Modern researches show that agilawood chromone HHHX-5 inhibits immune system diseases, inhibits autonomous and innate immunity by inhibiting STAT3 and NFKB channels, inhibits mast cell histamine release, reduces mouse injury behavior, and has a certain cell immunoregulation function.
The invention has the beneficial effects that:
(1) The invention is compatible with the whole prescription, the effects of invigorating qi and nourishing yin, strengthening spleen, moistening lung, tonifying kidney and replenishing essence are provided as monarch, the medlar and American ginseng are used as ministerial drugs for nourishing liver and kidney, replenishing vital essence and improving eyesight, the medlar and American ginseng are used as ministerial drugs for replenishing qi and nourishing yin, and clearing heat and promoting fluid production, the morinda officinalis is matched with the schisandra chinensis for astringing and astringing, tonifying kidney and calming heart as adjuvant, the agilawood is used for warming kidney and activating heart, and the qi are provided for enabling the whole prescription to tonify kidney and receive qi, nourishing liver and kidney, tonifying qi and nourishing yin, and no eighteen-effect and nineteen-effect are provided.
(2) By adopting the preparation method, the mixed water extract prepared from rhizoma polygonati, medlar and morinda officinalis in the prescription improves the dissolution of active substances such as crude polysaccharide, glycoside, aglycone and the like in the rhizoma polygonati, medlar and morinda officinalis; the mixed ethanol extract is prepared from the Chinese medicinal materials of the American ginseng and the schisandra chinensis, so that the dissolution of the medicinal components of the ginsenoside, the schizandrin A and the like is increased; the agilawood amount in the prescription is small, and the agilawood ethanol extract prepared from the agilawood in the prescription is singly extracted, so that the extraction rate of the agilawood active ingredients can be greatly improved; the classified extraction can fully dissolve water-soluble and alcohol-soluble components of various medicines, improves the extraction transfer rate of medicinal effect substances, promotes the medicinal components to fully play a synergistic effect, improves the immunity enhancing effect, and is better applied to preparing medicines or health-care foods for improving human immunity.
(3) By adopting the formula and the preparation method, the components are synergistic, the immunity enhancing effect is obviously improved, and the dosage of the traditional Chinese medicine raw materials can be effectively reduced.

Detailed Description
The invention is further described below with reference to examples and figures, but is not limited in any way, and any alterations or substitutions based on the teachings of the invention are within the scope of the invention.
The traditional Chinese medicine composition for enhancing immunity provided by the invention comprises, by weight, 30-70 parts of rhizoma polygonati, 20-60 parts of wolfberry fruit, 10-30 parts of morinda officinalis, 10-30 parts of American ginseng, 5-20 parts of schisandra chinensis and 3-15 parts of agilawood, and is prepared by the steps of pretreatment, extraction and post-treatment, and specifically comprises the following steps:
A. pretreating, namely drying and crushing raw materials of rhizoma polygonati, medlar and morinda officinalis in a formula ratio, sieving with a 50-mesh sieve to obtain a material a for standby use, drying and crushing raw materials of American ginseng and schisandra chinensis in a formula ratio, sieving with a 50-mesh sieve to obtain a material b for standby use, and drying and crushing agilawood with the ethanol extract content of 10% -20% and sieving with a 10-mesh sieve to obtain agilawood powder for standby use;
B. extracting: (1) Adding drinking water with the volume 6-8 times of the weight of the material a into the material a, reflux-extracting for 1-3 times at 95-100 ℃ for 30-90min each time, mixing the extracting solutions, concentrating under 0.060MPa at 60 ℃, and vacuum drying at 60 ℃ to obtain dry paste a; (2) Adding ethanol solution with volume percentage concentration of 30% -50% v/v, which is 6-8 times of the weight of the material b, into the material b, reflux-extracting for 1-3 times at 60-80 ℃ for 60-120min each time, mixing the extracting solutions, and concentrating under 0.060MPa at 60 ℃ to obtain dry paste b; (3) Adding lignum Aquilariae Resinatum powder into 80-100% v/v ethanol solution with 15-25 times of volume percentage concentration, soaking for 40-80min, reflux extracting at 60-80deg.C for 2-3 times each for 60-80min, mixing extractive solutions, and concentrating at 60 deg.C under 0.060MPa to obtain lignum Aquilariae Resinatum ethanol extract;
C. post-treatment: mixing and pulverizing the dry extract a, the dry extract b and the ethanol extract of eaglewood, and sieving with a 80-mesh sieve to obtain a material c, thereby preparing the target traditional Chinese medicine composition.
The preparation of the Chinese medicinal composition for enhancing immunity is prepared by adding acceptable auxiliary materials in food processing into the Chinese medicinal composition for enhancing immunity, and preparing powder, tablets, capsules, granules and pills. Adding proper amount of auxiliary materials into the material c, and uniformly mixing to prepare the target preparation. 20-30g of the traditional Chinese medicine composition is used in each 100g of the preparation.
The application of the agilawood-containing traditional Chinese medicine composition for enhancing immunity is the application of the agilawood-containing traditional Chinese medicine composition for enhancing immunity in preparation of medicines or health-care foods for enhancing human immunity.

Weighing 30g of rhizoma polygonati, 20g of medlar, 10g of morinda officinalis, 10g of American ginseng, 5g of shizandra berry and 3g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 6 times of drinking water, carrying out reflux extraction for 3 times at 95-100 ℃ for 60min each time, combining the filtrates, and concentrating under the conditions of 0.060MPa and 60 ℃ to obtain dry extract a; reflux extracting radix Panacis Quinquefolii and fructus Schisandrae chinensis with 7 times of 40% v/v ethanol solution at 60-80deg.C for 3 times, 60min each time, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding 20 times of 95% v/v ethanol solution, reflux extracting at 60-70deg.C for 2 times, soaking for 40min each time, reflux extracting for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain ethanol extract of lignum Aquilariae Resinatum; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.
Example 2
Weighing 70g of rhizoma polygonati, 60g of medlar, 30g of morinda officinalis, 30g of American ginseng, 20g of shizandra berry and 15g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 8 times of drinking water, carrying out reflux extraction for 3 times at 95-100 ℃ for 70min each time, combining the filtrates, and concentrating under the conditions of 0.060MPa and 60 ℃ to obtain dry extract a; adding radix Panacis Quinquefolii and fructus Schisandrae into 8 times of 50% v/v ethanol solution, reflux extracting at 60-80deg.C for 3 times each for 70min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding 20 times of 95% v/v ethanol solution, reflux extracting at 60-70deg.C for 3 times, soaking for 60min each time, reflux-extracting for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain ethanol extract of lignum Aquilariae Resinatum; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.
Example 3
Weighing 50g of rhizoma polygonati, 40g of medlar, 20g of morinda officinalis, 20g of American ginseng, 10g of shizandra berry and 6g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 5 times of drinking water, carrying out reflux extraction for 2 times at 95-100 ℃ for 30min each time, combining the filtrates, and concentrating at the temperature of 0.060MPa and 60 ℃ to obtain dry extract a; adding radix Panacis Quinquefolii and fructus Schisandrae into 8 times of 60% v/v ethanol solution, reflux extracting at 60-80deg.C for 3 times, 50min each time, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding lignum Aquilariae Resinatum powder into 95% v/v ethanol solution of 25 times weight, reflux extracting at 60-70deg.C for 2 times, soaking for 60min each time, reflux extracting for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain lignum Aquilariae Resinatum ethanol extract; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.

Weighing 60g of rhizoma polygonati, 50g of medlar, 20g of morinda officinalis, 20g of American ginseng, 20g of shizandra berry and 15g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 8 times of drinking water, carrying out reflux extraction for 3 times at 95-100 ℃ for 90min each time, combining the filtrates, and concentrating under the conditions of 0.060MPa and 60 ℃ to obtain dry extract a; adding radix Panacis Quinquefolii and fructus Schisandrae into 8 times of 50% v/v ethanol solution, reflux extracting at 60-80deg.C for 3 times each for 2 hr, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding lignum Aquilariae Resinatum powder into 90% v/v ethanol solution of 25 times weight, reflux extracting at 60-70deg.C for 3 times, soaking for 80min each time, reflux extracting for 80min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain lignum Aquilariae Resinatum ethanol extract; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.

Weighing 40g of rhizoma polygonati, 40g of medlar, 15g of morinda officinalis, 15g of American ginseng, 10g of shizandra berry and 12g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 8 times of drinking water, carrying out reflux extraction for 3 times at 95-100 ℃ for 45min each time, combining the filtrates, and concentrating at the temperature of 0.060MPa and 60 ℃ to obtain dry extract a; adding 6 times of 50% v/v ethanol solution into radix Panacis Quinquefolii and fructus Schisandrae, reflux extracting at 60-80deg.C for 2 times, each time for 90min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding 15 times of 85% v/v ethanol solution, reflux extracting at 60-70deg.C for 2 times, soaking for 70min each time, reflux extracting for 40min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain ethanol extract of lignum Aquilariae Resinatum; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.

Weighing 50g of rhizoma polygonati, 40g of medlar, 20g of morinda officinalis, 20g of American ginseng, 10g of shizandra berry and 6g of agilawood, crushing the rhizoma polygonati, the medlar and the morinda officinalis, sieving with a 50-mesh sieve, adding 7 times of drinking water, carrying out reflux extraction for 2 times at 95-100 ℃ for 60min each time, combining the filtrates, and concentrating under the conditions of 0.060MPa and 60 ℃ to obtain dry extract a; reflux extracting radix Panacis Quinquefolii and fructus Schisandrae chinensis with 7 times of 40% v/v ethanol solution at 60-80deg.C for 2 times each for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain dry extract b; adding 20 times of 90% v/v ethanol solution, reflux extracting at 60-70deg.C for 3 times, soaking for 50min each time, reflux extracting for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain ethanol extract of lignum Aquilariae Resinatum; pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain the target Chinese medicinal composition.
As shown in fig. 1, the prepared traditional Chinese medicine composition is added with dextrin and proper amount of 50% v/v ethanol to prepare soft materials, wet granules, drying, granule finishing and split charging in sequence, and the mass ratio of the granule to the traditional Chinese medicine composition is 100:25.

The following efficacy tests were performed on the traditional Chinese medicine composition prepared in example 6:
experimental animals: SPF-class male Balb/c mice weighing 20+ -2 g, were male and female halves, and were offered by Beijing Fu Biotechnology Co., ltd.
Experimental facilities, reagents: electronic balance, vernier caliper (precision 0.02 mm), sheep Red Blood Cell (SRBC), microinjector (50 μl), etc.
Sample: the granules for enhancing immunity prepared in example 6 are prepared by respectively taking 7.5g, 15g and 22.5g of pure water to 150mL, and fully mixing the granules to obtain low, medium and high dosage gastric juice.
The experimental method comprises the following steps: mice were randomly divided into 2 groups: i.e., low, medium, high, and control groups of 12 samples each. The gastric lavage amount of the sample group is 20 mL/kg.bw, the control group is given with an equal volume of physiological saline once a day, each mouse is intraperitoneally injected with 2% SRBC for 0.2mL for immunization on the 26 th day of continuous gastric lavage, after continuous gastric lavage for 4 days, the thickness of the left rear toe is measured, and the average value is obtained after 3 times of measurement; the measurement sites were then subcutaneously injected with 20% srbc, 20 μl each, and the left hind toe thickness was measured 24 hours after injection, and the average was taken 3 times. The results are shown in Table 1:

The experimental results show that: the toe swelling degree of the medium and high dose sample groups is remarkably different from that of the control group, which shows that the immunity enhancing granule prepared in the example 6 can enhance the immune response of mice and improve the immunity of the mice.
Example 8
The following efficacy tests were performed with the immunity-enhancing traditional Chinese medicine composition prepared in example 6:
experimental animals: SPF-class male Balb/c mice weighing 20+ -2 g, were male and female halves, and were offered by Beijing Fu Biotechnology Co., ltd.
Experimental facilities, reagents: carbon dioxide incubator, thermostatic water bath, centrifuge, surgical instrument, slide frame, 200 mesh screen, SRBC, complement (guinea pig serum), hank's solution, RPMI1640 culture solution, SA buffer solution, agarose.
Sample: the granules for enhancing immunity prepared in example 6 are prepared by respectively taking 7.5g, 15g and 22.5g of pure water to 150mL, and fully mixing the granules to obtain low, medium and high dosage gastric juice.
The experimental method comprises the following steps: mice were randomly divided into 2 groups: i.e., low, medium, high, and control groups of 12 samples each. The method comprises the steps of (1) performing gastric lavage on a sample group with the volume of 20mL/kg & bw, administering an equal volume of physiological saline to a control group once a day, performing intraperitoneal injection of 2% SRBC for 0.2mL of immunity on each mouse on the 30 th day of continuous gastric lavage, performing cervical dislocation for 4 days after continuous gastric lavage, killing, taking out spleen, placing the spleen in a small plate containing Hank's liquid, lightly grinding the spleen to prepare cell suspension, filtering the cell suspension by a 200-mesh screen or grinding the spleen by 4 layers of gauze, centrifuging (1000 r/min) for 10min, washing the cell suspension by Hank's liquid for 2 times, finally suspending the cell suspension in 5mL of RPM 1640 culture liquid, counting the cell, and adjusting the cell concentration to 5x10 6 And each mL. Cells may also be suspended in 8mL hank's solution.
Heating and dissolving surface culture medium (1 g agarose and double distilled water to 100 mL), placing in 45-50 ℃ water bath for heat preservation, mixing with an equal amount of Hank's liquid with pH of 7.2-7.4 and 2 times concentration, subpackaging into small test tubes, adding 50 mu L of 10% SRBC (v/v prepared by SA buffer solution) into each tube, rapidly and uniformly mixing 20 mu L of spleen cell suspension (5X 106/mL) or 25 mu L of spleen cell suspension into the tubes, pouring onto a glass slide with a brushed agarose thin layer, making parallel pieces, after agar is solidified, horizontally buckling the glass slide on a sheet frame, placing into a carbon dioxide incubator for incubation for 1-1.5 h, then adding complement (1:8) diluted by SA buffer solution into a groove of the glass slide frame, continuously incubating for 1-1.5 h, and counting the number of hemolysis plaques. The results are shown in Table 2:

The experimental results show that: the number of the mouse antibody-producing cells in the medium-high dose sample group is obviously different from that of the mouse antibody-producing cells in the control group, which indicates that the immunity-enhancing granule prepared in the example 6 is beneficial to enhancing the immunity of mice and improving the number of the mouse antibody-producing cells.
Example 9 comparative test study
(1) Preparation of Chinese medicine extract
Respectively pulverizing rhizoma Polygonati, fructus Lycii and radix Morindae officinalis, sieving with 50 mesh sieve, adding 5 times of purified water, reflux-extracting at 95-100deg.C for 2 times for 30min each time, mixing filtrates, concentrating at 60 deg.C under 0.060MPa, and respectively preparing rhizoma Polygonati water extract, fructus Lycii water extract and radix Morindae officinalis water extract;
respectively taking radix Panacis Quinquefolii and fructus Schisandrae, respectively adding 8 times of 60% v/v ethanol solution, reflux extracting at 60-80deg.C for 3 times, 50min each time, mixing filtrates, concentrating at 60 deg.C under 0.060MPa to obtain dry extract, and respectively preparing radix Panacis Quinquefolii ethanol extract and fructus Schisandrae ethanol extract;
adding lignum Aquilariae Resinatum powder into 95% v/v ethanol solution of 25 times weight, reflux extracting at 60-70deg.C for 2 times, soaking for 60min each time, reflux extracting for 60min, mixing filtrates, and concentrating at 60 deg.C under 0.060MPa to obtain lignum Aquilariae Resinatum ethanol extract.
(2) Preparation: referring to the preparation method of the granule of the embodiment 6, the rhizoma polygonati aqueous extract, the medlar aqueous extract, the morinda officinalis aqueous extract, the American ginseng ethanol extract, the schisandra chinensis ethanol extract and the agilawood ethanol extract are respectively prepared into granules to obtain rhizoma polygonati granules, medlar granules, morinda officinalis granules, american ginseng granules, schisandra chinensis granules and agilawood granules.
(3) Drug efficacy test study: the pharmacodynamic test study was conducted with reference to example 8, and the drug efficacy test study was conducted with the Polygonatum sibiricum granule, lycium barbarum granule, morinda officinalis granule, american ginseng granule, schisandra chinensis granule, and eaglewood granule, which were test groups 1-6 in sequence, 22.5g (high dose) of the corresponding granule was added with pure water to 150mL, and the mixture was thoroughly mixed, and test samples 1-6 were obtained in sequence. The experimental results show that: there was a significant difference in the number of mouse antibody-producing cells in test groups 1 to 6 compared with the high-dose group of the Chinese medicinal composition of example 6, and the number of mouse antibody-producing cells in test groups 1 to 6 was lower than that in the low-dose group (7.5 g) of the Chinese medicinal composition of example 6.
In conclusion, the components in the formula of the invention are synergistic, the immunity enhancing effect is obviously improved, and the dosage of the medicament is effectively reduced.

Claims (9)

1. The traditional Chinese medicine composition for enhancing immunity is characterized by being prepared from the following traditional Chinese medicine raw materials in parts by weight: 30-70 parts of rhizoma polygonati, 20-60 parts of medlar, 10-30 parts of morinda officinalis, 10-30 parts of American ginseng, 5-20 parts of schisandra chinensis and 3-15 parts of agilawood.

The traditional Chinese medicine composition for enhancing immunity according to claim 1, which is characterized by being prepared from the following traditional Chinese medicine raw materials in parts by weight: 50 parts of rhizoma polygonati, 40 parts of medlar, 20 parts of morinda officinalis, 20 parts of American ginseng, 10 parts of shizandra berry and 6 parts of agilawood.
3. The method of claim 1, wherein the step of preparing a mixed aqueous extract from the sealwort, wolfberry and morinda officinalis; preparing a mixed ethanol extract from the American ginseng and the schisandra chinensis; preparing the agilawood ethanol extract from the agilawood.
4. The Chinese medicinal composition for enhancing immunity as claimed in claim 3, wherein,
the volume percentage concentration of the ethanol solution used by the mixed ethanol extract is 30-50%;
the volume percentage concentration of the ethanol solution used by the agilawood ethanol extract is 80% -100%.
5. The method for preparing the traditional Chinese medicine composition for enhancing immunity according to claim 1, comprising the following steps:
A. pretreatment: weighing the raw materials of rhizoma polygonati, medlar, morinda officinalis, american ginseng, schisandra chinensis and agilawood respectively according to a formula, drying, crushing and sieving the rhizoma polygonati, the medlar and the morinda officinalis to obtain a material a for standby; drying, crushing and sieving American ginseng and schisandra chinensis to obtain a material b for standby; drying, crushing and sieving agilawood to obtain agilawood powder for later use;
B. extracting:
(1) Extracting the material a by water to obtain an extracting solution, concentrating and drying to obtain a dry paste a;
(2) Adding the material b into ethanol solution with the volume percentage concentration of 30% -50%, carrying out reflux extraction to obtain an extracting solution, concentrating and drying to obtain dry paste b;
(3) Adding the agilawood powder into an ethanol solution with the volume percentage concentration of 80% -100%, soaking, extracting under reflux to obtain an extracting solution, concentrating, and drying to obtain an agilawood ethanol extract;
C. post-treatment: mixing, pulverizing and mixing the dry extract a, the dry extract b and the ethanol extract of lignum Aquilariae Resinatum to obtain a material c, and making into the final product.

B. extracting:
(1) Adding drinking water with the volume 6-8 times of the weight of the material a into the material a, reflux-extracting for 1-3 times at 95-100 ℃ for 30-90min each time, mixing the extracting solutions, concentrating, and drying to obtain dry paste a;
(2) Adding ethanol solution with volume percentage concentration of 30% -50% and weight of 6-8 times of the material b into the material b, reflux-extracting for 1-3 times at 60-80 ℃ for 60-120min each time, mixing the extracting solutions, concentrating, and drying to obtain dry paste b;
