背景技术
适配体(aptamer)是合成的单链(ss)DNA或RNA分子,形成特定的二级和三级结构。它们可以特异性地结合天然折叠蛋白质、毒素或其他具有高亲和力和特异性的细胞靶标。它们是非免疫原性的,但与抗体一样,适配体可以激活或抑制受体功能。它们体积小、稳定性好、成本效益高、化学合成高度可控,使适配体成为极具吸引力的治疗剂。因此,适配体被认为是用于诊断和治疗目的的有前景的单克隆抗体合成替代品。
多特异性适配体是两个或两个以上的适配体连接在一起,设计用于以高特异性和亲和力来特异性结合不同的表位。多聚(multimeric)特异性开启了广泛的研究、诊断和临床应用,包括将细胞重新定向到另一种细胞类型(例如,T细胞或NK细胞到肿瘤细胞),阻断两种不同的信号通路,双重靶向不同的疾病介体,以及向特定细胞提供有效载荷。在这些应用中,精确的靶向性以及在某些情况下影响特定细胞功能的能力是成功研究、诊断和治疗应用的重要决定因素。
发明内容
本发明提供了一种工程化抗原结合分子,其包括两个或多个连接在一起的并且能够特异性结合一个或多个癌细胞抗原和一个或多个免疫效应细胞抗原的不同适配体部分。
本发明的一个方面是将关注的适配体连接在一起的方法。在一些实施方案中,这可以通过点击化学来实现。在一些实施方案中,所述连接体(linker)的长度、连接体赋予靶向部分的灵活性或移动性,并且连接体的类型可影响免疫效应器细胞功能或对影响亲和力、特异性和/或构象的靶向适配体部分进行干扰。在一些实施方案中,所述连接体的选择可影响多特异性适配体的药代动力学和药效学性质。在一些实施方案中,所述连接体的选择可影响活性和安全性(例如,免疫原性)。在一些实施方案中,所述多特异性适配体的抗原结合部分可识别具有高亲和力和特异性的特异性抗原。
本发明的另一方面是一种多特异性抗原分子,其包括两个或多个具有不同靶标结合特异性的连接适配体。在一些实施方案中,所述多特异性适配体可结合表达靶向抗原的邻近细胞并将其带入其中。
在一些实施方案中,所述多特异性适配体允许将免疫效应细胞重定向至癌细胞。反过来,所述工程化的多特异性适配体与各自的靶向表位的结合允许免疫效应细胞被激活并发挥其抗癌杀伤功能。
在一些实施方案中,所述多特异性适配体的抗原结合部分可将表达CD3、CD8、CD4或其他T细胞特异性抗原的免疫效应T细胞重定向至其他关注的细胞靶标,例如CD19、上皮细胞粘附分子、CD20、CD22、CD123、BCMA、B7H3、CEA、PSMA、Her2、CD33、CD38、DLL3、EGF-R、MHCI类相关蛋白MR1或间皮素。

1.一种基于适配体的多特异性抗原结合分子,包括1)两个或更多个具有不同靶标结合特异性的靶标结合适配体区域,以及2)一个或多个连接适配体区域的连接体。

基于适配体的多聚结合分子的连接部分可以是单个适配体之间的一个或多个共价键,或者可以是合成的或天然存在的聚合物,例如碳氢化合物、聚醚、多胺、聚酰胺、腙、硫醚、酯、三唑、核酸、肽、碳水化合物或脂质。在某些实施方案中,所述连接部分不是肽。在某些实施方案中,所述基于适配体的多特异性分子不含肽,且不含多肽和蛋白质。所述连接部分还可以采取纳米级结构(例如聚合物、蛋白质、纳米粒子、纳米管、纳米晶体、纳米线、纳米带、纳米晶体、胶束或脂质体)或微型结构(例如微珠或细胞)或更大的结构(例如固体载体)的形式。优选地,所述连接部分是可生物降解的聚合物。所述连接部分可以是线性、分支的、环状或这些结构的组合的聚合物。所述连接部分还可以用作树枝状大分子(dendrimeric)结构或枢纽(hub)或星形结构(例如两个或多个适配体结合的核心结构)的骨架。对于非共价结合,两个或多个单独的适配体可以通过适配体之间的非共价相互作用或通过与连接部分的相互作用结合。例如,所述非共价相互作用可以是一个或多个氢键、离子键、疏水键、范德华相互作用或其组合。高亲和力结合对,例如链霉亲和素-生物素,可用于非共价连接基于适配体的多聚结合分子中的适配体。

任何用于制备选择针对靶标的适配体的合适方法都可用于获得所述基于适配体的多特异性分子的组分适配体。例如,可以通过指数富集配体的系统进化(SELEX)来识别适配体。例如,美国专利5,270,163中描述了SELEX,该专利通过引用并入本文。简单地说,SELEX从多个核酸(即候选适配体序列)开始,这些核酸包括与靶标接触的各种核苷酸序列。未结合的核酸与形成适配体靶复合物的核酸分离。然后对适配体-靶标复合物进行解离,扩增核酸,并在所需的多个循环中重复结合、分离、解离和扩增步骤,以产生与靶标亲和力逐渐提高的适配体群体。选择和扩增的周期可以重复,直到进一步重复该循环后,结合亲和力没有显著改善。
在识别出单个适配体之前,选择和扩增的循环可以被中断。在这种情况下,一个适配体群体被鉴定出来,其可以提供有关序列、结构或基序的重要信息,从而允许适配体与靶结合。这样的候选适配体群体还可以告知适配体的哪些部分对靶向结合不是关键的。然后,这些信息可以引导产生其他指向同一目标的适配体。由此产生的适配体可作为新一轮SELEX的输入,可能产生具有更好结合亲和力或其他关注的特征的适配体。
在一些实施方案中,包括多聚适配体构建体的候选适配体序列被创建出来,例如基于候选适配体的多特异性分子,然后将其作为多聚结构体进行另一轮选择。多聚候选适配体结构体可以通过将单个候选适配体部分与连接部分连接,并任选地使用此类结构体作为输入进行一轮或多轮SELEX。在一些实施方案中,通过一轮或多轮SELEX独立选择单个适配体,并最终与连接部分连接在一起。因此,单体适配体以及多聚适配体结构体的多聚化可以在SELEX程序之前、期间或之后进行。
本技术进一步提供了细胞重定向适配体(例如,多价适配体),其可在基于适配体的CAR免疫治疗系统中用作适配体桥,以及用于细胞的体内或体外基因修饰。本技术的适配体桥、细胞、试剂盒和方法可用于多种用途,包括作为治疗癌症(例如血液学或非血液学、单个细胞或实体瘤)、自身免疫疾病(例如关节炎、重症肌无力、天疱疮)、神经炎性疾病的免疫疗法、眼科疾病、神经退行性疾病(如ALS、亨廷顿病、阿尔茨海默病)、神经肌肉疾病(包括杜氏肌营养不良症、SMA)、感染性疾病(如HIV、HSV、HPV、HBV、埃博拉、结核病、隐球菌)和代谢性疾病(如1型糖尿病)。它们还可用于提供诊断试剂、试剂盒和用于此类免疫疗法的方法,包括成像、细胞运输分析、新免疫疗法的研发,以及与干细胞疗法(如HSCT)结合时提供预防。
本发明中“嵌合抗原受体细胞”或“CAR细胞”是已在体外或体内被操纵的转基因细胞(例如,T细胞、NK细胞、单核细胞或其他),其表达单链可变结构域(scFv)抗体,所述抗体通过茎或跨膜结构域融合到受体(如CD3-TCR)的胞内结构域,从而赋予细胞识别和结合一种或多种特异性抗原的能力,并激活细胞免疫反应(如杀死癌细胞或摧毁病毒感染的细胞)。

本技术的多聚适配体或连接适配体包括两个或多个由连接部分共价或非共价结合的适配体。根据本技术的一个实施方案,两个或多个适配体可形成CAR结合部分和靶标结合部分,其中每个部分包括一个或多个适配体。CAR结合适配体与免疫细胞(例如T细胞)中表达的CAR结合,并且在一些实施方案中激活免疫细胞,而在其他实施方案中(例如,当充当“杀死”开关时)不激活免疫细胞。该靶标是免疫治疗的预期靶标,即用于消除的细胞。因此,表达CAR的细胞和适配体桥拟作为免疫治疗(如CAR-T细胞治疗)中的一个系统一起使用。所述适配体桥与CAR以及与靶标的结合优选为高亲和力结合。所述靶标可以是蛋白质(如细胞表面受体蛋白)、细胞、小分子或核酸。所述靶标优选地位于靶细胞(例如癌细胞)的表面上,并且可以在受试者的其他细胞(正常细胞)上找到,也可能找不到。

在优选实施方案中,选择CAR结合适配体或适配体以特异性结合到具有肽新表位(PNE)亲和力的CAR的胞外结构域,即抗PNE CAR。由于PNE是受试者体内不存在的表位,表达抗PNE CAR的免疫细胞不会被内源性生物分子激活,而是等待给受试者注射适配体桥,作为免疫细胞的“开启”开关,并将表达CAR的细胞靶向所需抗原或携带抗原的细胞类型。表达CAR的免疫细胞的免疫激活和体内扩增可通过向受试者施用含有PNE的肽或单体形式的桥的CAR结合适配体或靶结合适配体来关闭,其中任何一种都会终止靶细胞对表达CAR的免疫细胞的激活。

实施例
实施例1.针对PSMA和CD3的双特异性适配体的制备。
A10 RNA适配体(SEQ ID NO:8)是一个39个核苷酸长的序列,已针对人类前列腺特异性膜抗原(PSMA)进行选择,并用作siRNA的前列腺特异性递送剂(McNamara等人2006-Dassie等人2009)。

抗PSMAA10和抗CD3适配体通过铜催化的点击反应进行异源二聚化,根据制造商的说明,使用Oligo2 click kit L(baseclick,诺伊里德,德国)在45℃下进行60min的点击反应。反应产物在100V的1X TBE缓冲液(Invitrogen)中以3%琼脂糖凝胶电泳分离30min。使用Bio-Rad成像系统观察凝胶,结果如图4A和4B所示。从凝胶中切下对应于二聚体适配体的凝胶切片,并在25mM NaCl-TE缓冲液中通过被动洗脱在8℃下提取核酸72h。通过标准醋酸钠沉淀回收双特异性适配体二聚体,再悬浮在无菌水中,并在-20℃下储存,直至使用。
实施例2.PSMA专用适配体A10的功能稳定性。
A10 RNA适配体的稳定性在Dulbecco的磷酸盐缓冲盐水(DPBS)中测定,该盐水含有5%的FBS或仅含FBS。生物素化适配体在85℃下变性5min,然后立即在冰块上冷却至4℃持续5min。然后在添加5%FBS的DPB或纯FBS中将适配体稀释至2μM的最终浓度。样品在37℃下培养10min、30min、1h、2h、4h或24h;对照样品含有新制备的适配体,未在37℃下培养。然后将100nM链霉亲和素PE添加到每个溶液中,并将适配体与PSMA阳性LNCaP细胞(人前列腺癌-ATCC CRL-1740)一起培养。然后,基于荧光阳性细胞数随37℃下培养时间的变化,使用YL-1通道上的流式细胞术测定适配体A10在含有5%FBS的DPBS缓冲液或纯FBS中的半衰期。测定结果如图5所示。在含有5%血清的DPBS缓冲液中培养的适配体A10稳定24h以内。在纯血清中测试时,在培养的前2h内,一半的结合活性丧失。
实施例3.测定抗PSMA×抗CD3双特异性适配体对细胞上表达的靶标的亲和力和特
异性。
流式细胞术检测抗PSMA×抗CD3双特异性适配体对细胞表达的靶蛋白的亲和力和特异性。这些研究是在CD3阳性的Jurkat(急性T细胞白血病人类细胞系-ATCC TIB-152)和CD3阴性的Ramos(Burkitt淋巴瘤人类细胞系-ATCC CRL-1596)上进行的,PSMA阳性LNCaP(人前列腺癌-ATCC CRL-1740)和PSMA阴性PC-3(人前列腺癌-ATCC CRL-1435)细胞,通过在SELEX缓冲液中与生物素化RNA/DNA适配体或在DPBS缓冲液中与添加5%FBS的RNA/RNA适配体孵育。细胞在使用前在RPMI-1640培养基(Gibco Invitrogen)中培养,并添加10%FBS(Gibco Invitrogen)和1%青霉素/司托霉素(Gibco Invitrogen)。实验前,将Jurkat、Ramos、LNCaP和PC-3细胞(2.5×105细胞/孔)接种在96孔板中,并以2500rpm离心2min。丢弃上清液,用200μL在37℃下预热的SELEX或DPBS-5%FBS缓冲液清洗颗粒细胞两次。每个清洗步骤之后,在2500rpm下离心2min。适配体在85℃下变性5min,并立即放置在4℃的冰块上5min。随后以两种不同浓度稀释试样范围:3、10、30、100和300nM(CD3结合分析)和30、100和300nM(PSMA结合分析),然后向每个溶液中添加100nM藻红蛋白标记的链霉亲和素(链霉亲和素PE、eBioscience)。将Jurkat、Ramos、LNCaP和PC-3细胞重新悬浮在适配体稀释液(100μL/孔)中,并在37℃下在5%CO2增湿环境中培养30min。作为对照,将细胞与CD3单克隆抗体(PE标记、OKT3人抗CD3、Invitrogen)、PSMA单克隆抗体(Alexa Fluor488标记、GCP-05人抗PSMA、Invitrogen)、PE链霉亲和素、单体适配体或各自的缓冲液一起孵育,无需额外试剂。培养后,以2500rpm的转速离心细胞2min,并丢弃含有未结合序列的上清液。用SELEX或DPBS-5%FBS缓冲液(200μL/孔)清洗颗粒细胞,并离心两次,以去除所有弱附着和非特异性附着的序列。然后在37℃、5%CO2增湿的环境中,用1mg/mL鲑鱼精子DNA溶液(100μL/孔)清洗细胞。30min后,以2500rpm的转速离心2min,取出鲑鱼精子溶液,并用SELEX或DPBS-5%缓冲液(200μL/孔)额外清洗细胞两次,然后离心。然后固定带有附加DNA或RNA序列的Jurkat、Ramos、LNCaP和PC-3细胞(BD CellFIX solution#340181),并通过流式细胞术(AttunNXT;Invitrogen,Inc.)在YL-1通道上计数荧光阳性细胞。
与PSMA阳性细胞结合的研究结果如图6A和6B所示。分析了三种RNA/DNA适配体(A10×CELTIC_1s、A10×CELTIC_19s、A10×CELTIC_core)和两种RNA/RNA适配体(A10×ARACD3-370006和A10×ARACD3-0010209)以及A10单体适配体。为了进行比较,还测定了受试试剂与PSMA阴性PC-3细胞的结合情况。在最高测试浓度下,观察到A10与PSMA阳性LNCaP细胞的剂量依赖性结合,但信号未达到饱和。信号强度与抗体对照组一样强。A10单体与PC-3细胞的残余结合仅在最高测试浓度下观察到。所有双特异性PSMA×CD3适配体与A10单体具有相似的结合特性,但由于与PSMA阴性细胞的残余结合减少,对靶阳性细胞的特异性提高。对于每个测试浓度,双特异性适配体的信号强度都优于A10单体的信号强度,这表明异二聚作用导致了亲和力的提高。
在另一个实验中,研究了相同适配体与CD3阳性Jurkat和CD-3阴性Ramos细胞的结合。参见图7A和7B。正如预期的那样,A10适配体没有与这两个细胞系结合。抗CD3单体与Ramos细胞的残余结合仅在最高测试浓度下观察到。所有双特异性PSMA×CD3适配体均表现出类似的剂量依赖性结合,但由于与CD3阴性细胞的残余结合强烈减少,因此对靶阳性细胞具有更高的特异性。对于每个测试浓度,双特异性适配体的信号强度都低于抗CD3单体的信号强度,这表明异二聚体导致了亲和力降低。
总之,这些结果表明,在异源二聚化后,单独评估时针对不同靶标选择的适配体的结合特性不会因空间位阻而被破坏。根据所选的伴侣,二聚甚至可以提高各自靶标的特异性和亲和力。
实施例4.通过表面等离子体共振测定靶向PSMA和CD3的双特异性适配体的结合。
使用BIAcore T200仪(GE Healthcare)进行结合亲和力测定。为了分析适配体与CD3和PSMA蛋白质之间的相互作用,根据制造商的说明(GE Healthcare),将300个生物素化适配体共振单元固定在S系列传感器芯片SA(GE Healthcare)上。DPBS缓冲液用作运行缓冲液。在“单一动力学循环”模式下,以30μl/min的流速,通过注射不同浓度的人CD3ε/γ、CD3ε/δ、IgG1Fc和PSMA(Acro Biosystems)来测定相互作用。使用的最高适配体浓度为300nM。其他浓度通过3倍稀释获得。使用BIAcore T200评估软件评估相互作用的所有动力学数据。
预期比较单体和双特异性适配体的KD值会表明二聚化不会干扰每个亚单位对其特定靶标的结合特性。PSMA和CD3ε/γ的同时结合也可以通过手动注入模式以10μl/min的流速记录,并通过注入第一靶标溶液(饱和浓度),然后注入第二个靶标溶液(饱和浓度)。执行带有反向序(inverted sequence)的第二次注射。在这两个序列中,每次注射都会产生相同强度的反应,这表明当第一个抗原的结合位点被占据时,双特异性适配体的两个臂都能够结合第二个靶标。单体对两种靶溶液注射均无反应表明双特异性适配体可同时结合两种靶
实施例5.针对PSMA和CD3的双特异性适配体的生物活性。
对未受刺激的外周血单核细胞(PBMC)进行细胞毒性试验。新鲜制备的PBMC分离自从健康捐赠者(Etablissementdu Sang,Division)获得的血沉棕黄层(buffy coat)。在用DPBS稀释血液后,通过FICOLL密度梯度(FICOLL-PAQUE PREMIUM1.077 GE Healthcare)分离PBMC,用DPBS洗涤两次,重悬于RPMI-1640培养基(GibcoInvitrogen)中以获得5×106个细胞/ml的细胞密度。这些PBMC被用作效应细胞。
LNCaP靶细胞在37℃的细胞培养基中用2μM钙黄绿素AM(Trevigen Inc,盖瑟斯堡,马里兰州,美国)标记30min。钙黄绿素AM荧光色素是一种嵌入活LNCaP细胞内的染料,仅在定向裂解时释放。在细胞培养基中洗涤2次后,调整细胞密度在RPMI-1640培养基中为5×105个细胞/ml,每次测试反应使用100μl等分的50000个细胞。37℃/5%CO2下的标准反应持续4h,使用5×104个细胞钙黄绿素AM标记的靶细胞、5×105个PBMC(E/T比为1:10)和20μl双特异性适配体溶液,总体积为200μl。细胞毒性反应后,在荧光阅读器(VarioSkan Lux,ThermoFisher,Waltham,MA,USA)中对培养基中释放的染料进行定量,并将其与不存在细胞毒性化合物的对照反应地荧光信号以及测定完全裂解细胞荧光信号的反应中的荧光信号进行比较(适配体被从丹麦阿勒罗德Chemometec购买的A100试剂所取代)。根据这些读数,根据以下公式计算特定细胞毒性:[荧光(样品)-荧光(对照)]/[荧光(完全裂解)-荧光(对照)]×100。
图8示出了在100nM的适配体存在下以10:1的单个E:T比孵育4h后获得的细胞毒性试验结果。PSMA×CD3双特异性RNA/DNA适配体可观察到无效至弱特异性细胞杀伤活性(<10%)。用RNA/RNA适配体A10×ARACD3-3700006和A10×ARACD3-0010209测定其优越的特异性细胞毒性,其诱导40-50%的LNCaP细胞被杀死。缺少CD3结合部分的对照单体A10没有诱导任何细胞毒性。
这些结果表明,工程化适配体开关能够将效应T淋巴细胞招募到靶细胞,从而重新定向其细胞溶解机制,并消除特定的细胞群体。
实施例6.在临床前模型中用抗CD3×抗PSMA适配体治疗癌症。
对不同多聚适配体结构与单体适配体在小鼠体内的疗效和毒性进行了评估。对患有PSMA阳性肿瘤的成年小鼠给予特异性结合CD3和PSMA的适配体,在不同的小鼠组中,适配体可以是单体形式,也可以是多聚体形式。通过测定处理组与对照组的肿瘤大小、肿瘤生长和速率以及生存率来评估疗效。通过处理组与对照组的不良反应发生率来评估毒性。
实施例7.在临床前模型中用抗CD3×抗PSMA适配体治疗癌症。
对不同多聚适配体结构与单体适配体在小鼠体内的疗效和毒性进行了评估。对患有PSMA阳性肿瘤的成年小鼠施用特异性结合CD3和PSMA的适配体,在不同的小鼠组中,适配体为单体形式或多聚体形式。通过测定处理组与对照组的肿瘤大小、肿瘤生长和速率以及生存率来评估疗效。通过处理组与对照组的不良反应发生率来评估毒性。
实施例8.对PSMA和CAR-PNE具有特异性的双特异性适配体的制备。
ARAA-00100001和ARAA-01700001适配体购自baseclick(诺伊里德,德国),作为通过标准固相磷酰胺化学合成的HPLC-RP纯化的2’-F RNA低聚物。
A102’F-RNA适配体在其3’端用叠氮化物基团修饰,以进行随后的三唑核苷酸间二聚化。将生物素作为生物素TEG添加到A10适配体的5′端,在适配体序列和生物素标记之间引入一个16原子的混合极性间隔序列。ARAA-00100001和ARAA-01700001在其5’-端用炔基修饰,以进行随后的三唑核苷酸间二聚化。通过HPLC-MS验证分子量、纯度和完整性。
使用实施例1中描述的程序制备双特异性抗PSMAA10和抗CAR PNE适配体。使用Bio-Rad成像系统对凝胶进行可视化,结果如图4A所示。从凝胶中切下对应于二聚体适配体的凝胶切片,并在25mM NaCl-TE缓冲液中通过被动洗脱在8℃下提取核酸72h。通过标准醋酸钠沉淀来回收双特异性适配体二聚体,重悬在无菌水中,并在-20℃下储存,直至使用。
实施例9.测定抗PSMAx抗CAR-PNE双特异性适配体对细胞表达靶标的亲和力和特
异性。
通过流式细胞术评估抗PSMA×抗CAR PNE适配体与细胞上表达的靶蛋白的亲和力和特异性。这些研究是在含有5%FBS的DPBS缓冲液中对PSMA阳性LNCaP(人前列腺癌-ATCCCRL-1740)和PSMA阴性PC-3(人前列腺癌-ATCC CRL-1435)进行的,如实施例3所述。适配体在单个浓度范围内进行测试:30、100和300nM。
与PSMA阳性细胞结合的研究结果如图6A所示。对两种RNA/RNA适配体A10×ARAA-00100001和A10×ARAA-01700001以及A10单体适配体进行了分析。为了进行比较,还对受试试剂与PSMA阴性PC-3细胞的结合情况进行了测定。
使用A10观察到与PSMA阳性LNCaP细胞的剂量依赖性结合,但在最高测试浓度下信号未达到饱和。信号强度与抗体对照组一样强。仅在最高测试浓度下观察到了A10单体与PC-3细胞的残余结合。两种双特异性PSMA×CAR PNE适配体与A10单体具有相似的结合特性,但由于与PSMA阴性细胞的残余结合减少,其对靶阳性细胞的特异性提高。对于每个测试浓度,双特异性适配体的信号强度都优于A10单体的信号强度,这表明异二聚作用导致亲和力有所提高。
总之,实施例9和本实施例的结果表明,当分别评估时,针对不同靶标选择的适配体的异源二聚作用不会显著影响每个部分的结合性质。
实施例10.对CAR-PNE和PSMA具有特异性的双特异性适配体的生物活性
对未受刺激的外周血单核细胞(PBMC)进行细胞毒性试验。新鲜制备的PBMC分离自从健康捐赠者(Etablissementdu Sang,Division)获得的血沉棕黄层。在用DPBS稀释血液后,在FICOLL密度梯度(菲科尔-帕克PREMIUM 1.077 GEHealthcare)上分离PBMC,用DPBS洗涤两次,重悬于RPMI-1640培养基(Gibco Invitrogen)中以获得5×106个细胞/ml的细胞密度。这些PBMC用表达CAR-PNE受体的慢病毒载体进行转导。这些PBMC-CAR-PNE被用作效应细胞。
LNCaP靶细胞在37℃的细胞培养基中用2μM钙黄绿素AM(Trevigen Inc,盖瑟斯堡,马里兰州,美国)标记30min。钙黄绿素AM荧光色素是一种嵌入活LNCaP细胞内的染料,仅在定向裂解时释放。在细胞培养基中洗涤2次后,调整细胞密度在RPMI-1640培养基中为5×105个细胞/ml的,每次分析反应使用100μl等分的50000个细胞。37℃/5%CO2下的标准反应持续4h,使用5x104细胞钙黄绿素AM标记的靶细胞、5x105 PBMCs-CAR-PNE(E/T比为1:10)和20μl双特异性适配体溶液,1μM,总体积为200μl。细胞毒性反应后,在荧光读取器(VarioSkan Lux,ThermoFisher,Waltham,MA,USA)中对培养基中释放的染料进行定量,并与不存在细胞毒性化合物的对照反应的荧光信号以及测定完全裂解细胞的反应的荧光信号进行比较(适配体被从丹麦阿勒罗德Chemometec购买的A100试剂所取代)。根据这些读数,根据以下公式计算特定细胞毒性:[荧光(样品)-荧光(对照)]/[荧光(完全裂解)-荧光(对照)]×100。
细胞毒性试验的结果是在100nM适配体存在下以10:1的单个E:T比培养4h后获得的。用RNA/RNA适配体A10×CAR PNE测定特异性细胞毒性,其诱导超过30%的LNCaP细胞被杀死。还对缺少CAR-PNE结合部分的对照单体A10的细胞毒性进行了检查。
工程化适配体开关应该能够将效应T淋巴细胞招募到靶细胞,以重新定向其细胞溶解机制,并消除特定的细胞群体。
实施例11.在临床前模型中用CAR-T适配体开关治疗癌症
与单体适配体相比,对开关适配体结构体在小鼠体内的有效性和毒性进行了评估。将多聚适配体作为开启基于CAR T细胞疗法活性的开关进行制备。首先对携带肿瘤的成年小鼠用CAR-PNE转导的T细胞进行注射,然后注入由融合到PSMA、CD19、CD2或CD22肿瘤相关靶标的抗CAR-PNE适配体制成的多聚适配体。通过测定处理组与对照组的肿瘤大小、肿瘤生长和速率以及生存率来评估疗效。通过处理组与对照组的不良反应发生率来评估毒性。

Claims (13)
1.一种基于适配体的多特异性抗原结合分子,其包括1)两个或更多个具有不同靶标结合特异性的靶标结合适配体区域,以及2)一个或多个连接所述适配体区域的连接体。
2.根据权利要求1所述的基于适配体的多特异性抗原结合分子,其中所述连接体包括选自共价键、单链核酸、双链核酸、自组装互补寡核苷酸、肽、多肽、低聚糖、多糖、合成聚合物、腙、硫醚、酯、三唑、纳米粒子、胶束、脂质体、细胞、点击化学产物及其组合的连接部分,或由它们组成。
3.根据权利要求2所述的基于适配体的多特异性抗原结合分子,其中所述连接体包括点击化学产物。
4.根据前述任一项权利要求所述的基于适配体的多特异性抗原结合分子,其可结合到一个或多个人类细胞、免疫细胞、癌细胞、转基因细胞、细菌或病毒上的特定靶标。
5.根据前述任一项权利要求所述的基于适配体的多特异性抗原结合分子,其可将一种细胞类型的结合从一个靶细胞重定向到另一个靶细胞。
6.根据前述任一项权利要求所述的基于适配体的多特异性抗原结合分子,其可在免疫细胞和癌细胞之间形成桥梁。
7.根据前述任一项权利要求所述的基于适配体的多特异性抗原结合分子,其可刺激和激活免疫细胞。
8.根据权利要求7所述的基于适配体的多特异性抗原结合分子,其中所述免疫细胞为T细胞、NK细胞或巨噬细胞,且所述结合导致与基于适配体的多特异性抗原结合分子的靶标结合适配体结合的靶标细胞被破坏。
9.根据前述任一项权利要求所述的基于适配体的多特异性抗原结合分子,其中所述分子对于选自CD3、CD8、CD4、CD19、上皮细胞粘附分子、CD20、CD22、CD123、BCMA、B7H3、CEA、PSMA、Her2、CD33、CD38、DLL3、EGF-R、NKG2D配体、MHC I类相关蛋白MR1、间皮素、PD-1、PD-L1、CTLA04、Lag-3、TIM-3、OX40、CD47、VEGF、PRAME、NY-ESO-1、MAGEA4、MAGEA3/A6、MAGE A10和AFP的抗原具有结合特异性。
10.根据权利要求3所述的基于适配体的多特异性抗原结合分子,其中所述分子结合至表达CD3抗原的免疫细胞。
11.根据权利要求1所述的基于适配体的多特异性抗原结合分子,其中所述分子结合癌细胞上的PSMA抗原。
12.根据权利要求1所述的基于适配体的多特异性抗原结合分子,其包括一个或多个可结合到T细胞的CD3抗原结合区和一个或多个可结合到PSMA表达细胞的PSMA抗原结合区,其中所述CD3抗原结合区和所述PMSA抗原结合区通过一个或多个连接体连接。
13.根据权利要求12所述的基于适配体的多特异性抗原结合分子在治疗包括前列腺癌在内的表达PSMA的癌症中的用途。
