients with skin history such as psoriasis, eczema, atopic dermatitis, etc.; currently or in the last month oral or external use of anti-inflammatory drugs such as corticosteroids; currently or in the near month to other clinical experimenters; involuntarily participating, and cannot complete the required content on time; other clinical evaluations were considered unsuitable for the participating test subjects;
withdrawal criteria are subjects such as adverse events occurring during the study or violating the study protocol, which the researcher deems unsuitable for the subject to continue to complete the study; in other cases, the clinical study is exited while the reason for the exit is written.
Safety evaluation: all adverse skin reactions such as redness, stinging, itching, burning etc. occurring during the study period should be recorded and timely reported for serious adverse reactions.
Adverse reactions were judged according to the following three-level criteria:
mild-easy to tolerate, causing little discomfort, not affecting normal life.
Moderate-enough to affect normal life.
Severe-impeding normal life.
Relationship between adverse reactions and study:
irrelevant-obviously related to other factors; unlikely-most likely to be related to other factors; may-occur after use but may also be due to other factors; it is possible-that this occurs after use but cannot be explained by other factors; it is likely-that this occurs after use, but cannot be explained by other factors, and is present after use.
The subject: the female has the advantages that the female brings 32 persons into the female, falls off 2 persons, has the effective cases of 30 persons, has 0 men and 32 women; minimum age: 18 years of age, maximum age: age 57; average age 33.09 years.
(3) Measurement procedure
For the first visit, subjects were subjected to a test instruction and signed a written informed consent. The subjects were asked a series of questions regarding disease history, health status, etc. based on inclusion and exclusion criteria, etc., and compliance assessment was performed and recorded.
Before starting the test, the test center is required to provide a face cleaning product without any efficacy for the test part of the subject to be cleaned; clean skin is detected after sitting still in a satisfactory room for at least 30 minutes. For each return visit, the elements such as cleaning products and water temperature should be kept consistent.
Skin basal values before product use were assessed and tested, including instrument testing and standard image capture and data recorded, on the qualified subjects in the group. The same test was again performed on days 7 and 14 after the use of the product.
(4) Calculation result
Descriptive statistics: descriptive statistics are performed on the measured values of each test area, including number, mean, standard deviation, minimum, median, maximum.
Differential analysis: the rates of change of the TEWL averages on the sample application side and control side were calculated separately, and then the differences between the spot application area and control area at different measurement times were statistically analyzed using the rates of change. If the test data are normally distributed, adopting a t-test method to carry out statistical analysis; if the test data are in non-normal distribution, a rank sum test method is adopted for statistical analysis. The statistical methods all use a two-tailed test with a test level α=0.05.
(5) Result determination
Positive results: compared with the control side, the change rate of the TEWL value of the sample smearing side is significantly different, and P is smaller than 0.05, so that the tested sample has an effect of promoting the recovery of the transepidermal water loss of the barrier damaged skin.
Negative results: compared with the control side, the change rate of the TEWL value of the sample smearing side is not significantly different by more than 0.05, which indicates that the tested sample has no promoting effect on the recovery of the transepidermal water loss of the barrier damaged skin.
TABLE 2 TEWL value for the percutaneous Water loss of test samples at various times
The results of the experiment in Table 2 were statistically analyzed, and the comparative examples and examples were all reparative to different extents, but the rate of change of the TEWL value of the transepidermal water loss was much higher for all the examples of the experimental group than for all the comparative examples of the experimental group. Example 16 showed a 11% decrease in the trans-epidermal water loss value after 7 days of use, compared to before the sample, with a very significant difference, indicating that the composition had a repair effect after 7 days of use. While the comparative examples of the experimental groups had a certain reduction rate, the reduction degree did not have a significant difference. The control group data also show that the repairing effect is almost not found, and the repairing effect of all the examples of the experimental group is better than that of the comparative example, so that the repairing effect is greatly enhanced by the synergistic combination of various substances in the composition.
3. Test of soothing efficacy
The method is carried out by referring to the clinical evaluation standard method of the T/CNMIA 0015-2020 sensitivity-relieving functional skin care product.
1. Lactic acid stinging test
(1) Test sample: the compositions of examples 1 to 16, comparative examples 1 to 6 were added to a conventional emulsion external-use matrix to prepare test samples, in which a stable cleaning composition having a repairing and soothing effect accounted for 5% of the total mass of the emulsion.
(2) The testing method comprises the following steps: lactic acid stinging sensitive muscle screening: 50 μl of 10% lactic acid solution was dropped on a filter paper with a diameter of 8mm, placed in the nasal labial sulcus of both sides of the subject, and the degree of discomfort of itching, tingling, burning pain of the test site was evaluated by the subject at 30S, 2.5min and 5min, respectively. Scoring was performed by a score of 4 (no sensation at 0, mild at 1, moderate at 2, and heavy at 3). And the sum of the stinging scores of 2.5min and 5min is more than or equal to 3, and the stinging scores are positive to the lactic acid stinging. Subjects were equally divided into 22 groups (5 persons each) based on the lactic acid stinging screening results, each group being smeared with a different test sample. The test product was used 1 time a day in the morning and evening during the run of the cumulative trial. And the lactic acid stinging screening score is taken as a basic value, the lactic acid stinging test is carried out again after the product is used for 28 days, the SPSS is utilized to carry out statistical analysis on the lactic acid stinging score before and after the product is used, and the difference of P <0.05 is statistically significant.
TABLE 3 lactic acid stinging scores of test samples at various times
/>
As can be seen from the experimental results in table 3, examples 1 to 16 achieve the expected effect, can slow down the stimulation of lactic acid to skin, and through Wilcoxon pairing rank sum test, the test level α=0.05, p <0.001 is obtained, which indicates that the lactic acid stinging score of the subjects who continuously use the test samples of examples 1 to 16 for 28 days has a significant difference from the base value, and the stinging degree is greatly reduced; while the comparative examples 1 to 6 were improved over the prior art, the expected effect was not achieved, and the effect was only half that of the examples, indicating that the comparative examples may still be sensitive to a small proportion.
2. Hyaluronidase inhibition assay
Hyaluronidase is hydrolase for degrading hyaluronic acid, is a main component of extracellular matrix of connective tissue of an organism and is related to most allergic reactions, so that the in vitro inhibition experiment of the hyaluronidase can reflect the effects of products on relieving skin, resisting allergy, resisting inflammation and the like to a certain extent; hyaluronic acid is a higher polysaccharide composed of D-glucuronic acid and N-acetylglucosamine, and can undergo a color reaction according to the Elson-morgan reaction. The inhibition rate of the hyaluronidase of the sample can be determined by the reaction; the larger the inhibition rate is, the better the soothing, anti-sensitization or sensitization inhibition effect is.
(1) Test sample: the test samples were the samples of examples 1 to 16 and comparative examples 1 to 6, and the concentrations were 0.5%, 4% and 8%, respectively.
(2) The testing method comprises the following steps: the test was performed according to ELSON-MORGAN improvement.
Table 4 samples were assayed for hyaluronidase inhibition at various concentrations (unit:%)
/>
As can be seen from the experimental results of Table 4, the results of the hyaluronidase inhibition rates of examples 1 to 16 are significantly higher than those of comparative examples 1 to 6, indicating that the soothing, antiallergic or sensitization inhibition effects of examples 1 to 16 are better than those of comparative examples 1 to 6. Although the groups of comparative examples 1 to 6 also had a certain effect of relieving allergy, the effect of relieving was not remarkable, and far from the examples, the highest inhibition ratio of comparative example was 9.48% at 8% concentration, and the difference of example was 75.17%, which was almost 8-fold. The synergistic effect of the components and the content of the components is obvious and obvious.
3. Visia image analysis method
(2) Subject selection: subjects with lactic acid stinging pain
(3) Measurement procedure
The visual photographs of D0 before using the sample were taken first, and then the visual photographs of D7 and D14 were taken at 7 and 14 days, respectively. The visual facial red area picture was analyzed using Image-Pro Plus 7.0C to yield red area a values as shown in Table 5.
Table 5 test samples for facial red area a values at different times
/>
Statistical analysis of the experimental results of table 5, comparing the a values of examples 1-16 with 0 days for 7 days, 14 days, was significantly reduced with significant differences, P <0.001. The compositions of examples 1 to 16 are shown to have an effect of promoting recovery of facial inflammation. The recovery effect of comparative examples 1 to 6 on inflammation was not obvious, and the recovery effect of examples 1 to 16 on facial inflammation was significantly better than that of comparative examples 1 to 6, and the red area a value was reduced by 13% after 7 days of use of example 16 compared with that before the use of the sample, with a very significant difference, the sample had an obvious effect of relieving facial redness after 7 days of use, and the reduction rate had been reduced to 12.85 on day 14, and reached 34%. The effective relieving ability of the traditional Chinese medicine composition is proved, and the importance and the necessity of the synergistic effect of the components in solving the problem of skin inflammation are further demonstrated.
In summary, the invention is through the mutual cooperation and synergistic effect among several substances of caprylic acid/capric triglyceride, PEG-20 triglyceride triisostearate, olibanum resin extract, tripeptide-1 copper, camellia seed oil, jojoba seed oil, avocado oil, butter tree fruit fat, wild lacquer fruit wax and red rose seed oil, it can be seen from the results of the combination stability test, the maintenance efficacy test and the relief efficacy test that in the stability test, although the examples and the comparative examples are relatively stable, the stability of the examples is superior to that of the comparative examples; in the test of repairing efficacy and relieving efficacy, the effect of the composition is obviously better than that of the comparative example, the comparative example can be sensitive in a smaller proportion, and the effect of the example is enhanced by 8-10 times compared with the comparative example, so that the composition has obvious repairing and relieving effects.
