Abstract
本发明公开了一种无糖型安神补脑液及其制备方法,属于中药制药技术领域。包括如下组分:鹿茸2.5~3.2g、干姜10.5~13g、制何首乌60~63.5、淫羊藿49.5~51g、大枣12~13g、甘草55~60g、维生素B1 12.5~16g、低糖型掩味剂10~30g、羟丙基‑β‑环糊精5~10g、纯水适量;所述安神补脑液各组分混合均匀后的体积为1000mL。本发明用低糖型掩味剂代替传统处方中的蔗糖成分,已以满足糖尿病、肥胖、冠心病、高脂血症等忌食糖分的患者需求。另外,本发明选用甜菊素和甘草提取物作为低糖型掩味剂,甜菊素稳定性比较高,不会其他组分发生拮抗作用;甘草提取物作为甘草提取物,属于药用成分之一,都不会不会影响安神口服液原有成分的药效。

技术领域
本发明属于中药制药技术领域,尤其是一种无糖型安神补脑液及其制备方法。
背景技术
安神补脑液是由鹿茸、制何首乌、淫羊藿、干姜、甘草、大枣共6味中药及维生素B为原料药制成的非处方药,具有生精补髓、益气养血、安神健脑之功效,用于神经衰弱、失眠、健忘、头痛等症。其中的关键成分维生素B,又称硫胺素或抗神经炎素,具有维持神经正常功能的作用,对该制剂的临床疗效起着重要的作用,其含量作为本品质量控制的指标之一,需要严格控制其含量,避免降解。
另外,由于产品中原有掩味剂为蔗糖,在临床使用中无法满足对糖代谢有障碍的患者(例如,糖尿病、肥胖症、高脂血症、冠心病等忌食糖份的患者)的需求,因此进一步局限了该药物的应用。
发明内容
为了克服上述技术缺陷,本发明提供一种无糖型安神补脑液及其制备方法,以解决背景技术所涉及的问题。
本发明提供一种无糖型安神补脑液及其制备方法,包括:
鹿茸2.5~3.2g、干姜10.5~13g、制何首乌60~63.5、淫羊藿49.5~51g、大枣12~13g、甘草55~60g、维生素B112.5~16g、低糖型掩味剂10~30g、羟丙基-β-环糊精5~10g、纯水适量;
所述安神补脑液各组分混合均匀后的体积为1000mL。
优选地或可选地,所述低糖型掩味剂包括:0.3~1.0g的甜菊素和9.7~29g甘草提取物。
优选地或可选地,所述低糖型掩味剂包括:0.3~1.0g的甜菊素、6.4~20g的甘草酸、3~9g的甘草酸钾盐。
本发明还提供一所述的无糖型安神补脑液的制备方法,其特征在于,包括如下步骤:
干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,合并煎液,离心后,用超滤柱进行超滤,滤液备用;
鹿茸加水煎煮五次,分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液用超滤柱进行超滤,滤液备用;
将维生素B1用羟丙基-β-环糊精进行包和,备用;
将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
优选地或可选地,所述低糖型掩味剂的制备方法包括如下步骤:
取0.3~1.0g的甜菊素、6.4~20g的甘草酸、3~9g的甘草酸钾盐加入纯水,完全溶解、并充分混匀,并于低沸状态下保持1h,即得低糖型掩味剂。
优选地或可选地,所述超滤柱包括无机陶瓷膜或/和聚酰胺柱。
优选地或可选地,所述无机陶瓷膜的滤膜孔径为10nm~100nm,过滤压力为0.2~0.35mPa,过滤温度为40~50℃,膜通量为50~300L/m2·h。
优选地或可选地,所述通过无机陶瓷膜后的药液继续以0.5~2mL/s的流速通过聚酰胺柱,并用1-5倍聚酰胺柱体积的纯水洗脱,收集药液,然后用1-5聚酰胺柱体积的0.05~0.2%NaOH溶液洗脱,收集洗脱液调PH至4-6,获得干姜药渣、制何首乌、淫羊藿、大枣、甘草的混合提取液。
优选地或可选地,所述干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;
所述鹿茸加水煎煮五次,五次煎煮时间依次为4、4、3、3、2小时。
本发明涉及一种无糖型安神补脑液及其制备方法,相较于现有技术,具有如下有益效果:
1、本发明采用低糖型掩味剂代替传统处方中的蔗糖成分,已以满足糖尿病、肥胖、冠心病、高脂血症等忌食糖分的患者需求。
2、本发明通过将选用甜菊素和甘草提取物,作为掩味剂,在产品入口和留口时,无苦味,口感适合与大多数人群。而且,甜菊素稳定性比较高,不会其他组分发生拮抗作用;甘草提取物作为甘草提取物,属于药用成分之一,也不会影响安神口服液原有成分的药效。
3、通过替代流量传统蔗糖,不尽产品澄清度提高,而且无发酵性,因而可延长甜菊制品保质期,因此无需或仅需要少量加入苯甲酸钠、苯甲酸、羟苯乙酯等防腐剂,在保证安神补脑液保质期的前提下,进一步提高产品的口感。
4、本发明中将维生素B1用羟丙基-β-环糊精进行包和,一方面起到掩味作用,减轻刺激性气味挥发,降低患者服用的不适感,另一方面增加产品的稳定性,避免维生素B1降解,延长产品的保质期。
5、本发明通过采用多种过滤方式相结合的方式,过滤药液,从而达到物质的分离、除杂和精制的目的,避免大颗粒物质和杂质在存储过程中,发生聚沉,影响产品保质期。
6、本发明通过选用三种掩味剂,当甜菊素、甘草提取物和羟丙基-β-环糊精配合使用时,掩味最佳,而且还能适当延长产品的保质期,降低对防腐剂的依赖。

实施例11
该安神补脑液包括如下重量的原料:鹿茸3.0g、干姜12.5g、制何首乌63.5、淫羊藿50.0g、大枣12.5g、甘草56.5g、维生素B113.5g、低糖型掩味剂24g、羟丙基-β-环糊精10g、纯水适量;
所述低糖型掩味剂包括:0.5g的甜菊素、17g的甘草酸、6.5g的甘草酸钾盐;
所述安神补脑液的制备方法包括如下步骤:
步骤1、干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;合并煎液,离心后,过0.45微米滤膜,滤液备用;
步骤2、鹿茸加水煎煮五次,五次煎煮时间依次为4、4、3、3、2小时。分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液备用;
步骤3、将维生素B1用羟丙基-β-环糊精进行包和,备用;
步骤4、将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
实施例12
该安神补脑液的原料组成和低糖型掩味剂的原料组成同实施例11。
所述安神补脑液的制备方法包括如下步骤:
步骤1、干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;合并煎液,离心后,过0.45微米滤膜,用无机陶瓷膜进行超滤,滤液备用;所述无机陶瓷膜的滤膜孔径为100nm,过滤压力为0.3mPa,过滤温度为45℃,膜通量为100L/m2·h。
步骤2、鹿茸加水煎煮五次,五次煎煮时间依次为4、4、3、3、2小时。分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液用超滤柱进行超滤,滤液备用;所述无机陶瓷膜的滤膜孔径为100nm,过滤压力为0.3mPa,过滤温度为45℃,膜通量为100L/m2·h。
步骤3、将维生素B1用羟丙基-β-环糊精进行包和,备用;
步骤4、将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
实施例13
该安神补脑液的原料组成和低糖型掩味剂的原料组成同实施例11。
所述安神补脑液的制备方法包括如下步骤:
步骤1、干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;合并煎液,离心后,过0.45微米滤膜,用无机陶瓷膜和聚酰胺柱进行超滤,滤液备用;所述无机陶瓷膜的滤膜孔径为100nm,过滤压力为0.3mPa,过滤温度为45℃,膜通量为100L/m2·h。通过无机陶瓷膜后的滤液继续以1mL/s的流速通过聚酰胺柱,并用2倍于聚酰胺柱体积的纯水洗脱,收集药液,然后用2倍于聚酰胺柱体积的0.1%NaOH溶液洗脱,收集洗脱液调PH至5,获得干姜药渣、制何首乌、淫羊藿、大枣、甘草的混合提取液。
步骤2、鹿茸加水煎煮五次,五次煎煮时间依次为4、4、3、3、2小时。分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液用超滤柱进行超滤,滤液备用;所述无机陶瓷膜的滤膜孔径为100nm,过滤压力为0.3mPa,过滤温度为45℃,膜通量为100L/m2·h。
步骤3、将维生素B1用羟丙基-β-环糊精进行包和,备用;
步骤4、将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
实施例14
该安神补脑液的原料组成和低糖型掩味剂的原料组成同实施例11。
所述安神补脑液的制备方法包括如下步骤:
步骤1、干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;合并煎液,离心后,过0.45微米滤膜,用聚酰胺柱进行超滤,滤液备用;以1mL/s的流速通过聚酰胺柱,并用2倍于聚酰胺柱体积的纯水洗脱,收集药液,然后用2倍于聚酰胺柱体积的0.1%NaOH溶液洗脱,收集洗脱液调PH至5,获得干姜药渣、制何首乌、淫羊藿、大枣、甘草的混合提取液。
步骤2、鹿茸加水煎煮五次,五次煎煮时间依次为4、4、3、3、2小时。分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液用超滤柱进行超滤,滤液备用;所述无机陶瓷膜的滤膜孔径为100nm,过滤压力为0.3mPa,过滤温度为45℃,膜通量为100L/m2·h。
步骤3、将维生素B1用羟丙基-β-环糊精进行包和,备用;
步骤4、将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
实施例21~26
该安神补脑液包括如下重量的原料:鹿茸3.0g、干姜12.5g、制何首乌63.5、淫羊藿50.0g、大枣12.5g、甘草56.5g、维生素B113.5g、低糖型掩味剂24g、纯水适量;
所述安神补脑液的制备方法同实施例11。
各实施例中低糖型掩味剂的配方如下:
对比例1
该安神补脑液包括如下重量的原料:鹿茸3.0g、干姜12.5g、制何首乌63.5、淫羊藿50.0g、大枣12.5g、甘草56.5g、维生素B113.5g、低糖型掩味剂65.5g、纯水适量;
该安神补脑液的制备方法,包括如下步骤:干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时,合并煎液,滤液浓缩至浸膏,加3倍量乙醇,静止24小时后,滤过,滤液备用。
鹿茸加水煎煮五次,期间依次为4、4、3、3、2小时,分次滤过,合并滤液浓缩,加蜂蜡,煮沸静止至蜡层完全凝固,除去蜡层,抽滤,加乙醇,使含醇量为80%,室温静止48小时,滤过,滤液回收乙醇浓缩至适量,加乙醇,使含醇量为75%,室温静止24小时,滤过。滤液回收乙醇,加水和乙醇调节浓度(含醇量为20-30%)。
将上述药液、鹿茸提取液及单糖浆混匀,加入干姜挥发油,加维生素B1。搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
检测例1
常规外观检测:针对实施例11至实施例14、对比例1进行常规的外观检测、维生素B1含量检测,具体的检测结果参见下表:
讨论:本发明通过采用多种过滤方式相结合的方式,过滤药液,能够避免安神补脑液中的大颗粒物质和杂质在存储过程中,发生聚沉;另外,由于对比例1中并未加入防腐剂和羟丙基-β-环糊精,3个月后维生素B1含量变化较大,也侧面说明B1用羟丙基-β-环糊精进行包和,能够增加产品的稳定性,避免维生素B1降解,延长产品的保质期。
检测例2
本检测例采用随机、单盲法对样品进行测试,测试邀请25名志愿者(男性10名,女性15名)者为实施例11、实施例21至实施例27所制备的安神补脑液进行气味、口感等项目的评价。志愿者口尝供试品时,根据自己的口尝感受,按照从不苦到苦的顺序给供试品排序。
气味评价标准:Ⅴ级香味合适,有愉悦感;Ⅳ级香味过多,可接受;Ⅲ级没有香味,也没有其他异味,可接受;Ⅱ级淡淡的香味,夹杂其他异味,勉强接收;Ⅰ级有特殊臭味,难以接受。
味道评价标准:Ⅴ级味道很好,入口甜,无其他异味;Ⅳ级入口过甜,或有微弱的异味或苦味,可接受;Ⅲ级入口甜,有较明显的异味或苦味,可接受;Ⅱ级入口微甜或者不甜,有明显的异味或苦味,体验感差,仍可接受;Ⅰ级异味或苦味非常明显,恶心,几乎不能接受。

讨论:实施例21至实施例23的味道等级分别为Ⅱ级,Ⅱ级和Ⅰ级,均能获得了一定的掩味效果,实施例24至实施例26的味道等级分别为Ⅲ~Ⅳ级、Ⅲ级、Ⅱ~Ⅲ级,当两种掩味组分共同作用时,产品的苦味受到了比较好的掩盖,部分组合方式甚至接近于普通的蔗糖型安神补脑液的口感;当三种掩味剂,当甜菊素、甘草提取物和羟丙基-β-环糊精配合使用时,掩味最佳,大多数志愿者口感给出的评价甚至高于蔗糖类安神口服液的口感。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合。为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。

1.一种无糖型安神补脑液,其特征在于,包括如下组分:
鹿茸2.5~3.2g、干姜10.5~13g、制何首乌60~63.5、淫羊藿49.5~51g、大枣12~13g、甘草55~60g、维生素B112.5~16g、低糖型掩味剂10~30g、羟丙基-β-环糊精5~10g、纯水适量;
所述安神补脑液各组分混合均匀后的体积为1000mL。
2.根据权利要求1所述的无糖型安神补脑液,其特征在于,所述低糖型掩味剂包括:0.3~1.0g的甜菊素和9.7~29g甘草提取物。
3.根据权利要求2所述的无糖型安神补脑液,其特征在于,所述低糖型掩味剂包括:0.3~1.0g的甜菊素、6.4~20g的甘草酸、3~9g的甘草酸钾盐。
4.一种基于权利要求1至3任一项所述的无糖型安神补脑液的制备方法,其特征在于,包括如下步骤:
干姜提取挥发油,并将干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,合并煎液,离心后,过0.45微米滤膜,用超滤柱进行超滤,滤液备用;
鹿茸加水煎煮五次,分次过滤,合并滤液,离心后,过0.45微米滤膜,滤液用超滤柱进行超滤,滤液备用;
将维生素B1用羟丙基-β-环糊精进行包和,备用;
将上述药液、鹿茸提取液及低糖型掩味剂、混匀,加入干姜挥发油,加维生素B1、搅拌10分钟,静止滤过,加纯化水调整总量至1000ml。
5.根据权利要求4所述的无糖型安神补脑液的制备方法,其特征在于,所述低糖型掩味剂的制备方法包括如下步骤:
取0.3~1.0g的甜菊素、6.4~20g的甘草酸、3~9g的甘草酸钾盐加入纯水,完全溶解、并充分混匀,并于低沸状态下保持1h,即得低糖型掩味剂。
6.根据权利要求4所述的无糖型安神补脑液的制备方法,其特征在于,所述超滤柱包括无机陶瓷膜或/和聚酰胺柱。
7.根据权利要求6所述的无糖型安神补脑液的制备方法,其特征在于,所述无机陶瓷膜的滤膜孔径为10nm~100nm,过滤压力为0.2~0.35mPa,过滤温度为40~50℃,膜通量为50~300L/m2·h。
8.根据权利要求7所述的无糖型安神补脑液的制备方法,其特征在于,通过无机陶瓷膜后的滤液继续以0.5~2mL/s的流速通过聚酰胺柱,并用1-5倍聚酰胺柱体积的纯水洗脱,收集药液,然后用1-5聚酰胺柱体积的0.05~0.2%NaOH溶液洗脱,收集洗脱液调PH至4-6,获得干姜药渣、制何首乌、淫羊藿、大枣、甘草的混合提取液。
9.根据权利要求4所述的无糖型安神补脑液的制备方法,其特征在于,所述干姜药渣与制何首乌、淫羊藿、大枣、甘草加水煎煮三次,其中,第一次煎煮时间为2小时,第二次、第三次煎煮时间各1小时;
