Abstract
本发明涉及一种养阴生津、清热利咽金果饮及其制备方法,包括以下步骤:①陈皮单提,采用如下提取方法:陈皮加12倍量1.0‑2.0%碳酸钠溶液煎煮两次,每次20‑30分钟,收集合并煎液,加10‑30%氯化钙溶液,搅拌均匀,离心,离心液浓缩得陈皮清膏;②采用碟式离心技术,将麦冬、胖大海、南沙参、太子参等水提浓缩清膏离心;③采用碟式离心技术,将地黄、玄参、西青果、蝉蜕等醇沉上清液离心;④采用高速离心技术将麦冬等清膏、陈皮单提膏及地黄等清膏的合并液离心。本发明进一步提供一种改善口服液澄明度技术,配方如下:配料后加入稳定剂EDTA‑2Na及过滤采用过滤纸板及微孔滤膜组合高级过滤技术进行过滤除杂技术改善产品的澄明度,包装,即得。

技术领域
本发明属中药领域,本发明涉及一种养阴生津、清热利咽金果饮及其制备方法。
背景技术
金果饮由地黄、玄参、西青果、蝉蜕、麦冬、胖大海、南沙参、太子参、陈皮等多种中药材组成,具有养阴生津、清热利咽的功效。用于肺热阴伤所致的咽部红肿、咽痛、口干咽燥。处方中地黄来源于玄参科植物地黄Rehmannia glutinosa Libosch.的新鲜或干燥块根,具有清热凉血、养阴生津的功效,用于热入营血,温毒发斑,吐血衄血,热病伤阴,舌绛烦渴,津伤便秘,阴虚发热,骨蒸劳热,内热消渴;玄参来源于玄参科植物玄参Scrophularianingpoensis Hemsl.的干燥根,具有清热凉血,滋阴降火,解毒散结的功效,用于热入营血,温毒发斑,热病伤阴,舌绛烦渴,津伤便秘,骨蒸劳嗽,目赤,咽痛,白喉,瘰疬,痈肿疮毒;西青果来源于使君子科植物诃子Terminalia chebula Retz.的干燥幼果,具有清热生津、解毒的功效,用于阴虚白喉;蝉蜕来源于蝉科昆虫黑蚱CryptotympanapustulataFabricius的若虫羽化时脱落的皮壳,具有疏散风热、利咽、透疹、明目退翳、解痉的功效,用于风热感冒,咽痛音哑,麻疹不透,风瘆瘙痒,目赤翳障,惊风抽搐,破伤风;麦冬来源于百合科植物麦冬Ophiopogon japonicus(L.f)Ker-GawL的干燥块根,具有养阴生津、润肺清心的功效,用于肺燥干咳,阴虚痨嗽,喉痹咽痛,津伤口渴,内热消渴,心烦失眠,肠燥便秘;胖大海来源于梧桐科植物胖大海Sterculia lychnophora Hance的干燥成熟种子,具有清热润肺、利咽开音、润肠通便的功效,用于肺热声哑,干咳无痰,咽喉干痛,热结便闭,头痛目赤;南沙参来源于桔梗科植物轮叶沙参Adenophora tetraphylla(Thunb.)Fisch或沙参Adenophorastricta Miq.的干燥根,具有养阴清肺、益胃生津、化痰、益气的功效,用于肺热燥咳,阴虚劳嗽,干咳痰黏,胃阴不足,食少呕吐,气阴不足,烦热口干;太子参来源于石竹科植物孩儿参Pseudostellaria heterophylla(Miq.)Pax ex Pax et Hoffm.的干燥块根,具有益气健脾、生津润肺的功效,用于脾虚体倦,食欲不振,病后虚弱,气阴不足,自汗口渴,肺燥干咳;陈皮来源于芸香科植物橘Citrus reticulata Blanco及其栽培变种的干燥成熟果皮,具有理气健脾、燥湿化痰的功效,用于脘腹胀满,食少吐泻,咳嗽痰多。诸药合用,具有养阴生津、清热利咽之功效,是一副著名经验用方,具有疗效确切、无明显副作用和应用方便等优点。
金果饮配方和制备方法收载于《中国药典》2020年版一部。配方如下:地黄73g 玄参55g 西青果18g 蝉蜕27g 麦冬55g 胖大海18g 南沙参55g 太子参55g 陈皮36g 薄荷素油0.5ml。
现有的提取及制备方法如下:
以上九味地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅拌30分钟,静置24小时,取上清液滤过,滤液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用。麦冬、胖大海、南沙参、太子参、陈皮加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,静置24小时,取上清液滤过,滤液与上述清膏合并,加蔗糖200g,滤过,滤液加薄荷素油、枸橼酸1g、苯甲酸钠2g,搅拌30分钟,即得。
因现有的提取及制备方法采用传统的过滤工艺,致使生产效率偏低,且纯化程度低,致使成品澄明度不好;又现有的提取方法橙皮苷含量偏低,游走于法定标准线。故此本发明充分应用现代科学技术和手段对其进行改进,对制备方法进行设计和筛选。
发明内容
本发明的目的在于提供一种养阴生津、清热利咽的金果饮口服液的制备方法。本发明通过优化提取纯化工艺,使制剂成型更好且产品澄明度有较大改善。
本发明所述金果饮的配方如下:
地黄73g 玄参55g 西青果18g 蝉蜕27g 麦冬55g 胖大海18g 南沙参55g 太子参55g 陈皮36g 薄荷素油0.5ml,
制备方法,包括以下步骤:
(1)地黄、玄参、西青果、蝉蜕加4-8倍量水煎煮1-3次,滤过,滤液浓缩得清膏,加乙醇搅拌,静置,取上清液离心,离心液减压浓缩至得清膏,备用;
(2)麦冬、胖大海、南沙参、太子参加10-15倍量水煎煮1-3次,滤过,合并滤液,浓缩得清膏,离心,离心液备用;
(3)陈皮加12倍量1.0-2.0%碳酸钠溶液煎煮两次,每次20-30分钟,收集合并煎液,加10-30%氯化钙溶液,搅拌均匀,离心,离心液浓缩得陈皮清膏。
(4)陈皮清膏与上述两种清膏合并,加适量水离心,离心液加薄荷素油、以及辅料搅匀,加水至1000ml,灌封,灭菌,即得。
其中,陈皮提取步骤如下:陈皮加12倍量1-2%碳酸钠溶液煎煮2次,每次20-30分钟,收集合并煎液,加10-30%氯化钙溶液,搅拌均匀,离心,离心液浓缩得陈皮清膏。
其中,还包括配料后加入稳定剂0.1‰-0.30‰EDTA-2Na及过滤采用过滤纸板及微孔滤膜组合技术进行过滤除杂的步骤。
最优选的,本发明的制备方法,包括以下步骤:
(1)地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅匀,静置24小时,离心,离心液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用;
(2)麦冬、胖大海、南沙参、太子参等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,离心,离心液备用;
(3)陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩至相对密度为1.18(70℃测)的清膏,备用;
(4)陈皮清膏与另外两种清膏合并,加适量水离心,离心液加蔗糖200g,再加入0.1-0.3‰EDTA-2Na,加热煮沸30分钟,搅匀,通过0.65um微孔滤膜加纸板的板框过滤机过滤,滤液加薄荷素油0.5ml、枸橼酸lg、苯甲酸钠2g,搅匀,加水至1000ml,灌封,灭菌,即得。
采用本发明所述的方法及原料制成的口服液,与原工艺相比,本发明:①陈皮单提,采用如下提取方法:陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩至相对密度为1.18(70℃测)的清膏;②采用碟式离心技术,将麦冬、胖大海、南沙参、太子参等水提浓缩清膏离心;③采用碟式离心技术,将地黄、玄参、西青果、蝉蜕等醇沉上清液离心;④采用高速离心技术将麦冬等清膏、陈皮单提膏与地黄等清膏的合并液离心;⑤配料时加入稳定剂,配制后的药液通过0.65um微孔滤膜加纸板的板框过滤机过滤。这样不仅能除去液体中较小固体粒子,而且可截留多糖、蛋白质等大分子物质,且具有一次过滤完全,节省过滤时间,便于操作,且过滤效果更佳的优点。本法继承了现有处方的优势,保留了全方的有效成分,提高了药物的质量和稳定性,降低了生产成本,保证了药物的疗效;
下述实验例和实施例用于进一步说明但不限于本发明。
1、提取纯化方法的选择
中药口服液原料药材品种多,来源复杂,故提取液是多种成分的混合,既含有所需的有效成分,也含有一些高分子物质,如鞣质、淀粉、蛋白质、树胶、粘液质、多糖、色素等,按照传统的提取纯化方法提取配料后产品的质量不稳定,成品橙皮苷含量时高时低,且在生产贮存过程中经常出现沉淀,出现消费者投诉的情况。碟片式离心技术是一种高效的新型分离技术,与传统的过滤方法如板框过滤、布袋过滤等相比,其工艺简单,操作方便,分离效果好,分离时间短,是当前中药药液纯化分离的一个发展方向,而且产品不产生有害物质污染环境,保证了100%的纯天然性,符合当今寻找各开发节能环保的“中药开发绿色技术”的潮流。
拟采用以下三种提取纯化方法进行研究:
(1)以上九味地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅拌30分钟,静置24小时,取上清液离心,离心液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用;麦冬、胖大海、南沙参、太子参等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,趁热(药液温度不低于80℃)离心,收膏;陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩至相对密度为1.18(70℃测)的清膏;麦冬等清膏、陈皮单提膏与地黄等清膏合并;
(2)以上九味地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅拌30分钟,静置24小时,取上清液离心,离心液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用。麦冬、胖大海、南沙参、太子参、陈皮等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,趁热(药液温度不低于80℃)离心,离心液与上述清膏合并;
(3)以上九味地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅拌30分钟,静置24小时,取上清液滤过,滤液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用。麦冬、胖大海、南沙参、太子参、陈皮等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,静置24小时,滤过,滤液与上述清膏合并。考察结果如下:

结论:根据以上三种提取纯化方法的考察结果,可以看出方法一(①陈皮单提,采用如下提取方法:陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩至相对密度为1.18(70℃测)的清膏;②采用碟式离心技术,将麦冬、胖大海、南沙参、太子参等水提浓缩清膏离心;③采用碟式离心技术,将地黄、玄参、西青果、蝉蜕等醇沉上清液离心)的样品更为理想,地黄等清膏的沉淀量率方法一、方法二明显优于方法三,麦冬等清膏的沉淀量率方法一、方法二明显优于方法三,混合膏橙皮苷含量方法一优于方法二、方法三。故优选方法一作为本发明的方法。
2、过滤工艺优化
在本发明的制备方法中,所述过滤选自:微孔滤膜加过滤纸板过滤,其筛选过程如下:
2.1过滤介质的考察
使用板框压滤机进行过滤,并对微孔滤膜、帆布及过滤纸板三种介质进行考察。
将按配好待过滤的金果饮,平均分成三份(编号1,2,3),分别通过以0.65μm微孔滤膜、帆布、过滤纸板为过滤介质的板框压滤机过滤,考察药液过滤后液体的澄明度、鉴别、PH值、过滤的难易程度,结果见表2。(说明:澄明度的考察为样品经离心机4000转/min,离心10min后观察试管底部的沉淀情况。过滤难易程度的考察指标以过滤时板框压滤机的压力来判定,当过滤压力低于0.2MPa为容易过滤,在0.2MPa-0.25MPa时为较难过滤,超过0.25MPa为很难过滤。)

结果表明经微孔滤膜过滤后药液的澄明度均较好,但是比较难过滤,而较易过滤的过滤纸板和帆布过滤后的药液澄明度较差。各种过滤介质对药液的质量情况没有影响。
2.2微孔滤膜孔径的筛选
针对以上这种情况我们考虑将经过过滤纸板的药液平均分成两份(编号:4,5)分别通过以0.45μm微孔滤膜、0.65μm微孔滤膜为介质的过滤板框压滤机再次过滤,考察药液过滤后液体的澄明度、PH值、过滤的难易程度,结果见表3。(说明:澄明度的考察为样品经离心机4000转/min,离心10min后观察试管底部的沉淀情况。过滤难易程度的考察指标以过滤时板框压滤机的压力来判定,当过滤压力低于0.2MPa为容易过滤,在0.2MPa-0.25MPa时为较难过滤,超过0.25MPa为很难过滤。)

结果表明用过滤纸板过滤后再经微孔滤膜过滤后药液的澄明度均较好,而通过0.65μm的微孔滤膜过滤比较容易。
2.3组合过滤技术考察
由于过滤纸板及微孔滤膜均适合同一板框压滤机,因此我们考虑将微孔滤膜与纸板组合,将药液一次通过两种介质过滤,以期达到过滤速度及澄明度均好的效果。
将金果饮,通过以0.65μm微孔滤膜加过滤纸板为过滤介质的板框压滤机过滤,考察用先通过微孔滤膜加过滤纸板过滤效果(编号6)与先通过纸板再通过微孔滤膜的过滤效果(编号7)的药液液体的澄明度、PH值、过滤的难易程度,并将结果进行比较,结果见表4。(说明:澄明度的考察为样品经离心机4000转/min,离心10min后观察试管底部的沉淀情况。过滤难易程度的考察指标以过滤时板框压滤机的压力来判定,当过滤压力低于0.2MPa为容易过滤,在0.2MPa-0.25MPa时为较难过滤,超过0.25MPa为很难过滤。)

结果表明用微孔滤膜加过滤纸板过滤效果与先通过纸板再通过微孔滤膜两次过滤的效果无差别,该技术可行。
3、稳定剂用量考察
EDTA-2Na是强效螯合剂,防腐剂和抗氧化剂,能与碱金属以外的绝大多数大分子物质生成的络合物。EDTA-2Na的用量进行筛选,见表5

从上表的结果可以看出,加入极少量的EDTA-2Na对孕金果饮澄明度有明显澄清效果,当加入的量在0.1‰~0.30‰澄明度效果最好而且药液最澄清,随着量的增大,颜色变浅。
和现有技术相比本发明的优点如下

2、【制法】以上九味地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅拌30分钟,静置24小时,取上清液离心,离心液减压浓缩至相对密度为1.13~1.15(80℃)的清膏,备用;麦冬、胖大海、南沙参、太子参等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩至相对密度为1.03~1.08(80℃)的清膏,离心,离心液备用;陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩至相对密度为1.18(70℃测)的清膏;麦冬等清膏、陈皮单提膏与地黄等清膏合并,加适量水离心,离心液加蔗糖200g,再加入0.2‰EDTA-2Na,加热煮沸30分钟,搅匀,通过0.65um微孔滤膜加纸板的板框过滤机过滤,滤液加薄荷素油0.5ml、枸橼酸lg、苯甲酸钠2g,搅匀,加水至1000ml,灌封,灭菌,即得。

Claims (6)
1.一种养阴生津、清热利咽的金果饮口服液的制备方法,其特征在于,包括以下步骤:
配方:
地黄73g玄参55g西青果18g蝉蜕27g麦冬55g胖大海18g南沙参55g太子参55g陈皮36g薄荷素油0.5ml;
制备方法:
(1)地黄、玄参、西青果、蝉蜕加4-8倍量水煎煮1-3次,滤过,滤液浓缩得清膏,加乙醇搅拌,静置,取上清液离心,离心液减压浓缩至得清膏,备用;
(2)麦冬、胖大海、南沙参、太子参加10-15倍量水煎煮1-3次,滤过,合并滤液,浓缩得清膏,离心,离心液备用;
(3)陈皮加12倍量1.0-2.0%碳酸钠溶液煎煮两次,每次20-30分钟,收集合并煎液,加10-30%氯化钙溶液,搅拌均匀,离心,离心液浓缩得陈皮清膏;
(4)陈皮清膏与上述两种清膏合并,加薄荷素油、以及辅料搅匀,加水至1000ml,灌封,灭菌,即得。
2.根据权利要求1所述的制备方法,其特征在于,其中,陈皮提取步骤如下:陈皮加12倍量1-2%碳酸钠溶液煎煮2次,每次20-30分钟,收集合并煎液,加10-30%氯化钙溶液,搅拌均匀,离心,离心液浓缩得陈皮清膏。
3.根据权利要求1所述的制备方法,其特征在于,还包括配料后加入稳定剂0.1‰-0.30‰EDTA-2Na及过滤采用过滤纸板及微孔滤膜组合技术进行过滤除杂的步骤。
4.根据权利要求1所述的制备方法,其特征在于,包括以下步骤:
(1)地黄、玄参、西青果、蝉蜕加6倍量水煎煮二次,每次30分钟,滤过,滤液浓缩至相对密度为1.14~1.19(80℃)的清膏,加2倍量乙醇搅匀,静置24小时,离心,离心液减压浓缩得清膏,备用;
(2)麦冬、胖大海、南沙参、太子参等加12倍量水煎煮二次,第一次30分钟,第二次20分钟,滤过,合并滤液,浓缩得清膏,离心,离心液备用;
(3)陈皮加12倍量1.0%碳酸钠溶液煎煮两次,第一次30分钟,第二次20分钟,收集合并煎液,加20%氯化钙溶液57.5ml,搅拌均匀,离心,离心液浓缩得陈皮清膏,备用;
(4)将陈皮清膏与另外两种清膏合并,加适量水离心,离心液加蔗糖,再加入0.1‰EDTA-2Na,加热煮沸,搅匀,通过微孔滤膜加纸板的板框过滤机过滤,滤液加薄荷素油0.5ml、枸橼酸、苯甲酸钠,搅匀,加水至1000ml,灌封,灭菌,即得。
5.根据权利要求1所述的制备方法,其特征在于,包括以下步骤:

(4)陈皮清膏与另外两种清膏合并,加适量水离心,离心液加蔗糖200g,再加入0.3‰EDTA-2Na,加热煮沸30分钟,搅匀,通过0.65um微孔滤膜加纸板的板框过滤机过滤,滤液加薄荷素油0.5ml、枸橼酸lg、苯甲酸钠2g,搅匀,加水至1000ml,灌封,灭菌,即得。
