Preferably, the bacterial liquid in the step (2) is lactobacillus casei bacterial liquid, and the fermentation temperature is 37 ℃.

In a third aspect of the present invention, there is provided a method for preparing a toothpaste containing a rice extract, comprising the steps of:
(1) sequentially adding a friction agent and a thickening agent into a powder stirring bin for stirring; subsequently adding a foaming agent;
(2) adding solvent into the liquid material premixing tank, adding correctant, chelating agent, antiseptic and rice extract under stirring, and stirring;
(3) adding the humectant into a liquid material premixing tank and uniformly stirring;
(4) starting a vacuum paste making machine, starting a vacuum pump, sucking the liquid material in the liquid material premixing tank into the paste making machine, and starting a scraper and double stirring;
(5) controlling the vacuum degree of the vacuum paste making machine, and sucking the powder in the powder stirring bin into the vacuum paste making machine;
(6) after the powder in the powder stirring bin is sucked, improving the vacuum degree of the vacuum paste making machine and stirring;
(7) and (5) after the inspection is qualified, obtaining the paste.
Preferably, the abrasive in step (1) is selected from silica; most preferably, the abrasive is selected from one or more of silica type 302, silica type 201A; the foaming agent is selected from sodium dodecyl sulfate; the stirring time is 10 min.
Preferably, the solvent in step (2) is selected from water, most preferably purified water; said flavoring agent is selected from saccharin sodium; the chelating agent is selected from tetrasodium pyrophosphate; the preservative is selected from one or more of sodium benzoate and potassium sorbate; the stirring time is 10 min.
Preferably, the humectant in the step (3) is selected from one or more of sorbitol and polyethylene glycol; the stirring time is 1 min.
Preferably, the vacuum degree of the vacuum paste making machine is controlled to be-0.04 MPa to-0.08 MPa in the step (4).
Preferably, the vacuum degree of the vacuum paste making machine in the step (5) is controlled to be-0.05 MPa to-0.06 MPa.
Preferably, the vacuum degree of the vacuum paste making machine in the step (6) is controlled to be-0.090 to-0.098 MPa; the stirring time is 15 min.
Further, one or more of whitening agent and essence can be optionally added after the step (6); the whitening agent is selected from titanium dioxide.
Compared with the prior art, the invention has the following beneficial effects:
the invention takes rice as raw material, and adopts scientific method to extract active substance, so as to obtain rice extract as active ingredient of toothpaste. The prepared rice extract contains abundant and various micromolecular active oligopeptide, lactic acid, succinic acid, kojic acid and other beneficial components of human bodies, so that the remarkable effects of nourishing gum, protecting healthy teeth and keeping the teeth clean and tender can be achieved, the gum and the teeth are nourished and healthy from the root, the teeth are protected clearly, the oral cavity is healthy, the damage to the gum or the oral cavity caused by adopting a large amount of chemical active components is effectively reduced, and the potential safety hazard of the oral cavity is reduced.

Detailed Description
In order to make the objects, technical solutions and effects of the present invention clearer and clearer, the present invention is further described in detail below with reference to examples. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
Example 1
A toothpaste containing rice extract is prepared by the following steps:
(1) starting a powder dust remover, sequentially adding 5kg of silicon dioxide type 302, 0.5kg of sodium carboxymethylcellulose TH-10, 0.5kg of xanthan gum type RH and 10kg of silicon dioxide type 201A into a powder stirring bin, and starting stirring for 10 min; stopping stirring, and adding 3kg of sodium dodecyl sulfate;
(2) opening a purified water feeding valve, adding 28.4kg of purified water into the liquid material premixing tank, stirring, adding 0.05kg of saccharin sodium, 0.05kg of tetrasodium pyrophosphate, 0.1kg of sodium benzoate and 5kg of rice extract, stirring for 10min, and stopping stirring;
(3) weighing 42kg of sorbitol into a liquid material premixing tank; then adding 5kg of polyethylene glycol-400, and stirring for 1 min;
(4) starting a vacuum paste making machine, starting a vacuum pump, controlling the vacuum degree to be-0.04 Mpa to-0.08 Mpa, and sucking the liquid material in the liquid material premixing tank into the paste making machine; after all the inspection liquid materials are sucked into the paste making machine, starting a scraper and carrying out double stirring;
(5) when the vacuum degree is increased to-0.05 Mpa to-0.06 Mpa, the vacuum pump is stopped, and the powder in the powder stirring bin is sucked into the paste making machine; when the vacuum is reduced to-0.05 Mpa, the vacuum pump needs to be restarted;
(6) after the powder is detected to be completely sucked into the paste making machine, starting vacuum to the maximum, stirring for 15min in a timing manner, and stopping the vacuum pump when the vacuum degree reaches-0.090 MPa to-0.098 MPa;
(7) dissolving 0.2kg of titanium dioxide in water, stirring uniformly, adding into an essence tank, opening an aroma inlet valve, and sucking in the titanium dioxide solution;
(8) opening a fragrance inlet valve, sucking 0.2kg of rice essence, closing the fragrance inlet valve, and stirring for 2 min; starting a vacuum pump, and continuing stirring for 15 min;
(9) if the vacuum degree is not lower than-0.094 Mpa, stopping stirring and scraping, slowly opening to release gas, recovering normal pressure, sampling and inspecting; if the vacuum degree is lower than-0.094 Mpa, stopping stirring, and continuously opening the scraper plate to vacuumize until the vacuum degree meets the requirement;
(10) and (5) after the inspection is qualified, obtaining the paste.
The preparation method of the rice extract comprises the following steps:
(1) selecting high-quality rice as a fermentation substrate; boiling rice for 1h, cooling to room temperature, filtering with 200 mesh sieve, sterilizing at 115 deg.C for 20min, and collecting filtrate;
(2) after the filtrate is cooled to room temperature, inoculating a high-activity lactobacillus casei liquid, and fermenting at 37 ℃ until the pH value of the filtrate is below 4.0; after fermentation, breaking cell walls of the thalli by adopting ultrasonic waves, and adding lysozyme for enzymolysis for 60min at 30 ℃;
(3) after the enzymolysis is finished, adding trehalose as a protective agent, and carrying out thermal cracking treatment at 80 ℃;
(4) repeatedly extracting, concentrating, and cooling.
Verification example 1 physical and chemical Properties test
The rice extract prepared according to the preparation method of example 1 was used to test its physical and chemical properties, and the test results are shown in table 1 below:

Verification example 2 DPPH radical scavenging test
Adding 4.0mL of DPPH solution (0.0178 mmol/L) and rice extracts (0%, 2%, 4%, 6%, 8%, 10% and the solvent is purified water) with different concentrations prepared according to the invention in a 10mL colorimetric tube in sequence, adding absolute ethyl alcohol to the scales, uniformly mixing, immediately measuring the light absorption value at the wavelength of 517nm by using a 1cm cuvette, and marking the light absorption value as A1, then measuring the light absorption value at the wavelength of 517nm after storing for 30min in the dark at room temperature, and marking the light absorption value as A2, and measuring the light absorption value as A3 by using an ethanol solution only added with DPPH in a control group; the free radical clearance (K) was calculated as disclosed below:
K(%)=[1-(A1-A2)/A3]×100%。
the results of the tests are shown in fig. 1, and show that the rice extract prepared according to the present invention has a significant DPPH radical scavenging function, and the scavenging ability is concentration-dependent, i.e., the higher the concentration of the rice extract is, the stronger the scavenging ability is. The free radicals are mainly generated by human body oxidative metabolism, environmental pollution, bad life and rest and the like and are one of the main reasons for tooth and gum aging, so the toothpaste containing the rice extract prepared by the invention can effectively remove the free radicals generated by the human body, thereby obviously delaying the tooth and gum aging.
EXAMPLES 3 gingival hydration test
Selecting 20 subjects with age of 18-50 years, and randomly dividing the subjects into 2 groups of 10 persons; wherein group 1 was oral cavity cleaning using the toothpaste prepared according to the example of the present invention, and group 2 was oral cavity cleaning using the general commercially available toothpaste not containing the rice extract of the present invention, all once a day in the morning.
The gum was used as the test site, and the moisture content was measured every day for each subject, and the hydration rate of the gum was calculated, and the measurement results are shown in fig. 2. The results show that the experimental group using the toothpaste of the invention significantly increased the gingival moisture content of the subject during the test period, had significant moisturizing effect, and the difference was statistically significant, compared to the control group.
Verification example 4 oral cleaning test
Selecting 20 subjects with age of 18-50 years, and randomly dividing the subjects into 2 groups of 10 persons; wherein group 1 was oral cavity cleaning using the toothpaste prepared according to the example of the present invention, and group 2 was oral cavity cleaning using the general commercially available toothpaste not containing the rice extract of the present invention, all once a day in the morning.
The teeth were used as the test sites, and the brightness L and the melanin value MI were measured every day, and the results are shown in fig. 3. The results show that the experimental group using the toothpaste of the present invention can significantly improve the brightness of the subject's teeth during the test period, while reducing the melanin value, with a significant whitening effect, compared to the control group, with statistical significance of the difference.
Verification example 5 evaluation test of bacteriostatic Properties
(1) Respectively adding 90mL of sterile PBS and 10g of toothpaste sample prepared according to the invention in the 250mL of sterile triangular flask containing the glass beads, and shaking the triangular flask to fully disperse the toothpaste into a homogenate solution (10%) serving as a sample solution for later use; selecting common commercial toothpaste by the same method to prepare a reference solution;
(2) test bacteria (Streptococcus mutans (ATCC 700610), Porphyromonas gingivalis (ATCC 33277), and Fusobacterium nucleatum (ATCC 25586)) 24h plate cultures were washed with PBS to prepare a bacterial suspension (desired concentration: 100. mu.L was dropped in 5mL of PThe number of recovered bacteria in the BS control solution or 5mL sample solution was 1X 104 CFU/mL~9×104CFU/mL);
(3) Sucking 3 tubes of sample liquid (5 mL) and control liquid (5 mL), taking the bacterial suspension, respectively dripping 100 mu L of the bacterial suspension into each sample liquid and control sample or sample liquid, uniformly mixing, starting timing, fully and uniformly mixing after acting for 2min, properly diluting, then taking 2-3 dilutions, respectively sucking 1mL, placing in 2 plates, pouring 15mL of BHIA culture medium cooled to 40-45 ℃, rotating the plates to fully and uniformly, turning the plates after agar solidification, culturing for 48h at 35 +/-2 ℃, and counting viable bacteria colonies.
(4) The bacteriostatic rate was calculated as follows:

(5) Evaluation criteria
The bacteriostasis rate is more than or equal to 50-90 percent, and the product has bacteriostasis; the bacteriostasis rate is more than or equal to 90 percent, and the product has stronger bacteriostasis.
The results show that the toothpaste containing the rice extract prepared according to example 1 of the present invention has 95-99% inhibition against streptococcus mutans, porphyromonas gingivalis, and fusobacterium nucleatum, while the control solution has 40-77% inhibition. Therefore, the toothpaste containing the rice extract, which is prepared by the invention, has a remarkably improved antibacterial effect, can reduce oral bacteria and help to keep gum healthy.

Claims (10)
1. A toothpaste containing rice extract is characterized by comprising rice extract, an abrasive agent, a foaming agent, a thickening agent, a flavoring agent, a preservative, a chelating agent, a humectant and a solvent.
2. The toothpaste containing the rice extract as claimed in claim 1, which comprises the following components in percentage by mass:
0.01-10% of rice extract, 1-30% of abrasive, 0.5-5% of foaming agent, 0.05-6% of thickening agent, 0.05-1% of flavoring agent, 0.02-0.5% of preservative, 0.05-3% of chelating agent, 0.5-90% of humectant and 10-30% of solvent.
3. The toothpaste containing rice extract as claimed in claim 1, wherein the rice extract is prepared by a method comprising the steps of:
(1) selecting high-quality rice as a fermentation substrate; boiling rice, cooling to room temperature, filtering, sterilizing, and collecting filtrate;
(2) after the filtrate is cooled to room temperature, inoculating high-activity bacteria liquid for fermentation, crushing cell walls of the thalli by adopting ultrasonic waves after the fermentation is finished, and adding lysozyme for enzymolysis treatment;
(3) after the enzymolysis is finished, adding a protective agent for thermal cracking treatment;
(4) and (5) cooling to obtain the product.
4. The toothpaste containing rice extract as claimed in claim 3, wherein the boiling time in step (1) is 1h, and the sieving operation is performed by using a 200-mesh sieve, the sterilization temperature is 115 ℃, and the sterilization time is 20 min.
5. The toothpaste containing rice extract as claimed in claim 3, wherein the bacterial solution in the step (2) is Lactobacillus casei bacterial solution, and the fermentation temperature is 37 ℃.
6. The toothpaste containing rice extract as claimed in claim 3, wherein the judgment criterion for completion of fermentation in step (2) is: the pH of the filtrate is below 4.0; the enzymolysis temperature is 30-40 deg.C, and the enzymolysis time is 10-60 min.
7. The toothpaste containing rice extract as claimed in claim 3, wherein the protective agent in the step (3) is trehalose, and the cleavage temperature is 80-100 ℃.
8. The toothpaste containing rice extract as claimed in any one of claims 1 to 7, wherein the toothpaste further comprises one or more of a whitening agent and a perfume.
9. The method for preparing the toothpaste containing the rice extract as claimed in any one of claims 1 to 8, comprising the steps of:
(1) sequentially adding a friction agent and a thickening agent into a powder stirring bin for stirring; subsequently adding a foaming agent;
(2) adding solvent into the liquid material premixing tank, adding correctant, chelating agent, antiseptic and rice extract under stirring, and stirring;
(3) adding the humectant into a liquid material premixing tank and uniformly stirring;
(4) starting a vacuum paste making machine, starting a vacuum pump, sucking the liquid material in the liquid material premixing tank into the paste making machine, and starting a scraper and double stirring;
(5) controlling the vacuum degree of the vacuum paste making machine, and sucking the powder in the powder stirring bin into the vacuum paste making machine;
(6) after the powder in the powder stirring bin is sucked, improving the vacuum degree of the vacuum paste making machine and stirring;
(7) and (5) after the inspection is qualified, obtaining the paste.
10. The method of claim 9, wherein one or more of a whitening agent and a perfume may be optionally added after step (6).
