背景技术
全球经济的高速发展,促进了人们生活水平的不断提高和饮食结构变化,日常饮食摄入的营养物质不断增多,造成一部分人经常性的营养过剩,加之人口逐渐老龄化,导致了“富贵病”糖尿病数量迅速增加,糖尿病是一种由患者体内胰岛素不足,或机体对葡萄糖利用障碍引起的以糖代谢紊乱为主的内分泌代谢性疾病。已成为心血管疾病、肿瘤之后威胁人类健康的第三大疾病。
目前,糖尿病的治疗是以控制血糖消除症状为目的。虽然化学降糖药物的疗效非常肯定,但在改善机体对葡萄糖的利用方面都有局限性和不良反应,如长期使用会引起低血糖、肾功能降低、高血压等并发症及后期药物失效等。而糖尿病是慢性病,病程复杂,患者需要终生用药,西药的副作用对人体的损害因累积而难以克服。加上糖尿病由于体内血糖升高,中晚期将出现动脉硬化和周身广泛微血管病变,会引发多种慢性并发症如:糖尿病性神经病变、糖尿病性眼底病变、糖尿病合并高血压及高血脂症、肠胃功能紊乱、脑血管病、心血管病、肾病、神经末梢病变、糖尿病足等,给患者的健康造成了很大的威胁。因此,糖尿病的治疗应不仅在于降低血糖,纠正代谢紊乱,更重要的是减少糖尿病并发症和继发性西药失效。
白芸豆,其生物学名叫多花菜豆,因花色多样而得名。属豆科,蝶形花亚科,菜豆族菜豆属,白芸豆原产美洲的墨西哥和阿根廷,后经人工栽培驯化已适应冷凉潮湿的高原地带。种植面积较大的国家是美洲的阿根廷、美国、墨西哥,欧洲的英国,亚洲的中国、日本等。我国在16世纪末才开始引种栽培。现在中国的各个省区均有种植,面积较大的省份是云南、贵州、四川等,而云南的大理、丽江、兰坪种植面积较大。白芸豆营养丰富。据测,每百克芸豆含蛋白质23.1克、脂肪1.3克、碳水化合物56.9克、胡萝卜素0.24毫克、钙160毫克、磷410毫克、铁7.3毫克及丰富的B族维生素,鲜豆还含丰富的维生素C

本公开提供了一种白芸豆发酵提取物及其制备方法和降糖降压的用途,制备方法简单、原料来源广,制得的白芸豆发酵提取物具有很好的降血糖、降血压作用,安全无副作用,可以作用食疗产品、保健食品以及辅助和治疗药品的原料使用,具有广阔的应用前景。
本发明提供一种白芸豆发酵提取物的制备方法,包括将白云豆粉加入至含有魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌和米曲霉3.042的复合发酵菌液中发酵的步骤。
在一个实施方式中,在将白云豆粉加入复合发酵菌液之前进行了脱毒处理。示例性地,所述脱毒处理包括如下步骤:将白芸豆粉加入水中,煮沸第一时间段,过滤,豆渣加入乙醇-乙酸-水溶液中,继续煮沸第二时间段,过滤,干燥,得到脱毒白云豆粉
在一个实施方式中,在将白芸豆粉进行脱毒处理之前,对白芸豆进行预处理。示例性地,所述预处理包括将白芸豆洗净,干燥,打粉,过筛,得到白芸豆粉的步骤。
在一个实施方式中,复合发酵菌液的制备包括如下步骤:将魏斯氏菌(Weissellaconfusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于培养基中,活化培养,培养成菌种种子液,将三个种子液并稀释得到复合发酵菌液。
在一个实施方式中,培养基的制备包括如下步骤:将碳源、氮源、维生素、无机盐、水和中药提取物混合均匀,用PBS溶液调节其pH值为6.8-7.2,过滤,滤液用紫外线灭菌,得到培养基。
在一个实施方式中,白芸豆发酵提取物的制备方法还包括发酵后的提取步骤:将发酵后的产物进行过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
具体地,本发明提供一种白芸豆发酵提取物的制备方法,包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过筛,得到白芸豆粉;
S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,煮沸第一时间段,过滤,豆渣加入乙醇-乙酸-水溶液中,继续煮沸第二时间段,过滤,干燥,得到脱毒白云豆粉;
S3.培养基的制备:将碳源、氮源、维生素、无机盐、水和中药提取物混合均匀,用PBS溶液调节其pH值为6.8-7.2,过滤,滤液用紫外线灭菌,得到培养基;
S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,活化培养,培养成菌种种子液,将三个培养基混合并稀释得到复合发酵菌液,例如将三个培养基等体积混合;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,发酵,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
作为进一步的改进,步骤S1中所述过筛的筛网目数为150-200目。
作为进一步的改进,步骤S2中所述白芸豆和水的固液比为1:(3-5)g/mL;所述乙醇-乙酸-水溶液中乙醇、乙酸和水的体积比为(2-10):(5-12):50;所述豆渣与乙醇-乙酸-水溶液的固液比为1:(2-4)g/mL;所述第一时间段为15-30min;所述第二时间段为10-20min。
作为进一步的改进,步骤S3中所述碳源选自糖蜜、葡萄糖、麦芽糖、乳糖、蔗糖、果糖、淀粉中的至少一种;所述氮源选自氨水、尿素、铵盐、硝酸盐、氨基酸中的至少一种;所述维生素选自维生素C、维生素B1、维生素B2、维生素A、维生素K、维生素B12、维生素D、维生素E中的至少一种;所述无机盐选自氯化钠、氯化钾、氯化钙、硫酸镁、氯化铁、硫酸锌、硫酸铜、硫酸锰、氯化锌、氯化铜、氯化锰中的一种或几种混合;所述氨基酸选自甘氨酸、丝氨酸、苏氨酸、缬氨酸、色氨酸、亮氨酸、丙氨酸、半胱氨酸、甲硫氨酸、赖氨酸、异亮氨酸、苯丙氨酸中的一种或几种混合;所述碳源、氮源、维生素、无机盐、中药提取物和水的质量比为(10-20):(5-15):(0.1-0.5):(0.01-0.2):(10-20):100。
作为进一步的改进,所述中药提取物由以下原料按照重量份制备而成:罗布麻叶5-15份、野菊花5-10份、杜仲10-20份、黄芪5-10份、黄连2-5份。
作为进一步的改进,所述中药提取物的制备方法如下:将5-15重量份罗布麻叶、5-10重量份野菊花、10-20重量份杜仲、5-10重量份黄芪、2-5重量份黄连分别洗净,干燥,打粉,过100-200目筛,得到的中药粉,加入水中,加热煮沸,提取1-2h,过滤,重复提取一次,合并两次滤液,浓缩,干燥,得到中药提取物。
作为进一步的改进,步骤S4中所述活化培养的条件为35-40℃培养12-24h;所述菌种种子液的含菌量为107-108cfu/mL;所述稀释倍数为100-200倍;所述魏斯氏菌(Weissellaconfusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为2-4%、3-5%和1-3%。
作为进一步的改进,步骤S5中所述脱毒白云豆粉和复合发酵菌液的固液比为1:(5-10)g/mL;所述发酵的条件为35-40℃培养36-48h。
本发明进一步保护一种上述的制备方法制得的白芸豆发酵提取物。
本发明进一步保护一种上述的白芸豆发酵提取物在制备降血糖、降血压的食品、辅助或治疗降血糖、降血压的药品中的应用。
有益效果:本发明通过中药组合物包括罗布麻叶、野菊花、杜仲、黄芪、黄连进行提取得到的提取物加入培养基中,进行活化培养时,培养基内能产生大量的有益小分子营养物质,与后续加入白云豆粉进行混合后发酵后,能产生大量降血糖、降血压功能的营养组分,起到很好的协同降糖降压的效果;
本发明采用魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042对白芸豆粉进行发酵处理,不仅能有效降低白芸豆的抗营养因子以及有毒成分,破坏豆粉的植物细胞壁,使得其营养物质被大量溶出,还能够进一步发酵降解多肽、多糖等大分子应用物质,产生大量的氨基酸、短肽、寡糖、低聚糖以及小分子酸、酚、酯等营养物质,产生的复合营养物质能显著降低高血糖、高血压患者的血糖、血压,起到很好的药用功效。
本发明制备方法简单、原料来源广,制得的白芸豆发酵提取物具有很好的降血糖、降血压作用,安全无副作用,可以作用食疗产品、保健食品以及辅助和治疗药品的原料使用,具有广阔的应用前景。

实施例1
本实施例提供一种白芸豆发酵提取物的制备方法,包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过150目筛,得到白芸豆粉;
S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,固液比为1:3g/mL,煮沸15min,过滤,豆渣加入乙醇-乙酸-水溶液中,乙醇、乙酸和水的体积比为2:5:50,豆渣与乙醇-乙酸-水溶液的固液比为1:2g/mL,继续煮沸10min,过滤,干燥,得到脱毒白云豆粉;
S3.培养基的制备:将碳源、氮源、维生素、无机盐、水和中药提取物混合均匀,用PBS溶液调节其pH值为6.8,过滤,滤液用紫外线灭菌,得到培养基;所述碳源、氮源、维生素、无机盐、中药提取物和水的质量比为10:5:0.1:0.01:10:100;
中药提取物的制备方法如下:将5重量份罗布麻叶、5重量份野菊花、10重量份杜仲、5重量份黄芪、2重量份黄连分别洗净,干燥,打粉,过100目筛,得到的中药粉,加入水中,加热煮沸,提取1h,过滤,重复提取一次,合并两次滤液,浓缩,干燥,得到中药提取物;
S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为2%、3%和1%,35℃活化培养12h,培养成菌种种子液,含菌量为107cfu/mL,将三个培养基等体积混合并稀释100倍,得到复合发酵菌液;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,脱毒白云豆粉和复合发酵菌液的固液比为1:5g/mL,35℃发酵培养36h,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
实施例2
本实施例提供一种白芸豆发酵提取物的制备方法,包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过200目筛,得到白芸豆粉;
S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,固液比为1:5g/mL,煮沸30min,过滤,豆渣加入乙醇-乙酸-水溶液中,乙醇、乙酸和水的体积比为10:12:50,豆渣与乙醇-乙酸-水溶液的固液比为1:4g/mL,继续煮沸20min,过滤,干燥,得到脱毒白云豆粉;
S3.培养基的制备:将碳源、氮源、维生素、无机盐、水和中药提取物混合均匀,用PBS溶液调节培养基pH值为7.2,过滤,滤液用紫外线灭菌,得到培养基;所述碳源、氮源、维生素、无机盐、中药提取物和水的质量比为20:15:0.5:0.2:20:100;
中药提取物的制备方法如下:将15重量份罗布麻叶、10重量份野菊花、20重量份杜仲、10重量份黄芪、5重量份黄连分别洗净,干燥,打粉,过200目筛,得到的中药粉,加入水中,加热煮沸,提取2h,过滤,重复提取一次,合并两次滤液,浓缩,干燥,得到中药提取物;
S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为4%、5%和3%,40℃活化培养24h,培养成菌种种子液,含菌量为108cfu/mL,将三个培养基等体积混合并稀释200倍,得到复合发酵菌液;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,脱毒白云豆粉和复合发酵菌液的固液比为1:10g/mL,40℃发酵培养48h,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。

S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为3%、4%和2%,37℃活化培养18h,培养成菌种种子液,含菌量为108cfu/mL,将三个培养基等体积混合并稀释150倍,得到复合发酵菌液;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,脱毒白云豆粉和复合发酵菌液的固液比为1:7g/mL,37℃发酵培养42h,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
对比例1
与实施例3相比,步骤S4中未添加魏斯氏菌(Weissella confusa),其他条件均不改变。
包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过200目筛,得到白芸豆粉;
S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,固液比为1:4g/mL,煮沸22min,过滤,豆渣加入乙醇-乙酸-水溶液中,乙醇、乙酸和水的体积比为5:8:50,豆渣与乙醇-乙酸-水溶液的固液比为1:3g/mL,继续煮沸15min,过滤,干燥,得到脱毒白云豆粉;

包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过200目筛,得到白芸豆粉;

S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,固液比为1:4g/mL,煮沸22min,过滤,豆渣加入乙醇-乙酸-水溶液中,乙醇、乙酸和水的体积比为5:8:50,豆渣与乙醇-乙酸-水溶液的固液比为1:3g/mL,继续煮沸15min,过滤,干燥,得到脱毒白云豆粉;
S3.培养基的制备:将碳源、氮源、维生素、无机盐、水混合均匀,用PBS溶液调节培养基pH值为7,过滤,滤液用紫外线灭菌,得到培养基;所述碳源、氮源、维生素、无机盐和水的质量比为20:10:0.3:0.1:100;
S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为3%、4%和2%,37℃活化培养18h,培养成菌种种子液,含菌量为108cfu/mL,将三个培养基等体积混合并稀释150倍,得到复合发酵菌液;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,脱毒白云豆粉和复合发酵菌液的固液比为1:7g/mL,37℃发酵培养42h,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
测试例1小鼠降血糖研究
C57BL/6J小鼠10只(雌雄各半)作为对照组,将db/db小鼠48只(雌雄各半)按血糖值随机分为8组,分别为模型组、二甲双胍组、实施例1-3组、对比例1-3组。每天灌胃给药1次,实施例1-3组、对比例1-3组分别给予实施例1-3和对比例1-3制得的白芸豆发酵提取物,给药量20g·kg-1,对照组及模型组给予等量去离子水。连续给药12周后,在第13周给药后2h、24h测定小鼠的空腹血糖值,给药前,动物禁食4h。结果见表1。

由上表可知,本发明实施例1-3制得的白芸豆发酵提取物具有显著较好的降血糖的效果(图1和图2)。
测试例2
雄性Wistar大鼠10只,测量其血压值作为空白对照组,雄性SHR大鼠48只,测量其血压值,根据血压随机分为8组,分别为阴性对照组、阳性对照组、实施例1-3组、对比例1-3组。于分组当天开始给药,空白对照组和阴性对照组灌胃给予等量的生理盐水,阳性对照组灌胃给予氨氯地平混悬液(5mg/kg),实施例1-3组、对比例1-3组分别给予实施例1-3和对比例1-3制得的白芸豆发酵提取物,给药量20g·kg-1,每天给药1次,连续给药2周,每周测量动物血压1次。结果见表2。

由上表可知,本发明实施例1-3制得的白芸豆发酵提取物具有显著较好的降血压的效果。

Claims (13)
1.一种白芸豆发酵提取物的制备方法,其特征在于,包括将白云豆粉加入至含有魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌和米曲霉3.042的复合发酵菌液中发酵的步骤。
2.根据权利要求1所述的制备方法,其特征在于,在将白云豆粉加入复合发酵菌液之前进行了脱毒处理。
3.根据权利要求2所述的制备方法,其特征在于,在将白芸豆粉进行脱毒处理之前,对白芸豆进行预处理,例如,所述预处理包括将白芸豆洗净,干燥,打粉,过筛,得到白芸豆粉的步骤。
4.根据权利要求1所述的制备方法,其特征在于,包括以下步骤:
S1.白芸豆的处理:将白芸豆洗净,干燥,打粉,过筛,得到白芸豆粉;
S2.白芸豆粉的脱毒处理:将步骤S1制得的白芸豆粉加入水中,煮沸第一时间段,过滤,豆渣加入乙醇-乙酸-水溶液中,继续煮沸第二时间段,过滤,干燥,得到脱毒白云豆粉;
S3.培养基的制备:将碳源、氮源、维生素、无机盐、水和中药提取物混合均匀,用PBS溶液调节其pH值为6.8-7.2,过滤,滤液用紫外线灭菌,得到培养基;
S4.菌种的活化:将魏斯氏菌(Weissella confusa)、纳豆芽孢杆菌、米曲霉3.042分别接种于三份步骤S3制得的培养基中,活化培养,培养成菌种种子液,将三个培养基等体积混合并稀释得到复合发酵菌液;
S5.白芸豆发酵提取物的制备:将步骤S2制得的脱毒白云豆粉加入步骤S4制得的复合发酵菌液中,发酵,过滤,固体用无菌水洗涤,合并洗涤液和滤液,冷冻干燥,得到白芸豆发酵提取物。
5.根据权利要求4所述的制备方法,其特征在于,步骤S1中所述过筛的筛网目数为150-200目。
6.根据权利要求4所述的制备方法,其特征在于,步骤S2中所述白芸豆和水的固液比为1:(3-5)g/mL;所述乙醇-乙酸-水溶液中乙醇、乙酸和水的体积比为(2-10):(5-12):50;所述豆渣与乙醇-乙酸-水溶液的固液比为1:(2-4)g/mL;所述第一时间段为15-30min;所述第二时间段为10-20min。
7.根据权利要求4所述的制备方法,其特征在于,步骤S3中所述碳源选自糖蜜、葡萄糖、麦芽糖、乳糖、蔗糖、果糖、淀粉中的至少一种;所述氮源选自氨水、尿素、铵盐、硝酸盐、氨基酸中的至少一种;所述维生素选自维生素C、维生素B1、维生素B2、维生素A、维生素K、维生素B12、维生素D、维生素E中的至少一种;所述无机盐选自氯化钠、氯化钾、氯化钙、硫酸镁、氯化铁、硫酸锌、硫酸铜、硫酸锰、氯化锌、氯化铜、氯化锰中的一种或几种混合;所述氨基酸选自甘氨酸、丝氨酸、苏氨酸、缬氨酸、色氨酸、亮氨酸、丙氨酸、半胱氨酸、甲硫氨酸、赖氨酸、异亮氨酸、苯丙氨酸中的一种或几种混合;所述碳源、氮源、维生素、无机盐、中药提取物和水的质量比为(10-20):(5-15):(0.1-0.5):(0.01-0.2):(10-20):100。
8.根据权利要求4所述的制备方法,其特征在于,所述中药提取物由以下原料按照重量份制备而成:罗布麻叶5-15份、野菊花5-10份、杜仲10-20份、黄芪5-10份、黄连2-5份。
9.根据权利要求8所述的制备方法,其特征在于,所述中药提取物的制备方法如下:将5-15重量份罗布麻叶、5-10重量份野菊花、10-20重量份杜仲、5-10重量份黄芪、2-5重量份黄连分别洗净,干燥,打粉,过100-200目筛,得到的中药粉,加入水中,加热煮沸,提取1-2h,过滤,重复提取一次,合并两次滤液,浓缩,干燥,得到中药提取物。
10.根据权利要求4所述的制备方法,其特征在于,步骤S4中所述活化培养的条件为35-40℃培养12-24h;所述菌种种子液的含菌量为107-108cfu/mL;所述稀释倍数为100-200倍;所述魏斯氏菌(Weissel la confusa)、纳豆芽孢杆菌、米曲霉3.042的接种量分别为2-4%、3-5%和1-3%。
11.根据权利要求4所述的制备方法,其特征在于,步骤S5中所述脱毒白云豆粉和复合发酵菌液的固液比为1:(5-10)g/mL;所述发酵的条件为35-40℃培养36-48h。
12.一种如权利要求1-11任一项所述的制备方法制得的白芸豆发酵提取物。
13.一种如权利要求12所述的白芸豆发酵提取物在制备降血糖、降血压的食品、辅助或治疗降血糖、降血压的药品中的应用。
