1408 parts of prepared rehmannia root, 704 parts of wine dogwood fruit, 528 parts of moutan bark, 704 parts of Chinese yam, 528 parts of tuckahoe and 528 parts of rhizoma alismatis;
the preparation method comprises the following steps:
s21, crushing 110 parts of poria cocos to 70 meshes, adding water to the sieved part and the rest poria cocos, decocting for three times, each time for 25min, wherein the water is added 7 times, 4 times and 3 times respectively, mixing the filtrates, and concentrating to obtain a poria cocos thick paste;
s22, adding ethanol into the dogwood fruit pulp, performing reflux extraction twice, wherein the amount of the ethanol used in each time is 15mL, the ethanol used in each time is 55min, filtering, keeping medicine residues for later use, combining the filtrates, and concentrating to obtain a thick paste of the dogwood fruit pulp;

s31, crushing 110 parts of poria cocos to 90 meshes, adding water into the sieved part and the rest poria cocos, decocting for three times, 35min each time, wherein the amount of the water added for the first time is 5 times that of the rest poria cocos, the amount of the water added for the second time is 5 times that of the rest poria cocos, and the amount of the water added for the third time is 4 times that of the rest poria cocos, combining filtrates, and concentrating to obtain a poria cocos thick paste;
s32, adding ethanol into the dogwood pulp, performing reflux extraction twice, wherein the amount of the ethanol used in each time is 3000mL, the ethanol used in each time is 65min, filtering, keeping medicine residues for later use, combining the filtrates, and concentrating to obtain a thick paste of the dogwood pulp;
s33, distilling the tree peony bark with water vapor, adding 0.01mL0.8mol/L hydrochloric acid solution into the collected distillate to crystallize the tree peony bark extract, and sequentially filtering, washing, drying and grinding the tree peony bark extract crystals, wherein the drying temperature is controlled at 60 ℃, and the grinding is performed, and the materials are crushed to 100 meshes to obtain tree peony bark extract fine powder;
s34, mixing the water solution obtained after distilling the tree peony bark with tree peony bark dregs, dogwood dregs, prepared rehmannia root, chinese yam and rhizoma alismatis, adding water, decocting for three times, mixing decoction filtrates obtained in the three times, filtering, passing through macroporous adsorption resin, eluting by using 80% ethanol, wherein the using amount of the 80% ethanol is 1.5 times of the column volume, collecting eluent, and concentrating to obtain composition thick paste;
s35, mixing the composition thick paste, the poria cocos thick paste and the soft paste of the dogwood fruit pulp, drying under reduced pressure, crushing, adding the fine powder of the moutan bark extract and auxiliary materials, and encapsulating to obtain the six-ingredient rehmannia capsules.
Comparative example 1
The raw material medicinal materials comprise:
1408 parts of prepared rehmannia root, 704 parts of wine dogwood fruit, 528 parts of tree peony bark, 704 parts of Chinese yam, 528 parts of tuckahoe and 528 parts of rhizoma alismatis;
the preparation method comprises the following steps:
s1, crushing 110 parts of poria cocos to 80 meshes, adding water into the sieved part and the rest poria cocos, decocting for three times, 30min each time, wherein the amount of the water added for the first time is 5 times that of the rest poria cocos, the amount of the water added for the second time is 3 times that of the rest poria cocos, and the amount of the water added for the third time is 3 times that of the rest poria cocos, combining the filtrates, and concentrating to obtain a poria cocos thick paste;
s2, adding ethanol into the dogwood fruit, performing reflux extraction twice, wherein the amount of the ethanol used in each time is 2800mL, the ethanol used in each time is 60min, filtering, reserving dregs for later use, combining the filtrates, and concentrating to obtain a dogwood fruit thick paste;
s3, distilling the tree peony bark with water vapor, adding 0.01mL1.0mol/L hydrochloric acid solution into the collected distillate to crystallize the tree peony bark extract, and sequentially filtering, washing, drying and grinding the obtained tree peony bark extract crystals, wherein the drying temperature is controlled to be 60 ℃, and the grinding is performed, and the materials are ground to 100 meshes to obtain tree peony bark extract fine powder;
s4, mixing the water solution obtained after distilling the tree peony bark with tree peony bark dregs, dogwood dregs, prepared rehmannia root, chinese yam and rhizoma alismatis, adding water, decocting for three times, mixing decoction filtrates obtained in the three times, filtering, passing through macroporous adsorption resin, eluting by using 70% ethanol, wherein the using amount of the 70% ethanol is 3 times of the column volume, collecting eluent, and concentrating to obtain composition thick paste;
and S5, mixing the composition thick paste, the poria cocos thick paste and the soft paste of the dogwood fruit pulp, drying under reduced pressure, crushing, adding the fine powder of the moutan bark extract and auxiliary materials, and encapsulating to obtain the six-ingredient rehmannia capsules.
The invention respectively carries out finished product inspection on the Liuwei Dihuang capsules prepared in examples 1-3, and the inspection reports are shown in figures 1-3.
As can be seen from the figure, the indexes of the Liuwei Dihuang capsules prepared in the embodiments 1-3 of the invention, such as the property, the content and the like, all meet the corresponding quality standards.
Meanwhile, in order to further study the extraction rate of the effective components in each crude drug by the preparation method of the present invention, the effective components in the liuweidihuang capsules prepared in examples 1-3 and the comparative example were tested, and the test results are shown in tables 1-3.

As can be seen from tables 1 to 4, the content of the active ingredient in the liuweidihuang capsule prepared in example 1 of the present invention is significantly increased compared to the control example, which indicates that the preparation method of the present invention improves the bioavailability of the bulk drug and the purity of the drug. In addition, in the process of refining the decoction filtrate by using macroporous adsorption resin, the filtrate is firstly filtered, so that the dosage of ethanol in the elution process can be effectively reduced, and further, the production energy consumption and the pollution to the environment are reduced.
In contrast, as can be seen from comparative examples 1 to 3, when the macroporous adsorption resin is eluted, a 70% ethanol solution is preferred, and when 60% ethanol is used for extraction, the problem that paeonol cannot be effectively eluted exists, while the use of 80% ethanol increases the use amount of ethanol, which increases the production energy consumption and the pollution degree to the environment.
Finally, it should be noted that: the above embodiments are only used to illustrate the technical solution of the present invention, and not to limit the same; while the invention has been described in detail and with reference to the foregoing embodiments, it will be understood by those skilled in the art that: the technical solutions described in the foregoing embodiments may still be modified, or some or all of the technical features may be equivalently replaced; and these modifications or substitutions do not depart from the spirit of the corresponding technical solutions of the embodiments of the present invention.
Claims (10)
1. A preparation method of a capsule of six ingredients with rehmannia is characterized by comprising the following steps:
s1, adding water into poria cocos, decocting, and concentrating to obtain poria cocos thick paste;
s2, adding ethanol into the wine dogwood pulp, performing reflux extraction, and concentrating to obtain a wine dogwood pulp thick paste;
s3, distilling the tree peony bark by using steam, and crystallizing and drying distillate in sequence to obtain tree peony bark extract fine powder;
s4, mixing the water solution obtained after distilling the tree peony bark with tree peony bark dregs, dogwood dregs, prepared rehmannia root, chinese yam and rhizoma alismatis, adding water, decocting for three times, mixing decoction filtrates obtained in the three times, filtering, passing through macroporous adsorption resin, eluting by using 60-80% ethanol, collecting eluent, and concentrating to obtain composition thick paste;
s5, mixing the composition thick paste, the poria cocos thick paste and the soft paste of the dogwood fruit pulp, drying under reduced pressure, crushing, adding the fine powder of the moutan bark extract and auxiliary materials, and encapsulating to obtain a six-ingredient rehmannia capsule;
preferably, in step S4, during the decoction, the amount of water added for the first time is 5-7 times, the amount of water added for the second time is 3-5 times, and the amount of water added for the third time is 3-5 times;
wherein, the steps S1, S2 and S3 are not limited in sequence.
2. The preparation method according to claim 1, wherein the Liuwei Dihuang capsule comprises the following raw medicinal materials in parts by weight: 1350-1450 parts of prepared rehmannia root, 680-720 parts of dogwood fruit wine, 500-550 parts of tree peony bark, 680-720 parts of Chinese yam, 500-550 parts of tuckahoe and 500-550 parts of rhizoma alismatis.
3. The preparation method according to claim 1, wherein step S1 specifically comprises: pulverizing 100-120 parts of Poria to 70-90 mesh, decocting the rest Poria and rest Poria in water for three times (each for 25-35 min), mixing filtrates, and concentrating to obtain Poria soft extract;
preferably, during the decoction, the water is added in an amount which is 5 to 7 times that of the first time, 3 to 5 times that of the second time and 3 to 5 times that of the third time.
4. The preparation method according to claim 1, wherein the step S2 specifically comprises: extracting Corni fructus with ethanol under reflux twice, each for 55-65min, filtering, collecting residues, mixing filtrates, and concentrating to obtain soft extract.
5. The method of claim 4, wherein the amount of ethanol required per gram of wine dogwood is 8-10mL.

The method according to claim 1, wherein step S3 specifically comprises: distilling cortex moutan with steam, adding hydrochloric acid solution into the distillate to crystallize cortex moutan extract, filtering, washing, drying, and grinding the obtained cortex moutan extract crystal to obtain cortex moutan extract fine powder;
wherein, during the drying, the temperature is controlled to be 55-65 ℃;
during grinding, the materials are crushed to 90-110 meshes.
7. The preparation method of claim 6, wherein the amount of the hydrochloric acid solution required per gram of moutan cortex bark is 0.008-0.012mL, and the concentration of the hydrochloric acid solution is 0.5-1.5mol/L.
8. The method according to claim 1, wherein in step S4, the 60 to 80% ethanol is used in an amount of 1.5 to 2.5 column volumes;
the model of the macroporous adsorption resin is HPD-100.
9. The method of claim 1, wherein the reduced pressure drying and pulverization in step S5 can be replaced by boiling granulation.
10. A capsule of six ingredients with rehmannia, prepared by the method according to any one of claims 1 to 9.
