本发明公开了一种具有健脾胃益肠道功能的八珍益生菌组合物,所述八珍益生菌组合物包括中草药提取物1‑9份,可用于食品的肠道益生菌0.01‑2份,益生元0.1‑5份。本发明有效调节脾胃功能,达到健脾祛湿、益气养胃、滋肾益精之功效;加入可食用的肠道益生菌,改善肠道微生态环境,提高肠粘膜吸收能力;另加入有利于肠道健康的益生元,使肠道环境更适宜益生菌生存;益生菌、益生元协同中草药精华,从多方位更好地调理脾胃肠道功能,缓解脾胃虚弱、食欲减退、消化不良、腹痛腹泻等症状。

本发明属于中草药与营养食品领域,尤其涉及一种具有健脾胃益肠道功能的八珍益生菌组合物。
背景技术
“脾胃为后天之本”,在中医里,脾胃居重要的地位。脾胃在五行中属土,位于中焦,共同承担着将食物运化并生成气血的重任,周身的精、气、血、津液都必须依靠脾胃来供给。脾胃是健康的根本,若脾胃虚弱,会进一步影响其他脏器如心、肝、肺、肾的功能,引起五脏不调;并且脾胃虚弱易遭外邪入侵,感染幽门螺旋杆菌等细菌的风险升高,进而引起胃炎、胃溃疡等疾病,甚至引发胃癌。肠道是人体最大的消化器官,人体营养的99%需要靠肠道吸收,同时也是最易受到外界压力影响的器官。在抗生素治疗、旅行、饮食不佳以及快节奏的生活方式压力下,肠易激综合症(IBS)等肠道疾病正困扰着成年人肠道健康;同时,老年人随着年龄增长,肠道中的益生菌群比例也逐渐减少,肠道内微生态环境不良造成的腹痛、腹胀、腹泻、便秘随之出现。只有快速地重建肠道内微生态环境,才可以有效地缓解症状。研究证明中医脾胃与肠道正常菌群之间存在着十分密切的关系,脾胃虚弱反映了机体脾、胃、肠生理机能不足或异常,此时肠道的微生态系统会受到严重的破坏,大肠杆菌等条件性病原菌大量增殖,双歧杆菌、乳杆菌等益生菌数量显著下降;同时菌群失调又反过来影响机体对营养物质的吸收,降低机体免疫力,削弱肠道的屏障功能,进一步加重脾胃疾病。因此,脾、胃、肠作为一个有机整体为人体的健康提供有效保障,能够对其进行综合调理的健脾胃益肠道的产品是目前市场的需求。
“八珍”来源于传统中医方剂名“八珍汤”,最早八珍汤是传统中医流传下来的药方,最早出现记载于“正体类要”。八珍汤全方八药,实为四君子汤和四物汤的复方,即人参、白术、茯苓、甘草、当归、熟地黄、芍药及川芎。该方所治气血两虚证多由久病失治、或病后失调、或失血过多而致,病在心、脾、肝三脏。心主血,肝藏血,心肝血虚,故见面色苍白、头晕目眩、心悸怔忡、舌淡脉细;脾主运化而化生气血,脾气虚,故面黄肢倦、气短懒言、饮食减少、脉虚无力。治宜益气与养血并重。方中人参与熟地相配,益气养血,共为君药;白术、茯苓健脾渗湿,助人参益气补脾;当归、白芍养血和营,助熟地滋养心肝,均为臣药;川芎为佐,活血行气,使地、归、芍补而不滞;炙甘草为使,益气和中,调和诸药。用法中加入姜、枣为引,调和脾胃,以资生化气血,亦为佐使之用。该配方经过六百余年的检验,具有益气补血、健脾养胃之功效。近年来,国医大师吸收其精髓,在行医数十年中精心调配出健脾养胃的“八珍”配方,具有良好的健脾祛湿、益气养胃、滋肾益精的功效。
益生菌是一类对人和动物有益的活性微生物,定植于人体肠道、生殖系统内,可调节肠道微生物的平衡,增强免疫应答,有效抑制致病菌在肠道中的增殖,促进营养物质的消化和吸收,减少腹泻与肠道炎症,优化肠道环境,提高机体的免疫功能。益生元是益生菌有效的增殖因子,在通过上消化道时,大部分不被消化而能被肠道菌群所发酵,能刺激有益菌群的生长,从而有效改善肠道内环境,使之更适宜益生菌生存,并提高肠粘膜吸收能力。益生元在当前是功能性食品的热点,近年来我国开展的“国家公众营养改善项目”已把益生元作为重点产品进行推广。
中国专利申请第201610408040.9号公开了益生菌组合物及其制备方法,该组合物由下述重量百分比的物质制备而成:嗜酸乳杆菌菌粉1%-3%、干酪乳杆菌菌粉0.5%-1.5%、乳双歧杆菌菌粉1%-3%、甘露低聚糖5%-8%、圆苞车前子粉3%-5%、低聚果糖40%-60%、玉米低聚肽粉2%-5%、菊粉3%-5%、蓝莓粉5%-8%和麦芽糊精10%-25%。这一技术方案的缺陷:一是主要功效原料为益生菌和益生元,仅对肠道功能健康起作用,未能对脾胃功能进行调节;二是并无添加中草药原料,使得该配方功能性不强,调理效果不佳。
发明内容
本发明的目的是为了解决现有技术中存在的缺点,而提出的一种具有健脾胃益肠道功能的八珍益生菌组合物。
为了实现上述目的,本发明采用了如下技术方案:

益生菌:本发明所述的益生菌为青春双歧杆菌、动物双歧杆菌(乳双歧杆菌)、两歧双歧杆菌、短双歧杆菌、婴儿双歧杆菌、长双歧杆菌、嗜酸乳杆菌、干酪乳杆菌、卷曲乳杆菌、德氏乳杆菌、保加利亚亚种(保加利亚乳杆菌)、德氏乳杆菌乳亚种、发酵乳杆菌、格氏乳杆菌、瑞士乳杆菌、约氏乳杆菌、副干酪乳杆菌、植物乳杆菌、罗伊氏乳杆菌、鼠李糖乳杆菌、唾液乳杆菌、嗜热链球菌、乳酸片球菌。其作用是能够调节调节肠道菌群,抑制病原菌的生长,预防和减轻腹泻、便秘,改善肠道易激综合症(IBS)症状;还可以减少胆固醇的吸收,并且能够在肠道内合成维生素、氨基酸,提高机体对钙、磷、铁和维生素D的吸收利用;且能预防抗生素的副作用,提高宿主对放射线的耐受性,显著增加肠粘膜免疫球蛋白A(SIgA)含量,对机体肠粘膜局部免疫具有调节作用。
益生元:本发明所述益生元为菊粉、酵母、低聚果糖、低聚木糖、低聚半乳糖、低聚异麦芽糖、低聚乳果糖、大豆低聚糖、水苏糖、海藻糖。其作用是可以促益生菌肠道定植、肠道收敛,不被大多数的肠道腐败菌利用的同时促进人体内有益细菌(如乳杆菌、双歧杆菌)生长繁殖,从而可以抑制腐败菌生长,有助于改善和维持肠道正常功能;减轻肝脏负担、提高营养素的吸收率,特别是能够改善机体对钙、铁、锌等金属离子的吸收;此外还可代谢为酸性产物如醋酸盐和乳酸盐,使结肠内pH值降低,抑制肠道有害细菌的生长;使小肠内水电解质滞留在肠腔,产生高渗效果,刺激肠蠕动减少结肠运载时间,促进肠道内毒素及其他毒性物质的排出从而改善便秘症状。
具体实施方式
下面将结合具体实施例来进一步解说本发明。
具体实施例1
一种健脾胃益肠道的八珍益生菌组合物,由以下重量分数的组分混合均匀,即得。
上述组合物建议每次服用量10g,每日服用两次。
取上述八珍益生菌组合物,利用动物实验研究其对脾虚大鼠胃肠功能的调节作用。
1、八珍益生菌组合物对脾虚模型大鼠胃功能的影响
实验方法:
1)对胃液量的影响:取SD雄性大鼠40只,随机分成4组,每组10只,即模型对照组(NS),八珍益生菌组合物高、中、低剂量组(设计为人体推荐量的15倍、10倍、5倍,受试剂量分别为2.55g/kg·BW,1.7g/kg·BW,0.85g/kg·BW,下同)。每组按4g/kg·BW大黄水煎液连续灌胃7d,7d后各大鼠出现毛黄体瘦、便溏、尿黄、食量下降等脾虚症状,作为脾虚模型大鼠使用。分别对每组大鼠以同等剂量进行灌胃6d,末次给药后均禁食24h,但不禁水,第7d再灌胃一次,5h后用戊巴比妥钠(50mg/kg)麻醉,取全胃,倾出胃内容物并过滤、离心处理并精确记录胃液量。
2)对胃液总酸度、总酸排出量的影响:取上述方法制作的胃液1mL,加酚红指示剂1滴,用0.01mol/L NaOH滴定,直到胃液先呈黄色再转红色2s内不消失为终点。计算胃液总酸度及2h总酸排出量(总酸度mmol/L=消耗NaOH的量,2h总酸排出量mmol/L=总酸量×胃液量)。
3)对胃蛋白酶的影响:采用Mett毛细玻管法。将Mett毛细玻管洗净烤干,用新鲜蛋清打匀后用纱布过滤,将上述毛细玻管利用虹吸作用灌满蛋清(管内无气泡),然后放在85℃热水皿中使蛋白凝固,冷却后取出,用石蜡将管两端封固,贮冰箱中备用。取上清胃液1mL放入50mL三角瓶中,加0.5mol/L盐酸溶液15mL摇匀,放入已去两端石蜡封固部分的蛋白管连根,在37℃恒温箱中孵育24h,后用标尺测量蛋白管两端透明部分的长度(mm),以四端之值求其平均值。U(胃蛋白酶单位=平均值2)×16。
实验结果:
实验结论:与模型对照组比较,八珍益生菌组合物的高、中、低剂量组对脾虚模型大鼠的胃液分泌各项指标如总胃液量、总酸度、总酸排出量以及胃蛋白酶含量均有显著提高,作用明显。
2、八珍益生菌组合物对脾虚模型大鼠肠道功能的影响
实验方法:
1)对脾虚大鼠小肠运动功能的影响:取SD雄性大鼠40只,随机分成4组,每组10只,即模型对照组(NS),八珍益生菌组合物高、中、低剂量组(设计为人体推荐量的15倍、10倍、5倍,受试剂量分别为2.55g/kg·BW,1.7g/kg·BW,0.85g/kg·BW,下同)。每组按4g/kg·BW大黄水煎液连续灌胃7d后,各大鼠出现毛黄体瘦、便溏、尿黄、食量下降等脾虚症状,作为脾虚模型大鼠使用。停食24h后,模型对照组用含有10%碳末的NS灌胃,其余各组用含有10%碳末的等剂量受试物灌胃。灌胃30min后处死大鼠,打开腹腔,分离肠系膜,减去上端至油门、下端至回盲肠部的肠管,置于平台上,轻轻将小肠拉成直线,测量小肠总长度,从幽门至碳末前沿的长度作为碳末在肠内推进的距离,以下公式计算碳末推进率;碳末推百分率%=碳末在肠内推进的距离(cm)/小肠总长度(cm)。
2)对模型大鼠肠道菌群的影响:按上述方法制作脾虚模型大鼠,灌胃7d后无菌采大鼠新鲜粪便,用无菌NS稀释后取合适稀释度接种在相应的选择培养基上培养,通过菌落形态和革兰氏染色镜检来计数培养平板上的双歧杆菌、肠杆菌、乳杆菌、产气荚膜梭菌的数量,计算每g粪便4种指标菌的数量。
实验结果:
实验结论:与模型对照组比较,实施例1中的八珍益生菌组合物的高、中、低剂量组能促进脾虚模型大鼠的肠道推进功能,并且能促进肠道益生菌双歧杆菌、乳杆菌的定殖,降低肠道内有害菌如肠杆菌、产气荚膜梭菌的数量,效果显著。本实施例1的八珍益生菌组合物具有健脾胃益肠道的功效。
