Abstract
本发明提供了一种用于治疗肿瘤的盐酸纳曲酮微丸及其制备方法,涉及药物制剂领域。本发明将盐酸纳曲酮通过包衣工艺制备成符合要求的肠溶微丸,通过不同的肠溶包衣处方证明其能符合肠溶要求,且加速稳定性结果证明制备的肠溶微丸稳定性较好,这些处方和工艺证明了盐酸纳曲酮制备成肠溶微丸是确实可行且处方工艺均成熟;药效实验证明低剂量纳曲酮对结肠癌是有一定的治疗作用,这和纳曲酮作为常规非选择性阿片受体拮抗剂使用的药效是有一定的区别的,更加证实了低剂量纳曲酮是通过阿片类生长因子受体(OGFr)拮抗剂,阻断所有选择性激动剂阿片受体发挥抗肿瘤效果。

技术领域
本发明涉及药物制剂领域,具体涉及一种用于治疗肿瘤的盐酸纳曲酮微丸及其制备方法。
背景技术
结直肠癌是一种高发的恶性消化道肿瘤,由于结直肠癌患者的发病年龄向年轻化发展,这将会为国家带来沉重的负担。因此,无论预防还是治疗都应该加大对结直肠癌的投入。
目前,外科手术仍然是治疗结直肠癌的主要手段,化疗是辅助手段。在传统治疗中,手术仍是最有效的治疗方法,术后有指征的患者给予放化疗治疗,以降低复发转移几率。但一线化疗药物如奥沙利铂、氟尿嘧啶、伊立替康等都有明显的化疗副作用,如累积性神经病变以及肝脏毒性。靶向药物也参与了转移性结直肠癌的一线治疗,如检测结肠癌RAS和RAF野生型的患者可能在使用抗表皮生长因子受体药物中获益。而存在错配修复基因缺陷(dMMR)/高度微卫星不稳定(MSI-H)的结直肠患者可能从免疫检测点抑制剂中获益,但对于大多数的微卫星稳定的患者,则无明显获益。总体来说,结肠癌的靶向和免疫治疗均需要特定的基因类型才能产生疗效,但是这类药物的检测费用、药物本身的治疗费都是巨大的经济负担,因此迫切需要新的药物用于结直肠癌的治疗。
纳曲酮(Naltrexone,NTX)是一种阿片受体拮抗剂,于1984年美国食品和药品管理局(FDA)批准上市,用于酒精依赖、毒品戒断、阿片药物成瘾等领域的辅助治疗,临床应用剂量为50-100mg/d。但是随着研究的进展,发现在低剂量窗口即小于5mg/d,纳曲酮有免疫调节以及延缓肿瘤生长的效果,因此被称为低剂量纳曲酮(Low dose naltrexone,LDN)。LDN对肿瘤的影响最早的报道可追溯到1983年,Zagon等在Science发表文章证明LDN可以延缓神经母细胞瘤的进展,之后的研究发现,LDN在卵巢癌、乳腺癌、胰腺癌、非小细胞肺癌中均有抑制肿瘤生长的作用。
中国专利CN102784384A中公开了一种低剂量纳曲酮联合蛋氨酸脑啡肽作为制备抗癌药物的应用,该申请是通过与机体免疫系统细胞表面的阿片受体结合,刺激免疫细胞的活化,形成具有突破性疗效的肿瘤生物治疗新方法,该申请仅仅证明了低剂量纳曲酮有治疗癌症的作用,且是联合给药,没有涉及药物制剂以及剂型相关内容。
中国专利CN1166360C中公开了一种纳曲酮长效注射微球及其制备方法,其采用生物可降解材料乙交酯-丙交酯共聚物制备的微小球体,组合物可提供长达 25-35天的有效血药浓度,但是该申请公开的是注射微球。
中国专利CN105451740A公开了一种低剂量纳曲酮的新型治疗性应用,证明了纳曲酮作为Toll样受体9的拮抗剂发挥作用,慢性炎症和TLR9过表达是许多疾病的特征,包括癌症。
根据现有技术的记载可以看出,针对低剂量纳曲酮用于肿瘤治疗的文献较多,但是针对结直肠癌的治疗较少,将盐酸纳曲酮制备成肠溶微丸制剂的方法更是没有报道,因此亟需开发一种能够靶向针对结直肠癌的盐酸纳曲酮微丸,将盐酸纳曲酮制备成肠溶微丸,且符合肠溶要求,可以针对结直肠癌靶向给药。
发明内容
基于现有技术的不足,本发明提供了一种用于治疗肿瘤的盐酸纳曲酮微丸及其制备方法,该申请首次将盐酸纳曲酮通过包衣工艺制备成符合要求的肠溶微丸,通过不同的肠溶包衣处方证明其能符合肠溶要求,且加速稳定性结果证明制备的肠溶微丸稳定性较好,这些处方和工艺证明了盐酸纳曲酮制备成肠溶微丸是确实可行且处方工艺均成熟。
为实现上述目的,本发明采用的技术方案如下:
一种用于治疗肿瘤的盐酸纳曲酮微丸,所述的盐酸纳曲酮微丸包括盐酸纳曲酮含药层包衣、隔离层包衣和肠溶层包衣;
所述的盐酸纳曲酮含药层包衣包括载体100-500g、粘合剂2-20g、稀释剂 100-300g和药物有效成分3-20g;
所述的隔离层包衣包括粘合剂5-40g和稀释剂250-400g;
所述的肠溶层包衣包括肠溶聚合物10-40g、稳定剂10-35g、乳化剂1-10g、抗粘剂5-10g、增塑剂10-30g和稀释剂100-350g中的至少三种。
优选地,所述的盐酸纳曲酮含药层包衣包括载体200-400g、粘合剂5-10g、稀释剂150-200g和药物有效成分4-10g;
所述的隔离层包衣包括粘合剂10-25g和稀释剂270-350g;
所述的肠溶层包衣包括肠溶聚合物15-30g、稳定剂10-25g、乳化剂1-6g、抗粘剂5-8g、增塑剂12-25g和稀释剂120-250g中的至少三种。
所述的载体选自蔗糖丸芯、淀粉丸芯或微晶纤维素丸芯;
优选地,所述的载体为蔗糖丸芯。
所述的粘合剂为HPMC E5;
所述的稀释剂选自纯化水、95%乙醇或异丙醇;
优选地,所述的稀释剂为纯化水;
所述的药物有效成分为盐酸纳曲酮。
所述的肠溶聚合物选自Eudragit FS30D或Eudragit S100;
所述的抗粘剂选自滑石粉、单硬脂酸甘油酯或PlasACRYLTMT20;
所述的稳定剂为氨水;
所述的乳化剂为吐温80;
所述的增塑剂为枸橼酸三乙酯。
本发明还提供了上述盐酸纳曲酮含药层包衣、隔离层包衣和肠溶层包衣的制备方法。
所述的盐酸纳曲酮含药层包衣的方法为:将处方量的粘合剂用水过夜溶胀,加入处方量的盐酸纳曲酮,搅拌使其完全溶解;将载体放置于流化床的物料斗中进行上药操作,上药结束后,继续干燥0.5h,完成盐酸纳曲酮含药层包衣,得到含药丸芯1。
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速 0-20rpm,进风温度40-55℃,物料温度35-50℃;
优选地,所述的流化床包衣参数为雾化压力1-3kg,风量110-130m3/h,蠕动泵转速3-10rpm,进风温度50-55℃,物料温度35-40℃。
所述的隔离层包衣的方法为:先将处方量的粘合剂用水过夜溶胀,然后含药丸芯1放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,完成隔离层包衣,得到含药丸芯2。
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速0-20rpm,进风温度40-55℃,物料温度35-45℃;
优选地,所述的流化床包衣参数为雾化压力2-3kg,风量110-130m3/h,蠕动泵转速5-10rpm,进风温度40-50℃,物料温度37-40℃。
所述的肠溶层包衣的方法,包括如下步骤:
(1)取部分稀释剂加入肠溶聚合物,搅拌5-20min,确保肠溶聚合物完全浸湿,得到肠溶聚合物混悬液1;将稳定剂缓慢加入肠溶聚合物混悬液1中,搅拌60-120min,得到肠溶聚合物混悬液2;将增塑剂加入肠溶聚合物混悬液2中,搅拌60-120min,得到肠溶聚合物分散液;
(2)将抗粘剂加入剩余的稀释液中,使用高速分散机搅拌10-40min,使其混合均匀,得到抗粘剂混悬液;
(3)将步骤(2)中得到的抗粘剂混悬液加入肠溶聚合物分散液中搅拌均匀,将含药丸芯2放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,得到盐酸纳曲酮微丸。
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速 0-20rpm,进风温度40-55℃,物料温度30-45℃;
优选地,所述的流化床包衣参数为雾化压力1-3kg,风量100-120m3/h,蠕动泵转速10-15rpm,进风温度40-45℃,物料温度30-40℃。
本发明在实施过程中意外地发现肠溶包衣的种类会明显影响盐酸纳曲酮微丸的性能,本发明在实施过程中通过四种不同的肠溶包衣处方均制备了符合肠溶定位的低剂量纳曲酮的肠溶微丸,分别为:肠溶层包衣包括Eudragit S100,氨水,滑石粉,枸橼酸三乙酯和水;肠溶层包衣包括Eudragit FS30D,滑石粉,枸橼酸三乙酯和水;肠溶层包衣包括Eudragit FS30D,吐温80,单硬脂酸甘油酯,枸橼酸三乙酯和水;肠溶材料为EudragitFS30D,PlasACRYLTMT20和水;通过相关试验检测四个处方的溶出符合要求,稳定性较好,为低剂量纳曲酮肠溶微丸的产业化提供不同的方案。
本发明还提供了上述盐酸纳曲酮微丸在制备抗结肠癌药物中的应用。
与现有技术相比,本发明的有益效果在于:
(1)本发明首次将理论研究以及通路研究的低剂量纳曲酮的进行产业化,并且制备了符合肠溶要求的微丸制剂,并且通过药效证明低剂量纳曲酮对结肠癌的治疗作用;
(2)本发明将盐酸纳曲酮通过流化床包衣技术将空白丸芯制备成结肠定位的肠溶缓释微丸,达到一天一次的给药剂量;所选流化床包衣参数为风量 90-150m3/h,雾化压力1-5kg/cm2,蠕动泵转速1-30rpm/min,进风温度为30-55℃,物料温度为25-45℃;
(3)本发明创新通过四种不同的肠溶包衣处方均制备了符合肠溶定位的低剂量纳曲酮的肠溶微丸,通过相关试验检测四个处方的溶出符合要求,稳定性较好,为低剂量纳曲酮肠溶微丸的产业化提供不同的方案。
(4)本发明首次将盐酸纳曲酮通过包衣工艺制备成符合要求的肠溶微丸,通过不同的肠溶包衣处方证明其能符合肠溶要求,且加速稳定性结果证明制备的肠溶微丸稳定性较好,这些处方和工艺证明了盐酸纳曲酮制备成肠溶微丸是确实可行且处方工艺均成熟;药效实验证明低剂量纳曲酮对结肠癌是有一定的治疗作用,这和纳曲酮作为常规非选择性阿片受体拮抗剂使用的药效是有一定的区别的,更加证实了低剂量纳曲酮是通过阿片类生长因子受体(OGFr)拮抗剂,阻断所有选择性激动剂阿片受体发挥抗肿瘤效果的。

将制备得到的微丸筛分后于40℃熟化15-19小时后留存,并对微丸进行了外观和溶出度检测,具体参见图1-2。
根据图1可知:从扫描电镜中可以看出,制备得到的微丸大小均一,且表面外层致密,没有明显的孔洞和过多的滑石粉粘附在表面上。
根据图2可知:制备得到的微丸溶出曲线能达到肠溶的要求,即在2h酸中溶出较少,低于5%,在碱性环境中30min开始溶出,且基本上能完全溶出,符合药典对肠溶制剂溶出的要求。
对比例1
与实施例1的区别在于:肠溶层包衣配方中减少枸橼酸三乙酯的含量为8g,制备得到肠溶层包衣,但是检测发现得到的肠溶层包衣液成膜性降低,熟化后的处方抗酸性降低,抗酸实验(pH 1.0,2h,50rpm)的溶出量为21%,大于10% (药典规定肠溶制剂抗酸实验不得高于10%),且根据图3可以看出电镜下最外层膜有明显的孔洞,这个也是造成抗酸实验失败的原因。
对比例2
与实施例1的区别在于:肠溶层包衣配方中增加滑石粉的含量为12g,制备得到肠溶层包衣,但是在制备过程中造成堵枪的情况发生频繁,熟化后的处方抗酸实验(pH1.0,2h,50rpm)符合要求,但是根据图4可以看出电镜下滑石粉有明显残留。
实施例2一种盐酸纳曲酮微丸及其制备方法

根据图5可知:从扫描电镜中可以看出,制备得到的微丸大小均一,且表面外层致密光滑,没有明显的孔洞和滑石粉粘附在表面上。
根据图6可知:制备得到的微丸溶出曲线能达到肠溶的要求,即在2h酸中溶出较少,低于5%,在碱性环境中1h开始溶出,且基本上能完全溶出,符合药典对肠溶制剂溶出的要求。
对比例3
与实施例2的区别在于:肠溶层包衣配方中增加枸橼酸三乙酯的含量为5g,得到肠溶层包衣;但是枸橼酸三乙酯的含量增加会造成该肠溶层包衣成膜温度降低,从而导致聚合物析出,进而会造成堵枪等问题。
对比例4
与实施例2的区别在于:肠溶层包衣配方中降低滑石粉的含量为1g,得到肠溶层包衣;但是使用该得到肠溶层包衣制备的微丸粘连严重;熟化后微丸抗酸实验(pH 1.0,2h,50rpm)的溶出量为15%,大于10%(药典规定肠溶制剂抗酸实验不得高于10%),微丸粘连明显,根据图7可以看出最外层包衣明显有挤压情况。

(1)取部分稀释剂加入肠溶聚合物,搅拌10min,确保肠溶聚合物完全浸湿,得到肠溶聚合物混悬液;
(2)将剩余的稀释剂加热至90℃,将抗粘剂、乳化剂和增塑剂加入加热的稀释剂中,搅拌50min,得到单硬脂酸甘油酯乳液;
(3)将步骤(2)中得到的单硬脂酸甘油酯乳液加入肠溶聚合物分散液中搅拌均匀,将含药丸芯2放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,得到盐酸纳曲酮微丸。

将制备得到的微丸筛分后于40℃熟化15-19小时后留存,并对微丸进行了外观和溶出度检测,具体参见图8。
根据图8可知:从扫描电镜中可以看出,制备得到的微丸大小均一,且表面外层致密光滑,没有明显的孔洞。
实施例4一种盐酸纳曲酮微丸及其制备方法
1.1盐酸纳曲酮含药层包衣
配方组分:
制备方法:
将处方量的粘合剂用水过夜溶胀,加入处方量的盐酸纳曲酮,搅拌使其完全溶解;将载体放置于流化床的物料斗中进行上药操作,上药结束后,继续干燥 0.5h,完成盐酸纳曲酮含药层包衣,得到含药丸芯1。

1.3肠溶层包衣

制备方法:
(1)取部分稀释剂加入肠溶聚合物,搅拌10min,确保肠溶聚合物完全浸湿,得到肠溶聚合物混悬液;
(2)将抗粘剂加入剩余的稀释液中,使用高速分散机搅拌10min,使其混合均匀,得到抗粘剂混悬液;
(3)将步骤(2)中得到的抗粘剂混悬液加入肠溶聚合物分散液中搅拌均匀,后过不锈钢筛网;然后将含药丸芯2放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,得到盐酸纳曲酮微丸。

将制备得到的微丸筛分后于40℃熟化15-19小时后留存,并对微丸进行了外观和溶出度检测,具体参见图9-10。
根据图9可知:从扫描电镜中可以看出,制备得到的微丸大小均一,且表面外层致密光滑,没有明显的孔洞。
根据图10可知:制备得到的微丸溶出曲线能达到肠溶的要求,即在2h酸中溶出较少,低于5%,在碱性环境中1h开始溶出,且基本上能完全溶出,符合药典对肠溶制剂溶出的要求。
对比例5
与实施例4的区别在于:肠溶层包衣时进风温度调整到40℃,包衣液在喷枪中发生成膜现象,堵枪较严重。
试验例1稳定性试验
将实施例1处方制备的微丸于稳定性试验箱中(40±2℃,75%±5%RH)放置 30天,根据溶出考察处方的稳定性,检测结果见下表1和附图11。

根据上表1和附图11的检测结果可以看出,本发明实施例1制备的微球稳定性较好,符合要求。
试验例2低剂量盐酸纳曲酮(LDN)对结肠癌的抑制作用
造膜:C57BL/6小鼠皮下接种MC38细胞,建立鼠结肠癌皮下移植肿瘤模型。
实验分组:试验分为测试药LDN 0.1mg/kg QD组、LDN 0.5mg/kg QD组、 LDN 5mg/kg QD组、LDN 5mg/kg Q2D组、LDN 0.5mg/kg QD与PD-1抗体联合组及溶媒对照组,每组8只,口服灌胃给药,测试药每日给药一次,PD-1抗体每周给药两次,共给药三周。
根据相对肿瘤抑制率(TGI)进行疗效评价。
试验材料:盐酸纳曲酮、Anti-PD-1
试验动物:C57BL/6小鼠,雌性,6-8周(肿瘤细胞接种时的小鼠周龄),体重16.9-21.3g,110只。购自北京维通利华实验动物技术有限公司,动物合格证编号:110011211112329212。饲养环境:SPF级。
试验方法
(1)细胞培养
MC38细胞(细胞编号/批次:CL-01015/P9-210521)培养在含10%胎牛血清的DMEM培养液中。收集指数生长期的MC38细胞,PBS重悬至适合浓度用于小鼠皮下肿瘤接种。
(2)动物造模和随机分组
110只雌性C57BL/6小鼠右侧背部皮下接种1×106MC38细胞。定期观察肿瘤生长情况,待肿瘤生长至平均体积77mm3时,根据肿瘤大小随机分为7组,每组8只(见表1)。实验采用Study Director TM(版本号3.1.399.19,供应商 Studylog System,Inc.,S.SanFrancisco,CA,USA)软件“Matched distribution”分组的方法,以保证不同组别间的肿瘤体积相似。分组日期为2021年11月5日,分组当天定义为第0天,给药开始于分组当天。
(3)疗效评价标准
相对肿瘤增殖率,T/C%,即在某一时间点,治疗组和对照组相对肿瘤体积的百分比值。计算公式如下:
T/C%=TRTV/CRTV×100%(TRTV:治疗组平均RTV;CRTV:溶媒对照组平均RTV;RTV=Vt/V0,V0为分组时该动物的瘤体积,Vt为治疗后该动物的瘤体积)。
相对肿瘤抑制率,TGI(%),计算公式如下:TGI%=(1-T/C)×100%。(T 和C分别为治疗组和对照组在某一特定时间点的相对肿瘤体积(RTV))。
(4)实验终点
末次给药后终止实验,取瘤,称瘤重,拍照。
(5)试验结果:
溶媒对照组小鼠在给药开始后14天平均肿瘤体积为2121.5mm3。LDN(0.5 mg/kg)与PD-1抗体联合治疗组平均肿瘤体积为883.6mm3,相对肿瘤抑制率 TGI(%)为58.35%,与对照组有显著性差异(p<0.001)。测试药LDN各浓度单药治疗组表现出轻微肿瘤抑制趋势,TGI在-1.18%~30.98%之间,与对照组的差异无统计学意义(p>0.05),各浓度单药组间以中浓度组(0.5mg/kg)的肿瘤抑制趋势最明显,TGI为30.98%。
各治疗组和对照组肿瘤生长情况见表1、表2和附图12。

根据上表1-2和附图12可以看出,本发明可抑制结肠癌肿瘤生长,有明显的抗肿瘤作用。
综上,本发明将盐酸纳曲酮通过包衣工艺制备成符合要求的肠溶微丸,通过不同的肠溶包衣处方证明其能符合肠溶要求,且加速稳定性结果证明制备的肠溶微丸稳定性较好,这些处方和工艺证明了盐酸纳曲酮制备成肠溶微丸是确实可行且处方工艺均成熟;药效实验证明低剂量纳曲酮对结肠癌是有一定的治疗作用,这和纳曲酮作为常规非选择性阿片受体拮抗剂使用的药效是有一定的区别的,更加证实了低剂量纳曲酮是通过阿片类生长因子受体(OGFr)拮抗剂,阻断所有选择性激动剂阿片受体发挥抗肿瘤效果的。
以上所述仅为本发明的优选实施例而已,并不用于限制本发明,对于本领域的技术人员来说,本发明可以有各种更改和变化。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (10)
1.一种用于治疗肿瘤的盐酸纳曲酮微丸,所述的盐酸纳曲酮微丸包括盐酸纳曲酮含药层包衣、隔离层包衣和肠溶层包衣;其特征在于:所述的盐酸纳曲酮含药层包衣包括载体100-500g、粘合剂2-20g、稀释剂100-300g和药物有效成分3-20g;所述的隔离层包衣包括粘合剂5-40g和稀释剂250-400g;所述的肠溶层包衣包括肠溶聚合物10-40g、稳定剂10-35g、乳化剂1-10g、抗粘剂5-10g、增塑剂10-30g和稀释剂100-350g中的至少三种。
2.根据权利要求1所述的盐酸纳曲酮微丸,其特征在于:盐酸纳曲酮含药层包衣包括载体200-400g、粘合剂5-10g、稀释剂150-200g和药物有效成分4-10g;所述的隔离层包衣包括粘合剂10-25g和稀释剂270-350g;所述的肠溶层包衣包括肠溶聚合物15-30g、稳定剂10-25g、乳化剂1-6g、抗粘剂5-8g、增塑剂12-25g和稀释剂120-250g中的至少三种。
3.根据权利要求1所述的盐酸纳曲酮微丸,其特征在于:所述的载体选自蔗糖丸芯、淀粉丸芯或微晶纤维素丸芯;
所述的粘合剂为HPMC E5;
所述的稀释剂选自纯化水、95%乙醇或异丙醇;
所述的肠溶聚合物选自Eudragit FS30D或Eudragit S100;
所述的抗粘剂选自滑石粉、单硬脂酸甘油酯或PlasACRYLTMT20;
所述的稳定剂为氨水;
所述的乳化剂为吐温80;
所述的增塑剂为枸橼酸三乙酯。
4.根据权利要求3所述的盐酸纳曲酮微丸,其特征在于:所述的肠溶层包衣包括Eudragit S100,氨水,滑石粉,枸橼酸三乙酯和水;
或,所述的肠溶层包衣包括Eudragit FS30D,滑石粉,枸橼酸三乙酯和水;
或,所述的肠溶层包衣包括Eudragit FS30D,吐温80,单硬脂酸甘油酯,枸橼酸三乙酯和水;
或,所述的肠溶材料为Eudragit FS30D,PlasACRYLTMT20和水。
5.根据权利要求1所述的盐酸纳曲酮微丸,其特征在于:所述的盐酸纳曲酮含药层包衣的方法为:将处方量的粘合剂用水过夜溶胀,加入处方量的盐酸纳曲酮,搅拌使其完全溶解;将载体放置于流化床的物料斗中进行上药操作,上药结束后,继续干燥0.5h,完成盐酸纳曲酮含药层包衣,得到含药丸芯1;
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速0-20rpm,进风温度40-55℃,物料温度35-50℃。
6.根据权利要求5所述的盐酸纳曲酮微丸,其特征在于:所述的流化床包衣参数为雾化压力1-3kg,风量110-130m3/h,蠕动泵转速3-10rpm,进风温度50-55℃,物料温度35-40℃。
7.根据权利要求1所述的盐酸纳曲酮微丸,其特征在于:所述的隔离层包衣的方法为:先将处方量的粘合剂用水过夜溶胀,然后含药丸芯1放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,完成隔离层包衣,得到含药丸芯2;
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速0-20rpm,进风温度40-55℃,物料温度35-45℃。
8.根据权利要求7所述的盐酸纳曲酮微丸,其特征在于:所述的流化床包衣参数为雾化压力2-3kg,风量110-130m3/h,蠕动泵转速5-10rpm,进风温度40-50℃,物料温度37-40℃。
9.根据权利要求1所述的盐酸纳曲酮微丸,其特征在于:所述的肠溶层包衣的方法,包括如下步骤:
(1)取部分稀释剂加入肠溶聚合物,搅拌5-20min,确保肠溶聚合物完全浸湿,得到肠溶聚合物混悬液1;将稳定剂缓慢加入肠溶聚合物混悬液1中,搅拌60-120min,得到肠溶聚合物混悬液2;将增塑剂加入肠溶聚合物混悬液2中,搅拌60-120min,得到肠溶聚合物分散液;
(2)将抗粘剂加入剩余的稀释液中,使用高速分散机搅拌10-40min,使其混合均匀,得到抗粘剂混悬液;
(3)将步骤(2)中得到的抗粘剂混悬液加入肠溶聚合物分散液中搅拌均匀,将含药丸芯2放置流化床的物料斗中进行包衣操作,包衣结束后,继续干燥0.5h,得到盐酸纳曲酮微丸;
所述的流化床包衣参数为雾化压力1-5kg,风量90-150m3/h,蠕动泵转速0-20rpm,进风温度40-55℃,物料温度30-45℃。
10.根据权利要求9所述的盐酸纳曲酮微丸,其特征在于:所述的流化床包衣参数为雾化压力1-3kg,风量100-120m3/h,蠕动泵转速10-15rpm,进风温度40-45℃,物料温度30-40℃。
