Description
Technical Field
The invention belongs to the technical field of oral care, and particularly relates to a novel compound formula toothpaste containing fermented periplaneta peptide and a preparation method thereof.
Background
Toothpaste is one of tooth cleaning products, generally in a gel shape, and is usually applied to a toothbrush, and the mechanical friction of the toothbrush is used for cleaning the surfaces of teeth, cleaning the teeth and the periphery of the teeth and purifying and refreshing the oral cavity. Tooth cleaning is most important for making teeth beautiful, protecting gum health, and preventing bad breath, and toothpaste has become a necessity of human daily life.
The national oral environment research shows that the tooth brushing rate of the Chinese population is 85.6 percent, but the adult who achieves the good index of oral hygiene is only 0.22 percent, the morbidity of periodontal diseases reaches 99.4 percent, and the symptoms of gingival bleeding, gingival swelling and pain, oral ulcer, periodontal diseases and the like are the common oral diseases of many Chinese people. The traditional toothpaste which demands mothproofing, whitening and cleaning cannot meet the increasingly diverse and complex oral environment of Chinese people. When toothpaste is sold as a cosmetic, consumers pay more attention to the appearance, flavor and taste of the product, and some functionalities and characteristics that cannot be directly perceived are often overlooked.
Therefore, in recent years, in the new national standards for toothpaste (GB 8372 + 2008) and the light industrial standards for efficacy toothpaste (QB 2966 + 2008) issued by China, the traditional Chinese medicine is allowed to enter the visual field for improving the oral problems of people in China, and the traditional Chinese medicine is gradually accepted by people with the characteristics of safety, effectiveness, comprehensive oral care and the like. Therefore, it is always the pursuit of research and development personnel to find toothpaste prepared from natural, safe and environment-friendly components with comprehensive tooth-strengthening, whitening, bacteriostasis and repairing functions.

Table 1:
2. and (3) anti-allergy experiment: the perception dentin hypersensitivity is a subjective pain expression and has different responses to different stimuli, and the evaluation experiment adopts the evaluation from the aspects of subjective perception of dentin hypersensitivity pain, Schiff cold air sensitivity index and probing sensitivity improvement degree 3 of a subject every day, and records comparison before and after use.
Experimental materials: 100 toothpaste prepared in example 3 of the application;
control group: 30 Yunan white drug double-effect anti-allergy toothpastes;
subjects were eligible for inclusion criteria for healthy adult males and females: the group entry conditions are that the health condition of the whole body is good and no serious systemic diseases of the whole body exist; secondly, the testee appeals to cold, heat, acid and sweet stimulation sensitivity and pain caused by biting hard objects; age 18-60 years old. The subjects were confirmed by probing and cold water stimulation before the experiment.
90 qualified research objects are screened out according to the requirement of entering the group and divided into three groups (a blank group 30, a control group 30 and an experimental group 30), the experiment period, the morning and evening tooth brushing are carried out, the brushing is respectively tried for 2 weeks, 4 weeks and 6 weeks, and then the using effect is counted and recorded in the following table 2.
Table 2:
3. and (3) repairing experiment:
a rat model test is adopted, 12 rats are divided into two groups of blank control groups of 4 rats and an experimental group of 8 rats, 10% chloral hydrate is injected into the abdominal cavity for anesthesia and is fixed on a rat board, and model animals freely drink 10% high sugar water to form periodontitis. The blank control group had pure water. After confirming that periodontitis had formed in the rats, the rats were observed for red swelling and bleeding, and the toothpaste prepared in example 3 of the present application was applied as prescribed for 2 weeks.
And (3) effect detection: the gingival bleeding was observed by gently probing the inferior gingival margin with a periodontal probe and sliding, and scored according to the following scoring criteria. The upper left, mesial, palatal, and distal 3 sites of the rat were explored.
And (4) judging the standard:
1. the periodontal disease does not bleed, and the gum does not have red swelling;
2. the periodontal disease does not bleed, and the gum is slightly red and swollen;
3. periodontal punctate bleeding, gingival redness and swelling;
4. periodontal bleeding, gingival redness and swelling;
5. bleeding of the gums, which is a severe inflammation;
6. the gum bleeding automatically with ulcerated spots.

grouping | Rat group | 1 | 2 | 3 | 4 | 5 | 6 |
Blank space | 4 | 0 | 0 | 0 | 3 | 1 | 0 |
Example 3 | 8 | 4 | 2 | 2 | 0 | 0 | 0 |
4. The sanitation index is as follows: the toothpaste prepared in examples 1 to 7 were subjected to a hygiene index test, in which the test method was described with reference to GB8372 and the results of the detection of colonies contained therein were recorded in the following table;
the total number of colonies in the specified detection result is less than or equal to 500CFU/mL and is marked as reaching the standard;

Note: the antibacterial performance test of the compound use of the traditional Chinese medicine extract and the fermented probiotics is carried out before the experiment:
and (3) antibacterial test: selecting Streptococcus mutans ATCC25175 and Porphyromonas gingivalis ATCC 33277;
preparation of test samples: two components with antibacterial effect are selected to prepare a sample, the traditional Chinese medicine extracting solution and the fermented probiotics (1:1 compound), and the rest components are water. The agar plate dilution method was used.
Sample (I) | Concentration of | Streptococcus mutans | Porphyromonas gingivalis |
Blank space | 0 | ++ | ++ |
1 | Containing 20% of compound liquid | - | - |
2 | Containing 10% of compound liquid | - | - |
3 | Containing 5% of compound liquid | - | - |
