Abstract
本发明涉及一种枸杞纯化产品的制备方法,尤其是涉及一种黑果枸杞花色苷提取物纯化产品的制备方法和应用,其特征在于:该制备方法为首先制备黑果枸杞花色苷提取物,然后将所述黑果枸杞花色苷提取物依次加入XAD‑7、AB‑8等性质相似的大孔树脂,先经水洗脱、酸性醇溶液分别洗脱纯化后,再进行酸性醇洗脱液减压浓缩、最后干燥后既得得黑果枸杞花色苷纯化产品;该产品可以应用在制备降低体重制剂、制备降低体脂率制剂上、制备降低血脂水平制剂等;本发明制作方法独特、能有效控制肥胖小鼠的体重增加趋势,降低小鼠体内脂肪比例,减轻了小鼠肝脏的脂肪变性。

技术领域
本发明涉及一种枸杞纯化产品的制备方法,尤其是涉及一种黑果枸杞花色苷提取物纯化产品的制备方法和应用。
背景技术
《本草纲目》称枸杞,补肾生精,养肝,明目,坚精骨,去疲劳,易颜色,变白,明目安神,令人长寿;《四部医典》、《晶珠本草》等著作称黑果枸杞味甘、性平,清心热,用于治疗心热病、心脏病、月经不调、停经等病症。现代医学及营养学研究表明,黑果枸杞,果实黑色,富含天然活性成分—花色苷,含量高达4.1%(干果重),且花色苷结构独特,是黑果枸杞的主要活性成分之一,具有抗氧化等功效作用。
2017年国家卫计委将黑果枸杞列为普通食品。即食用黑果枸杞后无毒副作用,安全性较高,可开发成保健食品或相关大健康产品。
但是目前为止,还没有把黑枸杞花色苷提取物纯化产品作为减肥制剂上的应用。
发明内容
本发明的目的主要是克服现有技术的不足,提供一种制作方法独特、通过酸性醇提取方法及大孔树脂、凝胶色谱等获得黑果枸杞花色苷提取物,能有效控制肥胖小鼠的体重增加趋势,降低小鼠体内脂肪比例,减轻了小鼠肝脏的脂肪变性,具有制备减肥制剂的前景的一种黑果枸杞花色苷提取物纯化产品的制备方法。
本发明通过如下方式实现:
一种黑果枸杞花色苷提取物纯化产品的制备方法,其特征在于:该制备方法为首先制备黑果枸杞花色苷提取物,然后将所述黑果枸杞花色苷提取物依次加入XAD-7、AB-8等性质相似的大孔树脂,先经水洗脱、酸性醇溶液分别洗脱纯化后,再进行酸性醇洗脱液减压浓缩、最后干燥后既得得黑果枸杞花色苷纯化产品;
所述黑果枸杞花色苷提取物的制备方法为先将黑果枸杞果实粉碎,在pH值为1-6的酸性条件下,-4℃~45℃提取至果实基本无色,过滤收集提取液,低于45℃加热浓缩、冻干,得黑果枸杞花色苷提取物;
所述黑果枸杞花色苷提取物在制备减肥制剂上的应用;
所述的黑果枸杞花色苷提取物及其纯化产品在制备降低体重制剂上的应用;
所述的黑果枸杞花色苷提取物及其纯化产品在制备降低体脂率制剂上的应用;

本发明具有如下效果:
1)制备方法独特:本发明提供的方法为首先制备黑果枸杞花色苷提取物,然后将所述黑果枸杞花色苷提取物依次加入XAD-7、AB-8等性质相似的大孔树脂,先经水洗脱、酸性醇溶液分别洗脱纯化后,再进行酸性醇洗脱液减压浓缩、最后干燥后既得得黑果枸杞花色苷纯化产品;所述黑果枸杞花色苷提取物的制备方法为先将黑果枸杞果实粉碎,在pH值为1-6的酸性条件下,-4℃~45℃提取至果实基本无色,过滤收集提取液,低于45℃加热浓缩、冻干,得黑果枸杞花色苷提取物。
2)制备减肥制剂的前景:本发明提供的黑果枸杞花色苷为酸性醇提取物及其纯化产品,黑果枸杞提取物为酸性醇提取物及其进一步用大孔树脂、凝胶色谱纯化后产品,黑果枸杞花色苷可以有效控制肥胖小鼠的体重增加趋势,降低小鼠体内脂肪比例,减轻了小鼠肝脏的脂肪变性,具有制备减肥制剂的前景。
3)具有显著的减肥作用:本发明通过酸性醇提取方法及大孔树脂、凝胶色谱等获得黑果枸杞花色苷提取物,经动物实验证明具有显著的减肥作用。
4)通过动物试验证明本发明提供的黑果枸杞花色苷提取物及其纯化产品可以应用在制备降低体重制剂、制备降低体脂率制剂上、制备降低血脂水平制剂上的应用、制备减轻肝脏脂肪变性制剂等上。

具体实施方式
一种黑果枸杞花色苷提取物纯化产品的制备方法,该制备方法为首先制备黑果枸杞花色苷提取物,然后将所述黑果枸杞花色苷提取物依次加入XAD-7、AB-8等性质相似的大孔树脂,先经水洗脱、酸性醇溶液分别洗脱纯化后,再进行酸性醇洗脱液减压浓缩、最后干燥后既得得黑果枸杞花色苷纯化产品,上述黑果枸杞花色苷提取物的制备方法为先将黑果枸杞果实粉碎,在pH值为1-6的酸性条件下,-4℃~45℃提取至果实基本无色,过滤收提取液,低于45℃加热浓缩、冻干,得黑果枸杞花色苷提取物。
该产品的减肥效果,通过如下试验体现
1试验方法
1.1动物分组和饲养
14只C57BL/6雄性小鼠,8-10周龄,饲养于宁夏医科大学实验动物中心,适应性饲养3-5天后,将小鼠随机分为高脂组和花色苷组,高脂组给高脂饲料,正常饮水;花色苷组给高脂饲料,黑果枸杞花色苷饮水。饲养期间观察小鼠状态,每周测一次小鼠体重,摄食量和饮水量。
1.2生化指标检测
末次给药24h后结束实验心脏取血,按试剂盒要求利用酶学法测定小鼠血清TC,TG,HDL-C,LDL-C。
1.3小鼠肝脏生态学观察
取小鼠肝脏放入4%多聚甲醛中固定,24小时之后乙醇梯度脱水,二甲苯透明,使用石蜡包埋,切片之后进行苏木素-伊红(H&E)染色,观察各组细胞形态,用光学显微镜进行病理组织学检查。
1.4数据分析
使用SPSS 23.0软件进行统计分析,所有计量资料均用均数±标准差表示,多组均数比较用单因素方差分析,两两比较用LSD法,检验水准α=0.05。
2试验结果
2.1黑果枸杞花色苷对小鼠体重,摄食量和饮水量的影响。
饲养期间对小鼠体重,摄食量和饮水量进行测量如图1所示,实验结果表明,与高脂组相比,花色苷干预之后,小鼠体重明显下降,说明花色苷能够降低高脂饲料引起的小鼠肥胖。两组小鼠摄食量和饮水量无明显差异,说明花色苷并没有影响小鼠食欲。
2.2黑果枸杞花色苷对小鼠血清血脂的影响
黑果枸杞花色苷对高脂饲料喂养的小鼠血清血脂的影响如表1所示。与高脂组相比,花色苷干预后,小鼠血清中LDL-C和TG明显降低,而HDL-C和TC没有明显差异。

数据表示为X±SD(N=7),*p<0.05,**p<0.01.
2.3黑果枸杞花色苷对小鼠肝脏病理切片的影响
对肝脏进行H&E染色,结果如图2所示。对照组小鼠肝脏中出现大量脂肪空泡,与高脂组相比,花色苷干预之后,小鼠肝脏中脂肪空泡明显减少。
