Abstract
本发明属于医药生物技术领域,公开了一种用于前列腺癌辅助诊断的分子标志物——APOBEC3B。APOBEC3B在前列腺癌的癌组织中高表达,在癌旁正常组织中低表达;而且,APOBEC3B在前列腺癌细胞系中的表达水平显著高于正常前列腺细胞株;通过敲低APOBEC3B基因的表达水平,能明显抑制前列腺癌细胞的增殖、迁移和侵袭。因此,APOBEC3B可以作为前列腺癌辅助诊断的分子标志物;通过检测样本中APOBEC3B的表达水平,可以对前列腺癌进行辅助诊断,为临床医生对前列腺癌的诊断提供参考依据。

技术领域
本发明属于医药生物技术领域,具体涉及APOBEC3B在前列腺癌诊断、预后预测及治疗中的应用。
背景技术
前列腺癌(prostate cancer)是男性常见的恶性肿瘤之一,已成为世界范围内男性发病率第二的恶性肿瘤,仅次于肺癌。我国前列腺癌的发病率也呈明显升高趋势。因前列腺癌发病隐匿,早期多无症状,到发现时多已成为晚期转移性患者,预后较差。如果能够早期对前列腺癌进行明确诊断,对于前列腺癌患者接受个体化治疗、延长生存期、提高生存质量和降低社会医疗负担具有重要的意义。因此,寻找可用于前列腺癌早期精准诊断与靶标治疗的关键分子,是目前前列腺癌患者所亟需的。
研究证实前列腺癌细胞突变率的增加是疾病发生的驱动因素,并最终导致增殖、侵袭、转移、耐药性的发生。对全基因组和外显子组数据的分析表明,载脂蛋白B mRNA编辑酶催化多肽蛋白-3B(APOBEC3B)-胞苷脱氨酶突变模式的存在可能在致癌的体细胞突变中起重要作用。APOBEC3B属于载脂蛋白B mRNA编辑酶催化多肽(apolioprotein B mRNA-editingenzyme catalytic polypeptide,APOBEC)家族成员,APOBEC3B及其家族成员已经被证实对核酸上的胞苷有脱氨基作用,可以通过诱发靶基因上的胞嘧啶脱氨基成为尿嘧啶(C>U)导致基因组发生突变。大量研究显示,APOBEC3B在多种肿瘤(包括乳腺癌、肺癌、子宫内膜癌、肝癌等)中呈持续高水平表达,并且APOBEC3B表达通过经典癌基因的突变与癌症相关联已经得到广泛的证实。但是APOBEC3B在前列腺癌发生发展中的作用尚未见相关报道。
发明内容
针对现有技术中存在的问题和不足,本发明的目的旨在提供APOBEC3B在前列腺癌诊断、预后预测及治疗中的应用。
为实现发明目的,本发明采用的技术方案如下:
本发明第一方面提供了一种可用于前列腺癌辅助诊断及预后预测的生物标志物,所述分子标志物为APOBEC3B基因(NCBI基因ID:9582)或APOBEC3B蛋白(即APOBEC3B基因编码的蛋白质)。
通过免疫组织化学染色法检测APOBEC3B蛋白在前列腺癌的癌组织和癌旁正常组织中表达情况,发现APOBEC3B蛋白在前列腺癌的癌组织中的表达水平显著高于癌旁正常组织;说明APOBEC3B蛋白在前列腺癌组织中均有不同程度的表达上调。
通过实时荧光定量PCR(RT-qPCR)和Western blot检测APOBEC3B在前列腺癌细胞系LNCaP、22Rv1、Du145和PC-3以及正常前列腺细胞株RWPE-1中的表达情况,发现APOBEC3B在前列腺癌细胞系中的表达水平显著高于正常前列腺细胞系;说明APOBEC3B在前列腺癌细胞系中均有不同程度的表达上调。
对TCGA数据库中前列腺癌患者的生存信息进行分析发现,APOBEC3B低表达的前列腺癌患者的总体生存期(Overall survival,OS)优于APOBEC3B高表达APOBEC3B的前列腺癌患者,说明APOBEC3B基因能够用于辅助预测前列腺癌的预后。
本发明第二方面提供了APOBEC3B基因或蛋白的检测试剂在制备用于前列腺癌辅助诊断的产品中的应用。
根据上述的应用,优选地,所述产品通过RT-qPCR、基因芯片、高通量测序平台、免疫组织化学、Western blot或酶联免疫吸附检测样本中APOBEC3B基因或蛋白的表达水平。
根据上述的应用,优选地,所述产品中含有扩增APOBEC3B基因的特异性引物、与APOBEC3B基因杂交的探针与APOBEC3B蛋白特异性结合的抗体。

与现有技术相比,本发明取得的积极有益效果为:
(1)本发明首次发现APOBEC3B在前列腺癌的肿瘤组织中表达量较癌旁正常组织显著上调,APOBEC3B在前列腺癌细胞系中的表达水平显著高于正常前列腺细胞系,因此,APOBEC3B可以作为前列腺癌辅助诊断的分子标志物;通过检测样本中APOBEC3B的表达水平,实现前列腺癌的早期诊断,为临床医生对前列腺癌的诊断提供参考依据。
(2)本发明首次发现APOBEC3B低表达的前列腺癌患者的总体生存期(Overallsurvival,OS)明显优于APOBEC3B高表达APOBEC3B的前列腺癌患者,因此,APOBEC3B基因或APOBEC3B蛋白表达水平能够用于辅助预测前列腺癌的预后,从而为前列腺癌预后预测的判断提供了新途径,为临床医生对于前列腺癌病情分析提供了参考依据。
(3)本发明还发现APOBEC3B与前列腺癌细胞的增殖、迁移和侵袭能力相关,APOBEC3B能够促进前列腺癌细胞的增殖、迁移和侵袭,通过敲低APOBEC3B基因对前列腺癌细胞的增殖、迁移和侵袭具有明显的抑制作用;因此,通过抑制前列腺患者中APOBEC3B的表达水平,可以达到治疗前列腺癌的效果。
(3)本发明提供的特异靶向APOBEC3B基因的shRNA序列可以高效的抑制或敲低靶细胞中APOBEC3B的表达,抑制前列腺癌细胞的增殖、迁移和侵袭,进而抑制前列腺癌细胞的恶性进展,因此,可以用于治疗前列腺癌,在前列腺癌治疗中具有重要的意义。

下列实施例中未注明具体条件的实验方法,均采用本技术领域常规技术,或按照生产厂商所建议的条件;所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规产品。
为了使得本领域技术人员能够更加清楚地了解本发明的技术方案,以下将结合具体的实施例详细说明本发明的技术方案。
实施例一:生信数据分析APOBEC3B在前列腺癌中的表达
1.数据采集和整理
在TCGA公共数据库中系统检索并筛选与前列腺癌相关的数据集。对于基因芯片数据均下载CEL原始数据,转录组高通量测序(RNA sequencing,RNA-Seq)数据均下载表达计数(counts)矩阵。对下载后的数据进行质量控制、背景校正、归一化、基因注释等预处理。最终,从TCGA数据库中获得的499例前列腺癌肿瘤组织及52例正常前列腺组织样本的数据进行分析。
2.APOBEC3B在前列腺癌中的表达情况分析:
对步骤1中筛选出的前列腺癌组织样本和正常前列腺癌组织样本中APOBEC3B的表达情况进行统计分析,结果如图1所示。
由图1可知,在前列腺癌组织中APOBEC3B基因的总体水平较正常前列腺组织呈现明显升高,差异具有显著统计学意义(P=0.0002)。
3.APOBEC3B与前列腺癌预后的关系分析:
将步骤1中筛选出的有完整随访资料的前列腺癌患者分为APOBEC3B高表达和低表达组(以APOBEC3B的中位表达量为分界点),然后对APOBEC3B高表达和APOBEC3B低表达组的前列腺癌患者的总生存信息进行分析并绘制Kaplan-Meier生存曲线,结果图2所示。
由图2可知,APOBEC3B低表达的前列腺癌患者的总体生存期(Overall Survival,OS)优于那些高表达APOBEC3B的前列腺癌患者的总体生存时间。由此说明,APOBEC3B基因能够用于预测前列腺癌的预后。
实施例二:APOBEC3B在前列腺癌的肿瘤组织及癌旁正常组织中的表达情况研究
对前列腺癌组织芯片(包含25例前列腺癌癌旁正常组织样本和50例前列腺癌肿瘤组织样本)进行免疫组化染色分析,分析了APOBEC3B蛋白在前列腺癌和癌旁正常组织中的表达情况。
1.样本采集:
收集50例病理组织学确诊为原发性前列腺癌患者的肿瘤组织样本和25例前列腺癌患者癌旁正常组织样本。前列腺癌患者手术前均未进行内分泌治疗、放疗和化疗,手术是首选治疗方案。按照伦理审查委员会规定的制度,每个病人在取样前均签署了知情同意书。
2.实验方法
采用免疫组织化学染色法检测前列腺癌肿瘤组织和癌旁正常组织中APOBEC3B的蛋白表达水平,具体操作步骤如下:
(1)组织芯片免疫组化染色
1)石蜡切片脱蜡复水:将含有前列腺癌肿瘤组织和配对的癌旁正常组织的芯片依次置入二甲苯溶液中浸泡两次,每次15分钟,再用无水乙醇浸泡两次,每次5分钟,之后依次使用85%乙醇浸泡5分钟和75%乙醇浸泡5分钟,蒸馏水漂洗1次。
2)抗原修复:将脱蜡后的切片放置在用EDTA抗原修复液(pH 9.0)浸泡组织切片,然后将其放入微波炉内小火煮沸10-15分钟。待室温冷却后,用PBS洗涤切片三次(5分钟/次)。
3)抗原修复后的玻片上滴加3%的双氧水,孵育5分钟,置入水中终止反应,用PBS浸泡清洗三次(5分钟/次)。
4)封闭:使用吸水纸去除多余的PBS溶液,用5%山羊血清室温封闭30分钟。
5)一抗孵育:吸去封闭液并孵育APOBEC3B一抗,平放切片于湿盒内,在4℃条件下孵育12小时。
6)二抗孵育:切片置于PBS中并置于摇床上洗涤三次(5分钟/次)。去除多余PBS,加入HRP标记的山羊抗兔的二抗室温孵育2小时。
7)DAB显色:二抗孵育完毕后,将切片在PBS中漂洗三次(5分钟/次),使用DAB显色。
8)分化、返蓝:将显色后的组织芯片用PBS漂洗后,在苏木精中孵育2分钟,用蒸馏水清洗三次(5分钟/次),置于盐酸中孵育10秒钟,最后用蒸馏水冲洗切片15-20分钟。
9)脱水、透明:将切片依次放入70%、80%、90%、95%和100%酒精梯度缸中各5分钟,最后将切片置于二甲苯中孵育10分钟。
10)封片:将切片置于通风处晾干,其上覆盖中性树胶,用盖玻片封片,在室温下自然干燥。
11)使用组织切片扫描仪采集图像。
3.数据处理及分析
免疫染色结果由两位病理医师分别在显微镜下评估。评分标准:需综合考虑阳性细胞显色强度和百分比。免疫组织化学染色评分由染色阳性细胞百分比和染色强度两个指标得分相加得到。
其中,染色阳性细胞百分比的得分具体如下:0%的肿瘤细胞出现阳性染色为0分,(1%-30%)的肿瘤细胞出现阳性染色为1分,(30%-60%)的肿瘤细胞出现阳性染色为2分,(60%-100%)的肿瘤细胞出现阳性染色为3分。
染色强度的得分具体为:没有染色记为0分,黄色记为1分,棕黄色记为2分,红褐色记为3分。
以显色强度评分+百分比评分记为最终评分结果;其中,0分为阴性(-),1分为弱阳性(--+),2-4分阳性(+),5-6分强阳性(++)。
4.实验结果
50例前列腺癌肿瘤组织和25例癌旁正常组织免疫组化检测的结果如图3所示。由图3可知,与癌旁正常前列腺组织相比较,前列腺癌肿瘤组织中的染色阳性细胞数目明显增多,染色强度明显加深,提示前列腺癌中APOBEC3B表达水平相对较高,进一步根据免疫组织化学染色评分结果对正常组织和肿瘤组织中APOBEC3B蛋白的表达水平进行统计发现正常前列腺组织样本中APOBEC3B蛋白的表达水平明显低于前列腺癌肿瘤组织样本,其差异具有明显的统计学意义。
实施例三:APOBEC3B在前列腺癌细胞和正常前列腺细胞株中的表达情况研究
1.RT-qPCR检测APOBEC3B基因在前列腺癌细胞和正常前列腺细胞株中的表达
通过RT-qPCR实验,我们检测了正常前列腺上皮细胞系RWPE-1和前列腺癌细胞系LNCaP、22Rv1、Du145和PC-3中的APOBEC3B基因表达水平。
(1)细胞选取及培养:
培养人类正常前列腺细胞株RWPE-1,前列腺癌细胞系LNCaP、22RV1、DU145、PC-3;细胞常规培养于含10%的FBS、1%的青霉素和链霉素的1640细胞培养基中,其中RWPE-1用KM培养基,PC-3用F-12K培养基,而非1640培养基,五种细胞在37℃、5%CO2以及饱和湿度的条件下培养。细胞每2天换液,按1:3传代。
(2)实验方法:
采用RT-qPCR检测前列腺癌细胞和正常前列腺细胞株中APOBEC3B基因的表达水平,具体操作步骤如下:
1)细胞RNA提取:收集生长在6cm皿中处于对数期的细胞,弃培养基后加入1mLTrizol,充分裂解后,装入EP管中。加入200μL氯仿进行混合,室温下静置10分钟。将上述EP管移至离心机低温离心10分钟,12000rpm。吸取上清液体400μL,装入新EP管中。混入相同体积的异丙醇,室温静置15分钟,低温离心。离心后得到白色样沉淀,弃除上清。再向有沉淀的管中给予75%乙醇1mL以重悬溶解后低温离心。再次弃上清,晾干至白色沉淀变成透明状,加入适量Nuclease-free water,溶解沉淀。使用Nano Drop 2000检测总RNA的浓度和纯度。
2)逆转录合成cDNA:
a、取出HiScript III RT SuperMix for qPCR(+gDNA wiper)中的4×gDNA wiperMix、5×HiScript III qRT SuperMix,短暂离心,置于冰上。
按表1中组份配制去除基因组DNA的反应液,在冰上进行,配制好后用移液器轻轻吹打混匀。上机反应条件:42℃2分钟。

c、配置好的混合液轻柔混匀后,打开PCR仪,按下列循环设置好程序,将样品逐个放进仪器中,开始反转录反应,反转录反应程序为:37℃15分钟,85℃5秒。反应结束后及时取出反转录反应液即cDNA,1:10稀释用于qPCR反应(反转录反应液加入到下一步的RT-qPCR反应体系时,其加入量不要超过RT-qPCR反应体积的1/10(V/V)量)。
3)荧光定量检测:
以GAPDH为内参,通过RT-qPCR反应检测不同细胞株中APOBEC3B基因的相对表达量,比对之间的表达差异。
APOBEC3B的特异性扩增引物的核苷酸序列如下:
上游引物:5’-CGCCAGACCTACTTGTGCTAT-3’(SEQ ID NO.1);
下游引物:5’-CATTTGCAGCGCCTCCTTAT-3’(SEQ ID NO.2)。
GAPDH的特异性扩增引物的核苷酸序列如下:
上游引物:5’-CTGGGACGACATGGAGAAAA-3’;
下游引物:5’-AAGGAAGGCTGGAAGAGTGC-3’。
按表3的比例配置RT-qPCR反应体系(20μL体系)。

两步法进行RT-qPCR,并制作熔解曲线,程序设定如表4。

根据RT-qPCR原始检测结果,按照2^-ΔΔCт法计算APOBEC3B基因的相对表达水平,即前列腺癌细胞和正常前列腺细胞株,目的基因APOBEC3B转录水平的差异。
(3)数据处理分析
所有数据采用均数±标准差(mean±SD)表示。两组间比较采用双侧Student's t检验,三组及以上采用单因素方差分析。所有结果均采用GraphPad Prism 6软件进行绘图,以P<0.05为检验水准,当P<0.05时认为具有显著统计学意义。
(4)实验结果
RT-qPCR检测前列腺癌细胞和正常前列腺细胞中APOBEC3B基因的表达水平,其检测结果如图4所示。由图4可知,与前列腺正常细胞RWPE-1细胞相比,APOBEC3B基因在前列腺癌细胞DU145、PC-3、LNCaP、22Rv1表达显著升高,说明APOBEC3B基因在前列腺癌细胞中表达水平显著上调。
其中,以PC-3细胞中APOBEC3B基因的表达水平较高,因此,选择PC-3细胞系作为工具细胞用于后续的实验研究。
2.Western blot检测APOBEC3B蛋白在前列腺癌细胞和正常前列腺细胞株中的表达
通过Western blot检测了APOBEC3B蛋白在部分前列腺癌细胞系及正常前列腺上皮细胞中的表达情况。
(1)细胞选取及培养:
培养人类正常前列腺细胞株RWPE-1,前列腺癌细胞系LNCaP、22Rv1、DU145、PC-3;细胞常规培养于含10%的FBS、1%的青霉素和链霉素的1640细胞培养基中,其中RWPE-1用KM培养基,PC-3用F-12K培养基,而非1640培养基,五种细胞在37℃、5%CO2以及饱和湿度的条件下培养。细胞每2天换液,按1:3传代。
(2)实验方法:
采用Western blot检测前列腺癌细胞和正常前列腺细胞株中APOBEC3B的表达水平,具体操作步骤如下:
1)PAGE凝胶制备:取等体积下层胶溶液和下层胶缓冲液加入烧杯中混匀后加入促凝剂再次混匀,将下层凝胶混合液注入制胶玻璃板中。待下层胶凝固后,取等体积上层凝胶夜和上层凝胶缓冲液加入烧杯中混匀,混匀后加入促凝剂,再次混匀。将混匀后的上层凝胶混合液注入制胶玻璃板中,插入齿梳。
2)蛋白制样:收取上述成对数生长期的正常前列腺和前列腺癌细胞,先用PBS洗两遍,然后加入RIPA裂解液提取总蛋白,应用BCA蛋白定量试剂盒进行蛋白定量,然后往每一个待测孔中(包含标准孔和样品孔)各加入200μL,37℃孵育30分钟。用酶标仪测定570nm处样品孔的吸光度,根据OD值制作蛋白浓度标曲,最终计算出待测蛋白浓度。根据蛋白浓度的高低,选择加入5X上样缓冲液,充分混匀,95℃变性10分钟,-20℃短期保存进行后续实验。
3)跑胶:将凝胶固定在电泳装置上,加入电泳液,去除齿梳,在各个槽孔内依次加入15μL蛋白样。恒压70V至指示剂进入分离胶,120V电泳至结束。
4)转膜:把转膜装置、滤纸放进转膜液中浸湿。先把PVDF膜先放入甲醇中活化15-30秒,取出放在凝胶上,凝胶与PVDF膜之间不能有气泡产生,放入装置内,加入转膜液,将装置放在冰水混合的盆中,恒流250mA,转膜时间90分钟。
5)封闭:转膜完成后,取出凝胶和PVDF膜,用5%脱脂牛奶进行封闭,室温摇床封闭2小时。
6)一抗孵育:加入APOBEC3B抗体(1∶1000稀释),内参GAPDH(1:10000稀释),4℃摇床孵育过夜。
7)二抗孵育:TBST洗涤三次(10分钟/次);加入HRP标记的山羊抗兔及抗小鼠二抗(1:5000)室温孵育1小时。
8)曝光:TBST洗涤三次(10分钟/次);使用发光液进行蛋白显影。应用VisionWorks软件,以GAPDH为内参蛋白,计算目的蛋白相对表达量。
(3)实验结果:
Western blot检测前列腺癌细胞和正常前列腺细胞中APOBEC3B的表达水平的实验结果如图5所示。由图5可知,与前列腺正常细胞RWPE-1细胞相比,APOBEC3B蛋白在前列腺癌细胞DU145、PC-3、LNCaP、22Rv1表达显著升高,说明APOBEC3B蛋白在前列腺癌细胞中表达水平显著上调。
其中,以PC-3细胞中APOBEC3B蛋白的表达水平较高,因此,选择PC-3细胞系作为工具细胞用于后续的实验研究。
实施例四:APOBEC3B基因的敲减
1.细胞培养:
人前列腺癌细胞系PC-3,分别以含10%胎牛血清和1%的青霉素和链霉素的F-12K培养基在37℃、5%CO2、相对湿度为90%的培养箱中培养。细胞每2天换液1次,采用以消化酶进行消化传代。
2.shRNA设计
针对APOBEC3B基因的shRNA序列:
阴性对照shRNA(记作sh-Control)序列:
sh-Control:5’-CCGG-GAACTCGAGTTCACATGGTTC-CTCGA-GGAACCATGTGAACTCGAGTTCT-TTTTT-3’;

sh-APOBEC3B-2:5’-CCGG-GCAAAGCAATGTGCTCCTGAT-CTCGAG-ATCAGGAGCACATTGCTTTGC-TTTTTT-3’(SEQ ID NO.4)。
3.慢病毒包装:
1)将293T细胞消化离心,用新鲜的DMEM完全培养基将细胞沉淀重悬成单细胞悬液,细胞计数后将细胞浓度调整为2×106/mL后接种10cm培养皿中,每皿10mL细胞悬液。
2)37℃含有5%CO2培养箱中培养过夜后,细胞密度约70%时,弃去旧培养基,更换新鲜的完全培养基。
3)配制转染试剂:质粒与Lipofectamine 3000Reagent的质量体积比为1:3,先用750μL的Opti-MEM稀释Lipofectamine 3000Reagent,再分别将750μL的Opti-MEM,与18μg质粒(sh-APOBEC3B-1/2:8μg,PSPAX2:6μg,PMD2G:4μg)和稀释的Lipofectamine 3000Reagent一起混匀。室温静置15分钟。
4)将配好的转染试剂加入培养皿中,轻轻混匀后放置37℃含有5%CO2培养箱中。转染6小时后更换为新鲜的完全培养。
5)转染48小时后,收集含有慢病毒颗粒的细胞上清,4000g离心10分钟,去除细胞碎片,上清过0.45μm滤膜后收集至新的离心管中。分装,-80℃保存备用。
4.筛选稳转株细胞
1)将复苏后培养三代的PC-3细胞铺于6cm皿中,37℃含有5%CO2培养箱中培养过夜,细胞密度达到50%左右。
2)从-80℃冰箱中取出冻存的病毒于冰上缓慢融化,并准备细胞完全培养基和polybrene的混合物,polybrene的终浓度为8μg/mL。最后用混合物培养基与病毒制备1:2的病毒稀释液待用。
3)弃去培养皿中的旧培养基,加入病毒稀释液,摇匀后于37℃含有5%CO2培养箱中培养。
4)感染24小时后,吸去含有病毒的培养基,换上新鲜的完全培养基,继续置于37℃含有5%CO2培养箱中培养24小时。
5)筛选稳定感染的细胞克隆:弃去旧培养基,更换含有3μg/mL的puromycin的新鲜的完全培养基。每隔24小时换液一次,共三次(72小时)。
6)扩增培养:将细胞逐渐由6cm皿转入10cm皿中,用含有1μg/mL的puromycin的新鲜的完全培养基维持培养。
7)目的细胞鉴定与冻存:收细胞提RNA用荧光定量PCR验证目的基因的表达。若带有GFP,可与荧光显微镜下观察荧光的表达。若目的细胞鉴定正确,按照常规细胞冻存步骤,进行保种保存。
5.RT-qPCR检测APOBEC3B的表达水平:
(1)细胞RNA提取:
细胞RNA提取的具体操作与实施例二相同,在此不再赘述。
(2)逆转录合成cDNA:
逆转录合成cDNA的具体操作步骤与实施例一相同,在此不再赘述。
(3)荧光定量检测:
荧光定量检测的具体操作步骤与实施例一相同,在此不再赘述。
6.Western blot检测APOBEC3B的表达水平:
Western blot检测的具体操作步骤与实施例三相同,在此不再赘述。
7.实验结果:
RT-qPCR检测结果如图6所示。Western blot检测结果如图7所示。由图6和图7可知,与阴性对照组(sh-Control组)相比,在转染了sh-APOBEC3B-1和sh-APOBEC3B-2的PC-3细胞中APOBEC3B基因和蛋白水平显著下调。
实施例五:APOBEC3B对前列腺癌细胞的细胞增殖、迁移和侵袭的影响
1、CCK8检测细胞增殖实验
(1)细胞培养:
细胞培养的具体操作与实施例四相同,在此不再赘述。
(2)细胞转染:
细胞转染的具体操作与实施例四相同,在此不再赘述。
(3)CCK8检测细胞增殖
1)实验分为3组,分别为阴性对照组(sh-Control组)、sh-APOBEC3B-1组、sh-APOBEC3B-2组,每组均设5个复孔。
2)转染后经嘌呤霉素筛选过稳定敲低表达APOBEC3B的PC-3细胞分别用0.25%胰酶消化后进行细胞计数,将细胞接种于96孔板中,接种前浓度调整为5×103/mL,每孔加100μL。
3)8小时后弃掉每组5个孔的旧培养基,避光条件下,96孔板中每孔加入含有20μL的CCK8试剂新鲜培养基,轻轻晃动培养板,然后将培养板放入培养箱中继续培养,2小时后取出,在酶标仪450nm波长处测定OD值。并以此时间点同样的方法分别检测24小时、48小时及72小时的细胞的吸光度值。
(4)实验结果
CCK8检测细胞增殖的实验结果如图8所示。由图8可知,与阴性对照组(sh-Control组)相比,转染sh-APOBEC3B-1和sh-APOBEC3B-2的实验组细胞增殖受到明显的抑制(P<0.05)。由此说明,敲低APOBEC3B对PC-3细胞增殖能力有明显的抑制作用。
2、克隆形成实验
(1)细胞培养:
细胞培养的具体操作与实施例四相同,在此不再赘述。
(2)细胞转染:
细胞转染的具体操作与实施例四相同,在此不再赘述。
(3)克隆形成实验
1)实验分为3组,分别为阴性对照组(sh-Control组)、sh-APOBEC3B-1组、sh-APOBEC3B-2组,每组均设3个复孔。
2)转染后的PC-3细胞培养72小时,用0.25%胰酶消化进行细胞计数,将细胞接种于6孔板中,PC-3每孔铺1500个细胞,每组设置3个重复孔,每孔先加入2mL完全培养基,再根据计数浓度计算出每组需要的细胞量体积,对应加入相应孔中,混匀后放入37℃,5%CO2培养箱中培养,5天左右换一次液,2周后形成了肉眼可见的细胞集落。
3)去除培养基,用PBS清洗一次,每孔加入1mL预冷4%多聚甲醛固定15分钟,固定完后,去除固定液,加入0.5%结晶紫染色20分钟,染色完后,去除结晶紫,用清水轻轻冲洗残余结晶紫,晾干后,置于明亮光线下用摄像机进行拍照。
(4)实验结果
克隆形成实验结果如图9所示。由图9可知,与阴性对照组(sh-Control组)相比,转染sh-APOBEC3B-1、sh-APOBEC3B-2的实验组细胞增殖能力明显降低(P<0.05)。由此说明,敲低APOBEC3B对PC-3细胞增殖能力有明显的抑制作用。
3、Transwell小室迁移和侵袭实验
通过检测目的细胞在Transwell小室中向含血清培养基的迁移情况来验证目的基因对细胞迁移和侵袭能力的影响。
(1)实验方法:
1)无菌条件下Matrigel冰浴融化后用无血清培养基按1:7比例稀释Matrige1胶(迁移实验无需铺胶,侵袭实验需铺胶),在Transwell上室内部缓慢加入上室底部,铺胶后将再有Transwell孔的24孔板平移入37℃的细胞培养箱中孵育至其凝固成胶状。
2)实验分为3组,分别为阴性对照组(sh-Control组)、sh-APOBEC3B-1组、sh-APOBEC3B-2组,每组均设3个复孔,上室加入细胞数量为5×104个细胞不含血清的细胞悬液(迁移实验)、1×105个细胞不含血清的细胞悬液(侵袭实验),下室加入700μL含10%胎牛血清培养基,37℃下恒温培养箱内培养48小时。
3)取出Transwell小室,PBS清洗3遍,使用4%多聚甲醛固定30分钟后用PBS洗3遍,加入结晶紫染色30分钟后弃掉结晶紫溶液,显微镜下观察着色强度,用纯净水清洗,放入荧光显微镜下观察拍照并计数。
(2)数据处理及分析:
所有数据采用均数±标准差表示。两组间比较采用双侧Student's t检验,三组及以上采用单因素方差分析。所有结果均采用GraphPad Prism 6软件进行绘图,以P<0.05为检验水准,当P<0.05为差异有统计学意义。
(3)实验结果:
Transwell小室细胞迁移实验的检测结果如图10所示。由图10可知,与阴性对照组(sh-Control组)相比,转染sh-APOBEC3B-1、sh-APOBEC3B-2的实验组细胞迁移能力明显降低(P<0.05)。由此说明,敲低APOBEC3B对前列腺癌细胞PC-3细胞的迁移能力有明显抑制作用。
Transwell小室细胞侵袭实验的检测结果如图11所示。由图11可知,与阴性对照组(sh-Control组)相比,转染sh-APOBEC3B-1、sh-APOBEC3B-2的实验组细胞侵袭能力明显降低(P<0.05)。由此说明,敲低APOBEC3B对前列腺癌细胞PC-3细胞的侵袭能力有明显抑制作用。

Claims (10)
1. APOBEC3B基因或蛋白的检测试剂在制备用于前列腺癌辅助诊断的产品中的应用。
2. APOBEC3B基因或蛋白的检测试剂在制备预测前列腺癌预后的产品中的应用。
3.根据权利要求1或2所述的应用,其特征在于,所述产品通过实时定量PCR、基因芯片、高通量测序平台、免疫组织化学或酶联免疫吸附检测样本中APOBEC3B基因或蛋白的表达水平。
4.根据权利要求3所述的应用,其特征在于,所述产品中含有扩增APOBEC3B基因的特异性引物、APOBEC3B基因杂交的探针与APOBEC3B蛋白特异性结合的抗体。
5. 根据权利要求4所述的应用,其特征在于,扩增APOBEC3B基因的特异性引物的核苷酸序列如SEQ ID NO.1和SEQ ID NO.2;所述抗体为多克隆抗体、单克隆抗体或单域抗体。
6.根据权利要求5所述的应用,其特征在于,所述样本包括组织、细胞、血清;所述产品为芯片、制剂或试剂盒。
7.抑制APOBEC3B表达和/或功能的物质在如下任一中的应用:
(a1)制备用于治疗前列腺癌的产品中的应用;
(a2)制备用于抑制肿瘤生长的产品,或抑制肿瘤生长中的应用;所述肿瘤为前列腺癌;
(a3)制备用于抑制前列腺癌细胞增殖的产品,或抑制前列腺癌细胞增殖;
(a4)制备用于抑制前列腺癌细胞侵袭的产品,或抑制前列腺癌细胞侵袭;
(a5)制备用于抑制前列腺癌细胞迁移的产品,或抑制前列腺癌细胞迁移。
8.根据权利要求7所述的应用,其特征在于,所述抑制APOBEC3B表达和/或功能的物质包括特异靶向APOBEC3B基因的shRNA或特异靶向APOBEC3B基因的siRNA或APOBEC3B蛋白的抑制剂。
9. 根据权利要求8所述的应用,其特征在于,所述特异靶向APOBEC3B基因的shRNA核苷酸序列如SEQ ID NO.3或SEQ ID NO.4所示。
10.一种治疗前列腺癌的药物,其特征在于,所述药物中含有抑制APOBEC3B表达和/或功能的物质;所述抑制APOBEC3B表达和/或功能的物质包括特异靶向APOBEC3B基因的shRNA或特异靶向APOBEC3B基因的siRNA或APOBEC3B蛋白的抑制剂。
