Abstract
本发明属于生物化工及化妆品技术领域,公开了一种保湿角质活化护肤液及其制备方法。制备方法包括:(1)分别收集水蛭粘液和孵化后的卵茧壳、囊,合并后加入ε‑聚赖氨酸冻干,粉碎得到水蛭提取物;(2)将水蛭提取物与蟹壳混合,粉碎研磨,加入甘露醇、EDTA钠盐,用盐酸调pH值至中性,即得保湿角质活化护肤液。该保湿角质活化护肤液能够保持皮肤湿润,同时持续释放N‑乙酰氨基葡萄糖,活化更新皮肤角质层。

技术领域
本发明涉及生物化工及化妆品领域,具体涉及一种保湿角质活化护肤液及其制备方法。
背景技术
水蛭体表会分泌凝胶状的粘液。水蛭体表粘液的主要作用是:湿润体表、维持呼吸、调节身体水分、抗菌抗寄生虫。另外水蛭在孵化出壳前会由口器分泌大量的几丁质酶用于破壳,因此,卵茧壳、囊里也富含几丁质酶。水蛭粘液、卵茧壳、囊是水蛭养殖和水蛭素提取的副产物,通常情况下被直接作为生物垃圾处理,不能获得很好的利用。
发明内容
有鉴于此,本发明目的在于提供一种保湿角质活化护肤液及其制备方法。用该法制备的护肤液能够保持皮肤湿润,同时持续释放N-乙酰氨基葡萄糖,活化更新皮肤角质层。
本发明采用的技术方案如下:
一种保湿角质活化护肤液的制备方法,包括如下步骤:
S1:收集水蛭粘液及孵化后的卵茧壳、囊,加入ε-聚赖氨酸冻干后,粉碎得到水蛭提取物;
S2:将所述水蛭提取物与蟹壳混合,研磨得粉末,所述粉末中加入甘露醇和EDTA二钠,调pH值至6.5-7.4,即得保湿角质活化护肤液。所述优选采用行星球磨机进行研磨。
优选的,步骤S1中,所述的水蛭粘液、孵化后的卵茧壳、囊和ε-聚赖氨酸的比例为1mL:(50-230)g:(2-5)g,最优为1mL:120g:3g。
优选的,步骤S2中,所述的水蛭提取物与蟹壳、甘露醇、EDTA二钠的比例为100g:(3-8)g:(1-5)g:(0.2-0.5)g,最优为100g:4g:3g:0.3g。
优选的,步骤S2中,所述的研磨细度为100-300nm,最优为220nm。
优选的,步骤S2中,所述调pH值采用质量分数浓度为10%-28%的盐酸调pH值。
优选的,步骤S2中,所述的混合、粉碎研磨,调pH值均在0-8℃下进行,最优为4℃。
本发明还提供了一种上述制备方法制备得到的保湿角质活化护肤液。
与现有技术相比,本发明利用水蛭卵茧壳、囊中的几丁质酶在皮肤角质层原位催化水解蟹壳中的甲壳素,获得N-乙酰氨基葡萄糖,活化更新角质层。借助水蛭粘液保湿作用防止保湿液水分蒸发,为几丁质酶的催化水解提供湿环境;加入ε-聚赖氨酸既作为抑菌剂,防止皮肤表面微生物分泌的蛋白酶对几丁质酶的破坏,又能为几丁质酶催化提供pH6.5-7.4的缓冲体系,此缓冲范围恰好是几丁质酶的最适反应酸碱度范围。通过营造有利于几丁质酶反应的微环境(最适pH的溶液环境),有效催化甲壳素原位水解,获得N-乙酰氨基葡萄糖的缓慢释放。

具体实施方式
根据下述实施例,可以更好地理解本发明。然而,本领域的技术人员容易理解,实施例所描述的内容仅用于说明本发明,而不应当也不会限制权利要求书中所详细描述的本发明。
实施例1
1、收集1mL水蛭粘液及120g孵化后的卵茧壳、囊,加入3gε-聚赖氨酸,冻干后,粉碎得到水蛭提取物;
2、在4℃环境下,将100g步骤1制备的水蛭提取物与4g蟹壳混合,加入到行星式球磨机中进行研磨,粉末细度为220nm,加入3g甘露醇、0.3gEDTA二钠,用质量分数为15%盐酸调pH值至6.8,即得保湿角质活化护肤液。
实施例2
1、收集1mL水蛭粘液及50g孵化后的卵茧壳、囊,加入2gε-聚赖氨酸,冻干后,粉碎得到水蛭提取物;
2、在0℃环境下,将100g步骤1制备的水蛭提取物与8g蟹壳混合,加入到行星式球磨机中进行研磨,粉末细度为100nm,加入5g甘露醇、0.5gEDTA二钠,用质量分数为10%盐酸调pH值至7.4,即得保湿角质活化护肤液。
实施例3
1、收集1mL水蛭粘液及230g孵化后的卵茧壳、囊,加入5gε-聚赖氨酸,冻干后,粉碎得到水蛭提取物;
2、在8℃环境下,将100g步骤1制备的水蛭提取物与3g蟹壳混合,加入到行星式球磨机中进行研磨,粉末细度为300nm,加入1g甘露醇、0.2gEDTA二钠,用质量分数为28%盐酸调pH值至6.5,即得保湿角质活化护肤液。
N-乙酰氨基葡萄糖的测定:
采用Hedera NH2色谱柱(250mm×4.6mm,5μm),流动相为乙腈-磷酸缓冲液(65:35),流速为1.0mL/min,柱温30℃,检测波长195nm,进样体积20μL。
取新鲜大鼠背部全层皮肤5cm×5cm,放入恒温恒湿室内,37℃相对湿度60±10%保持24h,之后取实施例1-3的保湿角质活化护肤液5mL,用硅胶刷均匀涂刷在皮肤表面,在0、1、2、3、4、5、6、7、8h分别取样1cm×1cm全层皮肤,浸没在30mL无菌水中提取30min,取出皮肤后将水冻干,再加入0.2mL无菌水充分溶解,测定N-乙酰氨基葡萄糖的含量,结果如图1所示。
如图1可见,随着时间的延长,实施例的保湿角质活化护肤液中的N-乙酰氨基葡萄糖含量逐渐增加。在皮肤上使用该护肤液,这些释放的N-乙酰氨基葡萄糖能够不断地活化更新皮肤角质层。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (8)
1.一种保湿角质活化护肤液的制备方法,其特征在于,包括如下步骤:
S1:收集水蛭粘液及孵化后的卵茧壳、囊,加入ε-聚赖氨酸冻干后,粉碎得到水蛭提取物;
S2:将所述水蛭提取物与蟹壳混合,粉碎研磨得粉末,所述粉末中加入甘露醇和EDTA二钠,调pH值至6.5-7.4,即得保湿角质活化护肤液。
2.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S1中,所述的水蛭粘液、所述孵化后的卵茧壳、囊和所述ε-聚赖氨酸的用量比为1mL:(50-230)g:(2-5)g。
3.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S2中,所述的水蛭提取物、蟹壳、甘露醇和EDTA二钠的质量比为100:(3-8):(1-5):(0.2-0.5)。
4.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S2中,所述研磨为:加入行星球磨机中研磨。
5.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S2中,所述研磨的研磨细度为100-300nm。
6.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S2中,所述调pH值具体为:采用浓度为10%-28wt%的盐酸调pH值。
7.根据权利要求1所述的一种保湿角质活化护肤液的制备方法,其特征在于,步骤S2中,所述的混合、粉碎研磨,调pH值均在0-8℃下进行。
8.权利要求1-7任一项所述的制备方法制备得到的保湿角质活化护肤液。
