s1, preparing a nano analgesic: weighing 20 parts by weight of frankincense, 10 parts by weight of striga asiatica, 30 parts by weight of long pepper and 15 parts by weight of pepper, cleaning, baking and frying for 40min at the temperature of 110 ℃, then putting the materials into a grinder to grind to 50-80 meshes, adding water with the volume 5 times of that of the materials, boiling for 4h, filtering, carrying out steam flow concentration on the filtrate, adding the concentrated solution into a high-pressure homogenizer with the pressure of 12MPa, and homogenizing for 2h to obtain the nano analgesic drug;
s2, preparing a nano auxiliary medicament: weighing 10 parts of vitamin C, 5 parts of vitamin B complex, 10 parts of iron agent and 10 parts of medlar in parts by weight, drying the medlar, crushing the dried medlar into 60-80 meshes, adding the crushed medlar into a ball mill, adding the vitamin C, the vitamin B complex and the iron agent simultaneously, and carrying out ball milling for 3 hours to obtain a nano auxiliary medicament;

S4, preparing a nano preparation: mixing the nanometer analgesic medicine, the nanometer therapeutic medicine and the nanometer adjuvant medicine at a mass ratio of 2:2:4, stirring, and drying in a low temperature vacuum drying oven at 55 deg.C to obtain nanometer preparation. The rest of the procedure was the same as in example 2.
And (3) clinical trials:
220 oral patients (110 men and 110 women) aged between 20 and 55 years and continuously suffered from persistent diseases caused by excessive fire due to kidney deficiency, exuberance of stomach fire and the like for more than 2 weeks are recruited, 55 patients suffer from toothache and dental ulcer at the same time, 99 patients suffer from dental ulcer and 66 patients suffer from dental ulcer, the patients are randomly divided into 11 experimental groups, each group comprises 20 patients, each group simultaneously suffers from 5 toothache and dental ulcer, and 9 patients suffer from dental ulcer and 6 toothache, the preparations prepared in the examples 1-3 and the comparative examples 1-8 are respectively adopted for treatment twice a day with the dosage of 0.5g each time, the preparations are respectively used after morning getting up and before night sleep, the treatment is continuously carried out for 15 days, the preparations are not used and then continuously observed for 15 days, and the other patients are all normally taken and eaten. Clinical trial results the healing profile is shown in table 1, and the analgesic effect, the healing profile of the canker sores and the recurrence profile are shown in table 2.
Test investigation indexes are as follows:

and (4) invalidation: toothache symptoms are not improved and pain is hard to endure, oral ulcer is not healed after 15 days, and the appetite is poor.

The results in table 1 show that the nano preparation prepared by the nano preparation formula and the preparation method of the embodiments 1-3 of the invention can effectively treat toothache and canker sore diseases caused by kidney deficiency and exuberant fire, exuberant stomach fire and the like, the total effective rate reaches 100%, the cure rate reaches 90.9%, the wound healing speed is high, and no recurrence phenomenon exists in the near term.
The results of the comparative example 2 and the comparative examples 1 to 6 show that the nano preparation obtained by using the nano preparation has the advantages of obviously reduced treatment effect and the most obvious reduction of the number of cured people due to excessive or insufficient analgesic drugs, excessive or insufficient auxiliary drugs and excessive or insufficient treatment drugs, which indicates that the selected drugs are totally effective, but the optimal effect cannot be achieved due to unreasonable proportion, and the rehabilitation speed and the relapse condition are influenced.
The results of the comparative example 2 and the comparative examples 7 to 8 show that the preparation of the traditional Chinese medicine raw materials in the comparative example 7 after direct extraction has the advantages that the total effective rate is obviously reduced compared with the example 2, and the cure rate is 0; comparative example 8, the traditional Chinese medicine components are directly mixed after being prepared into the nano raw materials, and the therapeutic effect of releasing the traditional Chinese medicine components together is not as good as that of example 2, which shows that the preparation method of the invention can improve the absorption and utilization effects of the active ingredients of the medicine.

As can be seen from the results in table 2, the nano-preparation prepared by the nano-preparation formulation and the preparation method of the embodiments 1 to 3 of the present invention can relieve pain within 2.5 to 3.3min after use, can completely heal the oral ulcer wound within 4d, and does not recur after drug withdrawal, which indicates that the nano-preparation can rapidly relieve pain and promote wound healing, can effectively solve the symptoms of kidney deficiency, exuberant fire, exuberant stomach fire, etc., of the body, and can improve the disease resistance of the body.
Comparative example 1 has excessive pain-stopping drugs, and although the pain-stopping speed is high, the oral ulcer healing is slow, and the recurrence rate is high; in comparative example 2, the analgesic drug was too little, and although the recurrence rate was relatively low, the analgesic rate was significantly slowed; in comparative example 3, the adjuvant drug was excessive, and although the recurrence rate was relatively low, the analgesic rate and the healing rate of oral ulcer were slow; in comparative example 4, the recurrence rate is obviously increased due to the fact that the auxiliary drugs are too little; comparative example 5, which is overdose with a slow analgesic rate, although the recurrence rate is relatively low and the oral cavity heals faster; in comparative example 6, the pain-relieving time, the oral healing time and the recurrence rate of the drug are obviously increased when the drug is too little; the nano preparation formula can quickly relieve pain and promote wound healing, and can effectively solve the symptoms of toothache and oral ulcer.
In the comparative example 7, the pain relieving time, the oral healing time and the recurrence rate of the traditional Chinese medicine raw materials are obviously increased compared with those of the example 2; the traditional Chinese medicine components in the comparative example 8 are prepared into the nanometer raw materials and then directly mixed, so that the pain relieving time, the oral healing time and the recurrence rate are all improved compared with those of the example 2, but the effect is better compared with that of the comparative example 7, which shows that the nanometer preparation prepared by the preparation method of the invention is easier to be absorbed by organisms, and the absorption effect is better through layered release of the medicine.
In conclusion, the invention strictly controls the formula of each effective component of the nano preparation, and combines the preparation method of the invention, the obtained nano preparation has the total effective rate of 100 percent for treating toothache and oral ulcer diseases caused by internal kidney deficiency, exuberant fire, exuberant stomach fire and other reasons, the pain relieving speed and the wound healing speed are high, the recent relapse is avoided, and the effect is obvious.
Finally, it should be emphasized that the above-described preferred embodiments of the present invention are merely examples of implementations, rather than limitations, and that many variations and modifications of the invention are possible to those skilled in the art, without departing from the spirit and scope of the invention.
