Abstract
本发明涉及生物信息学技术领域,尤其涉及一种鉴别检疫实蝇有效热处理的方法及其应用。所述方法包括:通过代谢物的含量的差异鉴别经过检疫和未经过检疫的实蝇;所述代谢物包括酪氨酸、3‑羟基犬尿氨酸、4‑羟基马尿酸、腺苷酸、多巴、海藻糖、苹果酸、高丝氨酸、葡萄糖、延胡索酸、黄尿酸、甘露醇、琥珀酸、肌醇、苏糖酸、松二糖、尿酸、甲硫氨酸、丝氨酸、油酸、硬脂酸、棕榈油酸、羟基乙酸、甘氨酸、棕榈酸、腐胺、乳酸、氨基丙二酸、β‑羟基丁酸、α‑羟基戊二酸、景天庚酮糖‑7‑磷酸或腺苷中的一种或多种。本发明提供了多个可以用于鉴别检疫实蝇的标志物,其可以有效分辨出实蝇是否经过热处理。
Description
技术领域
本发明涉及生物信息学技术领域,尤其涉及一种鉴别检疫实蝇有效热处理的方法及其应用。
背景技术
截止目前,热处理技术因其环保、无残留、安全性高,逐渐重新引起人们的关注。热处理技术也随之开始了新一轮的蓬勃发展。热处理技术包括热水、蒸热、强制热空气和干热处理技术等。这些技术也逐步应用于检疫处理中。但是对于番石榴实蝇,南亚果实蝇、粉蚧等的热处理的检疫标准,目前尚未有明确的标准。
所以,快速判别幼虫是否进行了检疫处理,以较好地防范外来有害生物的入侵,对于保障生物安全有重要的理论意义和实用价值。需要一种快速可靠的方法来鉴定待检疫的货物是否进行过检疫处理。
发明内容
为了解决现有技术存在的问题,本发明的目提供一种鉴别检疫实蝇有效热处理的方法及其应用。
第一方面,本发明提供一种鉴别检疫实蝇有效热处理的方法,包括:
通过代谢物的含量的差异鉴别经过检疫和未经过检疫的实蝇;
所述代谢物包括酪氨酸、3-羟基犬尿氨酸、4-羟基马尿酸、腺苷酸、多巴、海藻糖、苹果酸、高丝氨酸、葡萄糖、延胡索酸、黄尿酸、甘露醇、琥珀酸、肌醇、苏糖酸、松二糖、尿酸、甲硫氨酸、丝氨酸、油酸、硬脂酸、棕榈油酸、羟基乙酸、甘氨酸、棕榈酸、腐胺、乳酸、氨基丙二酸、β-羟基丁酸、α-羟基戊二酸、景天庚酮糖-7-磷酸或腺苷中的一种或多种。
进一步地,所述经过检疫为经过合格热处理,所述未经过检疫包括未经过热处理以及热处理程度不合格。热处理程度不合格的情况下,会导致检疫结果不准确,因此只有经过合格热处理流程的实蝇才被认为是经过检疫的实蝇。
进一步地,所述代谢物为酪氨酸和/或3-羟基犬尿氨酸。
进一步地,采用提取液处理待鉴定实蝇,后进行代谢组学检测,根据检测结果判断所述待鉴定实蝇是否经过检疫;
所述提取液包括甲醇、乙腈、水和所述代谢物的同位素标记内标混合物。
进一步地,所述提取液中甲醇、乙腈和水的体积比为(2~4):(2~4):(1~2)。
进一步地,所述采用提取液处理待鉴定实蝇包括:
将所述带鉴定实蝇在38~40℃的条件下处理4~6小时得到检测样品;
后将所述检测样品加入所述提取液研磨处理,后进行超声和离心;
所述检测样品和所述提取液的质量体积比为1:(10~100)。
进一步地,所述根据检测结果判断所述待鉴定实蝇是否经过检疫包括:
比较所述待鉴定实蝇和未经过检疫的实蝇的代谢组学检测结果,若检测结果中所述代谢物的含量有显著异性差异,即判断所述述待鉴定实蝇为经过检疫的实蝇。
进一步地,通过t检验、ROC曲线和/或Log2折叠倍数判断检测结果中所述代谢物的含量之间是否有显著性差异。
进一步地,所述代谢组学检测结果为超高效液相色谱串联质谱的检测结果。
进一步地,所述检疫为经过热处理技术的检疫流程。
本发明进一步提供所述方法在鉴别实蝇是否经过热处理中的应用。
本发明具有如下有益效果:
本发明将代谢组学检测应用于检疫实蝇的鉴别,即通过检测几种代谢物质的含量,即可鉴别实蝇是否经过有效热处理,进而可以确认实蝇是否经过检疫,可以快速判别实蝇幼虫是否经过了检疫处理,以较好地防范外来有害生物的入侵,在保障生物安全的领域有重要的理论意义和实用价值。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。
实施例1
1、检疫性实蝇幼虫热处理
将实蝇3龄幼虫放入含有保湿滤纸的玻璃瓶中,封口后放入38℃的水浴锅中,持续5h。对照组幼虫为裸虫,置于25℃的饲养环境中。然后用液氮将热处理组和对照组的幼虫速冻,置于-80℃冰箱储存。
2、样本的代谢组前处理
称取50mg样品,加入1000μL提取液(甲醇:乙腈:水=2:2:1(V/V),含同位素标记内标混合物);
35Hz研磨处理4min,超声5min(冰水浴);
重复研磨-超声步骤2~3次;
-40℃静置1h;
将样品4℃,12000rpm(离心力13800(×g),半径8.6cm)离心15min;
取上清于进样瓶中上机检测;
所有样品另取等量上清混合成QC样品上机检测。
3、上机检测
本发明使用Vanquish(Thermo Fisher Scientific)超高效液相色谱仪,通过Waters ACQUITY UPLC BEH Amide(2.1mm×100mm,1.7μm)液相色谱柱对目标化合物进行色谱分离。液相色谱A相为水相,含25mmol/L乙酸铵和25mmol/L氨水,B相为乙腈。样品盘温度:4℃,进样体积:3μL。
Thermo Q Exactive HFX质谱仪能够在控制软件(Xcalibur,Thermo)控制下进行一级、二级质谱数据采集。详细参数如下:Sheath gas flow rate:30Arb,Aux gas flowrate:25Arb,Capillary temperature:350℃,Full ms resolution:60000,MS/MSresolution:7500,Collision energy:10/30/60 in NCE mode,Spray Voltage:3.6kV(positive)或-3.2kV(negative)。
4、数据处理
原始数据经ProteoWizard软件转成mzXML格式后,使用自主编写的R程序包(内核为XCMS)进行峰识别、峰提取、峰对齐和积分等处理,然后与BiotreeDB(V2.1)自建二级质谱数据库匹配进行物质注释,算法打分的Cutoff值设为0.3。
5、正交偏最小二乘方判别分析(OPLS-DA)
采用正交偏最小二乘方判别分析(OPLS-DA)过滤与模型分类不相关信号即正交信号,建立可靠的OPLS-DA模型。模型质量参数为:R2Y(cum)=0.998,Q2(cum)=0.4,模型质量良好。OPLS-DA得分图如图1所示,过滤掉与分类不相关的噪音信号后,两组样本在PC1(即t[1]P)上具有很好的代谢谱分离,即两组样本分别处于主成分(PC1,即t[1]P)的正负两侧。
6、差异性代谢物及其结构鉴定
由于过滤掉了不相关的正交信号,因而获得的差异性代谢物更加可靠。采用OPLS-DA模型第一主成分的VIP(Variable Importance in the Projection)值(阈值>1),并结合t检验(t-test)的p值(阈值0.05)来寻找差异性表达代谢物。差异性代谢物的定性方法为:搜索NIST商业数据库(比较质谱和色谱保留时间RT或保留指数RI),以及采用标准物质数据比对确定。
结果显示,本发明提供的方法可以将热处理组与对照组之间的差异性分开,灵敏度和特异性高,代谢物有32个,其中19个代谢物下降,13个代谢物上升(表1),确认了热处理组和对照组的代谢差异,可用于橘小实蝇幼虫热处理有效性检测。在以后的检测中,只需要针对表1中的这32个代谢物进行以上代谢组学检测以及OPLS-DA分析即可。

*热处理组与对照组均值之比的对数值(以2为底),正号表示热处理组相对于对照组上升,负号表示下降。
在以上方案的基础上,本发明从这32种差异性代谢物之中进一步筛选出能够代表有效热处理的生物标志物,以在保证检测准确性的基础上,更进一步地简化检测程序。
7、代谢组学GC-MS分析后物质峰值鉴别热处理的ROC曲线分析及生物标志物质鉴定
根据代谢组学检测结果,热处理的诊断采用t-test检测,筛选对照与热处理组间有显著差异(P<0.05),以Log2为底,折叠倍数(Fold change)大于1的代谢物。同时,以(1-特异性)为横坐标,敏感度为纵坐标绘制ROC曲线。ROC曲线分析将某试验的灵敏度及特异性联系起来,是一种全面的、科学的评价检测项目的方法。曲线下面积(AUC)越大,诊断的价值越大,AUC接近0.5时,无诊断意义;AUC<0.7,表示诊断准确率较低;AUC在0.7-0.9,表示诊断准确性中等;AUC>0.9时,表示诊断有较高的准确性。
如表2所示,热处理组中,代谢物质酪氨酸(tyrosine)和3-羟基犬尿氨酸(3-hydroxykynurenine)经过ROC分析后AUC分别为1.00和0.97,且t-test检验P值<0.5,Log2的折叠倍数分别为1.53和1.30。该两种物质是橘小实蝇热处理的生物标志物。因此,判别是否进行热处理,且热处理是否有效,需要对比待检测组与对照组酪氨酸和3-羟基犬尿氨酸是否有显著差异,且该两种物质的峰面积或物质浓度的Log2的折叠倍数是否>1。

因此,在实际应用过程中,可仅仅检测酪氨酸和3-羟基犬尿氨酸的含量,并通过t检验、ROC曲线峰面积,以及Log2折叠倍数来判断是否有显著差异。当t检验结果P<0.05,为差异显著,ROC曲线的AUC>0.7,Log2折叠倍数大于1或小于-1时,则可判定样品经历了有效热处理。
实施例2
橘小实蝇幼虫热处理:将实蝇3龄幼虫放入含有保湿滤纸的玻璃瓶中,封口后放入38℃的水浴锅中,分别持续1、2、3、5h。对照组幼虫为裸虫,置于25℃的饲养环境中。然后用液氮将热处理组和对照组的幼虫速冻,置于-80℃冰箱储存。分别测定酪氨酸和3-羟基犬尿氨酸的含量后,进行t检验和Log2折叠倍数分析。各组数据分析结果见下表:

实验结果表明,只有经过38℃热水处理5h,酪氨酸和3-羟基犬尿氨酸的分析结果满足P<0.05且Log2折叠倍数大于1,证明样品经历了有效热处理。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (10)
1.一种鉴别检疫实蝇有效热处理的方法,其特征在于,包括:
通过代谢物的含量的差异鉴别经过检疫和未经过检疫的实蝇;
所述代谢物包括酪氨酸、3-羟基犬尿氨酸、4-羟基马尿酸、腺苷酸、多巴、海藻糖、苹果酸、高丝氨酸、葡萄糖、延胡索酸、黄尿酸、甘露醇、琥珀酸、肌醇、苏糖酸、松二糖、尿酸、甲硫氨酸、丝氨酸、油酸、硬脂酸、棕榈油酸、羟基乙酸、甘氨酸、棕榈酸、腐胺、乳酸、氨基丙二酸、β-羟基丁酸、α-羟基戊二酸、景天庚酮糖-7-磷酸或腺苷中的一种或多种。
2.根据权利要求1所述的方法,其特征在于,所述代谢物为酪氨酸和/或3-羟基犬尿氨酸。
3.根据权利要求1或2所述的方法,其特征在于,包括:
采用提取液处理待鉴定实蝇,后进行代谢组学检测,根据检测结果判断所述待鉴定实蝇是否经过检疫;
所述提取液包括甲醇、乙腈、水和所述代谢物的同位素标记内标混合物。
4.根据权利要求3所述的方法,其特征在于,所述提取液中甲醇、乙腈和水的体积比为(2~4):(2~4):(1~2)。
5.根据权利要求3或4所述的方法,其特征在于,所述采用提取液处理待鉴定实蝇包括:
将所述带鉴定实蝇在38~40℃的条件下处理4~6小时得到检测样品;
后将所述检测样品加入所述提取液研磨处理,后进行超声和离心;
所述检测样品和所述提取液的质量体积比为1:(10~100)。
6.根据权利要求3所述的方法,其特征在于,所述根据检测结果判断所述待鉴定实蝇是否经过检疫包括:
比较所述待鉴定实蝇和未经过检疫的实蝇的代谢组学检测结果,若检测结果中所述代谢物的含量有显著异性差异,即判断所述述待鉴定实蝇为经过检疫的实蝇。
7.根据权利要求6所述的方法,其特征在于,通过t检验、ROC曲线和/或Log2折叠倍数判断检测结果中所述代谢物的含量之间是否有显著性差异。
8.根据权利要求1-7任一项所述的方法,其特征在于,所述检疫为经过热处理技术的检疫流程。
9.根据权利要求6所述的方法,其特征在于,所述代谢组学检测结果为超高效液相色谱串联质谱的检测结果。
10.权利要求1-9任一项所述的方法在鉴别实蝇是否经过热处理中的应用。
