随着现代都市生活节奏的加快,人们饮食结构不合理、肠道菌群失调、精神心理压力大、久坐不动等现象逐渐凸显出来,导致便秘成为困扰众多现代人的健康问题。便秘若不加以防治,病程6个月以上者视为慢性便秘,慢性便秘可继发痔疮、肛裂等肛肠疾病,增加结肠癌、心血管疾病、阿尔茨海默病等疾病的发生风险,增加患者的经济和心理负担。
目前长期便秘的治疗主流是以药物为主的经验治疗,而长期药物通便会导致恶心、腹胀、腹部痉挛和呕吐等不良反应,另外还会产生疗效减退、医疗资源耗费等问题,因此探寻出一种安全、有效缓解或治疗便秘的非药物的组合物具有重大的社会意义。
发明内容
本发明的目的,旨在提供一种具有缓解便秘功能的组合物,以减少药物治疗便秘带来的副作用,改善肠道健康,有效缓解便秘。
本发明的另一目的,是提供上述具有缓解便秘功能的组合物的制备方法,以令制备出的组合物通便效果更佳。
为了实现上述目的,本发明采用的技术方案是:
一种具有缓解便秘功能的组合物,制成它的有效成分的原料,以重量份数计包括:乌梅粉15~25份、硫酸镁0.5~2份、可溶性大豆多糖6~10份、白萝卜部位粉45~55份、秋葵粉5~15份、低聚异麦芽糖8~12份、复配益生菌粉1~5份;其中,所述白萝卜部位粉为白萝卜叶粉、白萝卜籽粉、白萝卜粉中的至少一种;所述复配益生菌由植物乳杆菌、乳双歧杆菌和鼠李糖乳杆菌按质量比为1:0.5~3:0.5~3复配而成。
白萝卜,又称莱菔,既是人们喜食的蔬菜又具有很高的营养价值和药用价值。萝卜的种子、叶子和根均具有多种药理活性。白萝卜中的膳食纤维含量是非常可观的,尤其是叶子中含有的植物纤维更是丰富。这些植物纤维可以促进肠胃的蠕动,消除便秘。经分析白萝卜叶的营养价值和药用功能绝不逊色于萝卜。但是,现阶段白萝卜叶不是被菜农扔掉,就是被用于加工为饲料,附加值很低,进一步开发利用的市场前景广阔。
乌梅里含有茶酸、苹果酸能促进肠蠕动,把适量的水分导引到大肠,形成粪便排出体外;可溶性大豆多糖是双歧杆菌促进因子,能促进人体肠道内双歧杆菌的繁殖增长,调节肠内菌群,促进肠道正常蠕动,预防和治疗便秘;硫酸镁因在肠道很少被吸收,因此有良好的导泻功能;秋葵营养丰富,含有大量的维生素、胡萝卜素,叶酸、膳食纤维等营养成份,可在肠胃内吸收水分膨胀,刺激肠胃蠕动,减少食物残渣在肠道内的停留时间,起到润肠通便的作用。
作为对发明的一种限定,所述植物乳杆菌活菌数为1010~1011CFU/g;
所述乳双歧杆菌活菌数为1010~1011CFU/g;所述鼠李糖乳杆菌活菌数为1010~1011CFU/g。
本发明还提供了一种具有缓解便秘功能的组合物的制备方法,该制备方法包括依次进行的以下步骤:
S1:称取乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜部位粉、秋葵粉、低聚异麦芽糖、植物乳杆菌、乳双歧杆菌、鼠李糖乳杆菌,搅拌混合均匀,制得物料A;
S2:物料A加入润湿剂溶液,过筛造粒,制得物料B;
S3.物料B进行冷冻干燥,即得具有缓解便秘功能的组合物。
作为对制备方法的一种限定,所述步骤S2中润湿剂为浓度为0.5~2%的灭菌的胶溶液;物料A与灭菌的胶溶液的质量比为10:0.5~1.5。
作为对制备方法的进一步限定,所述灭菌的胶溶液由果胶、明胶、阿拉伯胶、瓜尔胶中的至少一种与水配制而成。
作为对制备方法的另一种进一步限定,所述步骤S2中物料B的粒径为20~30目。
由于采用了上述技术方案,本发明与现有技术相比,所取得的技术进步在于:
①本发明通过大量实验得到了一种缓解便秘功能的组合物,该组合物属于食品、保健食品领域,安全有效,无药物治疗带来的不良反应和毒副作用;
②本发明提供的具有缓解便秘功能的组合物中添加了适宜比例的植物乳杆菌、乳双歧杆菌、鼠李糖乳杆菌三种益生菌,添加了可溶性大豆多糖、低聚异麦芽糖作为益生元,添加了富含膳食纤维的秋葵粉和促进肠道蠕动的乌梅粉,添加了具消食导滞作用的食源性植物白萝卜的不同部分粉,还添加了兼具营养强化剂和导泻功能的镁盐,经精心组方、科学配伍,各种原料相互协同改善患者肠道微生态,共奏缓解便秘之功效;
③本发明提供的具有缓解便秘功能的组合物的制备方法中采用胶体包埋造粒技术,令菌体与外界隔离,可在一定程度上保护益生菌免受胃酸环境和相关因素影响,改善益生菌存活率,进而保证益生菌的生物活性。
综上所述,本发明提供食用安全的缓解便秘功能的组合物经精心组方、科学配伍,各种原料相互协同改善患者肠道微生态,共奏缓解便秘之功效;制备时采用胶体包埋技术,不仅令工艺简单、合理,还可改善益生菌存活率,保证益生菌的生物活性。
本发明适用于制备具有缓解便秘功能的组合物,产品适用于儿童、成人、老年人食用。
具体实施方式
下面通过具体实施例对本发明作进一步详细说明。应当理解所描述的实施例仅用于解释本发明,并不限定本发明。
实施例1-6一种具有缓解便秘功能的组合物
实施例1-6分别为一种具有缓解便秘功能的组合物,制成其有效成分的原料相同,不同之处在于不同实施例中原料的用量不同,不同实施例原料配比具体详见表1。

实施例1-6一种具有缓解便秘功能的组合物的制备方法按照以下步骤进行:
S1.称取乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜部位粉、秋葵粉、低聚异麦芽糖、植物乳杆菌、乳双歧杆菌、鼠李糖乳杆菌,搅拌混合均匀,制得物料A;
S2:物料A加入灭菌的胶溶液,过筛造粒,制得物料B;
S3.物料B进行冷冻干燥,即得具有缓解便秘功能的组合物。
制备一种具有缓解便秘功能的组合物的过程中,不同制备步骤各项工艺参数见下表2。

实施例7具有缓解便秘功能的组合物耐酸特性试验
本实施例为具有缓解便秘功能的组合物耐酸特性试验,具体方法如下:
S1.人工胃液的制备
取10%的盐酸加去离子水稀释,分别调节pH为2.0、3.0、4.0,加入胃蛋白酶使其终浓度为1%,再用盐酸调整溶液的pH为2.0、3.0、4.0。在无菌操作台中,用0.22μm的微孔滤膜过滤后备用;
S2.对比实施例的制备
对比实施例1与实施例4的配方相同,其不同之处在于步骤S2造粒时选用10kg灭菌水代替,其余制备步骤和工艺参数不做变动。
S3.耐酸性实验
分别取0.5g实施例4和对比实施1制备的产品,分别与4.5mL的不同pH值的人工胃液混合,于37℃培养箱中静置培养,分别于2h、4h后取样,稀释涂布平板后37℃培养24h,测定菌落数。以未经人工胃液处理的菌落数作为对照,计算存活率,不同制备工艺制备的具有缓解便秘功能的组合物耐酸性实验结果见下表表3。

由表3实验结果可知,制备时选用胶体包埋技术,益生菌被包裹在胶体内,在口服消化时避免菌体与胃酸接触,能有效提高益生菌活菌数及其稳定性。两种造粒方式对比,pH=2时胶体造粒较水造粒制备的产品益生菌存活率有数量级级别的提升;在pH=3和pH=4时,实施例4制备的产品益生菌存活率均超过90%,较对比实施例1制备的产品益生菌存活率有很大提升。
实施例8具有缓解便秘功能的组合物对便秘小鼠的影响
本实施例是为了验证配方中添加的乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜部位粉、秋葵粉、低聚异麦芽糖和复配益生菌的协同增效作用,同时考察白萝卜的不同部位在本配方中对于缓解便秘作用是否有差异,具体方法如下:
本实施例共制作五组对比实施例,其中对比实施例1为在实施例4的配方基础上仅不添加低聚异麦芽糖和复配益生菌,其余原料的种类、数量和制备方法不做变动;对比实施例2为在实施例4的配方基础上仅不添加乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜叶粉、秋葵粉,其余原料的种类、数量和制备方法不做变动;比实施例3为在实施例4的配方基础上仅不添加硫酸镁、秋葵粉、白萝卜叶粉,其余原料的种类、数量和制备方法不做变动;对比实施例4为在实施例4的配方基础上将白萝卜叶粉替换为白萝卜粉,其余原料的种类、数量和制备方法不做变动;对比实施例5为在实施例4的配方基础上将白萝卜叶粉替换为白萝卜籽粉,其余原料的种类、数量和制备方法不做变动。
本实施例选取纯种健康SD雄性大鼠90只,其中,SD雄性大鼠为3~4周周龄、体重60±10g;大鼠经适应饲养7天后,随机分为9组(每组10只),其中组1为正常组、组2为模型组、组3为阳性对照组,组4-组9为实验1组-实验6组。
除正常组外,其他各组每天按剂量5mg/kg腹腔注射洛哌丁胺,连续5天,建立小鼠便秘模型;其余各组以常规饲料喂养,组3灌胃培菲康1g/kg;组4-组9灌胃对比实施例1-5、实施例4制备的产品,灌胃剂量1g/kg;正常组和模型组灌胃等剂量的生理盐水,1次/天。喂食3天后,各组小鼠灌胃碳末混悬液(10mL/kg)再灌胃相应药物或产品,记录每只小鼠首粒黑便的时间,6h内排便总数和黑便数,测定粪便含水率,具体结果见表4。

由上表4可知,6组实验组灌胃小鼠后均能起到缓解便秘的作用。比较实验1组、实验2组和实验3组实验数据,可知益生菌的加入能缩短排黑便时间,增加排便粒数,提高粪便含水率;比较实验2组、实验3组、实验4组实验数据可知,在本实验范围内随着乌梅粉、大豆多糖、硫酸镁、白萝卜叶粉和秋葵粉添加量的增加,小鼠排黑便时间逐渐缩短,排便粒数增加,粪便含水率提高;比较实验4组、实验5组、实验6组和阳性对照组实验数据可知,三种产品与阳性对照组结果无明显差异性,实验4组、实验5组和实验6组采用萝卜不同部位,对缓解小鼠便秘效果无明显差异,可见萝卜根、籽、叶各部位均能有效缓解小鼠便秘。
实施例9便秘人群试服具有缓解便秘功能的组合物试验
本实施例为便秘人群试服试验,其目的在于考察对比实施例与缓解便秘功能组合物缓解便秘的功效,具体方法如下:
S1.确定入选标准
年龄18-60岁;符合便秘罗马Ⅲ标准;病程最少半年;近一周内未服用影响胃肠道动力药物;诊断IBS依据不足;结肠镜或结肠造影检查排除肠道器质性病变及由全身性疾病引起的便秘;近三月满足以下两项或两项以上症状:①至少25%的排便感到费力②至少25%的排便为干球状便或硬便③至少25%的排便有不尽感④至少25%的排便有肛门直肠的阻塞感⑤排便次数每周少于3次。
S2.试服方法
选取符合入选标准的试服人员110人,随机分3组,3组受试者每天早饭30min后温水冲服:A组(38人,口服对比实施例1产品),B组(31人,口服对比实施例3产品),C组(41人,口服实施例4产品),每次一袋,3g/袋,连续服用14天。试服期间要求受试者每天根据自身情况填写试服情况记录表。
S3.结果分析
通过试服后,受试者每周排便次数大于3次,每次排便时间小于15min,粪便硬度偏软,排便感觉轻松,视为试服有效结果;试服人员的试服结果经统计见表5。

由表5试验结果可知,虽对比实施例1中仅添加乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜叶粉、秋葵粉的对比实施例1,对比实施例3中仅添加乌梅粉、可溶性大豆多糖、低聚异麦芽糖、复配益生菌粉,这2种产品也能使患者便秘症状得到缓解,但有效率分别为71.05%和80.65%,可见有效率偏低;实施例4制备的产品能有效缓解患者便秘症状,有效率高达92.68%,有效率显著高于两对比实施例。
Claims (4)
1.一种具有缓解便秘功能的组合物,其特征在于,制成它的有效成分的原料,以重量份数计包括:乌梅粉15~25份、硫酸镁0.5~2份、可溶性大豆多糖6~10份、白萝卜部位粉45~55份、秋葵粉5~15份、低聚异麦芽糖8~12份、复配益生菌粉1~5份;
所述白萝卜部位粉为白萝卜叶粉、白萝卜籽粉、白萝卜粉中的至少一种;
所述复配益生菌由植物乳杆菌、乳双歧杆菌和鼠李糖乳杆菌按质量比为1:0.5~3:0.5~3复配而成;
其中,所述植物乳杆菌活菌数为1010~1011CFU/g;
所述乳双歧杆菌活菌数为1010~1011CFU/g;
所述鼠李糖乳杆菌活菌数为1010~1011CFU/g;
所述组合物的制备方法包括依次进行的以下步骤:
S1:称取乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜部位粉、秋葵粉、低聚异麦芽糖、植物乳杆菌、乳双歧杆菌、鼠李糖乳杆菌,搅拌混合均匀,制得物料A;
S2:向物料A中加入润湿剂溶液,过筛造粒,制得物料B;
S3.物料B进行冷冻干燥,即得所述具有缓解便秘功能的组合物;
所述步骤S2中润湿剂为浓度为0.5~2%的灭菌的胶溶液;
物料A与灭菌的胶溶液的质量比为10:0.5~1.5。
2.根据权利要求1所述的一种具有缓解便秘功能的组合物的一种制备方法,其特征在于,该制备方法包括依次进行的以下步骤:
S1:称取乌梅粉、硫酸镁、可溶性大豆多糖、白萝卜部位粉、秋葵粉、低聚异麦芽糖、植物乳杆菌、乳双歧杆菌、鼠李糖乳杆菌,搅拌混合均匀,制得物料A;
S2:向物料A中加入润湿剂溶液,过筛造粒,制得物料B;
S3:物料B进行冷冻干燥,即得所述具有缓解便秘功能的组合物;
所述步骤S2中润湿剂为浓度为0.5~2%的灭菌的胶溶液;
物料A与灭菌的胶溶液的质量比为10:0.5~1.5。
3.根据权利要求2所述的具有缓解便秘功能的组合物的一种制备方法,其特征在于,所述胶为果胶、明胶、阿拉伯胶、瓜尔胶中的至少一种。
4.根据权利要求2所述的具有缓解便秘功能的组合物的一种制备方法,其特征在于,所述步骤S2中物料B的粒径为20~30目。
